Elevated extracellular glutamate levels increased the formation of hydroxyl radical in the striatum of anesthetized rat.

Results from various in vitro experiments have indicated that excitotoxicity and oxidative stress are two interrelated major mechanisms in causing neuronal damage in brain disorders such as cerebral ischemia/reperfusion. Thus, we have conducted experiments to investigate whether in the striatum of anesthetized rats the elevated brain extracellular concentrations of glutamate could increase the formation of hydroxyl radical. Elevation of glutamate levels and trapping of hydroxyl radical were accomplished by perfusing Ringer solutions containing both glutamate and salicylic acid through microdia

Reduction of MPP(+)-induced hydroxyl radical formation and nigrostriatal MPTP toxicity by inhibiting nitric oxide synthase.

N-Methyl, 4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) produces experimental parkinsonism after oxidation to N-methylpyridinium ion (MPP+), accumulation in dopamine neurons and concentration in mitochondria. Inhibition by MPP+ of mitochondrial electron transport impairs respiratory function, but the molecular mechanisms of cell death are not clear. We tested the hypothesis that locally produced nitric oxide is a key component in MPTP toxicity by providing a necessary intermediate in the production of hydroxyl free radicals. Inhibition of nitric oxide synthase reduced MPP(+)-induced hydroxyl radi

D-mannitol, a specific hydroxyl free radical scavenger, reduces the developmental toxicity of hydroxyurea in rabbits.

Hydroxyurea (HU) is a potent mammalian teratogen. Within 2-4 hours after maternal injection, HU causes 1) a rapid episode of embryonic cell death and 2) profound inhibition of embryonic DNA synthesis. A variety of antioxidants delays the onset of embryonic cell death and reduces the incidence of birth defects. Antioxidants do not block the inhibition of DNA synthesis, indicating that early embryonic cell death is not caused by inhibited DNA synthesis. We have suggested that some HU molecules may react within the embryo to produce H2O2 and subsequent free radicals, including the very reactive h

Hydroxyl radical footprints reveal novel structural features around the NF I binding site in adenovirus DNA.

We have identified a number of as yet unknown structural abnormalities of the NF I-DNA binding site within the inverted terminal repetition of adenovirus DNA by probing it with a hydroxyl radical footprinting technique. NF I binding alters the accessibility of the deoxyribose moieties to hydroxyl radicals both at the 3' and at the 5' side of the recognition sequence 5'-TGG(N)6GCCAA-3'. A smooth bend at the 5' side of the binding sequence is already present in naked linear DNA and it is further enhanced by protein binding. This could be demonstrated not only by hydroxyl radical footprinting but

Comparative studies of hydroxyl radical and sulfate radical-mediated degradation of aromatic compounds: Changes in electron-donating moieties and disinfection byproduct formation potential.

Aromatic compounds contribute the majority of UV254, electron-donating moieties (EDM), and disinfection byproduct formation potential (DBP-FP) for natural organic matter (NOM). This study selected 4-ethylphenol (4-EP), 1-(4-hydroxyphenyl)ethanol (4-HPE), 1-(4-hydroxyphenyl)ethenone (4-HPA), and 4-hydroxyphenylacetic acid (4-HPAA) as model compounds representing aromatic hydrocarbons, alcohols, ketones, and carboxylic acids, respectively. 4-HPA behaves differently from the other three: its electron-withdrawing carbonyl group substantially increases UV254 while decreasing EDM. During oxidation,

Theoretical insight into reaction mechanisms of 2,4-dinitroanisole with hydroxyl radicals for advanced oxidation processes.

The detailed degradation mechanism of an insensitive explosive, 2,4-dinitroanisole (DNAN), in advanced oxidation processes (AOPs) was investigated computationally at the M06-2X/6-311 + G(d,p)/SMD level of theory. Results obtained show that the addition-elimination reaction is the dominant mechanism. The phenol products formed can continue to be oxidized to benzoquinone radicals that are often detected by experiments and may be the initial reactants of ring-opening reactions. The H-abstraction reaction is an unavoidable competing mechanism; the intermediate generated can also undergo the proces

Isoprene photo-oxidation products quantify the effect of pollution on hydroxyl radicals over Amazonia.

Nitrogen oxides (NO x ) emitted from human activities are believed to regulate the atmospheric oxidation capacity of the troposphere. However, observational evidence is limited for the low-to-median NO x concentrations prevalent outside of polluted regions. Directly measuring oxidation capacity, represented primarily by hydroxyl radicals (OH), is challenging, and the span in NO x concentrations at a single observation site is often not wide. Concentrations of isoprene and its photo-oxidation products were used to infer the equivalent noontime OH concentrations. The fetch at an observation site

Cleavage of human transferrin by Porphyromonas gingivalis gingipains promotes growth and formation of hydroxyl radicals.

Porphyromonas gingivalis, a gram-negative anaerobic bacterium associated with active lesions of chronic periodontitis, produces several proteinases which are presumably involved in host colonization, perturbation of the immune system, and tissue destruction. The aims of this study were to investigate the degradation of human transferrin by gingipain cysteine proteinases of P. gingivalis and to demonstrate the production of toxic hydroxyl radicals (HO*) catalyzed by the iron-containing transferrin fragments generated or by release of iron itself. Analysis by polyacrylamide gel electrophoresis a

Effects of nicotine on hydroxyl free radical formation in vitro and on MPTP-induced neurotoxicity in vivo.

Parkinson's disease (PD) is one of the most frequent disorders of the basal ganglia. From epidemiological studies there is a controversial discussion on the question whether tobacco smoking is correlated with a decreased incidence of PD. The present study aimed to elucidate the role of nicotine and its potential neuroprotective effects in a rodent model of PD. These effects may be related to an altered hydroxyl radical formation; this possibility was studied in vitro. Nicotine and alpha-phenyl-N-tert-butyl nitrone (PBN) were examined in a cell-free in vitro Fenton system (Fe3+/EDTA + H2O2) for

Determination of hydroxyl radicals using salicylate as a trapping agent by gas chromatography-mass spectrometry.

OBJECTIVES: To establish a sensitive method for measuring hydroxyl radical formation in biological systems using salicylate as probe. METHODS: Salicylate hydroxylation products and related aromatic compounds were extracted and converted to trimethylsilyl (TMS) derivatives. The derivatives were analyzed by gas chromatography-mass spectrometry (GC-MS). Quantitation was achieved by selected-ion-recording (SIR) with benzoic acid (ring-D5) as an internal standard. RESULTS: All compounds were well separated and specifically quantitated by a GC-MS procedure. Standard curves were linear in the conce

Production of hydroxyl radicals and their disassociation from myocardial cell injury during calcium paradox.

The production of hydroxyl radicals during calcium paradox injury was investigated by measuring the production of 2,5-dihydroxybenzoic acid (2,5-DHBA) from salicylate. Four groups of rats were analyzed. In the first group, isolated hearts were perfused with calcium-free medium for 10 minutes followed by perfusion with medium containing Ca++ for 10 minutes. In the other groups, 0.25 microM N,N'-diphenyl-1,3-phenylenediamine (DPPD), 80 microM cytochrome c, or 450 U/ml catalase was added. Coronary effluent was analyzed for the presence of 2,5-DHBA, and tissue sections were examined using light mi

Ab initio kinetics and thermochemistry of hydrogen atom abstraction from solketal by hydroxyl radicals.

As oxygenated biofuels gain prominence in sustainable energy strategies, understanding their reaction kinetics with hydroxyl (˙OH) radicals is essential for predicting combustion behavior. In this work, all six hydrogen-atom abstraction (HAA) pathways in the solketal (2,2-dimethyl-1,3-dioxolane-4-methanol) + ˙OH reaction were characterized using high-level ab initio calculations and variational transition state theory (VTST). Molecular geometries and vibrational frequencies were obtained at the M06-2X/6-311++G(d,p) level, with single-point energies refined at the QCISD(T)/CBS level. Low-freque

Production and prediction of hydroxyl radicals in distinct redox-fluctuation zones of the Yellow River Estuary.

Hydroxyl radicals (·OH) produced in subsurface sediments play an important role in biogeochemical cycles. One of the major sources of·OH in sediments is associated with reduced compounds (e.g., iron and organic matter) oxygenation. Moreover, the properties of iron forms and dissolved organic matter (DOM) components varied significantly across redox-fluctuation zones of estuaries. However, the influence of these variations on mechanisms of·OH production in estuaries remains unexplored. Herein, sediments from riparian zones, wetlands, and rice fields in the Yellow River Estuary were collected to

VUV Activated Fe(VI) by Promoting the Generation of Intermediate Valent Iron and Hydroxyl Radicals.

In this study, vacuum ultraviolet (VUV) was first proposed to activate ferrate (Fe(VI)) for degrading micropollutants (e.g., carbamazepine (CBZ)). Results indicated that VUV/Fe(VI) could significantly facilitate the CBZ degradation, and the removal efficiencies of VUV/Fe(VI) were 30.9-83.4% higher than those of Fe(VI) at pH = 7.0-9.0. Correspondingly, the degradation rate constants of VUV/Fe(VI) were 2.3-36.0-fold faster than those of Fe(VI). Free radical quenching and probe experiments revealed that the dominant active species of VUV/Fe(VI) were •OH and Fe(V)/Fe(IV), whose contribution ratios

Insights into phenanthrene attenuation by hydroxyl radicals from reduced iron-bearing mineral oxygenation.

The oxygenation of Fe(II)-bearing minerals for hydroxyl radicals (HO•) formation and contaminant attenuation receive increasing attention, while the mechanisms for specific Fe(II) species in manipulating HO• formation and contaminant attenuation are unclear. Herein, a total of four Fe(III)-bearing minerals were applied in the reduction-oxygenation processes to produce HO•. Results showed that the total HO• generated from the Fe-(oxyhydr)oxides were significantly higher than those from the Fe-silicates, with the order of goethite and hematite (~1500 μmol kg-1) > Fe-montmorillonite (~550 μmol kg

Reactions of methyl, hydroxyl and peroxyl radicals with the DOTA chelating agent used in medical imaging.

The mechanism of reaction of DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) with ·CH3, CH3O2· and ·OH radicals were studied. The radicals were formed in situ radiolytically. The methyl radicals react orders of magnitude slower with DOTA and with MIII(DOTA)- than the hydroxyl radicals. The various final products were identified and mechanisms for their formation are proposed. CH3O2· radicals do not react, or react too slowly to be observed, with DOTA and with MIII(DOTA)- as long as the central cation is not oxidized by the peroxyl radical. The results imply that synthesis of th

Iron-generated hydroxyl radicals kill retinal cells in vivo: effect of ferulic acid.

Siderosis bulbi is vision threatening. An investigation into its mechanisms and management is crucial. Experimental siderosis was established by intravitreous administration of an iron particle (chronic) or FeSO(4) (acute). After siderosis, there was a significant dose-responsive reduction in eletroretinogram (a/b-wave) amplitude, and an increase in OH level, greater when caused by 24 mM FeSO(4) than that by 8 mM FeSO(4). Furthermore, the FeSO(4)-induced oxidative stress was significantly blunted by 100 microM ferulic acid (FA). Siderosis also resulted in an excessive glutamate release, increa

Block of cardiac ATP-sensitive K(+) channels reduces hydroxyl radicals in the rat myocardium.

The present study examined whether opening of an ATP-sensitive K(+) (K(ATP)) channel can induce hydroxyl free radical ((*)OH) generation in the rat myocardium. Sodium salicylate in Ringer's solution (0.5 nmol/microl/min) was infused directly through a microdialysis probe to detect the generation of (*)OH as reflected by the nonenzymatic formation of 2,3-dihydroxybenzoic acid (DHBA). Induction of cromakalim (100 microM), a K(ATP) channel opener, through the microdialysis probe significantly increased the level of 2,3-DHBA. Another K(ATP) channel opener, nicorandil, also increased the level of 2
