Host genetics and gut microbiota jointly regulate blood biochemical indicators in chickens.

Blood biochemical indicators play a crucial role in assessing an individual's overall health status and metabolic function. In this study, we measured five blood biochemical indicators, including total cholesterol (CHOL), low-density lipoprotein cholesterol (LDL-CH), triglycerides (TG), high-density lipoprotein cholesterol (HDL-CH), and blood glucose (BG), as well as 19 growth traits of 206 male chickens. By integrating host whole-genome information and 16S rRNA sequencing of the duodenum, jejunum, ileum, cecum, and feces microbiota, we assessed the contributions of host genetics and gut micro

Advances and perspectives in genetically encoded biosensors for biomarker detection and theranostic circuits.

Effective biomarker detection is critical for timely and accurate clinical disease diagnosis. However, conventional analytical methodologies are often constrained by laborious procedures, reliance on specialized equipment, and an inherent inability to perform continuous in situ monitoring. To circumvent these limitations, synthetic biology has pioneered the development of genetically encoded biosensors as a rapid, highly sensitive, and cost-effective alternative. This review specifically focuses on transcriptional regulation-based biosensors, primarily those utilizing allosteric transcription

Emerging insights in gastric cancer pathogenesis.

PURPOSE OF THE REVIEW: Gastric cancer (GC) is a leading cause of cancer-related mortality worldwide. Although Helicobacter pylori (Hp) is a well known, leading risk factor for GC, not all patients with Hp develop malignancy, highlighting the multifactorial mechanisms involved in gastric carcinogenesis. This review summarizes recent insights into GC pathogenesis and the implications for risk stratification, screening surveillance, and prevention. RECENT FINDINGS: Emerging evidence suggests complex interactions between microbial, genetic, environmental, and immune factors in GC pathogenesis. Ch

Probiotics in Celiac Disease.

Recently, the interest in the human microbiome and its interplay with the host has exploded and provided new insights on its role in conferring host protection and regulating host physiology, including the correct development of immunity. However, in the presence of microbial imbalance and particular genetic settings, the microbiome may contribute to the dysfunction of host metabolism and physiology, leading to pathogenesis and/or the progression of several diseases. Celiac disease (CD) is a chronic autoimmune enteropathy triggered by dietary gluten exposure in genetically predisposed individu

Linking the Human Gut Microbiome to Inflammatory Cytokine Production Capacity.

Gut microbial dysbioses are linked to aberrant immune responses, which are often accompanied by abnormal production of inflammatory cytokines. As part of the Human Functional Genomics Project (HFGP), we investigate how differences in composition and function of gut microbial communities may contribute to inter-individual variation in cytokine responses to microbial stimulations in healthy humans. We observe microbiome-cytokine interaction patterns that are stimulus specific, cytokine specific, and cytokine and stimulus specific. Validation of two predicted host-microbial interactions reveal th

Pathogenic microorganism DNA high-throughput genetic sequencing to diagnose peritoneal dialysis-associated peritonitis due to Mycobacterium tuberculosis infection.

Peritoneal dialysis-associated peritonitis is a serious complication of peritoneal dialysis, and the prevention and treatment of this condition are important for improving the long-term survival and quality of life of patients. However, peritoneal dialysis-associated peritonitis due to Mycobacterium tuberculosis infection is relatively rare and not easily diagnosed. Here, we present a case of peritoneal dialysis-associated peritonitis caused by Mycobacterium tuberculosis identified by pathogenic microbial DNA high-throughput genetic sequencing. This case demonstrates that pathogenic microbial

The human skin virome: Ecological dynamics, aberrant profiles, and therapeutic opportunities.

Human skin harbors a complex microbial ecosystem in which viruses, including eukaryotic viruses, bacteriophages, and endogenous retroviruses, are integral yet comparatively understudied. Shaped by host physiology, genetics, and environmental exposures across the lifespan, the cutaneous virome interacts closely with resident bacteria, fungi, and immune cells. In this review, we synthesize current knowledge of virome composition, biogeography, and dynamics, with an emphasis on how bacteriophages influence bacterial ecology, gene flow, and antimicrobial resistance on the skin. We further discuss

Microbial functions in skin inflammation: a Mendelian randomization study.

Inflammatory skin diseases (ISDs) represent a significant global health burden, yet the role of the human microbiome in their pathogenesis remains unclear. This study employed a comprehensive two-sample Mendelian randomization (MR) framework to investigate potential causal associations between multi-site microbiota (3117 oral, 412 gut, and 150 skin microbial features) and five major ISDs: psoriasis, atopic dermatitis, acne, seborrheic dermatitis, and urticaria. Genetic instruments were selected from large-scale genome-wide association studies (GWAS) of microbiota composition and ISD outcomes.

Interleukin23 Receptor Genetic Variants Associate With Crohn's Disease Risk and Microbiome Changes in Healthy First-Degree Relatives.

BACKGROUND & AIMS: Single nucleotide polymorphisms in the interleukin23 receptor gene are associated with Crohn's disease, suggesting a role in pathogenesis, and several biologic agents targeting this pathway are now established therapies. Interleukin23 has been suggested to be involved in regulation of intestinal barrier function and may impact gut microbial composition. We investigated whether interleukin23 receptor genetic variants predict Crohn's disease risk and influence gut barrier function and microbiome composition in healthy first-degree relatives. METHODS: A total of 3055 healthy f

An Integrated Model Based on Gut Microbiota and APOE Genotype for Predicting Dementia Risk.

BACKGROUND: Dementia develops through the combined influence of genetic vulnerability, biological processes, and environmental exposures. The apolipoprotein E (APOE) ε4 allele is a well-known genetic contributor to dementia risk, and growing evidence links gut microbial alterations to cognitive decline and cerebrovascular-related pathology. Nevertheless, studies jointly evaluating genetic, microbiome, and clinical information remain relatively scarce. This study examined an integrated framework combining APOE genotype and gut microbiome data for cross-sectional dementia classification. METHOD

Integrated metagenomic and soil chemical analyses revealed shifts of microbial nutrient cycling with poplar plantation age.

INTRODUCTION: Poplar (Populus spp.) is widely recognized as an ideal model system for studying plant-microbial interactions due to its rapid growth, genetic tractability, and ecological importance in afforestation programs. Leveraging these advantages, we investigated how poplar cultivation reshapes soil microbial communities and their nutrient cycling functions. Although plant roots are known to profoundly influence microbial community structure and functionality, comprehensive studies systematically linking poplar-induced microbiome shifts to nutrient cycling remain limited. METHODS: Here,

Can attention to the intestinal microbiota improve understanding and treatment of anorexia nervosa?

Anorexia nervosa (AN) is characterized by severe dietary restriction or other weight loss behaviors and exhibits the highest mortality rate of any psychiatric disorder. Therapeutic renourishment in AN is founded primarily on clinical opinion and guidelines, with a weak evidence base. Genetic factors do not fully account for the etiology of AN, and non-genetic factors that contribute to the onset and persistence of this disease warrant investigation. Compelling evidence that the intestinal microbiota regulates adiposity and metabolism, and more recently, anxiety behavior, provides a strong rati

The microbiota-metabolite-immune axis in colorectal cancer: mechanistic insights and emerging clinical applications.

Colorectal cancer (CRC) results from a complex interplay of host genetics, environmental factors, and gut microbiota. Increasing evidence suggests that intestinal microorganisms significantly affect the initiation and progression of CRC through metabolic and immunological reprogramming. Dysbiosis, defined as an imbalance between beneficial and harmful microbial species, leads to chronic inflammation, genotoxic stress, and disruption of epithelial homeostasis. Microbial metabolites, such as short-chain fatty acids, secondary bile acids, and tryptophan derivatives, function as signaling molecule

Characterizing the metabolomic signature of attention-deficit hyperactivity disorder in twins.

Emerging evidence implicate the gut microbiota as a potential susceptibility factor in attention-deficit hyperactivity disorder (ADHD), a common multifactorial neurodevelopmental condition. However, little is known about the biochemical signature of ADHD, including the metabolic contribution of the microbiota via the gut-brain axis, and the relative contribution of genetics and environmental factors. Here, we perform unbiased metabolomic profiling of urine and fecal samples collected from a well-characterized Swedish twin cohort enriched for ADHD (33 ADHD, 79 non-ADHD), using 1H nuclear magnet

The Microbiota-Gut-Brain Axis.

The importance of the gut-brain axis in maintaining homeostasis has long been appreciated. However, the past 15 yr have seen the emergence of the microbiota (the trillions of microorganisms within and on our bodies) as one of the key regulators of gut-brain function and has led to the appreciation of the importance of a distinct microbiota-gut-brain axis. This axis is gaining ever more traction in fields investigating the biological and physiological basis of psychiatric, neurodevelopmental, age-related, and neurodegenerative disorders. The microbiota and the brain communicate with each other

Deterministic processes structure bacterial genetic communities across an urban landscape.

Land-use change is predicted to act as a driver of zoonotic disease emergence through human exposure to novel microbial diversity, but evidence for the effects of environmental change on microbial communities in vertebrates is lacking. We sample wild birds at 99 wildlife-livestock-human interfaces across Nairobi, Kenya, and use whole genome sequencing to characterise bacterial genes known to be carried on mobile genetic elements (MGEs) within avian-borne Escherichia coli (n = 241). By modelling the diversity of bacterial genes encoding virulence and antimicrobial resistance (AMR) against ecolo

Metagenomic sequencing reveals altered gut microbial compositions and gene functions in patients with non-segmental vitiligo.

BACKGROUND: Vitiligo has been correlated with an abnormal gut microbiota. We aimed to systematically identify characteristics of the gut microbial compositions, genetic functions, and potential metabolic features in patients with non-segmental vitiligo. METHODS: Twenty-five patients with non-segmental vitiligo and 25 matched healthy controls (HCs) were enrolled. Metagenomic sequencing and bioinformatic analysis were performed to determine the gut microbiota profiles. Differences in gut microbiota diversity and composition between patients with vitiligo and HCs were analyzed. Gene functions an

Exploring the Gut Microbiome as a Promising Frontier in Alzheimer's Disease Therapy.

Alzheimer's Disease (AD) is a major global health challenge, particularly in ageing populations, and current therapies offer limited modification of disease progression. Emerging evidence indicates that the gut microbiome contributes to AD pathogenesis through metabolic, immune, and neuroendocrine mechanisms. Microbial metabolites, including Short-Chain Fatty Acids (SCFAs), bile acids, and trimethylamine-N-oxide (TMAO), regulate neuronal signalling and blood-brain barrier integrity, and dysbiosis has been linked to amyloid-β (Aβ) accumulation, tau hyperphosphorylation, chronic neuroinflammatio
