Cytomorphological aspects on the response of the branchial heart complex of Sepia officinalis L. (cephalopoda) to xenobiotics and bacterial infection.

The ovoid cells of the branchial heart complex of Sepia officinalis L. were investigated with respect to their role in detoxification processes. The electron microscopical localization of in situ injected ferritin in the endocytotic-lysosomal system, and the fluorescence microscopical localization of protein-bound Evans blue in the ovoid cells of in vivo incubated animals, indicate that foreign materials are eliminated from the hemolymph by the branchial heart tissue. In addition to the non-circulating ovoid cells of the branchial heart, hemocytes in the circulating blood and in the wall of th

BgTEP: An Antiprotease Involved in Innate Immune Sensing in Biomphalaria glabrata.

Insect thioester-containing protein (iTEP) is the most recently defined group among the thioester-containing protein (TEP) superfamily. TEPs are key components of the immune system, and iTEPs from flies and mosquitoes were shown to be major immune weapons. Initially characterized from insects, TEP genes homologous to iTEP were further described from several other invertebrates including arthropods, cniderians, and mollusks albeit with few functional characterizations. In the freshwater snail Biomphalaria glabrata, a vector of the schistosomiasis disease, the presence of a TEP protein (BgTEP) w

Genetics of a Drosophila phenoloxidase.

An electrophoretic mobility variant of phenoloxidase in a lz stock of Drosophila melanogaster was identified as the A3 component of the phenoloxidase complex by using two different activators to study enzyme activity-natural activator isolated from pupae and 50% 2-propanol. The structural gene for the A3 proenzyme, Dox-3, was not associated with lz on the X chromosome; it mapped to the right of rdo (53.1) and left of M(2)m in the second linkage group. The lz locus affects the differentiation of the crystal cell, the type of hemocyte that carries prophenoloxidase(s) in paracrystalline form. All

Immunological functional differentiation of two transmembrane variants of Dscam in Chinese mitten crab.

Down syndrome cell adhesion molecule (Dscam), also called hypervariable Dscam (Dscam-hv), is an important player in arthropod alternative splicing that connects neurons and immune regulation, acting as a pathogen-specific recognition molecule. Dscam-hv has two forms: transmembrane (TM) Dscam (mDscam) and soluble Dscam (sDscam). Herein, we investigated two transmembrane variants of mDscam resulting from alternative splicing of the transmembrane domain, focusing on differences in their immune regulation. We characterized the Dscam[TM1] and Dscam[TM2] genes of Chinese mitten crab (Eriocheir sinen

Nerve degeneration and regeneration in the cephalopod mollusc Octopus vulgaris: the case of the pallial nerve.

Regeneration is a process that restores structure and function of tissues damaged by injury or disease. In mammals complete regeneration is often unsuccessful, while most of the low phyla animals can re-grow many parts of their body after amputation. Cephalopod molluscs, and in particular Octopus vulgaris, are well known for their capacity to regenerate their arms and other body parts, including central and peripheral nervous system. To better understand the mechanism of recovery following nerve injury in this species we investigated the process of axon regrowth and nerve regeneration after co

Tributyltin-sulfhydryl interaction as a cause of immunotoxicity in phagocytes of tunicates.

We reported elsewhere that tributyltin (TBT) has detrimental effects on the immune system of the colonial ascidian Botryllus schlosseri, through interaction with calmodulin and alteration of Ca2+ homeostasis. Here, we studied the capability of TBT to react with intracellular thiols. After exposure to 0.1 microM TBT, a significant decrease in B. schlosseri hemocytes stained for total thiols and reduced glutathione (GSH) was detected. Exogenous sulfhydryl and sulfide compounds can prevent TBT-induced cell morphology alterations and decrease the percentage of tin-containing hemocytes, indicating

A Carbonic Anhydrase Serves as an Important Acid-Base Regulator in Pacific Oyster Crassostrea gigas Exposed to Elevated CO2: Implication for Physiological Responses of Mollusk to Ocean Acidification.

Carbonic anhydrases (CAs) have been demonstrated to play an important role in acid-base regulation in vertebrates. However, the classification and modulatory function of CAs in marine invertebrates, especially their responses to ocean acidification remain largely unknown. Here, a cytosolic α-CA (designated as CgCAII-1) was characterized from Pacific oyster Crassostrea gigas and its molecular activities against CO2 exposure were investigated. CgCAII-1 possessed a conserved CA catalytic domain, with high similarity to invertebrate cytoplasmic or mitochondrial α-CAs. Recombinant CgCAII-1 could co

Dynamics of haemocytes from Pseudosuccinea columella circulating infected by Fasciola hepatica.

The lymnaeids are important in the epidemiology of Fasciola hepatica, a neglected and endemic zoonosis. The interaction between the internal defense system of Pseudosuccinea columella and F. hepatica has been little studied. In the present study the effect of infection by F. hepatica on P. columella circulating haemocytes was investigated. Changes in the average number of total circulating haemocytes have been observed at 30 minutes post-infection and 1, 7, 10, 14, 21, 28 and 50 days post-infection (dpi). Miracidia were observed head-foot and mantle at 30 minutes post-infection. Miracidia/Spor

TNF-NF-κB signaling mediates immune-biomineralization crosstalk during shell repair under ocean acidification in Mytilus edulis.

Ocean acidification (OA) impairs biomineralization in bivalves, but its effects on immune-biomineralization crosstalk during shell repair remain unknown. Here, we exposed adult Mytilus edulis bearing standardized shell perforations to three pH levels (8.1, 7.9, and 7.7) for up to 40 days. OA slowed early repair and caused microstructural disorganization and an approximately 87% reduction of compressive strength at pH 7.7, yet the damaged area appeared largely closed by day 15, suggesting a decoupling between morphological closure and functional recovery. In addition, transcriptomic profiling o

Disk abalone (Haliotis discus discus) expresses a novel antistasin-like serine protease inhibitor: Molecular cloning and immune response against bacterial infection.

A novel antistasin-like cDNA homologue named as Ab-Antistasin was isolated from the disk abalone Haliotis discus discus normalized cDNA library. The Ab-Antistasin (1398-bp) consisted of an 1185-bp open reading frame encoding 395 amino acid (aa) residues. The predicted molecular mass and isoelectric point of Ab-Antistasin was 44 kDa and 8.5, respectively, and showed highest identity (23.1%) to Hydra magnipapillata antistasin. The most striking feature of Ab-Antistasin is the 12-fold internal repeats (IR) of an antistasin-like domain. Ten of the 12 IR domains (26-27 aa) are highly conserved, wit

Sex-specific survival and absence of Wolbachia-mediated protection during invertebrate iridescent virus 31 infection in Armadillidium vulgare.

Invertebrate Iridescent Virus 31 (IIV-31, or Iridovirus armadillidium1) is a large double-stranded DNA virus that induces iridescence and high mortality in terrestrial isopods. Despite its broad distribution, its pathogenic mechanisms and interactions with host immunity and endosymbionts are poorly understood. We examined the effects of IIV-31 infection on survival, hemocyte concentration and viability, septicemia, and viral load in Armadillidium vulgare, while testing for protective effects of the feminizing endosymbiont Wolbachia (wVulC strain). Virgin adults from asymbiotic (males and femal

H+ channels in embryonic Biomphalaria glabrata cell membranes: Putative roles in snail host-schistosome interactions.

The human blood fluke Schistosoma mansoni causes intestinal schistosomiasis, a widespread neglected tropical disease. Infection of freshwater snails Biomphalaria spp. is an essential step in the transmission of S. mansoni to humans, although the physiological interactions between the parasite and its obligate snail host that determine success or failure are still poorly understood. In the present study, the B. glabrata embryonic (Bge) cell line, a widely used in vitro model for hemocyte-like activity, was used to investigate membrane properties, and assess the impact of larval transformation p

Differences in cellular immune competence explain parasitoid resistance for two coleopteran species.

The immune defence of an organism is evolving continuously, causing counteradaptations in interacting species, which in turn affect other ecological and evolutionary processes. Until recently comparative studies of species interactions and immunity, combining information from both ecological and immunological fields, have been rare. The cellular immune defense in insects, mainly mediated by circulating hemocytes, has been studied primarily in Lepidoptera and Diptera, whereas corresponding information about coleopteran species is still scarce. In the study presented here, we used two closely re

Restrained Wnt Signaling Pathway by Enhanced EsGSK3β Activity Facilitates the Infection of Spiroplasma and Leads to Neuropathic Diseases in Crustaceans.

Spiroplasma eriocheiris has been identified as a lethal pathogen of Eriocheir sinensis tremor disease (TD), one neurological disease with typically paroxysmal tremors of the pereiopod. But, the pathogenic mechanism of TD was still unknown. First, in the current study, the phosphoproteomic changes of E. sinensis thoracic ganglion after S. eriocheiris infection were obtained using TMT labeling and affinity enrichment followed by high-resolution LC-MS/MS analysis. A total of 349 phosphorylation proteins are upregulated, and 331 phosphorylation proteins are downregulated when compared to the contr

Iron Regulatory Protein 1 Inhibits Ferritin Translation Responding to OsHV-1 Infection in Ark Clams, Scapharca Broughtonii.

Elemental iron is an indispensable prosthetic group of DNA replication relative enzymes. The upregulation of ferritin translation by iron regulatory proteins (IRP1) in host cells is a nutritional immune strategy to sequester available iron to pathogens. The efficient replication of Ostreid herpesvirus 1 (OsHV-1), a lethal dsDNA virus among bivalves, depends on available iron. OsHV-1 infection was found to trigger iron limitation in ark clams; however, it is still an enigma how OsHV-1 successfully conducted rapid replication, escaping host iron limitations. In this study, we identified the IRP1

Heat shock proteins (HSP 90, 70, 60, and 27) in Galleria mellonella (Lepidoptera) hemolymph are affected by infection with Conidiobolus coronatus (Entomophthorales).

Invertebrates are becoming more popular models for research on the immune system. The innate immunity possessed by insects shows both structural and functional similarity to the resistance displayed by mammals, and many processes occurring in insect hemocytes are similar to those that occur in mammals. The humoral immune response in insects acts by melanization, clotting and the production of reactive oxygen species and antimicrobial peptides, while the cellular immunity system is based on nodulation, encapsulation and phagocytosis. An increasingly popular insect model in biological research i
