Development of time-resolved chemiluminescence for the determination of antu in river water, wheat, barley, and oat grain samples.

A novel chemiluminescence method for the determination of antu has been developed based on the reaction between potassium permanganate in acid medium with this rat-poison in the presence of formaldehyde as an emission enhancer. The main feature of the system used is that the recording of the whole chemiluminescence intensity-vs-time profiles can be obtained, using the stopped-flow technique in a continuous-flow system. This enables the use of three quantitative parameters adjustable via software settings, one of them a typically kinetic parameter, such as rate of the light-decay reaction, and

Oats and barley toxicity in coeliac patients.

The effect of addition of oats and barley to the glutenfree diets of a group of patients with coeliac disease has been studied using the oral 5 g xylose excretion test to assess small bowel function before and after these supplementations. Both oats and barley were found to be potentially harmful to coeliac patients although the toxic effect of barley was more readily produced than that of oats. On the basis of their results, the authors recommend the exclusion of both barley and oats, in addition to wheat and rye, from the coeliac diet. In view of the well recognized risk of the development o

Conserved protein folds underpin the diversification of secreted proteins in a fungal pathogen.

BACKGROUND: During host colonization, fungal plant pathogens secrete effector-like proteins that alter host cell physiology and target plant-associated microbes. However, rapid evolution and low sequence conservation hinder the study and characterization of these proteins. The fungus Zymoseptoria passerinii infects Hordeum spp. and includes lineages adapted to wild and domesticated barley. To date, the evolution of effector-like proteins in this species has not been addressed. RESULTS: We combined multiple structure-based and network analyses to unravel the secretome of Z. passerinii. We firs…

A haplotype-based approach to explore genotype-by-environment interactions for barley yield.

Climate change poses a significant threat to cereal production, making it increasingly important to understand the role of genotype-by-environment interactions (GEIs) in crops. Traditional breeding approaches consider GEIs at a whole genotype level missing valuable breeding potential that can be dissected by exploring this relationship at a chromosome segment level. A robust commercial dataset consisting of 961 spring barley (Hordeum vulgare L.) genotypes grown across 12 sites was used to explore these questions and to co-locate haplotypes under GEIs with key physiology genes to better underst…

Targeted knockout of barley Ycf54 demonstrates its essential function in the Mg-protoporphyrin IX monomethyl ester cyclase involved in chlorophyll biosynthesis.

BACKGROUND: The Mg-protoporphyrin IX monomethyl ester cyclase is one of the 15 enzymes required for biosynthesis of chlorophyll in plants. The Ycf54 protein is a component associated with this enzyme and probably functions as a chaperone. Due to the lack of mutants in the Ycf54 gene, we targeted this gene in barley (Hordeum vulgare L.) by RNA-guided Cas9 genome editing to dissect its role in chlorophyll biosynthesis. RESULTS: Two guide RNAs were designed and delivered via Agrobacterium-mediated transformation. We generated four homozygous mutant alleles harboring deletions of 1, 2, 6 and 27 b…

Discovery and Evolutionary Analysis of a Novel Genus of Endogenous Pararetroviruses with Long Terminal Repeats.

Despite being widespread in plants, endogenous pararetroviruses (EPRVs) are still poorly understood in barley and many other cereal crops. In this study, the barley reference genome was examined and a new EPRV was identified and named Hvu-EPRV. In contrast to all EPRVs identified thus far, Hvu-EPRV contains long terminal repeats (LTRs) which are similar to LTR retrotransposons. Homologous sequences of Hvu-EPRV were found in a wide range of plants, however, only those in 17 grasses belonging to the six tribes contain LTRs. The insertion times of nested LTR retrotransposons indicated that Hvu-EP

Boosting photosynthesis opens new opportunities for agriculture sustainability and circular economy: The BEST-CROP research and innovation action.

There is a need for ground-breaking technologies to boost crop yield, both grains and biomass, and their processing into economically competitive materials. Novel cereals with enhanced photosynthesis and assimilation of greenhouse gasses, such as carbon dioxide and ozone, and tailored straw suitable for industrial manufacturing, open a new perspective for the circular economy. Here we describe the vision, strategies, and objectives of BEST-CROP, a Horizon-Europe and United Kingdom Research and Innovation (UKRI) funded project that relies on an alliance of academic plant scientists teaming up w

Analysis of growth physiological changes and metabolome of highland barley seedlings under cadmium (II) stress.

This study aims to investigate the physiological changes in growth and metabolic response mechanisms of highland barley under different concentrations of cadmium. To achieve this, cadmium stress was applied to green barley at levels of 20, 40, and 80 mg/L. The results revealed that, under Cd(II) stress, the chlorophyll content and photosynthesis in leaves of highland barley seedlings were inhibited to some extent. Additionally, the malondialdehyde (MDA) content and superoxide dismutase activity increased significantly, indicating that the seedlings were affected by oxidative stress. In additio

Biochar and metal-tolerant bacteria in alleviating ZnO nanoparticles toxicity in barley.

The constant use of zinc oxide nanoparticles (ZnO NPs) in agriculture could increase their concentration in soil, and cause a threat to sustainable crop production. The present study was designed to determine the role of spore-forming and metal-tolerant bacteria, and biochar in alleviating the toxic effects of a high dose of ZnO NPs (2000 mg kg-1) spiked to the soil (Haplic Chernozem) on barley (Hordeum sativum L). The mobile compounds of Zn in soil and their accumulation in H. sativum tissues were increased significantly. The addition of biochar (2.5% of total soil) and bacteria (1010 CFU kg-…

Bioaccessibility of phenolic acids in Canadian hulless barley varieties.

In order to gain understanding of bioaccessibility of phenolic acids in food-grade barley, an investigation was conducted using four cooked whole-grain, hulless, barley varieties. An in vitro digestion model was used to mimic human upper gastrointestinal digestion. Boiling enhanced the extractability of bound phenolic acids while digestion increased the level of free phenolic acids. The high bioaccessibilities observed were likely due to the release of bound phenolic acids during cooking and digestion. The major bioaccessible phenolics were ferulic and p-coumaric acids with bioaccessibility ra

What is gluten?

Gluten is the main storage protein of wheat grains. Gluten is a complex mixture of hundreds of related but distinct proteins, mainly gliadin and glutenin. Similar storage proteins exist as secalin in rye, hordein in barley, and avenins in oats and are collectively referred to as "gluten." The objective was to discuss the biochemical and functional properties of the gluten proteins, including structure, sources, and dietary intakes. Literature was reviewed from food science and nutrition journals. The gluten protein networks vary because of different components and sizes, and variability caused

Acceleration of plastoquinone pool reduction by alternative pathways precedes a decrease in photosynthetic CO2 assimilation in preheated barley leaves.

Heat stress causes inhibition of photosynthetic CO(2) assimilation, affects light photosynthetic reactions and accelerates alternative pathways of plastoquinone pool reduction (APPR). We have studied all these heat-sensitive processes after preheating to a broad range of physiological temperatures (24-46 degrees C) to explore a role of these alternative pathways during heat stress. Primarily, the effective quantum yield of PSII photochemistry was reduced (at 40 degrees C). This PSII downregulation was accompanied by the stimulation of APPR and preceded reduction of photosynthetic CO(2) assimil

Morphological and physiological responses of barley plants grown on soils characterised by metal toxicity and metal deficiency.

Aim of this work was to investigate which are the effects on barley crops grown on two different soils: a soil lacking in Cu, an essential micronutrient (A) and a naturally polluted soil rich in lead, zinc, copper (B). In particular we investigated the relationship between some ecophysiological parameters such as biomass, chlorophyll concentration and guaiacolo peroxidase activity and the chemical-physical properties of the soils like pH, organic matter and heavy metal content. Because metals uptake by plants is strongly correlated with the bioavailable fraction rather then their total amount

Decline of pesticide residues from barley to malt.

The fate of dinitroaniline herbicides (pendimethalin and trifluralin), organophosphous insecticides (fenitrothion and malathion), and pyrimidine (nuarimol) and triazole (myclobutanil and propiconazole) fungicides from barley to malt was determined. Several samples for residue analysis were taken after each stage of malting (steeping, germination and kilning). Pesticide residue analysis was carried out by GC/ITMS in selected ion monitoring mode. Pesticides decline along the process, although in different proportions. The carryover of residues after steeping was 45-85%. A good correlation (r > 0

Is carbonyl sulfide a precursor for carbon disulfide in vegetation and soil? Interconversion of carbonyl sulfide and carbon disulfide in fresh grain tissues in vitro.

The interconversion of carbonyl sulfide (COS) and carbon disulfide (CS(2)) was studied in the roots and shoots of barley and chickpeas. Ratios of conversion gases, K, 40 h after the addition of COS or CS(2) are recorded. The proportion of COS converted to each of CS(2), CO, and H(2)S and the proportion of CS(2) converted to COS were greater in roots than in shoots. More COS was converted to CS(2) than CS(2) to COS in roots and shoots of barley and chickpeas. The amount of COS converted to H(2)S and CO was 8 times the amount converted to CS(2) in barley and 3-4 times the amount in chickpeas. Ca

Endonucleases for UV-irradiated and depurinated DNA in barley chloroplasts.

Lysates of barley chloroplasts release more radioactivity into acid soluble form from UV-irradiated and alkylated-depurinated E. coli [3H] DNA than from intact DNA. By means of affinity chromatography on depurinated DNA-cellulose and/or UV irradiated DNA-cellulose and by electrophoresis in polyacrylamide gels, four activities on depurinated DNA were separated. One of these contained activity against heavily UV-irradiated /270 J.m-2/ native DNA. In addition, two other nucleases specific towards UV-DNA were separated. One of them was active on native and heat denatured DNA irradiated with 10 J .

Evidence of biological nitrification inhibition in barley to support sustainable nitrogen management.

Nitrification drives nitrogen loss in agricultural systems, resulting in leached nitrate and increased emissions of the greenhouse gas nitrous oxide, thereby reducing nitrogen use efficiency (NUE). Biological nitrification inhibition (BNI) provides a promising nature-based solution to low-NUE by suppressing nitrifying microorganisms via plant-exuded bioactive metabolites. Although BNI is well documented in several major cereal grasses, evidence of BNI in barley (Hordeum vulgare) is lacking. This study demonstrates barley BNI activity through suppression of rhizosphere ammonia-oxidizer abundanc…

Bi-Parental Mapping and Validation of Barley Yellow Dwarf Virus Resistance Locus in Barley Cultivar 'Lightning'.

Barley Yellow Dwarf Virus (BYDV), caused by multiple viruses within the Luteoviridae family and primarily transmitted by aphids, is among the most economically significant viral diseases impacting barley (Hordeum vulgare subsp. vulgare) production worldwide, resulting in stunted growth, leaf discoloration, and considerable reductions in grain yield. To validate previously reported BYDV resistance quantitative trait loci (QTL) in the barley cultivar 'Lightning', a doubled haploid (DH) population (n = 131) derived from a cross between 'Lightning' and the susceptible line 'DH141222' was evaluated…

Carbohydrate-Mediated Cellular Uptake of Boronic Acid Hybrids In Cellulo and Insulin-Deficient Zebrafish: Live Imaging and Application of Multiphoton FLIM.

Understanding how synthetic molecular probes interact with biological carbohydrates is essential for theranostics design and discovery. We report new supramolecular polysaccharide complexes, comprising simultaneously functionalised boronic acids conjugated to xanthate-based fluorescent tags bound to saccharides such as glucose or β-D-glucan (a natural product from Hordeum Vulgare). These hybrids were investigated in solution and in the cellular milieu using advanced biophysical methods, including multiphoton fluorescence lifetime imaging microscopy (MP-FLIM). A new type of fluorescent hybrid n…

Metabolic Profiling Analysis Reveals the Potential Contribution of Barley Sprouts against Oxidative Stress and Related Liver Cell Damage in Habitual Alcohol Drinkers.

Chronic excessive alcohol consumption is associated with multiple liver defects, such as steatosis and cirrhosis, mainly attributable to excessive reactive oxygen species (ROS) production. Barley sprouts (Hordeum vulgare L.) contain high levels of polyphenols that may serve as potential antioxidants. This study aimed to investigate whether barley sprouts extract powder (BSE) relieves alcohol-induced oxidative stress and related hepatic damages in habitual alcohol drinkers with fatty liver. In a 12-week randomized controlled trial with two arms (placebo or 480 mg/day BSE; n = 76), we measured c…
