Physico-chemical properties, antioxidant activities and antihypertensive effects of walnut protein and its hydrolysate.

BACKGROUND: Some food proteins hydrolysates are found to possess multiple health effects. In this study, walnut protein (WP) was enzymatically hydrolysed by alcalase and trypsin under optimal conditions. The physico-chemical properties, antioxidant and angiotensin-converting enzyme (ACE) inhibitory activities of WP, alcalase-generated walnut protein hydrolysate (AWPH) and trypsin-generated walnut protein hydrolysate (TWPH) were comparatively studied. Stability properties of the walnut protein hydrolysate (WPH) and the antihypertensive activity in spontaneously hypertensive rats (SHRs) were als

Multi-site evaluation of Chilean native rhizobacteria for the sustainable management of the walnut root-lesion nematode Pratylenchus vulnus (Tylenchida: Pratylenchidae).

Pratylenchus vulnus is the most damaging nematode affecting walnut (Juglans regia L.) worldwide, but evidence on the performance of biological control agents under commercial orchard conditions remains limited. We evaluated two native Chilean rhizobacteria (Bacillus mycoides 25 and Oerskovia sp. UCH-1055), previously shown to achieve >98% in vitro mortality and 76% biocontrol efficacy under greenhouse conditions, using application strategies combining inoculum concentrations of 106-108 CFU mL-1, with or without reinoculation after 30 or 60 days, alongside untreated and chemical controls. Treat…

Multi-targeted antimicrobial action of Juglone against Proteus mirabilis biofilm and virulence.

Proteus mirabilis is a uropathogen frequently implicated in catheter-associated urinary tract infections (CAUTIs), largely due to its ability to form biofilms and express multiple virulence factors. The limitations of conventional antibiotics, along with increasing resistance rates, necessitate the exploration of natural compounds with broader antimicrobial mechanisms against such pathogens. This study investigated the antibiofilm and anti-virulence potential of Juglone, a phytochemical derived from Juglans regia, against P. mirabilis. A series of in vitro assays demonstrated that Juglone not …

Juglone prevents metabolic endotoxemia-induced hepatitis and neuroinflammation via suppressing TLR4/NF-κB signaling pathway in high-fat diet rats.

Juglone as a natural production mainly extracted from green walnut husks of Juglans mandshurica has been defined as the functional composition among a series of compounds. It showed powerful protective effect in various diseases by inhibiting inflammation and tumor cells growth. However, studies on its anti-inflammatory effect based on high-fat diet-induced hepatitis and neuroinflammation are still not available. In this regard, we first investigated whether juglone suppresses high-fat diet-stimulated liver injury, hypothalamus inflammation and underlying mechanisms by which they may recover t…

Radioprotective effects of ethanolic extract from walnut kernel internal septum on lethal X-ray exposure in Wistar rats.

This study assessed the radioprotective efficacy of an ethanolic extract from the walnut kernel internal septum (Juglans regia L.) in Wistar rats exposed to a lethal X-ray dose of 6.73 Gy. The extract was administered at 400, 1600, or 6400 mg/kg, either 2 h before (pre-treatment, n = 7 per group) or after (post-treatment, n = 9 per group) irradiation, with survival monitored over 30 and 60 days. In pre-treated rats, the high dose (6400 mg/kg) consistently provided full protection, surpassing radiation-only controls which showed significant mortality (42.9 ± 7 % at 30 days and 28.5 ± 7 % at 60 …

Walnut supplementation increases levels of UCP1 and CD36 in brown adipose tissue independently of diet type.

Dietary interventions that modulate the brown adipose tissue (BAT) thermogenic activity could represent a promising therapy for metabolic disorders. In order to examine if dietary walnuts intake regulates the expression of BAT thermogenic markers levels in healthy and metabolically challenged (fructose fed) animals, rats were initially divided into the control and fructose-fed groups. After nine weeks, these groups were subdivided into the one kept on the original regimens and the other supplemented with walnuts. High-fructose diet resulted in an increased relative BAT mass and no change in UC

Comparative Transcriptomics and Metabolomics Analysis Revealed the Mechanism of Exogenous Salicylic Acid Improving the Cold Tolerance of Walnut.

Walnut (Juglans regia) is an economically significant woody oil tree species widely cultivated in China. However, its production is increasingly threatened by extreme low-temperature events, such as unseasonal frosts and late-spring cold. Salicylic acid (SA) is a key phytohormone known to enhance cold tolerance in plants, yet its underlying mechanism in walnut remains unclear. In this study, we present the first integrated analysis combining physiological measurements, transcriptomics, and metabolomics to investigate how exogenous SA improves cold tolerance in walnut leaves. Our results showed…

Development of high protein 3D printed dysphagia salmon analogs based on walnut protein-egg white protein emulsion gels.

Dysphagia in the elderly causes inadequate nutrient intake, particularly protein. 3D food printing offers a promising solution by producing customized nutrient-rich foods. To develop 3D-printed salmon analogs, walnut protein (WP)-egg white protein (EWP) emulsion gels were used. Higher EWP content improved printability due to increased mechanical strength and a denser microstructure formed through hydrogen bonding. Subsequently, the effects of pre-processing (baking and steaming) on 3D-printed analogs were investigated. Products baked for 3 min scored highest in sensory attributes (salmon-like

Active packaging application of walnut kernels using ethyl cellulose-based antioxidant electrospun films containing carbon dots.

Sustainable active food packaging films have recently emerged as a critical focus in prolonging food's shelf life. Herein, the study focuses on the biopolymers ethyl cellulose (EC) and polyethylene oxide (PEO) for fabricating fibrous films through a simple electrospinning technique. The FE-SEM analysis showed the uniform morphology of the EC/PEO films with fiber diameters ranging from 0.58 μm to 1.81 μm by varying the amount of PEO in EC during the electrospinning process. Biomass-based sustainable carbon dots (CDs) were made from banana stems and were encapsulated into fibrous films through b

Adsorption-oxidation of hydrogen sulfide on Fe/walnut-shell activated carbon surface modified by NH3-plasma.

Walnut-shell activated carbon (WSAC) supported ferric oxide was modified by non-thermal plasma (NTP), and the removal efficiency for hydrogen sulfide over Fe/WSAC modified by dielectric barrier discharge (DBD) was significantly promoted. The sample modified for 10min and 6.8kV output (30V input voltage) maintained 100% H2S conversion over a long reaction time of 390min. The surface properties of adsorbents modified by NTP under different conditions were evaluated by the methods of X-ray photoelectron spectroscopy (XPS), Brunauer-Emmett-Teller (BET) analysis and in-situ Fourier transform infrar

Detection by real time PCR of walnut allergen coding sequences in processed foods.

A quantitative real-time PCR (RT-PCR) method, employing novel primer sets designed on Jug r 1, Jug r 3, and Jug r 4 allergen-coding sequences, was set up and validated. Its specificity, sensitivity, and applicability were evaluated. The DNA extraction method based on CTAB-phenol-chloroform was best for walnut. RT-PCR allowed a specific and accurate amplification of allergen sequence, and the limit of detection was 2.5pg of walnut DNA. The method sensitivity and robustness were confirmed with spiked samples, and Jug r 3 primers detected up to 100mg/kg of raw walnut (LOD 0.01%, LOQ 0.05%). Therm

Phenolic analysis and erectogenic function of African Walnut (Tetracarpidium conophorum) seeds: The impact of the seed shell on biological activity.

This study investigated the erectogenic potential of African walnut seed (AWS). The extract from AWS cooked with/without shell interacted with phosphodiesterase-5 (PDE-5), arginase, angiotensin-I converting enzymes (ACE), and acetylcholinesterase (AChE); enzymes associated with erectile dysfunction (ED) and Fe2+ -induced malonaldehyde (MDA) production in the isolated penile tissue. The results showed that the extracts inhibited the enzymes and MDA production, but Walnut cooked with shell had the highest effect. This agreed with increased phenolic acids and flavonoids, found in the AWS cooked w

Distinct haplotypes and reversed dominance at a single-gene balanced polymorphism controlling heterodichogamy in two genera of wingnuts.

In the angiosperm mating system of heterodichogamy, two hermaphroditic morphs temporally alternate between male and female flowering phases, promoting disassortative mating between morphs. Heterodichogamy is widespread within Juglandaceae and is controlled by distinct ancient structural variants in Juglans and Carya. Here, we identify a novel inherited basis in two additional Juglandaceae genera, Pterocarya and Cyclocarya. In Pterocarya, we map dichogamy types to an ∼70 kb trans-species structural variant overlapping a single candidate gene, GFAFL, with a dominant haplotype for female-first fl…
