The complex mechanistic network of periprosthetic osteolysis: Integration from inflammatory response and tissue stress to signaling pathways.

Periprosthetic osteolysis (PPO) is the main pathophysiological mechanism responsible for aseptic loosening of a prosthesis following a total joint arthroplasty, adversely affecting long-term prosthetic fixation and clinical outcomes. The pathogenesis is primarily rooted in a cascade of biological responses induced by wear particles produced at the prosthesis-bone interface. These particles are identified by pattern recognition receptors (PRRs) on the surface of macrophages, which trigger the activation of signal transduction pathways, including the TLR/NF-κB pathway and the NLRP3 inflammasome.

Integrated physiological and transcriptomic analyses reveal coordinated gill responses to heat stress in pikeperch (Sander lucioperca).

Climate change-driven warming of aquatic environments has made thermal stress an increasingly important factor influencing fish physiological homeostasis. Given their central roles in respiration and osmoregulation, gills are particularly responsive to variations in ambient temperature. Histological examination, physiological measurements, and transcriptome profiling were integrated to investigate the mechanisms associated with heat stress-induced gill injury in pikeperch (Sander lucioperca). Histological analysis revealed that exposure to 29 °C directly caused structural damage to the gills o

Integrated transcriptomics and metabolomics reveal key metabolic pathway responses in Pistia stratiotes under Cd stress.

Cadmium (Cd) can interfere with plant gene expression, change the content of metabolites and affect plant growth. In this study, untargeted metabolomics (LC-MS) and RNA-Seq sequencing were performed on root tissues of Pistia stratiotes exposed to Cd stress. The results showed that cadmium stress affected the accumulation and transport of cadmium in plants and increased the content of soluble sugar, the activities of ascorbate peroxidase (APX), and peroxidase (POD) by 34.89%, 41.45%, and 6.71% on average, and decreased the activity of superoxide dismutase (SOD) by 51.51% on average. A new factor controlling cell envelope integrity in Alphaproteobacteria in the context of cell cycle, stress response and infection.

The bacterial envelope is a remarkable and complex compartment of the prokaryotic cell in which many essential functions take place. The article by Herrou and collaborators (Herrou et al., in press), by a clever combination of structural analysis, genetics and functional characterization in free-living bacterial cells and during infection in animal models, elucidates a new factor, named EipA, that plays a major role in Brucella spp envelope biogenesis and cell division. The authors demonstrate a genetic connection between eipA and lipopolysaccharide synthesis, specifically genes involved in th

Integrated biomarker response-based investigation of oxidative stress in Hemolymph, gills, and digestive glands of freshwater mussels (Unio delicatus) exposed to Etoxazole.

Pesticides extensively used in agriculture pose risks to non-target aquatic organisms. This study assessed the acute and sublethal effects of etoxazole (ETX), an isoxazoline-derived acaricide, on the freshwater mussel Unio delicatus. The 96 h LC₅₀ was calculated as 463.97 mg/L (316.29-662.47 mg/L, 95 % CI). Mussels were exposed to sublethal ETX concentrations for 96 h and 7 days. Total hemocyte counts (THCs), total antioxidant status (TAS), and total oxidant status (TOS) were measured in hemolymph. In this study, elevated total hemocyte counts (THCs) indicated activation of cellular immune res

A New Insight into the Mechanism of Atrazine-Induced Neurotoxicity: Triggering Neural Stem Cell Senescence by Activating the Integrated Stress Response Pathway.

Atrazine (AT), a widely utilized chemical herbicide, causes widespread contamination of agricultural water bodies. Recently, exposure to AT … ture/intact neurons. Mechanistically, AT induces senescence in these recruited HtNSCs by activating integrated stress response signaling. This consequently hinders the repair of damaged neurons by inhibiting HtNSC proliferation and differentiation. Overall, our findings underscore the pivotal role of the integrated stress response pathway in AT-induced HtNSC senescence and …

Targeting the integrated stress response or Ataxin-2 alleviates neurodegeneration in PolyGR models of C9orf72 associated frontotemporal dementia and amyotrophic lateral sclerosis.

Frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS) are fatal, early-onset neurodegenerative diseases. The most common genetic cause of FTD and ALS is a G4C2 hexanucleotide repeat expansion in the C9orf72 gene. This mutation leads to the production of toxic dipeptide repeat proteins (DPRs), via repeat-associated non-AUG (RAN) translation. These DPRs disrupt stress granule (SG) dynamics, with SG regulators such as Ataxin-2 (ATXN2) implicated in disease risk. The integrated stress response (ISR), a key driver of SG formation via eIF2α phosphorylation, has been linked to C9orf72…

Current Knowledge of the Integrated Stress Response in the Development and Management of Acute Myeloid Leukemia: A Novel Target with Encouraging Progress.

Primary or acquired resistance to standard chemotherapy and novel targeted therapies remains a common cause of relapsed/refractory acute myeloid leukemia (AML). The five-year overall survival rate for AML patients remains poor. Exploring novel therapeutic pathways may offer effective strategies to address this challenge. The Integrated Stress Response (ISR) is a signaling pathway that maintains cellular homeostasis by reducing global protein synthesis in response to external and internal stressors. Recent studies have demonstrated that ISR exerts a dual role in AML. Moderate activation of ISR …

Integrated physiological, transcriptomics, and metabolite analysis reveal photosynthetic toxicity related to the bisphenol A stress response mechanism in pepper (Capsicum annuum L.).

Bisphenol A (BPA), an emerging endocrine-disrupting chemical found in irrigation water, soil, and agricultural plastic mulch, presents potential exposure risks to pepper plants (Capsicum annuum L.). This study investigated BPA impact, observing that BPA accumulates in a concentration-dependent manner, with highest levels in roots and lowest in fruit. High BPA exposure decreased photosynthetic pigment content, reduced stomatal size, caused chloroplast dilation, and increased reactive oxygen species and malondialdehyde accumulation, disrupting hormone balance in leaves with a 2.3-fold increase i

GCN2 drives diurnal patterns in the hepatic integrated stress response and maintains circadian rhythms in whole body metabolism during amino acid insufficiency.

Disruptions in circadian rhythms are associated with an increased risk of developing metabolic diseases. General control nonderepressible 2 (GCN2), a primary sensor of amino acid insufficiency and activator of the integrated stress response (ISR), has emerged as a conserved regulator of the circadian clock in multiple organisms. The objective of this study was to examine diurnal patterns in hepatic ISR activation in the liver and whole body rhythms in metabolism. We hypothesized that GCN2 activation cues hepatic ISR signaling over a natural 24-h feeding-fasting cycle. To address our objective,…

Integrated stress response inhibition restores hsa-miR-145-5p levels after IFN-β stimulation in salivary gland epithelial cells. Association between cellular stress and miRNA biogenesis in Sjögren's disease.

Labial salivary glands (LSG) from Sjögren's disease (SjD) patients are characterized by increased levels of pro-inflammatory cytokines, such as type I interferons (IFN-I). These LSG also show activation of the integrated stress response (ISR) with overexpression of protein kinase R (PKR), a known IFN-stimulated gene. In vitro, IFN-I stimulation reproduces the downregulation of hsa-miR-145-5p, which is associated with TLR4 overexpression observed in LSG of SjD patients. MicroRNA levels depend on its biogenesis, which is a multi-step process involving several protein complexes. It is not known w…

Integrative analysis of morphological, transcriptomic, and metabolomic approaches to uncover the function of flavonoids in the salt stress response of Alhagi camelorum.

Salinity or salt stress significantly influences plant productivity, growth, and development, including that of Alhagi camelorum. In the current study, transcriptomic characterization discovered DEGs among the samples of four paired groups. During the metabolomic profiling, the top ten identified primary metabolites are amino acids and their derivatives, fatty acids, alcohols and amines, lipids, GP, heterocyclic compounds, and flavonoids. An analysis of comparative metabolic pathways and KEGG enrichment indicated that salt stress disrupts the biosynthesis of sesquiterpenoids, triterpenoids, an

Integrative genomics resolves rice loci associated with acidic soil-related aluminum, cadmium, and manganese stress responses.

Meta-QTL and transcriptomic integration prioritized transport, redox, and hormonal modules, while contrasting genotypes revealed coordinated differences in metal partitioning, cell-wall-associated spectral profiles and gene expression under combined stress. Rice in acidic and metal-affected soils may encounter aluminum (Al), cadmium (Cd), and manganese (Mn), whose distinct toxicities have partially overlapping effects on ion and redox homeostasis. We combined meta-quantitative trait locus (M-QTL) analysis with 14 public RNA-sequencing datasets and experimental evaluation of contrasting genotyp
