Tetrazine Functionalized Graphene Enables Capture of Ultra-Low Concentrations of Biomacromolecules.

Monolayer graphene has great potential for a range of technological and biomedical applications. To achieve this potential, however, selective functionalization of the surface using mild chemical methods that yield spatially and chemically well-defined functional groups is needed. Here, we describe a method to densely and uniformly modify the surface of graphene using a diazonium salt containing a tetrazine moiety, a functional group amenable to bioorthogonal chemistry. The resulting functionalized graphene efficiently undergoes inverse electron demand Diels-Alder reactions with trans-cyclooct

The Hidden Influences of Macromolecular Crowding: A New Frontier in Cellular Biology and Medicine.

Macromolecular crowding, the dense packing of macromolecules within cells, significantly influences enzyme kinetics, cell signaling, liquid-liquid phase separation, and disease progression. This review explores the multifaceted effects of crowding on osmotic balance, intracellular compartmentalization, and metabolic efficiency. We highlight the exclusion volume effect's role in protein folding, stability, and osmotic homeostasis. Crowding also modulates enzyme activity and kinetics, contributing to metabolic regulation, and plays a crucial role in forming membraneless organelles. Dysregulation

An Intrinsic Photothermal Supramolecular Hydrogel with Robust Mechanical Strength and NIR-Responsive Shape Memory.

Near-infrared (NIR)-triggered shape memory hydrogels with promising mechanical strength hold immense potential in the field of biomedical applications and soft actuators. However, the optical and mechanical properties of currently reported hydrogels usually suffer from limited solubility and dispersion of commonly used photothermal additives in hydrogels, thus restricting their practical implementations. Here,, a set of NIR-responsive shape memory hydrogels synthesized by polyaddition of diisocyanate-terminated poly(ethylene glycol), imidazolidinyl urea (IU), and p-benzoquinone dioxime (BQDO)

Diarylethene-Powered Light-Induced Folding of Supramolecular Polymers.

Helical folding of randomly coiled linear polymers is an essential organization process not only for biological polypeptides but also for synthetic functional polymers. Realization of this dynamic process in supramolecular polymers (SPs) is, however, a formidable challenge because of their inherent lability of main chains upon changing an external environment that can drive the folding process (e.g., solvent, concentration, and temperature). We herein report a photoinduced reversible folding/unfolding of rosette-based SPs driven by photoisomerization of a diarylethene (DAE). Temperature-contro

Thickness dependent self limiting 1-D tin oxide nanowire arrays by nanosecond pulsed laser irradiation.

Fast, sensitive and discriminating detection of hydrogen at room temperature is crucial for storage, transportation, and distribution of hydrogen as an energy source. One dimensional nanowires of SnO2 are potential candidates for improved H2 sensor performance. The single directional conducting continuous nanowires can decrease electrical noise, and their large active surface area could improve the response and recovery time of the sensor. In this work we discuss synthesis and characterization of nanowire arrays made using nanosecond ultraviolet wavelength (266 nm) laser interference processin

Modeling high adsorption capacity and kinetics of organic macromolecules on super-powdered activated carbon.

The capacity to adsorb natural organic matter (NOM) and polystyrene sulfonates (PSSs) on small particle-size activated carbon (super-powdered activated carbon, SPAC) is higher than that on larger particle-size activated carbon (powdered-activated carbon, PAC). Increased adsorption capacity is likely attributable to the larger external surface area because the NOM and PSS molecules do not completely penetrate the adsorbent particle; they preferentially adsorb near the outer surface of the particle. In this study, we propose a new isotherm equation, the Shell Adsorption Model (SAM), to explain t

[Covalent-bound aggregates of equine venezuelan encephalomyelitis virus induced by UV-irradiation].

Formation of Venecuelan equine encephalomyelitis virus (VEE) aggregates induced by UV-light has been studied. The high doses of UV-irradiation induced the protein-protein cross-links resulting in formation of fast sedimenting viral structures. The latter structures are supposed to be presented by the aggregates of several virions linked by the UV-light induced RNA-protein and protein-protein covalent bonds. The lesions in the fine structure of virion envelope was registered by the electron microscopy technique.

Introduction of macromolecules into mammalian cells by cell fusion.

Proteins with molecular weights of up to 500K can be enclosed in erythrocyte ghosts by exposing the ghosts to hypotonic solution containing these proteins. The proteins can then be introduced into recipient cells by fusing the ghosts with the cells using HVJ, PEG, or influenza virus. Some applications of this method are described. By an improved method, 15 kbp DNA and IgM (900 kDa) can be entrapped in erythrocyte membranes and these are then treated with liposomes containing gangliosides and HVJ. These treated membranes containing large macromolecules fuse with almost 100% of the recipient cel

[Immunochemical and electron microscopic analysis of the high-molecular structures in the tick-borne encephalitis virus].

High-molecular structures of tick-borne encephalitis virus were sedimented by centrifugation from virus-containing culture fluid and separated by sucrose concentration gradient centrifugation into 3 main fractions: rapidly sedimenting virions, the fraction sedimenting at a moderate rate, represented by "stellate" structures, and the fraction found on the top of the gradient and represented by structures similar in size and shape to virions, designated slowly sedimenting virions. The latter are first described in the paper. In immunodiffusion, the rapidly sedimenting virions form 1 of the 2 pre

Ferritin: design and formation of an iron-storage molecule.

Although essential for most forms of life, too much iron is harmful. To cope with these antagonistic phenomena an iron-storage molecule, ferritin, has evolved. The structure of horse spleen apoferritin, which has recently been refined, consists of 24 symmetrically related subunits forming a near-spherical hollow shell. In ferritin the central cavity is occupied by an iron core of 'ferrihydrite', a geologically ephemeral mineral found in hot or cold springs and in mine workings, or produced in the laboratory by heating solutions of ferric salts. Ferritin itself forms most readily from apoferrit

Viscosity studies of deoxyhemoglobin S: evidence for formation of microaggregates during the lag phase.

To look for evidence for the formation of deoxy S microaggregates during the lag phase, the kinetics of gelation of purified deoxy S in dilute phosphate buffer were continuously monitored by a Wells-Brookfield cone and plate microviscometer. Three separate manipulations were performed during the lag phase. (1) The rotating cone of the viscometer was stopped for varying intervals and then restarted. (2) Samples were withdrawn from runs in mid-to-late lag phase ("poised") and reintroduced early in the lag phases of other runs with appropriate volume, temperature, and concentration adjustments. (

Assembly of fibrin. A light scattering study.

Using stopped flow light scattering, we show that assembly of fibrin following activation with non-rate-limiting amounts of thrombin or reptilase occurs in two steps, of which the first is end-to-end polymerization of fibrin monomers to protofibrils and the second is lateral association of protofibrils to fibers, in agreement with Ferry's original proposal. Polymerization is found to proceed as a bimolecular association of bifunctional monomers; the overall rate varies as the inverse first power of the concentration; end-to-end association of two monomers, of a monomer and an oligomer, and of

Self-association of band-protein from human erythrocyte membranes in aqueous solutions.

Band 3, the main integral protein of the human erythrocyte membrane, was solubilized and purified in high concentrations of acetic acid. After removal of the organic solvent by dialysis, the self-association of the protein in aqueous solutions was studied by analytical ultracentrifugation. Sedimentation velocity and sedimentation equilibrium experiments clearly demonstrate that, under appropriate conditions of protein preparation, at protein concentrations c less than 200 micrograms/ml, ionic strengths 2 less than 10mM and pH values remote from the isoelectric pH of the protein, band 3 shows a

Near-ultraviolet tyrosyl circular dichroism of pig insulin monomers, dimers, and hexamers. Dipole-dipole coupling calculations in the monopole approximation.

The tyrosyl circular dichroism (CD) has been calculated using the conformation of pig insulin observed in rhombohedral crystals containing 2 zinc atoms per hexamer. These calculations predict that the tyrosyl CD at 275 nm will be enhanced disproportionally as monomers interact to form dimers and as dimers interact to form hexamers. This enhanced tyrosyl CD (delta epsilon per 5800 molecular weight) results from new coupling interactions generated in the regions of contact between monomers and between dimers. These calculations illustrate that a large CD enhancement may accompany aggregation eve

Macroamylasemia: clinical and laboratory features in 7 patients.

The clinical features of patients with macroamylasemia are diverse. A certain correlation between macroamylasemia and the disease of the pancreas or hepato-biliary system may exist, because among 7 patients in this series 4 patients were diagnosed as acute pancreatitis or relapsing pancreatitis and another patient was performed choledochotomy and manipulation of papilla of Vater may had a effect on pancreas. Among other 2 patients, one was completely symptomless and had no abnormal findings and in another the action of anticancer drugs is suspected. The course of macroamylasemia is usually chr

Molecular Crowding by Computational Approaches.

Molecular crowding plays a crucial role in biological and medicinal systems, impacting the structure, behavior, and function of biomolecules within the densely packed environments of cells. This chapter provides an overview of the implications of molecular crowding, exploring how the high concentration of macromolecules such as proteins, nucleic acids, and other biological entities impacts biochemical reactions and cellular processes. The discussion highlights the challenges associated with experimental studies of molecular crowding, including challenges in creating accurate in vitro models, c
