Immunoinformatics analysis of the proteins MPT83 and MPT51 to design a possible chimeric vaccine against Mycobacterium tuberculosis.

Mycobacterium tuberculosis (Mtb) is the pathogen that causes tuberculosis (TB). This disease affects one-third of the world's population, mainly in its latent form. The use of reverse vaccinology and immunoinformatics stands out for the production of vaccines based on peptides or proteins, since they are more specific, safe, effective and economical. The present study evaluated the immunological potential of the proteins MPT83 and MPT51 for vaccine production, comparing them with MPT64. To do this, the sequences of these proteins from MTB H37Rv were downloaded and analyzed. The prediction of T

Designing of a Multi-epitope Vaccine against the Structural Proteins of Marburg Virus Exploiting the Immunoinformatics Approach.

Marburg virus disease (MVD) caused by the Marburg virus (MARV) generally appears with flu-like symptoms and leads to severe hemorrhagic fever. It spreads via direct contact with infected individuals or animals. Despite being considered to be less threatening in terms of appearances and the number of infected patients, the high fatality rate of this pathogenic virus is a major concern. Until now, no vaccine has been developed to combat this deadly virus. Therefore, vaccination for this virus is necessary to reduce its mortality. Our current investigation focuses on the design and formulation of

Design of a novel epitope-based tetravalent subunit vaccine against dengue virus: An immunoinformatic approach.

Dengue imposes a profound global impact, with millions affected annually. Its transmission by Aedes mosquitoes poses significant challenges to combat, aggravated by urbanization and climate change. Despite efforts, no impeccable antiviral treatment exists to date, highlighting the urgency for a vaccine. Developing one encounters hurdles like the four distinctive serotypes of the virus and complex immune responses. In this study, we employed an immunoinformatics approach to design an epitope-based tetravalent subunit vaccine aimed at confronting all DENV serotypes. The study contemplates epitop

Immunoinformatic Design and Evaluation of a Multi-Epitope mRNA Vaccine RP14914P Targeting Latent Tuberculosis Infection.

Background: Latent tuberculosis infection (LTBI) is the principal reservoir for active tuberculosis, with >85% of cases attributable to reactivation. Bacillus Calmette-Guérin fails to block this transition, leaving a critical gap in prevention. Methods: An immunoinformatics/reverse-vaccinology pipeline was applied to seven dormancy-related antigens retrieved from Mycobrowser. T-cell epitopes were predicted with NetMHCI/IIpan-4.1 and B-cell epitopes with ABCpred; antigenicity, allergenicity, and toxicity were evaluated with VaxiJen, AllerTOP, and ToxinPred. Secondary/tertiary structures were mo

A novel immunoinformatic approach for design and evaluation of heptavalent multiepitope foot-and-mouth disease virus vaccine.

BACKGROUND: Foot-and-mouth disease virus (FMDV) vaccine development can be a laborious task due to the existence of various serotypes and lineages and its quasi-species nature. Immunoinformatics provide effective and promising avenue for the development of multiepitope vaccines against such complex pathogens. In this study, we developed an immunoinformatic pipeline to design a heptavalent multi-epitope vaccine targeting circulating FMDV isolates in Egypt. RESULT: B and T-cell epitopes were predicted and selected epitopes were proved to be non-allergenic, non-toxic, with high antigenicity, and

Molecular Mimicry between Toxoplasma gondii B-Cell Epitopes and Neurodevelopmental Proteins: An Immunoinformatic Approach.

Epidemiological studies and meta-analyses have shown a strong association between high seroprevalence of Toxoplasma gondii (T. gondii) and schizophrenia. Schizophrenic patients showed higher levels of anti-Toxoplasma immunoglobulins M and G (IgM and IgG) when compared to healthy controls. Previously, in a rat model, we demonstrated that the progeny of mothers immunized with T. gondii lysates before gestation had behavioral and social impairments during adulthood. Therefore, we suggested that T. gondii infection can trigger autoreactivity by molecularly mimicking host brain proteins. Here, we a

Immunoinformatics Studies and Design of a Potential Multi-Epitope Peptide Vaccine to Combat the Fatal Visceral Leishmaniasis.

Leishmaniasis is a neglected tropical disease caused by parasitic intracellular protozoa of the genus Leishmania. The visceral form of this disease caused by Leishmania donovani continues to constitute a major public health crisis, especially in countries of endemicity. In some cases, it is asymptomatic and comes with acute and chronic clinical outcomes such as weight loss, pancytopenia, hepatosplenomegaly, and death if left untreated. Over the years, the treatment of VL has relied solely on chemotherapeutic agents, but unfortunately, these drugs are now faced with challenges. Despite all effo

Human immune responses to H. pylori HLA Class II epitopes identified by immunoinformatic methods.

H. pylori persists in the human stomach over decades and promotes several adverse clinical sequelae including gastritis, peptic ulcers and gastric cancer that are linked to the induction and subsequent evasion of chronic gastric inflammation. Emerging evidence indicates that H. pylori infection may also protect against asthma and some other immune-mediated conditions through regulatory T cell effects outside the stomach. To characterize the complexity of the CD4+ T cell response generated during H. pylori infection, computational methods were previously used to generate a panel of 90 predicted

Immunoinformatics-driven multi-epitope vaccine design as a promising strategy against multidrug-resistant pathogens: a comprehensive review.

Infections caused by multidrug-resistant (MDR) pathogens, both gram-negative and gram-positive organisms, have transformed into a silent global pandemic. These pathogens, especially ESKAPE pathogens, exhibit wide resistance patterns to several clinically significant antibiotics. This has decreased the effectiveness of already available antibiotics, increasing mortality, morbidity, economic burden and prolonged hospital stay. As the discovery of new antibiotics is an extensive and time-consuming process, an urgent need for innovative and practical preventive strategies arises. This review empha

Design of Multi-Epitope Cancer Vaccine Targeting p53 Tumor Antigen using Myotis lucifugus Sequencers: An Immunoinformatics Approach.

The p53 tumor suppressor protein is dysregulated in more than 50% of human cancers, representing an attractive target for immunotherapy. However, immune tolerance to the self-antigen limits the effectiveness of traditional p53-based vaccines. The presented study is based on a comprehensive in silico approach to design a multi-epitope cancer vaccine using the p53 sequence from Myotis lucifugus (little brown bat) that would break the self-tolerance without losing cross-reactive immunogenicity. Human and bat p53 amino acid sequences were aligned using claustral omega, with a highly conserved core

Unveiling reverse vaccinology and immunoinformatics toward Saint Louis encephalitis virus: a ray of hope for vaccine development.

INTRODUCTION: Infectious diseases continue to challenge human health with high incidence and mortality rates worldwide. Notably, the adaptability of RNA viruses, highlighted by outbreaks of SARS, MERS, and COVID-19, emphasizes the timely need for effective therapeutics. Saint Louis encephalitis virus (SLEV) belonging to the Flaviviridae family is an RNA virus that mostly affects the central nervous system (CNS) of humans. Although supportive care treatments such as antiemetics and painkillers are being used against SLEV infection, it still lacks potential therapeutics for the effective treatme

A novel mRNA-based multi-epitope vaccine for rabies virus computationally designed via reverse vaccinology and immunoinformatics.

The current research investigated the development of a multi-epitope mRNA vaccine against the rabies virus on the basis of viral proteomes via the use of bioinformatic tools and reverse vaccinology. The aim of this study was to address the limitations of the currently available rabies vaccine by eliciting strong and long-lasting humoral and cellular immune responses. The cytotoxic T lymphocytes (CTLs), helper T lymphocytes (HTLs), and linear B-cell epitopes (LBLs) were mapped and prioritized from four top-ranking vaccine targets (nucleoprotein, phosphoprotein, matrix, and glycoprotein) that we

Immunoinformatics Predictions on Variable Mycobacterium tuberculosis Lineage 6 T Cell Epitopes and HLA Interactions in West Africa.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a global health challenge. The human-adapted TB-causing bacteria are distributed into ten lineages with distinct global distributions and clinical outcomes. Mtb lineages 4 (L4) and L6 are good prototypes of these differences, because L4 is globally prevalent, whereas L6 is geographically restricted to West Africa and associated with slower disease progression. Given the fundamental role of T cells for the control of TB, we questioned whether Mtb L4 or L6 antigens and HLA interactions would be disrupted in West African hosts
