[Effect of maternal health and prenatal environmental exposure factors on tooth development].

Odontogenesis is a consequence of a complex series of reciprocal signal interactions between odontogenic epithelium and neural crest-derived odontotgenic mesenchyme. These interactions result from a complex interplay of genetic and environmental factors. Given that a fetus develops in the mother, maternal health and environmental exposures have a great influence on tooth development. In this review, we focused on the key issues in the developmental defects of teeth induced by various types of maternal environmental factors, including environmental endocrine disruptors, joint action of two or m

Maternal high estradiol exposure alters CDKN1C and IGF2 expression in human placenta.

INTRODUCTION: The increased maternal estradiol (E2) concentrations induced by assisted reproductive technology (ART) result in lower birth weight of offspring, which is associated with increased risk of adult diseases. However, the exact mechanism remains unknown. The present study investigated the effect of high E2 exposure on the expression of imprinted genes CDKN1C and IGF2 in human placentas and the DNA methylation status of their differential methylation regions (DMRs). METHODS: The mRNA expression of CDKN1C and IGF2 in human placentas and the human trophoblast cells (HTR8) treated with

Intergenerational Effect of Maternal Exposure to Childhood Maltreatment on Newborn Brain Anatomy.

BACKGROUND: Childhood maltreatment (CM) confers deleterious long-term consequences, and growing evidence suggests some of these effects may be transmitted across generations. We examined the intergenerational effect of maternal CM exposure on child brain structure and also addressed the hypothesis that this effect may start during the child's intrauterine period of life. METHODS: A prospective longitudinal study was conducted in a clinical convenience sample of 80 mother-child dyads. Maternal CM exposure was assessed using the Childhood Trauma Questionnaire. Structural magnetic resonance imag

Maternal Iodine Exposure: A Case of Fetal Goiter and Neonatal Hearing Loss.

A 27-year-old gravid 1 at 27 weeks 6 days with a history of hypothyroidism had an ultrasound that demonstrated a 3.9 × 3.2 × 3.3-cm well-circumscribed anterior neck mass, an extended fetal head, and polyhydramnios. Further characterization by magnetic resonance imaging (MRI) showed a fetal goiter. During her evaluation for the underlying cause of the fetal goiter, the patient revealed she was taking nutritional iodine supplements for treatment of her hypothyroidism. She was ingesting 62.5 times the recommended amount of daily iodine in pregnancy. The excessive iodine consumption caused suppres

Maternal exposure to traffic-related air pollution and birth defects in Massachusetts.

Exposures to particulate matter with diameter of 2.5µm or less (PM2.5) may influence risk of birth defects. We estimated associations between maternal exposure to prenatal traffic-related air pollution and risk of cardiac, orofacial, and neural tube defects among Massachusetts births conceived 2001 through 2008. Our analyses included 2729 cardiac, 255 neural tube, and 729 orofacial defects. We used satellite remote sensing, meteorological and land use data to assess PM2.5 and traffic-related exposures (distance to roads and traffic density) at geocoded birth addresses. We calculated adjusted o…

Chronic binge alcohol exposure during pregnancy impairs rat maternal uterine vascular function.

BACKGROUND: Alcohol exposure during pregnancy results in an array of structural and functional abnormalities called fetal alcohol spectrum disorders (FASD). Alcohol dysregulates the exquisite coordination and regulation of gestational adaptations at the level of the uterine vasculature. We herein hypothesized that chronic binge-like alcohol results in uterine vascular dysfunction and impairs maternal uterine artery reactivity to vasoconstrictors and dilators. METHODS: We utilized a once-daily binge alcohol (4.5 g/kg body weight) exposure paradigm (gestational day 7 to 17) in a pregnant rat mo

Exposure to biological maternal sounds improves cardiorespiratory regulation in extremely preterm infants.

OBJECTIVE: Preterm infants experience frequent cardiorespiratory events (CREs) including multiple episodes of apnea and bradycardia per day. This physiological instability is due to their immature autonomic nervous system and limited capacity for self-regulation. This study examined whether systematic exposure to maternal sounds can reduce the frequency of CREs in NICU infants. METHODS: Fourteen preterm infants (26-32 weeks gestation) served as their own controls as we measured the frequency of adverse CREs during exposure to either Maternal Sound Stimulation (MSS) or Routine Hospital Sounds

Fetal and maternal immune responses to methylmercury exposure: a cross-sectional study.

Methylmercury (MeHg) is a ubiquitous environmental contaminant with known neurodevelopmental effects. In humans, prenatal exposures primarily occur through maternal consumption of contaminated fish. In this study, we evaluated the association between prenatal exposure to MeHg and titers of total immunoglobulins (Ig) and specific autoantibodies in both mothers and fetuses by analyzing maternal and cord blood serum samples. We examined multiple immunoglobulin isotypes to determine if these biomarkers could inform as to fetal or maternal responses since IgG but not IgM can cross the placenta. Fin

[Maternal sulfur dioxide exposure and the risk of low birth-weight babies].

The relationship between maternal exposure to sulfur dioxide (SO2) and the risk of low birth-weight (LBW) babies which has been reported in literatures remained inconclusive. In this study, the data on SO2 emission, meteorology, topography as well as LBW and control babies in the Dallas-Fort Worth area of the United States in 1997 were collected to re-examine the associations between maternal SO2 exposure and the risk of LBW in babies. A combination of air dispersion modeling, Geographic Information System (GIS) spatial interpolation and geo-statistical analysis, as well as logistic regression…

Effects of maternal tobacco-smoke exposure on fetal growth and neonatal size.

Exposure to tobacco smoke, through both active and passive measures, has a significant impact on women's health, including effects on the cardiovascular, pulmonary and reproductive systems. Of particular interest is the effect of smoking on pregnancy outcomes. One crucial outcome that has been linked to the subsequent development of both neonatal and adult disease is intrauterine or fetal growth restriction. In this article, we will summarize the effects of smoking on newborn size and fetal growth. We will review evidence showing that tobacco consumption during pregnancy leads to a reduction i

Optic nerve dysfunction in a child following low-dose maternal warfarin exposure.

Because the embryopathy associated with maternal warfarin use seems dose-dependent, some physicians advocate low-dose warfarin for pregnant women requiring anticoagulation. The current case, however, highlights that optic nerve dysfunction (as well as other signs of warfarin embryopathy) can occur after low-dose maternal warfarin exposure.

11β-HSD2 buffers fetal glucocorticoid exposure inducing Per1 expression under maternal stress.

Glucocorticoids (GCs) have been proposed as maternal-fetal communication signals. However, fetal circadian rhythms are initially shielded from maternal entrainment, in addition to delayed circadian clock emergence due to CLOCK suppression. Premature CLOCK/BMAL1 activation disrupts Hes7-driven somite-like structure in gastruloids. Given the genomic proximity of Per1 to Hes7 and their transcriptional ripple effect, the physiological significance of delayed cell-autonomous circadian clock development and the temporal program of maternal-fetal communication during the developmental process has rem

High glucose exposure induces differential pathophysiological changes in feto-maternal interface cells.

Gestational diabetes mellitus, a hyperglycemic condition during pregnancy, increases the risk of macrosomia and preterm birth (PTB). Nutrient-sensing pathways, particularly mTOR in placental trophoblast cells (PTCs), promote fetal overgrowth. NF-κB, oxidative stress, and p38 MAPK pathways in fetal membranes and decidua (DEC) contribute to PTB. However, the impact of hyperglycemia on these compartments remains unclear. We hypothesized that hyperglycemia differentially affects these maternal, placental, and fetal membrane interface cells, inducing macrosomia-associated pathways and perturbing ho

Maternal tributyltin exposure is associated with male-biased immune dysregulation across generations.

Tributyltin (TBT) is an environmental obesogen and endocrine-disrupting chemical that promotes adipogenesis and transgenerational metabolic dysfunction in mice. Although developmental TBT exposure has been linked to male-biased adiposity, insulin dysregulation, and hepatic pathology, its effects on the immune system remain unclear. We investigated whether maternal TBT exposure is associated with persistent immune dysregulation in F1 and F3 offspring. Peripheral blood immunophenotyping data generated by the German Mouse Clinic were reanalyzed in F1 offspring of control- and TBT-exposed dams. Ma

Maternal alcohol exposure and atopic dermatitis in offspring: A scoping review protocol.

Maternal alcohol use is a modifiable, yet significant public health concern, causing fetal alcohol syndrome and neurodevelopmental disorders. Research has predominantly focused on the neurodevelopmental consequences of maternal alcohol consumption, but growing evidence suggests that these effects may extend to the developing immune system, potentially increasing the risk of allergic and inflammatory conditions such as atopic dermatitis in offspring, which remains underexplored. This scoping review aims to map the available literature on the association between maternal alcohol exposure (precon
