Identification of a homozygous splice site mutation in the dynein axonemal light chain 4 gene on 22q13.1 in a large consanguineous family from Pakistan with congenital mirror movement disorder.

Mirror movements (MRMV) are involuntary movements on one side of the body that mirror voluntary movements on the opposite side. Congenital mirror movement disorder is a rare, typically autosomal-dominant disorder, although it has been suspected that some sporadic cases may be due to recessive inheritance. Using a linkage analysis and a candidate gene approach, two genes have been implicated in congenital MRMV disorder to date: DCC on 18q21.2 (MRMV1), which encodes a netrin receptor, and RAD51 on 15q15.1 (MRMV2), which is involved in the maintenance of genomic integrity. Here, we describe a lar

Object presence modulates activity within the somatosensory component of the action observation network.

In the present study, we used functional magnetic resonance imaging to investigate the influence that the presence of an object, intended as the target for an action, might have on the neural circuit underlying the observation of the movement of others (action observation network [AON]). This system entails a simulation process of the observed movement occurring via the recruitment of the motor and somatosensory cortices involved in the execution of such movement. Here, participants were requested to observe grasping actions ending behind a partition knowing in advance whether a target-object

The effects of prior exposure to prism lenses on de novo motor skill learning.

Motor learning involves plasticity in a network of brain areas across the cortex and cerebellum. Such traces of learning have the potential to affect subsequent learning of other tasks. In some cases, prior learning can interfere with subsequent learning, but it may be possible to potentiate learning of one task with a prior task if they are sufficiently different. Because prism adaptation involves extensive neuroplasticity, we reasoned that the elevated excitability of neurons could increase their readiness to undergo structural changes, and in turn, create an optimal state for learning a sub

Dynamic belief representation and updating through learned attractor-like dynamics in the frontal cortex.

UNLABELLED: To act adaptively, animals must infer hidden states of the world from incomplete sensory information and update beliefs as new observations accrue. While dopamine signals are well explained by reinforcement learning models that incorporate belief states, how the brain implements belief-state inference remains unknown. Prior modeling work showed that recurrent neural networks trained to predict value (Value-RNN) develop task-specific, attractor-like dynamics that mirror evolution of beliefs. Here we performed high-density electrophysiological recordings from orbitofrontal (OFC) and

Deciphering Peripheral Taste Neuron Diversity: Using Genetic Identity to Bridge Taste Bud Innervation Patterns and Functional Responses.

Peripheral taste neurons exhibit functional, genetic, and morphological diversity, yet understanding how or if these attributes combine into taste neuron types remains unclear. In this study, we used male and female mice to relate taste bud innervation patterns to the function of a subset of proenkephalin-expressing (Penk+) taste neurons. We found that taste arbors (the portion of the axon within the taste bud) stemming from Penk+ neurons displayed diverse branching patterns and lacked stereotypical endings. The range in complexity observed for individual taste arbors from Penk+ neurons mirror

The Expression of Tubb2b Undergoes a Developmental Transition in Murine Cortical Neurons.

The development of the mammalian brain requires the generation, migration, and differentiation of neurons, cellular processes that are dependent on a dynamic microtubule cytoskeleton. Mutations in tubulin genes, which encode for the structural subunits of microtubules, cause detrimental neurological disorders known as the tubulinopathies. The disease spectra associated with different tubulin genes are overlapping but distinct, an observation believed to reflect functional specification of this multigene family. Perturbation of the β-tubulin TUBB2B is known to cause polymicrogyria, pachygyria,

Caffeine protects neuronal cells against injury caused by hyperoxia in the immature brain.

Caffeine administered to preterm infants has been shown to reduce rates of cerebral palsy and cognitive delay, compared to placebo. We investigated the neuroprotective potential of caffeine for the developing brain in a neonatal rat model featuring transient systemic hyperoxia. Using 6-day-old rat pups, we found that after 24 and 48h of 80% oxygen exposure, apoptotic (TUNEL(+)) cell numbers increased in the cortex, hippocampus, and central gray matter, but not in the hippocampus or dentate gyrus. In the dentate gyrus, high oxygen exposure led to a decrease in the number of proliferating (Ki67(

A ketamine/methionine exposure paradigm, alone or combined with Lactobacillus rhamnosus GG, assessing schizophrenia-relevant behavioral, oxidative stress, and molecular endpoints in adult zebrafish.

BACKGROUND: Schizophrenia (SCZ) is a multifactorial neuropsychiatric disorder involving glutamatergic dysfunction, oxidative imbalance, impaired cellular plasticity, and neuron-glia interactions. Adult zebrafish provide a translational vertebrate model for investigating SCZ-relevant behavioral and biological domains. This study evaluated the effects of repeated ketamine (KET) and methionine (MET) exposure, administered alone or combined with Lactobacillus rhamnosus GG (LGG), on behavioral, oxidative, transcriptional, histopathological, and immunofluorescence (IF) endpoints. METHODS: Adult zeb

Development of rostral inferior parietal lobule area functional connectivity from late childhood to early adulthood.

Although the mirror neuron system (MNS) has been extensively studied in monkeys and adult humans, very little is known about its development. Previous studies suggest that the MNS is present by infancy and that the brain and MNS-related cognitive abilities (such as language, empathy, and imitation learning) continue to develop after childhood. In humans, the PFt area of the inferior parietal lobule (IPL) seems to particularly correlate with the functional properties of the PF area in primates, which contains mirror neurons. However, little is known about the functional connectivity (FC) of the

Visual feedback therapy for restoration of upper limb function of stroke patients.

OBJECTIVE: To investigate the effects of mirror neuron theory-based visual feedback therapy (VFT) on restoration of upper limb function of stroke patients and motor-related cortical function using functional magnetic resonance imaging (fMRI). METHODS: Hemiplegic stroke patients were randomly divided into two groups: a VFT group and a control (CTL) group. Sixteen patients in the VFT group received conventional rehabilitation (CR) and VFT for 8 weeks, while 15 patients in the CTL group received only CR. The Barthel Index (BI) was used to assess the activities of daily living at baseline and the

How effective is therapy for childhood stuttering? Dissecting and reinterpreting the evidence in light of spontaneous recovery rates.

BACKGROUND: Similar positive results (e.g. immediate decreases in stuttering frequency and a 60-80% recovery rate from stuttering) have been reported for numerous therapeutic protocols for treating childhood stuttering, many of which have been diametrically opposite in their orientations and implementations. For example, Johnson advocated indirect treatments that simply advocated refraining from drawing any negative attention to childhood disfluencies as persistent and chronic stuttering was thought to progress via negative parental reactions to normal disfluencies. In contrast, direct interve

Action viewing and language in adolescents with autism spectrum disorder.

The mirror neuron system consists of fronto-parietal regions and responds to both goal-directed action execution and observation. The broader action observation network is specifically involved in observation of actions and is thought to play a role in understanding the goals of the motor act, the intention of others, empathy, and language. Many, but not all, studies have found mirror neuron system or action observation network dysfunction in autism spectrum disorder. The objective of this study was to use observation of a goal-directed action fMRI paradigm to examine the action observation ne

Resting state brain subnetwork relates to prosociality and compassion in adolescents.

Adolescence is a crucial time for social development, especially for helping (prosocial) and compassionate behaviors; yet brain networks involved in adolescent prosociality and compassion currently remain underexplored. Here, we sought to evaluate a recently proposed domain-general developmental (Do-GooD) network model of prosocial cognition by relating adolescent functional and structural brain networks with prosocial and compassionate disposition. We acquired resting state fMRI and diffusion MRI from 95 adolescents (ages 14-19  years; 46 males; 49 females) along with self-report questionnair

Effects of tRNS on perception of static and dynamic displays of social-emotional communication.

Accurately decoding emotions from facial and vocal cues is essential to successful social interaction. The human mirror neuron system (hMNS) is thought to support this through sensorimotor simulation of observed emotional expressions. While prior studies linked hMNS activity-indexed by mu rhythm desynchronization (mu-ERD)-to emotional action perception, causal evidence with dynamic, multimodal social stimuli remains limited. We investigated whether transcranial random noise stimulation (tRNS) to the inferior frontal cortex (IFC), a key node of the hMNS, enhances perception of static and dynami

Investigating visuo-tactile mirror properties in borderline personality disorder: A TMS-EEG study.

OBJECTIVES: Patients with borderline personality disorder (pw-BPD) have decreased levels of cognitive empathy, which may be subtended by mirror-like mechanisms in the somatosensory cortices, i.e., the Tactile Mirror System (TaMS). Here, we aimed to shed light on the TaMS and empathic deficits in pw-BPD focusing on connectivity, using transcranial magnetic stimulation and electroencephalography (TMS-EEG). METHODS: After study preregistration, we collected self-report measures of empathic abilities, behavioral performance in a visuo-tactile spatial congruency task investigating TaMS activity, a

Peripheral nerve axons contain machinery for co-translational secretion of axonally-generated proteins.

The axonal compartment of developing neurons and mature peripheral nervous system (PNS) neurons has the capacity to locally synthesize proteins. Axonally-synthesized proteins have been shown to facilitate axonal pathfinding and maintenance in developing central nervous system (CNS) and PNS neurons, and to facilitate the regeneration of mature PNS neurons. RNA-profiling studies of the axons of cultured neurons have shown a surprisingly complex population of mRNAs that encode proteins for a myriad of functions. Although classic-appearing rough endoplasmic reticulum (RER), smooth endoplasmic reti

A human DCC variant causing mirror movement disorder reveals that the WAVE regulatory complex mediates axon guidance by netrin-1-DCC.

The axon guidance cue netrin-1 signals through its receptor DCC (deleted in colorectal cancer) to attract commissural axons to the midline. Variants in DCC are frequently associated with congenital mirror movements (CMMs). A CMM-associated variant in the cytoplasmic tail of DCC is located in a conserved motif predicted to bind to a regulator of actin dynamics called the WAVE (Wiskott-Aldrich syndrome protein-family verprolin homologous protein) regulatory complex (WRC). Here, we explored how this variant affects DCC function and may contribute to CMM. We found that a conserved WRC-interacting

Pathogenic Role of FGFR3 Autoantibodies in Small Fiber Neuropathy.

Sensory neuronopathies (SNN) and small fiber neuropathies (SFN) are debilitating disorders associated with neuropathic pain, yet their underlying mechanisms remain poorly understood. Autoantibodies against fibroblast growth factor receptor 3 define a subset of patients with consistent reports of neuropathic pain harboring a distinct clinical phenotype characterized by small-fiber and non-length-dependent neuropathy, suggesting dorsal root ganglia (DRG) dysfunction. FGFR3-autoantibody-positive sera (FGFR3-AbS) bind to sensory neurons within dorsal root ganglia (DRG). The target of autoantibodie

Microglial CR3 promotes neuron ferroptosis via NOX2-mediated iron deposition in rotenone-induced experimental models of Parkinson's disease.

The activation of complement receptor 3 (CR3) in microglia contributes to neurodegeneration in neurological disorders, including Parkinson's disease (PD). However, it remains unclear for mechanistic knowledge on how CR3 mediates neuronal damage. In this study, the expression of CR3 and its ligands iC3b and ICAM-1 was found to be up-regulated in the midbrain of rotenone PD mice, which was associated with elevation of iron content and disruption of balance of iron metabolism proteins. Interestingly, genetic deletion of CR3 blunted iron accumulation and recovered the expression of iron metabolism
