Recombinant production of the insecticidal scorpion toxin BjαIT in Escherichia coli.

Scorpion long-chain insect neurotoxins have important potential application value in agricultural pest control. The difficulty of obtaining natural toxins is the major obstacle preventing analyses of their insecticidal activity against more agricultural insect pests. Here we cloned the insect neurotoxin BjαIT gene into the pET32 expression vector and expressed the resulting thioredoxin (Trx)-BjαIT fusion protein in Escherichia coli. Soluble Trx-BjαIT was expressed at a high level when induced at 18 °C with 0.1 mM isopropyl β-d-1-thiogalactopyranoside, and it was purified by Ni2+-nitriloacetic

Isobutyl amides--potent compounds for controlling Diatraea saccharalis.

BACKGROUND: A dichloromethane-methanol extract of the seeds of Piper tuberculatum Jacq. (Piperaceae) and two isobutyl amides, 4,5-dihydropiperlonguminine (1) and pellitorine (2), which were isolated by chromatographic methods, were assayed for their lethality against the sugarcane borer Diatraea saccharalis F. (Lepidoptera: Pyralidae). RESULTS: Bioassays were carried out with fourth-instar caterpillars through topical application of test solutions to the dorsal surface of the prothorax, and dose-response correlations were determined. Significant insect mortalities were observed 24, 48 and 72

Isolation and characterization of a novel lepidopteran-selective toxin from the venom of South Indian red scorpion, Mesobuthus tamulus.

BACKGROUND: Scorpion venom contains insect and mammal selective toxins. We investigated the venom of the South Indian red scorpion, Mesobuthus tamulus for the purpose of identifying potent insecticidal peptide toxins. RESULTS: A lepidopteran-selective toxin (Buthus tamulus insect toxin; ButaIT) has been isolated from this venom. The primary structure analysis reveals that it is a single polypeptide composed of 37 amino acids cross-linked by four disulfide bridges with high sequence homology to other short toxins such as Peptide I, neurotoxin P2, Lqh-8/6, chlorotoxin, insectotoxin I5A, insect

[Vaccination against two vector-borne diseases: bluetongue and West Nile].

In 1999 and 2006, two viral diseases emerged massively and unexpectedly in the United States (West Nile disease) and northern Europe (bluetongue disease). Control of infectious diseases transmitted by insect vectors is based on a variety of approaches (including sanitary measures), but primarily on vaccination. Vaccination is more efficient and less expensive than monitoring of insect vectors. The dynamics and epidemiology of two arboviral diseases (West Nile and bluetongue) are described, together with the different vaccines and vaccination methods.

Removal of gut symbiotic bacteria negatively affects life history traits of the shield bug, Graphosoma lineatum.

The shield bug, Graphosoma lineatum (Heteroptera, Pentatomidae), harbors extracellular Pantoea-like symbiont in the enclosed crypts of the midgut. The symbiotic bacteria are essential for normal longevity and fecundity of this insect. In this study, life table analysis was used to assess the biological importance of the gut symbiont in G. lineatum. Considering vertical transmission of the bacterial symbiont through the egg surface contamination, we used surface sterilization of the eggs to remove the symbiont. The symbiont population was decreased in the newborn nymphs hatched from the surface

Mountain Plover responses to deltamethrin treatments on prairie dog colonies in Montana.

Pyrethroid insecticides containing deltamethrin provide broad spectrum insect control that can adversely affect food supplies of insectivorous birds. I hypothesized that this could result in lowered nest survival for a ground-nesting insectivorous bird, the Mountain Plover (Charadrius montanus), which preferentially nests on prairie dog colonies. I studied Mountain Plover nest survival in 2003-2010 at a small cluster of black-tailed prairie dog (Cynomys ludovicianus) colonies in north-central Montana. Three colonies were treated with deltamethrin to control fleas and limit the spread of plague…

The insects as an assessment tool of ecotoxicology associated with metal toxic plants.

In this article, the assessment of lethal effects of Copper (Cu) on Luffa acutangula and Spinacia oleracea plants investigated in relation to the presence of insect species Oxycarenus hyalinipennis. The analysis of Cu-treated plants displays the information of rapid growth of Oxycarenus hyalinipennis species in triplicate. However, results showed that the impact of metal toxicity appeared as the reduced growth rate of plants, and dense growth of the insect species Oxycarenus halinipennis followed by the chewing/degradation of the toxic plant. The insect's inductees into polluted plants were ju

The effect of ULV-based mosquito control on target and non-target organisms in Hungary: An experimental field study.

Ultra-low volume (ULV) insecticide spraying with deltamethrin as the active ingredient is widely used in mosquito control programs, yet its effectiveness against target mosquitoes and its ecological side effects remain poorly quantified under field conditions in Central Europe. Here, we experimentally evaluated the short-term impact of ground ULV spraying (using deltamethrin + Chrysanthemum cinerariaefolium extract) on both mosquito populations and non-target flying insects in Hungary using a paired before-after-control-impact (BACI) design. Mosquitoes were sampled with BG Sentinel traps, whil

Sterile Insect Technique in a n-patch System with Allee Effect and Mass Trapping: Modelling, Analysis and Simulations.

The sterile insect technique (SIT) is a biological control method aimed at reducing or eliminating populations of pests and disease vectors. This technique involves releasing sterilised insects which, by mating with wild individuals, will reduce the target population. In this study, spatial aspects are incorporated through an explicit metapopulation model, in which both wild and sterile insects disperse across multiple patches. We derive a general sufficient condition ensuring elimination of the wild population in all patches through SIT. When a strong Allee effect is naturally present in each

The 'migratory connectivity' concept, and its applicability to insect migrants.

Migratory connectivity describes the degree of linkage between different parts of an animal's migratory range due to the movement trajectories of individuals. High connectivity occurs when individuals from one particular part of the migratory range move almost exclusively to another localized part of the migratory range with little mixing with individuals from other regions. Conversely, low migratory connectivity describes the situation where individuals spread over a wide area during migration and experience a large degree of mixing with individuals from elsewhere. The migratory connectivity

Bust Stimulation for Sustained Locomotion Control and Autonomous Navigation of Terrestrial Cyborg Beetles.

Terrestrial cyborg insects have become a leading candidate for insect-scale robots. These biohybrid systems utilize living insects as mobile platforms while controlling their locomotion via electrical stimulation of sensory organs. Despite remarkable advancement in locomotion control, the deployment of these cyborgs is hindered by their deterioration in locomotory responses to electrical stimulation, which typically involves prolonged, continuous stimuli. This study proposes burst stimulation as an alternative to the conventional stimulation protocol to achieve more sustained responses of cybo

Detection and identification of insect processed animal proteins in microscopic images using deep learning.

The introduction of terrestrial invertebrate processed animal proteins (PAPs) from insects in animal feed has introduced challenges for official controls in the European Union. Currently, their detection relies only on light microscopy and is entirely dependent on the expertise of trained operators. Recent studies have highlighted two primary problems regarding insect PAP detection: specificity issues due to the misidentification of non-insect structures as insect derived ones, and the difficulty in categorizing a wide morphological diversity of particles resulting from the grinding of whole i

Exposure to artificial light at night (ALAN) alters RNA:DNA ratios in a sandy beach coleopteran insect.

Coastal habitats worldwide, including sandy beaches, are becoming increasingly exposed to Artificial Light at Night (ALAN). Despite the spread of this global stressor, research assessing ALAN potential impacts remain scarce, particularly at the molecular level. This study addressed this gap by assessing the influence of ALAN on the physiological condition of the sandy beach insect Phalerisida maculata Kulzer (Coleoptera, Tenebrionidae). RNA:DNA ratios were used here as a proxy of the insect's nutritional condition in laboratory trials that lasted 20 d. Insects were exposed to two representativ

Caffeine and related methylxanthines: possible naturally occurring pesticides.

Natural and synthetic methylxanthines inhibit insect feeding and are pesticidal at concentrations known to occur in plants. These effects are due primarily to inhibition of phosphodiesterase activity and to an increase in intracellular cyclic adenosine monophosphate. At lower concentrations, methylxanthines are potent synergists of other pesticides known to activate adenylate cyclase in insects. These data suggest that methylxanthines may function as natural insecticides and that phosphodiesterase inhibitors, alone or in combination with other compounds, may be useful in insect control.

Effectiveness of Naturally Occurring Substances Added to Duck Litter in Reducing Emergence and Landing of Adult Musca domestica (Diptera: Muscidae).

Poultry barns provide an ideal breeding environment for Musca domestica L. (Diptera: Muscidae) owing to the controlled temperatures and litter (manure plus bedding material) accumulation. Management techniques that render the litter less suitable for immature stage development, such as the addition of naturally occurring insecticidal substances, could reduce fly infestations. The effectiveness of acetic, boric, and citric acids; diatomaceous earth; and hydrated lime at restricting female landing and growth of house flies in duck litter was evaluated. Litter treated with 1.9% and 4.7% by weight

Studies on the control of plague in the western United States: laboratory trials of six insecticide dust formulations applied to soil for the control of the plague vector Oropsylla montana (Siphonaptera: Ceratophyllidae).

In this study, six insecticide dust formulations were laboratory-tested in soil mixtures ranging in concentration from 40 to 1,280 parts per million (ppm) against a proven vector of bubonic plague, Oropsylla montana (Baker). Pulicidal effects of the six insecticides, which included three organophosphates, two carbamates, and a pyrethroid, were compared by LD(50) values at both ambient and 75% RH. Permethrin (Pyraperm 455 dust), a pyrethroid with a LD(50) less than 40 ppm, demonstrated the most effective pulicidal action for the 13-wk trial period, followed in order by chlorpyrifos, bendiocarb,

Onchocerciasis.

Onchocerciasis is a devastating blinding disease caused by the parasite Onchocerca volvulus that infects about 80 million people, causing blindness and visual impairment in 1-2 million people. In hyperendemic areas, more than half of the population will become blind from onchocerciasis before they die. Blindness is the most important effect of the disease and results, in part, from direct invasion of the eye by microfilariae. The recent development of ivermectin has revolutionized our ability to treat this disease. An annual oral dose of only 150 mg/kg completely suppresses the disease manifes
