A prefrontal cortex-nucleus accumbens circuit attenuates cocaine-conditioned place preference memories.

The infralimbic (IL) subregion of the prefrontal cortex (PFC), via its descending projection to the nucleus accumbens (NAc), inhibits cue-induced drug seeking and reinstatement, but the underlying mechanisms are not fully understood. Here we show that the activity of IL layer 5 pyramidal neurons projecting to the NAc shell (IL-NAcSh neurons) suppresses cocaine-associated memories. Following repeated cocaine exposures in a conditioned place preference paradigm, IL-NAcSh neurons anatomically traced by fluorescent Retrobeads undergo prolonged decrease of membrane excitability, lasting for at leas

TRPA1 antagonism reveals sex-specific mechanisms in morphine withdrawal-induced negative emotional behavior via dopaminergic pathway.

Chronic opioid use facilitates changes in brain reward and affective systems. Cessation of opioid use induces negative affective states characterized by increased anxiety- and despair-like behaviors. In this study, the potential role of transient receptor potential ankyrin 1 channel (TRPA1) was assessed in morphine-induced dependence and negative emotional behaviors. Male and female C57BL/6J mice were exposed to escalating doses of morphine to induce dependence. Naloxone was administered to precipitate withdrawal symptoms. TRPA1 antagonism with A967079 treatment reduced withdrawal syndrome in

Review of neurotransmitters and their role in alcoholism treatment.

Studies on the genetic basis of addiction indicate that the tendency to develop alcoholism is inherited. In addition, alcoholism appears to be associated with a specific neurochemical disorder. Research has focused on the mesolimbic system, which is associated with the ability to feel pleasure (i.e. hypothalamic control centres are related to daily survival activities, and the medial forebrain bundle is involved in the positive reinforcement of addictive drugs). Current findings support the hypothesis that a neurochemical deficiency causes alcohol-dependent individuals to drink. Thus, pharmaco…

Ghrelin receptor antagonism attenuates cocaine- and amphetamine-induced locomotor stimulation, accumbal dopamine release, and conditioned place preference.

INTRODUCTION: Recently we demonstrated that genetic or pharmacological suppression of the central ghrelin signaling system, involving the growth hormone secretagogue receptor 1A (GHS-R1A), lead to a reduced reward profile from alcohol. As the target circuits for ghrelin in the brain include a mesolimbic reward pathway that is intimately associated with reward-seeking behaviour, we sought to determine whether the central ghrelin signaling system is required for reward from drugs of abuse other than alcohol, namely cocaine or amphetamine. RESULTS: We found that amphetamine-as well as cocaine-in

[Role of the monoamine system in the brain on the development of psychological dependence on toluene].

Abuse of volatile organic solvents among youth remains a major social problem in Japan. Organic solvents are cheap and relatively easy to obtain, so they carry the risk of becoming a so-called "gate-way drug" for users. Psychological dependence assessment systems have been established for drug inhalation using the conditioned place preference (CPP) procedure. We found toluene produced the rewarding effect in this new CPP system. The mesolimbic dopamine pathway, which includes dopaminergic neurons in the ventral tegmental area (VTA) of the midbrain and their targets in the limbic forebrain, esp

[Behavioral changes following lesioning of the nucleus accumbens (ACB) and effects of centrally acting drugs in rats (author's transl)].

ACB is one of the sites containing nerve terminals of the mesolimbic dopamine system. We have found that all the male Sprague-Dawley rats with electrolytic lesions of bilateral ACB showed locomotor hyperactivity and hyperemotionality. Muricide was also observed in about 40% of these lesioned rats. Hyperemotionality and muricide were maximum during the first 2-3 days after the lesioning, then gradually declined. Locomotor hyperactivity lasted invariably for over 30 days. Hyperemotionality and muricide were both inhibited by the following drugs given i.p., chlorpromazine; haloperidol; diazepam;

Histone H3 dopaminylation in nucleus accumbens, but not medial prefrontal cortex, contributes to cocaine-seeking following prolonged abstinence.

Enduring patterns of epigenomic and transcriptional plasticity within the mesolimbic dopamine system contribute importantly to persistent behavioral adaptations that characterize substance use disorders (SUD). While drug addiction has long been thought of as a disorder of dopamine (DA) neurotransmission, therapeutic interventions targeting receptor mediated DA-signaling have not yet resulted in efficacious treatments. Our laboratory recently identified a non-canonical, neurotransmission-independent signaling moiety for DA in brain, termed dopaminylation, whereby DA itself acts as a donor sourc

CD81-induced behavioural changes during chronic cocaine administration: in vivo gene delivery with regulatable lentivirus.

CD81, a tetraspanin transmembrane protein involved in cell adhesion, is up-regulated in the mesolimbic dopaminergic pathway 24 h following acute administration of high doses of cocaine [Brenz-Verca et al., (2001) Mol. Cell. Neurosci., 17, 303-316]. Further evidence consecutive with this observation and based on microarray analysis are presented here. In addition, a regulatable lentivirus was developed bearing the rat CD81 gene under the control of a tetracycline inducible system. This lentivirus vector was stereotaxically injected into the ventral tegmental area (VTA) of two groups of animals,

Psychostimulant-induced plasticity of intrinsic neuronal excitability in ventral subiculum.

Psychostimulant drugs such as amphetamine are prescribed to increase vigilance, suppress appetite, and treat attention disorders, but they powerfully activate the dopamine system and have serious abuse potential. Repeated psychostimulant exposure induces neuronal plasticity within the mesolimbic dopamine system. Here we present evidence that repeated amphetamine exposure results in a suppression of intrinsic neuronal excitability in the ventral subiculum, a hippocampal region that activates dopamine neurotransmission. We used patch-clamp recordings from brain slices obtained at different times

Chronic inflammatory pain suppresses alcohol intake and accumbal dopamine response.

Alcohol use disorders (AUDs) are influenced by factors that initiate, maintain, and/or induce relapse. Chronic pain is both a risk factor for and consequence of AUD, sharing neurological pathways that affect the mesolimbic dopaminergic system. This study examines how inflammatory pain impacts long-term alcohol intake and mesolimbic dopamine transmission in alcohol-naïve rats. Inflammatory pain was induced in eight-week-old Sprague Dawley rats using complete Freund adjuvant (CFA), while controls received saline. Two protocols were followed: one group had continuous access to 20 % ethanol for on

From Satiety to Substance Use: Neural Mechanisms of Glucagon-Like Peptide-1 Signaling in Appetite and Reward.

The central glucagon-like peptide-1 (GLP-1) system encompasses homeostatic and hedonic neural circuits. GLP-1-producing neurons within the brainstem project broadly to GLP-1 receptor (GLP-1R)-containing brainstem, hypothalamic, limbic, and mesolimbic nuclei. The diverse expression of the GLP-1R throughout metabolic and reward nuclei positions this system to integrate motivated behavior with physiological state. GLP-1 acts as a state-dependent behavior-ending signal that suppresses appetite, promotes satiety, and dampens hedonic and motivational reward value-effects that extend beyond palatable

Role of melanin-concentrating hormone in drug use disorders.

Melanin-concentrating hormone (MCH) is a neuropeptide primarily transcribed in the lateral hypothalamus (LH), with vast projections to many areas throughout the central nervous system that play an important role in motivated behaviors and drug use. Anatomical, pharmacological and genetic studies implicate MCH in mediating the intake and reinforcement of commonly abused substances, acting by influencing several systems including the mesolimbic dopaminergic system, glutamatergic as well as GABAergic signaling and being modulated by inflammatory neuroimmune pathways. Further support for the role

Neuroendocrinology of reward in anorexia nervosa and bulimia nervosa: Beyond leptin and ghrelin.

The pathophysiology of anorexia nervosa (AN) and bulimia nervosa (BN) are still poorly understood, but psychobiological models have proposed a key role for disturbances in the neuroendocrines that signal hunger and satiety and maintain energy homeostasis. Mounting evidence suggests that many neuroendocrines involved in the regulation of homeostasis and body weight also play integral roles in food reward valuation and learning via their interactions with the mesolimbic dopamine system. Neuroimaging data have associated altered brain reward responses in this system with the dietary restriction a

A unified circuit for social behavior.

Recent advances in circuit manipulation technologies have enabled the association of distinct neural circuits with complex social behaviors. The brain areas identified through historical anatomical characterizations as mediators of sexual and parental behaviors can now be functionally linked to adult social behaviors within a unified circuit. In vivo electrophysiology, optogenetics and chemogenetics have been used to follow the processing of social sensory stimuli from perception by the olfactory system to valence detection by the amygdala and mesolimbic dopamine system to integration by the c

Medication treatment of different types of alcoholism.

Alcoholism remains a serious cause of morbidity and mortality despite progress through neurobiological research in identifying new pharmacological strategies for its treatment. Drugs that affect neural pathways that modulate the activity of the cortico-mesolimbic dopamine system have been shown to alter drinking behavior, presumably because this dopaminergic system is closely associated with rewarding behavior. Ondansetron, naltrexone, topiramate, and baclofen are examples. Subtyping alcoholism in adults into an early-onset type, with chronic symptoms and a strong biological predisposition to

Effects of dopaminergic drugs on locomotor activity in teleost fish of the genus Oreochromis (Cichlidae): involvement of the telencephalon.

Single Oreochromis niloticus and O. mossambicus were placed in an unfamiliar white basin for 21 min, and their activity in this open-field situation was recorded from overhead on video. Apomorphine added to the water (2-8 mg/liter) caused a significant increase in locomotor activity, as assessed by the frequency that a fish swam over a rectilinear array of lines drawn on the base. This effect was attenuated by chlorpromazine (2 mg/liter) and abolished by the D1 antagonist SCH-23390 (1 mg/liter); the D2 antagonist metoclopramide (8 mg/liter) had no effect. Removal of both hemispheres of the tel

An innate brainstem self-other system involving orienting, affective responding, and polyvalent relational seeking: Some clinical implications for a "Deep Brain Reorienting" trauma psychotherapy approach.

Underlying any complex relational intersubjectivity there is an inherent urge to connect, to have proximity, to engage in an experience of interpersonal contact. The hypothesis set out here is that this most basic urge to connect is dependent on circuits based in three main components: the midbrain superior colliculi (SC), the midbrain periaqueductal gray (PAG), and the mesolimbic and mesocortical dopamine systems originating in the midbrain ventral tegmental area. Firstly, there is orienting towards or away from interpersonal contact, dependent on approach and/or defensive/withdrawal areas of

Treatment-related associations of nucleus accumbens connectivity within mesocorticolimbic circuits in depression.

Pharmacological treatment remains a mainstay in managing depression, yet the neural correlates associated with treatment response remain incompletely understood. This study used multimodal neuroimaging to examine nucleus accumbens (NAc)-centered structural and functional alterations associated with fluoxetine and Shugan Jieyu Capsule (SG), a traditional Chinese medicine, in patients with mild-to-moderate depression (MMD). Sixty patients were randomized to an 8-week course of fluoxetine or SG. Depression severity was assessed using the 24-item Hamilton Depression Rating Scale (HAMD-24), and str

Opioid withdrawal: role in addiction and neural mechanisms.

Withdrawal from opioids involves a negative affective state that promotes maintenance of drug-seeking behavior and relapse. As such, understanding the neurobiological mechanisms underlying withdrawal from opioid drugs is critical as scientists and clinicians seek to develop new treatments and therapies. In this review, we focus on the neural systems known to mediate the affective and somatic signs and symptoms of opioid withdrawal, including the mesolimbic dopaminergic system, basolateral amygdala, extended amygdala, and brain and hormonal stress systems. Evidence from preclinical studies sugg
