Discovery of Plasmodium modulators by genome-wide analysis of circulating hemocytes in Anopheles gambiae.

Insect hemocytes mediate important cellular immune responses including phagocytosis and encapsulation and also secrete immune factors such as opsonins, melanization factors, and antimicrobial peptides. However, the molecular composition of these important immune cells has not been elucidated in depth, because of their scarcity in the circulating hemolymph, their adhesion to multiple tissues and the lack of primary culture methods to produce sufficient material for a genome-wide analysis. In this study, we report a genome-wide molecular characterization of circulating hemocytes collected from t

Vibrio parahaemolyticus infectious for both humans and edible mollusk abalone.

The aims of this study are to report evidence of the first laboratory-acquired infection of Vibrio parahaemolyticus associated with handling experimentally infected abalones and to describe the virulence of the two bacterial strains tested in these animals. Two strains of V. parahaemolyticus, one from the stool of a patient with acute gastroenteritis (strain 880713) and the other from the hemolymph of a diseased small abalone Haliotis diversicolor supertexta (strain 880915), were identified and characterized. Both strains were lethal to small abalone, with similar LD(50) values (8.36-8.41 x 10

Friend or foe? Effects of host immune activation on the gut microbiome in the caterpillar Manduca sexta.

For many animals, the gut microbiome plays an essential role in immunity and digestion. However, certain animals, such as the caterpillar Manduca sexta, do not have a resident gut microbiome. Although these animals do have bacteria that pass through their gut from their natural environment, the absence of such bacteria does not reduce growth or survival. We hypothesized that M. sexta would sterilize their gut as a protective measure against secondary infection when faced with a gut infection or exposure to heat-killed bacteria in the blood (haemolymph). However, we found that gut sterilization

The stress-immunity axis in shellfish.

It is a difficult task to describe what constitutes a 'Работает' shellfish (e.g., crustacean, bivalve). Visible defects such as discolouration, missing limbs or spines, fouling, lesions, and exoskeletal fractures can be indicative of underlying issues, senescence, or a 'stressed' animal. The absence of such symptoms is not evidence of a disease-free or a stress-free state. Now, more than ever, aquatic invertebrates must cope with acute and chronic environmental perturbations, such as, heatwaves and cold shocks, xenobiotic contaminants, intoxication events, and promiscuous pathogens expanding th

Semi-field studies on biochemical markers of honey bee workers (Apis mellifera) after exposure to pesticides and their mixtures.

Due to the fact that many different pesticides are used in crop production and their residues can accumulate in the environment, bees are in contact with various pesticides at the same time. Most studies on their influence on honey bees focus on single substances in concentrations higher than those found in the environment. Our study assessed the chronic effects of commonly used pesticides and their mixtures on selected biochemical markers in worker bee hemolymph. Workers developed in the hive and were provisioned with to pesticides in concentrations corresponding to residues detected in polle

Seroprevalence of nasal myiasis in camels determined by indirect enzyme-linked immunosorbent assay utilizing the most diagnostic Cephalopina titillator larval antigens.

BACKGROUND AND AIM: Nasal myiasis is a serious parasitic disease among camels caused by Cephalopina titillator larvae that negatively affect animal health and production globally. The diagnosis of the infestation relies on postmortem examination of the head region, which considers a cause impeding treatment of live animals and may be misdiagnosed as central nervous system disorders. This study aimed to identify the most diagnostic larval antigen with the capacity for monitoring C. titillator infestation, and to estimate the seroprevalence of nasal myiasis in camels in Egypt, using indirect enz

In vivo direct cell-penetrating peptide mediated protein transduction system in Acyrthosiphon pisum.

OBJECTIVE: The principal delivery method for CRISPR-based genome editing in insects is now based on microinjection into single cells or embryos. The direct protein transduction systems cannot be employed in aphids because oogenesis occurs without apparent vitellogenesis. Given the limited timing of injection into the embryonic stage in oviparous aphids, a protein delivery system from the hemolymph to the germline and embryos would be a useful tool for genome editing. This study reports a newly developed direct protein delivery system for aphids using cell-penetrating peptides (CPPs). CPPs are

Vermilion and cinnabar are involved in ommochrome pigment biosynthesis in eyes but not wings of Bicyclus anynana butterflies.

If the same pigment is found in different tissues in a body, it is natural to assume that the same metabolic pathways are deployed similarly in each tissue. Here we show that this is not the case for ommochromes, the red and orange pigments found in the eyes and wings of butterflies. We tested the expression and function of vermilion and cinnabar, two known fly genes in the ommochrome pathway, in the development of pigments in the eyes and in the wings of Bicyclus anynana butterflies, both traits having reddish/orange pigments. By using fluorescent in-situ hybridization (HCR3.0) we localized t

Data from expressed sequence tags from the organs and embryos of parthenogenetic Haemaphysalis longicornis.

OBJECTIVES: Haemaphysalis longicornis is the most important tick species in Japan and has a wide range of vector capacity. Due to its veterinary and medical importance, this tick species has been used as a model for tick/vector biological studies. To identify the key molecules associated with physiological processes during blood feeding and embryogenesis, full-length cDNA libraries were constructed using the fat body, hemocytes-containing hemolymph, midgut, ovary and salivary glands of fed females and embryos of the laboratory colony of parthenogenetic H. longicornis. The sequences of cDNA fro

Lipophorin levels in the yellow fever mosquito, Aedes aegypti, and the effect of feeding.

High density lipophorin (HDLp) is the major lipid transport vehicle in insect hemolymph. Using an indirect ELISA, levels of HDLp were measured in the yellow fever mosquito, Aedes aegypti. The level of lipophorin, when normalized to the total weight of the insect, was similar in the different developmental stages. Starvation (access to water only) of adult females did not affect the level of HDLp nor its density when compared to sugar-fed females. On the other hand, blood feeding (of normally sugar-fed females) resulted in a three-fold increase of the HDLp level at 40 h after feeding. This incr

Effects of low salinity exposure on osmotic regulation, physiological metabolism, oxidative stress, health status, and apoptosis in Haliotis diversicolor.

Low salinity exposure triggers a series of physiological responses in marine mollusks, whereas the underlying regulatory mechanism in Haliotis diversicolor-a stenohaline marine gastropod with an optimal growth salinity of 30-35‰-under low-salinity conditions remain unclear. In this study, H. diversicolor was exposed to acute low-salinity conditions for 96 h at salinities of 18, 20, and 22‰, with natural seawater at 30‰ as the control. Physiological and histological indices were determined at 12, 24, 48, 72, and 96 h, and multivariate analyses were performed using PCA and DFA.Low salinity expos

A Study on the Bio-responses of a Freshwater Snail (Biomphalaria alexandrina) to Fungal-derived Compounds.

BACKGROUND: Biomphalaria alexandrina snails, as transitional hosts of schistosomiasis, plays an essential part in the spread of the illness. Control of these snails by the substance molluscicides antagonistically influences the oceanic climate, causing poisonous and cancer-causing consequences for non-target life forms. OBJECTIVE: Looking for new naturally safe substances that can treat schistosomiasis disease with minimal side effects on the environment and plants, fish wealth and do not affect vital human functions. METHODS: Fifty fungal species were used to evaluate their activity against

Effect of schistosome infection on protein, glycogen and glucose contents in Biomphalaria arabica and Bulinus truncatus.

B. arabica and B. truncatus the snail intermediate hosts of intestinal and urinary schistosomiasis in Yemen Arab Republic, were bred in the laboratory and infected with S. mansoni and S. haematobium respectively. Schistosome infection caused a marked decrease in the glycogen and protein contents in the tissue of B. arabica and B. truncatus. The glucose content in the hemolymph of both infected snails was lower than that in the normal snails.

Biofilm disruption vs. rapid resistance: Evaluating the role of Calliphora vicina defensin in Staphylococcus aureus clearance.

Defensins are evolutionarily conserved antimicrobial peptides (AMPs) considered a cornerstone of innate immunity. However, the rapid emergence of resistance in pathogens like Staphylococcus aureus challenges our understanding of defensin efficacy in vivo. We used the blowfly Calliphora vicina-an insect adapted to pathogen-rich environments-to investigate S. aureus resistance dynamics. During an active immune response, defensin concentrations in larval hemolymph reach 11-16 μM. At these levels, purified defensin effectively eradicates both planktonic cells and pre-formed S. aureus biofilms (MBE

Evaluation of the Effect of Adipokinetic Hormone/Corazonin-Related Peptide (ACP) on Ovarian Development in the Mud Crab, Scylla paramamosain.

In this study, we identified Sp-ACP and its putative receptor from the mud crab S. paramamosain and explored its potential role in ovarian development. RT-PCR results suggested Sp-ACP was extensively expressed in nervous tissues, the ovary, the middle gut, and the Y-organ, while Sp-ACPR was highly expressed in the ovary. The expression level of Sp-ACP in the ovary, eyestalk, and cerebral ganglia gradually increased during ovarian development, whereas its receptor exhibited an opposite expression pattern in the ovary. Immunofluorescence revealed that ACP was specifically localized in the follic

Antibiotics-induced dysbiosis in gut microbiota affects bumblebee health via regulating host amino acid metabolism.

The gut bacteria can provide nutrition for the host, and regulate host physiological functions and host behavior. In this study, we specifically examined the important roles of free amino acids in the gut microbiota-host interaction. Bumblebees were treated with different concentrations of antibiotics (ampicillin combined with low/high concentrations of tetracycline). Then the effect of antibiotic treatments on the host body weight, gut microbiota, and the free amino acid profiles in the hindgut, hemolymph and brain of bees was evaluated. The results showed that antibiotic treatments resulted

Molecular cloning of cDNAs for two pro-phenol oxidase subunits from the malaria vector, Anopheles gambiae.

Phenol oxidase exists in insect hemolymph as a zymogen, pro-phenol oxidase (pro-PO), which is activated by specific proteolysis in response to infection or wounding. Phenol oxidase catalyses the synthesis of quinones that polymerize to form melanin deposits, which encapsulate parasites and help to seal wounds. Antibodies to pro-PO from Manduca sexta bound to 76, 72, and 71 kDa polypeptide bands from hemolymph of Anopheles gambiae larvae. This antiserum was used to screen a cDNA library from A. gambiae fourth-instar larvae. Full-length clones were isolated for two different pro-POs, designated
