The genesis of neurons and glia in the developing retina.

The retina is a highly complex neural organ that captures light and converts it into electrochemical signals that are transmitted to the brain to generate vision. There are five basic types of neurons that can be further divided into many specific subtypes, and one type of glial cell, all of which are generated during development from a common progenitor cell. The process by which this diversity of neurons is generated during development has been the subject of active investigation for over 40 years, and this review attempts to summarize the key concepts that have emerged. For example, many of

Neuroglial responses to bacterial, viral, and fungal neuroinfections.

Evidence regarding the host's response to peripheral pathogens in humans abound, whereas studies on the pathogenesis of central nervous system-penetrating infections are relatively scarce. However, given the spate of epidemic and pandemic neuroinfections in the 21st century, the field has experienced a renewed interest lately. This chapter discusses a timely and exciting topic on the roles of glial cells, mainly microglia and astrocytes, in neuroinvasive infections. This chapter considered fungal, viral, and bacterial neuroinfections, X-raying their neuroinvasiveness, neurotropism, and neurovi

Purkinje cell dopaminergic inputs to astrocytes regulate cerebellar-dependent behavior.

Dopamine has a significant role in motor and cognitive function. The dopaminergic pathways originating from the midbrain have received the most attention; however, the relevance of the cerebellar dopaminergic system is largely undiscovered. Here, we show that the major cerebellar astrocyte type Bergmann glial cells express D1 receptors. Dopamine can be synthesized in Purkinje cells by cytochrome P450 and released in an activity-dependent fashion. We demonstrate that activation of D1 receptors induces membrane depolarization and Ca2+ release from the internal store. These astrocytic activities

Inhibition of ERK/MAPK signaling as potential therapy to prevent optic pathway glioma in infants with neurofibromatosis type 1.

Pediatric low-grade gliomas (pLGGs) arise primarily at early stages of development. The molecular mechanisms of pLGG gliomagenesis are unclear, as is the progenitor cell of origin. In this issue of Developmental Cell, Jecrois et al. show that NF1-associated optic pathway gliomas originate from migrating glial progenitors that have distinct MEK/ERK dependency.

The contribution of glial cells to Huntington's disease pathogenesis.

Glial cells play critical roles in the normal development and function of neural circuits, but in many neurodegenerative diseases, they become dysregulated and may contribute to the development of brain pathology. In Huntington's disease (HD), glial cells both lose normal functions and gain neuropathic phenotypes. In addition, cell-autonomous dysfunction elicited by mutant huntingtin (mHTT) expression in specific glial cell types is sufficient to induce both pathology and Huntington's disease-related impairments in motor and cognitive performance, suggesting that these cells may drive the deve

Glial Cell Contributions to Auditory Brainstem Development.

Glial cells, previously thought to have generally supporting roles in the central nervous system, are emerging as essential contributors to multiple aspects of neuronal circuit function and development. This review focuses on the contributions of glial cells to the development of auditory pathways in the brainstem. These pathways display specialized synapses and an unusually high degree of precision in circuitry that enables sound source localization. The development of these pathways thus requires highly coordinated molecular and cellular mechanisms. Several classes of glial cells, including

Radial glia defines boundaries and subdivisions in the embryonic midbrain of the lizard Gallotia galloti.

We have studied the organization of the midbrain radial glia in embryos of Gallotia galloti using the fluorescent lipophilic dye 1,1'-dioctadecyl-3,3,3',3'-tetramethyl-indocarbocyanine perchlorate (DiI) and the antibodies H5 and RC2. Our goal was to verify if the radial glia takes part in the midbrain boundaries formation and if it defines different zones. Our exam reveals two clear limits, anterior or mesencephalic-diencephalic (m/d) and posterior or mesencephalic-rhombencephalic (m/r), that can be defined as the borders where the midbrain radial glia processes end. Moreover, fasciculate radi

Trophic effects of purines in neurons and glial cells.

In addition to their well known roles within cells, purine nucleotides such as adenosine 5' triphosphate (ATP) and guanosine 5' triphosphate (GTP), nucleosides such as adenosine and guanosine and bases, such as adenine and guanine and their metabolic products xanthine and hypoxanthine are released into the extracellular space where they act as intercellular signaling molecules. In the nervous system they mediate both immediate effects, such as neurotransmission, and trophic effects which induce changes in cell metabolism, structure and function and therefore have a longer time course. Some tro

Low, but not high, dose triptolide controls neuroinflammation and improves behavioral deficits in toxic model of multiple sclerosis by dampening of NF-κB activation and acceleration of intrinsic myelin repair.

Cuprizone (Cup) is a copper chelating agent frequently used to study factors that affect oligodendrocytes (OLGs) death and acute demyelination. Triptolide (TP), a nuclear factor-kappaB (NF-κB) blocker, is a major bioactive component of Tripterygium wilfordii Hook f. (TWHf) with various therapeutic activities. In this study, we examined the effects of TP on neuroglia activation, inflammation, apoptosis, demyelination, and behavioral deficits in the Cup-induced toxic model of multiple sclerosis (MS). C57BL/6 J mice were fed with chow containing 0.2% Cup for 6 weeks to induce detectable neuroinfl…

Glial cells in dementia: From cellular dysfunction to therapeutic frontiers.

Neurodegenerative dementias, including Alzheimer's disease and vascular dementia, have long been viewed through a neuron-centric lens. However, growing evidence highlights the indispensable and multifaceted roles of glial cells, astrocytes, microglia, and oligodendrocytes in both the onset and progression of these disorders. While prior reviews have cataloged glial dysfunction in isolation, this review offers a novel, integrative framework that maps the interconnected roles of glial subtypes across molecular, cellular, and circuit-level pathology in dementia. We critically synthesize recent ad

Glial Contributions to Neuropathic Pain: Central and Peripheral Strategies in Postherpetic Neuralgia Pathogenesis and Therapeutics.

Neuropathic pain (NP), particularly postherpetic neuralgia (PHN), involves intricate mechanisms that extend far beyond neuronal dysfunction. In recent years, glial cell-mediated neuro-immune-inflammatory circuits mediated by glial cells (including spinal microglia, astrocytes, and dorsal root ganglion satellite glia) have been regarded as one of the key pathological mechanisms for the occurrence and maintenance of NP. Glial cells exacerbate pain pathogenesis by driving neuroinflammation, amplifying nociceptive signaling, and modulating ion channels and pain-related signaling molecules. A syste

Friend or foe? Glial-vascular interactions in health and neurodegenerative disease.

Dysfunction of glial and vascular cells is increasingly recognized as a central feature of neurodegenerative diseases. Growing evidence points to disruptions in glial-vascular interactions, which are critical for maintaining the functions of the neurogliovascular unit throughout the lifespan, as key contributors to disease initiation and progression. However, the mechanisms governing this complex intercellular crosstalk and its potential role in disease pathogenesis remain incompletely understood. In this review, we summarize the current understanding of glial-vascular communication across hea

Neuroglial Advances: New Roles for Established Players.

Neuroglial cells perform numerous physiological functions and contribute to the pathogenesis of all diseases of the nervous system. Neuroglial neuroprotection defines the resilience of the nervous tissue to exo- and endogenous pathological challenges, while neuroglial defence determines the progression and outcome of neurological disorders. IN this paper, we overview previously unknown but recently discovered roles of various types of neuroglial cells in diverse physiological and pathological processes. First, we describe the role of ependymal glia in the regulation of cerebrospinal fluid flow…

Glial-modulating agents for the treatment of pain: a systematic review.

Preclinical research supports a critical role for nervous system glia in pain pathophysiology. This systematic review of human trials of potential glia-modulating drugs for the prevention or treatment of pain followed a predefined search strategy and protocol registration. We searched for English language, randomized, double-blind trials comparing putative glia-modulating drugs to placebo or other comparators. The primary outcomes included validated participant-reported measures of pain intensity or relief and, in studies of opioid administration, measures of opioid consumption and/or opioid-r

[The role of enteric glial cells in intestinal neuroimmunomodulation].

As a type of special glial cells, enteric glial cells (EGCs) have long been considered capable of only supporting and nourishing enteric neurons. With the deepening of brain-gut research, a large number of latest research results have altered this understanding, and revealed a series of important functions of EGCs in the development and plasticity of enteric nervous system, as well as enteric neuroimmunomodulation. EGCs can respond to immune regulation signals such as cytokines, bacteria and neurotransmitters in the extracellular environment. By autocrine or paracrine regulation, glial cell li

Autophagy in Myelinating Glia.

Autophagy is the cellular process involved in transportation and degradation of membrane, proteins, pathogens, and organelles. This fundamental cellular process is vital in development, plasticity, and response to disease and injury. Compared with neurons, little information is available on autophagy in glia, but it is paramount for glia to perform their critical responses to nervous system disease and injury, including active tissue remodeling and phagocytosis. In myelinating glia, autophagy has expanded roles, particularly in phagocytosis of mature myelin and in generating the vast amounts o
