This study investigated the pleiotropy and the underlying biology linking COVID-19 outcomes (infection, hospitalization, and severe symptoms) to brain structure and function, leveraging genome-wide data from the COVID-19 Host Genetics Initiative (up to 122,616 cases and 2,475,240 controls) and 3,935 brain imaging-derived phenotypes (IDP) from the UK Biobank (up to 33,224 participants). COVID-19 outcomes were genetically correlated with 11 IDPs related to brain connectivity, cortical structures, and white matter. With respect to these, we also identified local genetic correlations within five l

BACKGROUND: Observational studies have revealed a link between major depressive disorder (MDD) and a higher chance of developing atrial fibrillation (AF). It is still uncertain whether or not this correlation indicates a causal relationship. This research set out to evaluate the causal impact of MDD on AF.
METHODS: To evaluate the causal relationship between MDD and AF, we employed a two-sample Mendelian randomization (MR) method. A new genome-wide association study (GWAS) with 500,199 participants was used to obtain an overview of the association of genetic variations with MDD. An additional

The observation of genetic correlations between disparate human traits has been interpreted as evidence of widespread pleiotropy. Here, we introduce cross-trait assortative mating (xAM) as an alternative explanation. We observe that xAM affects many phenotypes and that phenotypic cross-mate correlation estimates are strongly associated with genetic correlation estimates (R2=74%). We demonstrate that existing xAM plausibly accounts for substantial fractions of genetic correlation estimates and that previously reported genetic correlation estimates between some pairs of psychiatric disorders are

PROBLEM: Polycystic ovary syndrome (PCOS) is known to be associated with immune response dysregulation, resembling autoimmune diseases. However, the causal relationship between PCOS and autoimmune diseases remains unclear.
METHODS: A two-sample bidirectional Mendelian randomization (MR) analysis was conducted using publicly available genome-wide association study (GWAS) data. The primary causal effects were estimated using the inverse-variance weighted (IVW) method, complemented by the weighted median, weighted mode, and MR-Egger regression approaches. Horizontal pleiotropy was assessed using

Mendelian randomization (MR) uses genetic variants as instruments to study causal relationships between traits. Standard unidirectional MR approaches are vulnerable to pleiotropy and typically assume that pleiotropic effects are independent of instrument strength, assumptions that may fail when causation operates in both directions. We propose BayBiMR, a Bayesian bidirectional MR framework that jointly estimates causal effects in both directions while modeling both correlated and uncorrelated pleiotropy through a hierarchical spike-and-slab prior on pleiotropic effects and an Inverse-Wishart p

In a standard analysis, pleiotropic variants are identified by running separate genome-wide association studies (GWAS) and combining results across traits. But such statistical approach based on marginal summary statistics may lead to spurious results. We propose a new statistical approach, Debiased-regularized Factor Analysis Regression Model (DrFARM), through a joint regression model for simultaneous analysis of high-dimensional genetic variants and multilevel dependencies. This joint modeling strategy controls overall error to permit universal false discovery rate (FDR) control. DrFARM uses

BACKGROUND: There is a growing interest in the study of the genetic origins of comorbidity, a direct consequence of the recent findings of genetic loci that are seemingly linked to more than one disorder. There are several potential causes for these shared regions of linkage, but one possibility is that these loci may harbor genes with manifold effects. The established genetic correlation between reading disability (RD) and attention-deficit/hyperactivity disorder (ADHD) suggests that their comorbidity is due at least in part to genes that have an impact on several phenotypes, a phenomenon kno

The role of micronutrients in autoimmune thyroiditis (AIT) has been controversial and lacks sufficient genetic evidence. This study aimed to assess the relationships between various micronutrients and AIT by Mendelian randomization (MR). The single nucleotide polymorphisms (SNPs) of micronutrients and AIT were acquired from IEU Open GWAS project and FinnGen, and were screened according to the basic assumptions of MR. Subsequently, inverse variance weighted was set as the primary tool for MR analysis, with weighted median, simple mode, MR-Egger, and weighted mode set as secondary tools. Next, t

Epidemiological studies have recognized a genetic diathesis for suicidal behavior, which is independent of other psychiatric disorders. Genome-wide association studies (GWAS) on suicide attempt (SA) and ideation have failed to identify specific genetic variants. Here, we conduct further GWAS and for the first time, use polygenic score analysis in cohorts of patients with mood disorders, to test for common genetic variants for mood disorders and suicide phenotypes. Genome-wide studies for SA were conducted in the RADIANT and GSK-Munich recurrent depression samples and London Bipolar Affective D
