Induction and measurement of the early stage of a host-parasite interaction using a combined optical trapping and Raman microspectroscopy system.

Understanding and quantifying the temporal acquisition of host cell molecules by intracellular pathogens is fundamentally important in biology. In this study, a recently developed holographic optical trapping (HOT)-based Raman microspectroscopy (RMS) instrument is applied to detect, characterize and monitor in real time the molecular trafficking of a specific molecular species (isotope-labeled phenylalanine (L-Phe(D8)) at the single cell level. This approach enables simultaneous measurement of the chemical composition of human cerebrovascular endothelial cells and the protozoan parasite Toxopl

Cortisol influences the host-parasite interaction between the rainbow trout (Oncorhynchus mykiss) and the crustacean ectoparasite Argulus japonicus.

The host-parasite interaction between the rainbow trout Oncorhynchus mykiss and the fish louse Argulus japonicus was investigated by administering low levels of dietary cortisol before infecting the fish with low numbers of the parasite. After 24 h, the dietary cortisol treatment elevated blood cortisol and glucose levels and stimulated the synthesis of secretory granules in the upper layer of skin cells. Infection with 6 lice per fish caused skin infiltration by lymphocytes, also in areas without parasites. The lymphocyte numbers in the blood at 48 h post-parasite infection were reduced. Othe

De Novo transcriptome assembly of Ligula intestinalis (Linnaeus, 1758) Gmelin, 1790 (Cestoda: Diphyllobothriidae) plerocercoids and their host, the common bream Abramis Brama L. (Cypriniformes: Leuciscidae): model for studying host-parasite interactions.

BACKGROUND: Ligula intestinalisis a parasitic tapeworm with a complex life cycle. During the procercoid stage, they parasitize copepods. Later, the cestodes develop into plerocercoids in the body cavity of plankton-feeding fish before finally maturing within the intestines of piscivorous birds. The impact of plerocercoid infection on hosts may be dramatic, including organ compression, disrupted maturation and reproduction, and alter behavior. These effects, which are mediated by neuroendocrine and immunomodulatory mechanisms, can also lead to a mass fish kill. L. intestinalis and their host fi

Shotgun proteomics of Strongyloides venezuelensis infective third stage larvae: Insights into host-parasite interaction and novel targets for diagnostics.

Strongyloides venezuelensis is an important alternative source of antigen for the serologic diagnosis of human strongyloidiasis. Proteomics techniques applied to the analysis of the protein content of infective third stage larvae (iL3) of S. venezuelensis provide a powerful tool for the discovery of new candidates for immunodiagnosis. This study presents an overview of the protein iL3 S. venezuelensis focusing on the diagnosis of strongyloidiasis. A total of 877 proteins were identified by shotgun proteomics. Many of these proteins are involved in different cellular processes, metabolic as wel

Impacts of artificial light at night on interactions between four trematode parasites and their marine invertebrate hosts: species-specific effects?

Parasitism is influenced by anthropogenic stressors, among which artificial light at night (ALAN) remains understudied. With the expansion of urban areas and the development of lighting technology, the range and intensity of ALAN's effects on host-parasite interactions are increasing. Trematodes, with their complex life cycles, are particularly sensitive to anthropogenic stressors, especially during the transmission of motile cercariae (infective stages) from the first to the second intermediate host while exposed to external conditions. We examined how three types of ALAN (warm white LED, col…

Majority of the erythrocyte binding proteins of the Pvfam "a" family of Plasmodium vivax interact with Basigin to assist parasite entry into the host cell.

Molecular mechanisms of red cell invasion by the Plasmodium vivax parasite remain obscure since information on receptor-ligand interaction is scarce. Several proteins of the P. vivax Pvfam "a" family are known to bind with host erythrocytes. Some of them share their erythrocyte receptors with each other and vice versa, but the identification of these receptors is awaited with the exception of PvTRAg38. Here, we demonstrate by using solid-phase binding assay and surface plasmon resonance that majority (7 out of 10) of these erythrocyte binding proteins (PvTRAg, PvTRAg33.5, PvTRAg35.2, PvTRAg34,

Proteomic Analysis of Leishmania donovani Membrane Components Reveals the Role of Activated Protein C Kinase in Host-Parasite Interaction.

Visceral leishmaniasis (VL), mainly caused by the Leishmania donovani parasitic infection, constitutes a potentially fatal disease, for which treatment is primarily dependent on chemotherapy. The emergence of a resistant parasite towards current antileishmanial agents and increasing reports of relapses are the major concerns. Detailed research on the molecular interaction at the host-parasite interface may provide the identification of the parasite and the host-related factors operating during disease development. Genomic and proteomic studies highlighted several essential secretory and cytoso

Trichomonas vaginalis and Tritrichomonas foetus: interaction with fibroblasts and muscle cells - new insights into parasite-mediated host cell cytotoxicity.

Trichomonas vaginalis and Tritrichomonas foetus are parasitic, flagellated protists that inhabit the urogenital tract of humans and bovines, respectively. T. vaginalis causes the most prevalent non-viral sexually transmitted disease worldwide and has been associated with an increased risk for human immunodeficiency virus-1 infection in humans. Infections by T. foetus cause significant losses to the beef industry worldwide due to infertility and spontaneous abortion in cows. Several studies have shown a close association between trichomonads and the epithelium of the urogenital tract. However,

Mucosal immune and stress responses of Neoparamoeba perurans-infected Atlantic salmon (Salmo salar) treated with peracetic acid shed light on the host-parasite-oxidant interactions.

Treatment development for parasitic infestation is often limited to disease resolution as an endpoint response, and physiological and immunological consequences are not thoroughly considered. Here, we report the impact of exposing Atlantic salmon affected with amoebic gill disease (AGD) to peracetic acid (PAA), an oxidative chemotherapeutic. AGD-affected fish were treated with PAA either by exposing them to 5 ppm for 30 min or 10 ppm for 15 min. Unexposed fish from both infected and uninfected groups were also included. Samples for molecular, biochemical, and histological evaluations were coll

Tick-crocodilian interactions: a review, with the first record of tick (Acari: Ixodidae) infestation in the saltwater crocodile (Crocodylus porosus), and a concise host-parasite index.

Interactions between ticks and crocodilians (crocodiles, alligators, caiman, and gharials) are poorly studied but may have significant bearing on the ecology and health of these reptiles. The first record of tick infestation of the saltwater crocodile (Crocodylus porosus) is reported along with the first case of infestation by Amblyomma cordiferum on Cuvier's dwarf caiman (Paleosuchus palpebrosus). A review is also provided of tick-crocodilian interactions with a concise host-parasite index.

paraCell: a novel software tool for the interactive analysis and visualization of standard and dual host-parasite single-cell RNA-seq data.

Advances in sequencing technology have led to a dramatic increase in the number of single-cell transcriptomic datasets. In the field of parasitology, these datasets typically describe the gene expression patterns of a given parasite species at the single-cell level under experimental conditions, in specific hosts or tissues, or at different life cycle stages. However, while this wealth of available data represents a significant resource, analysing these datasets often requires expert computational skills, preventing a considerable proportion of the parasitology community from meaningfully inte

Host-Parasite Interaction Between the Nematoda Procamallanus (Spirocamallanus) paraensis and Fish Hoplias malabaricus From the Natural Environment in the Brazilian Amazon.

Histopathological studies of parasitic infections in fish from the natural environment of Brazilian Amazon, are quite scarce. In this study, we investigated the histopathological changes of the proximal intestine of specimens of the Amazonian fish Hoplias malabaricus infected by the hematophagous nematode Procamallanus (Spirocamallanus) paraensis. The prevalence of the infection was 60%, with an average abundance of 1.46 and an average intensity of 2.43 parasites/fish. No significant difference was observed in the prevalence of infection and fish size or sex, but larger fish showed greater inf

Parasite-Host Interaction and Pathophysiology Studies of the Human Relapsing Malarias Plasmodium vivax and Plasmodium ovale Infections in Non-Human Primates.

Malaria remains a serious health concern across the globe. Historically neglected, non-Falciparum human malarias were put back on the agenda by a paradigm shift in the fight against malaria from malaria control to malaria eradication. Here, we review the modeling of the relapsing parasites Plasmodium vivax (P. vivax) and Plasmodium ovale (P. ovale) in non-human primates with a specific focus on the contribution of these models to our current understanding of the factors that govern parasite-host interactions in P. vivax and P. ovale parasite biology and pathophysiology.

Host-parasite interaction: selective Pv-fam-a family proteins of Plasmodium vivax bind to a restricted number of human erythrocyte receptors.

BACKGROUND: Plasmodium vivax synthesizes the largest number of 36 tryptophan-rich proteins belonging to the Pv-fam-a family. These parasite proteins need to be characterized for their biological function because tryptophan-rich proteins from other Plasmodium species have been proposed as vaccine candidates. METHODS: Recombinant P. vivax tryptophan-rich antigens (PvTRAgs) were used to determine their erythrocyte-binding activity by a cell-based enzyme-linked immunosorbent assay, flow cytometry, and a rosetting assay. RESULTS: Only 4 (PvTRAg26.3, PvTRAg34, PvTRAg36, and PvTRAg36.6) of 21 PvTRA

Electron tomography and cryo-SEM characterization reveals novel ultrastructural features of host-parasite interaction during Chlamydia abortus infection.

Chlamydia (C.) abortus is a widely spread pathogen among ruminants that can be transmitted to women during pregnancy leading to severe systemic infection with consecutive abortion. As a member of the Chlamydiaceae, C. abortus shares the characteristic feature of an obligate intracellular biphasic developmental cycle with two morphological forms including elementary bodies (EBs) and reticulate bodies (RBs). In contrast to other chlamydial species, C. abortus ultrastructure has not been investigated yet. To do so, samples were fixed by high-pressure freezing and processed by different electron m

Host-parasite interaction networks between nematodes and non-volant small mammals in Brazil: A systematic review and ecological network analysis.

Rodents and didelphids are highly diverse and abundant groups that can be found in different ecological niches, including anthropized environments. This can favor a high diversity of zoonotic agents, constituting a complex network of ecological interactions. Nematode parasites can have adaptative flexibility to emerge as new diseases, considering its ecology and diversity, so we aimed to evaluate the functional role of a network interaction between small mammals and nematodes parasites. We used a literature search to construct our network. A total of 52 articles were selected for analysis, fro

Host-parasite interaction between trematode, Clinostomum marginatum (Clinostomidae) and armoured catfish, Pterygoplichthys pardalis (Loricariidae) from Brazilian Amazon.

Clinostomid trematodes are only widely studied due to the ability to infect their metacercariae, which can affect amphibians, fish, snakes and occasionally mammals, with occasional records in humans. The Loricariids constitute the most diverse family of neotropical fish, with more than 800 registered species. They present a large heterogeneity of colors and body forms that reflect its high degree of ecological specialization and importance on economic aspects such as ornamentation and food. In spite of these factors, studies involving the diversity of parasitic fauna that affect in these fish
