Cyrene™ as a tyrosinase inhibitor and anti-browning agent.

The tyrosinase pathway takes part in the enzymatic process of food browning and is primarily responsible for food spoilage - manifesting itself from a decrease in its nutritional value to a deterioration of taste, which consequently leads to a gradual loss of shelf life. Finding safe and bio-based tyrosinase inhibitors and anti-browning agents may be of great importance in agriculture and food industries. Herein, we showed that Cyrene™ exhibits tyrosinase inhibitory activity (IC50: 268.2 µM), the 1.44 times higher than ascorbic acid (IC50: 386.5 μM). Binding mode studies demonstrated that the

Tracking thermal degradation on passion fruit juice through Nuclear Magnetic Resonance and chemometrics.

Thermal food processing mainly aims to control microorganism in order to extend its shelf life. However, it may induce chemical and nutritional changes in foodstuff. The Nuclear Magnetic Resonance (NMR) coupled to multivariate analysis was used to evaluate the effect of different thermal processing conditions (85 and 140°C for 4; 15; 30; and 60s) on the passion fruit juice using an Armfield pasteurizer. Through this approach it was possible to identify the changes in the juice composition. The temperature and the time lead to a hydrolysis of the sucrose to glucose and fructose. Additionally, j

Nutritional aspects of food preservatives.

Despite the benefits attributed to food preservatives, some concern still remains regarding their safety and possible influence on nutrients. Surprisingly, there is quite a lack of scientific knowledge in this field. In order to describe a few examples, the effects of the extensively used sulphite on thiamine, folates, pyridoxal and other nutrients have been reported. Among its antibrowning effects, inhibition of ascorbic acid browning is also considered. As far as sorbic acid is concerned, notwithstanding its easy reaction with protein, probably the acid environment of the stomach determines

Effect of Simultaneous Administration of Dihydroxyacetone on the Diffusion of Lawsone Through Various In Vitro Skin Models.

Unprotected sunlight exposure is a risk factor for a variety of cutaneous cancers. Topically used dihydroxyacetone (DHA) creates, via Maillard reaction, chemically fixed keratin sunscreen in the stratum corneum with significant protection against UVA/Soret radiation. When used in conjunction with naphthoquinones a naphthoquinone-modified DHA Maillard reaction is produced that provides protection across the UVB/UVA/Soret spectra lasting up to 1 week, resisting sweating and contact removal. The aim of this study was to examine a simplified version of this formulation for effect on UV transmissio…

Glycation of human beta 2-microglobulin in patients with hemodialysis-associated amyloidosis: identification of the glycated sites.

beta 2-Microglobulin (beta 2M) is a major component forming amyloid deposits in patients with hemodialysis-associated amyloidosis (HAA), a serious complication of long-term hemodialysis. Recently, we demonstrated that beta 2M modified with the Maillard reaction is a definite constituent of amyloid deposits in patients with HAA. Our further study demonstrated that this modified beta 2M induces not only chemotaxis of monocytes but also secretion of tumor necrosis factor-alpha, interleukin-1 beta, and interleukin-6 from macrophages, suggesting the potential link of glycation of beta 2M by the Mai…

Some toxic compounds produced in food by cooking and processing.

Both cooking and processing of food can produce toxic compounds in food, if the appropriate precursors are present. N-Nitrosamines, polycyclic aromatic hydrocarbons and phenolic compounds, lipid polymerisation products resulting from deep-fat frying, lipid oxidation products, Maillard-browning products and other products of protein reactions are discussed as well as their formation, concentration and control. The conclusion points out that contamination of human food is almost impossible to avoid and must be considered together with the beneficial effects of food processing on food safety, fla

Insights into the flavor enhancement mechanism during dried bonito processing: An integrated analysis of flavoromics and untargeted lipidomics.

This study aimed to unravel the flavor enhancement mechanisms during dried bonito processing by focusing on five critical stages. Taste char … caramel-like aromas. The dynamic changes in volatile compounds were influenced by lipid metabolism, Maillard reactions, and the transformation of phenolic compounds, which collectively construct the unique fatty-roasted-smoky composite flavor profile of dried bonito products. Untargeted lipidomics results showed that lipid oxidation, primarily driven by phospholipi …

First Evidence for Marked Reducing Sugar Accumulation in Lotus Roots During Postharvest Storage as the Key Driver of Acrylamide Formation in Air-Fried Lotus Root Chips.

Acrylamide is a potentially toxic and neurotoxic compound primarily formed in foods during high-temperature cooking and processing through Maillard reactions involving naturally occurring asparagine and carbonyl compounds. This study investigated the marked variation in acrylamide formation in air-fried lotus root chips prepared from lotus roots obtained from different market sources and examined whether postharvest storage contributes to this variability. Differences in acrylamide levels among market-sourced lotus roots were significantly associated with variation in total reducing sugar cont…

Site-Specific Glycation of Human Heat Shock Protein (Hsp27) Enhances Its Chaperone Activity.

Non-enzymatic posttranslational modifications are believed to affect at least 30% of human proteins, commonly termed glycation. Many of these modifications are implicated in various pathological conditions, e.g., cataract, diabetes, neurodegenerative diseases, and cancer. Chemical protein synthesis enables access to full-length proteins carrying site-specific modifications. One such modification, argpyrimidine (Apy), has been detected in human small heat shock protein Hsp27 and closely related proteins in patient-derived tissues. Thus far, studies have looked into only artificial mixtures of A

Alcoholic Liver Disease Is Associated with Elevated Plasma Levels of Novel Advanced Glycation End-Products: A Preliminary Study.

Elucidating the biochemical mechanisms associated with the progression of alcoholic liver disease (ALD) to more advanced stages such as alcoholic hepatitis (AH) remains an important clinical and scientific challenge. Several hypotheses point to the involvement of advanced glycation end-products (AGEs) in alcohol-associated liver injuries. Recently, we determined the structure of a synthetic, melibiose-derived AGE (MAGE), which was an analog of the novel AGE subgroup AGE10. The primary objective of our study was to determine whether AGE10 was associated with alcoholic hepatitis. The secondary o

Trends in pretreatment and determination methods for furfurals in foods: Update since 2017.

BACKGROUND: Furfurals, key derivatives of Maillard reactions commonly found in everyday foods, have been identified as having significant toxic effects on human health. Excessive intake of furfurals can lead symptoms such as weight loss, poor nutrient metabolism which followed by disease occurrence. The severe carcinogenicity, mutagenicity and genotoxicity of furfurals were well recognized recently. In response, international organizations have established strict limits on the allowable levels of furfurals in food products. Therefore, it is of paramount importance to implement control which me…

Regulating the structure of soybean protein isolate-dextran maillard conjugate through cold plasma treatment: enhancing the storage and processing stability of oil-in-water emulsion loaded with curcumin.

The performance of soybean protein isolate-dextran (SPI-Dex) Maillard conjugates as O/W emulsion stabilizers is fundamentally limited by their structure and modification degree. However, cold plasma (CP) technology offers a compelling approach to overcome these limitations. Therefore, this study investigates the impact of CP treatment time on the structural and emulsifying properties of SPI-Dex conjugates to develop a high-performance, protein-based emulsion delivery system. Results indicated that compared with the control group (0 min), a CP treatment for 5 min (SPI-Dex5) reduced the disulfid

Cysteine-Induced pH-Dependent Formation of Thiols and Sulfides or 2-Acetylthiazole and Pyrazines during Thermal Treatment of N-(1-Deoxy-d-xylulos-1-yl)-alanine.

The influence of pH was studied on volatile flavor formation during thermal treatment of an Amadori rearrangement product (ARP) with or without the addition of cysteine (Cys). The formation of thiols and sulfides or 2-acetylthiazole and pyrazines induced by Cys during thermal degradation of ARP was pH-dependent. At low pH levels, the hydrolysis of Cys to hydrogen sulfide (H2S) was promoted, giving rise to the increase of thiols and sulfides with an obvious meaty aroma. However, alkaline conditions were beneficial for enhancing the cyclization or transformation of imine to the enol structure, w

Glycine-driven abiotic humification: Differential mediation of montmorillonite and kaolinite on the structural evolution of humic-like substances.

Abiotic humification via the Maillard reaction represents an indispensable geochemical process driving humic-like substance (HLS) formation and potential soil organic carbon (C) stabilization. Clay minerals act as critical modulators of this process, yet the differential mediating mechanisms of montmorillonite (Mont) and kaolinite (Kao)-two dominant soil clay minerals with distinct crystal structures-on glycine (Gly)-driven structural evolution of HLS remain poorly understood. Specifically, how Gly dosage couples with clay mineral types to modulate condensation reactions and structural differe…

Advances in molecular, biochemical mechanisms and innovative technologies for browning regulation and quality preservation of fresh-cut lotus (Nelumbo nucifera Gaertn.) rhizome.

Fresh-cut lotus (Nelumbo nucifera Gaertn.) rhizome (FCLR) is highly susceptible to enzymatic browning and non-enzymatic blue/black staining following mechanical wounding, limiting its commercial shelf-life to 3-5 d under refrigeration. Despite extensive empirical research on preservation strategies, a systematic synthesis integrating molecular wounding mechanisms with the design principles of emerging technologies remains lacking. This review critically examines recent progress in browning control and quality preservation of FCLR, dissecting physiological wound responses including reactive oxy
