Arginine methylation-enabled FUS phase separation with SMN contributes to neuronal granule formation.

Various ribonucleoprotein complexes (RNPs) often function in the form of membraneless organelles derived from multivalence-driven liquid-liquid phase separation (LLPS). Post-translational modifications, such as phosphorylation and arginine methylation, govern the assembly and disassembly of membraneless organelles. This study reveals that asymmetric dimethylation of arginine can create extra binding sites for multivalent Tudor domain-containing proteins like survival of motor neuron (SMN) protein, thereby lowering the threshold for LLPS of RNPs, such as fused in sarcoma (FUS). Accordingly, FUS…

Phase separation of DDX21 promotes colorectal cancer metastasis via MCM5-dependent EMT pathway.

RNA binding proteins (RBPs) contributes to cancer progression, but the underlying mechanism reminds unclear. Here, we find that DDX21, a rep … C metastasis. Phase-separated DDX21 highly binds on MCM5 gene locus, which is markedly reduced when phase separation is disrupted by mutations on its intrinsically disordered region (IDR). The impaired metastatic ability of CRC upon DDX21 loss is restored by ectopic expression of MCM5, indicating MCM5 is a key downstream target of DDX21 for CRC metastasis. Further …

TOR signaling regulates liquid phase separation of the SMN complex governing snRNP biogenesis.

The activity of the SMN complex in promoting the assembly of pre-mRNA processing UsnRNPs correlates with condensation of the complex in nucl … ion of human SMN controls the capacity of the complex to condense in Cajal bodies via liquid-liquid phase separation. Our findings link SMN complex condensation and UsnRNP biogenesis to cellular energy levels and suggest modulation of TOR signaling as a rational concept for therapy of the SMN-linked neuromuscular disorder spinal muscular atrophy.

Membraneless condensates by Rapsn phase separation as a platform for neuromuscular junction formation.

Our daily life depends on muscle contraction, a process that is controlled by the neuromuscular junction (NMJ). However, the mechanisms of NMJ assembly remain unclear. Here we show that Rapsn, a protein critical for NMJ formation, undergoes liquid-liquid phase separation (LLPS) and condensates into liquid-like assemblies. Such assemblies can recruit acetylcholine receptors (AChRs), cytoskeletal proteins, and signaling proteins for postsynaptic differentiation. Rapsn LLPS requires multivalent binding of tetratricopeptide repeats (TPRs) and is increased by Musk signaling. The capacity of Rapsn t…

Separation of the two enantiomers of naproxcinod by chiral normal-phase liquid chromatography.

A normal-phase enantioselective high-performance liquid chromatographic method was developed for the enantiomeric resolution of naproxcinod, the most advanced cyclooxygenase-inhibiting nitric oxide donator of anti-inflammatory drugs designed for treatment of osteoarthritis. The enantiomers of naproxcinod were resolved on a Chiralpak AD-H (250 × 4.6 mm, 5 μm) column using a mobile phase system containing n-hexane and 2-propanol (95:5, v/v). The resolution between the enantiomers was found to be more than 2.0. The limit of detection and limit of quantitation of (R)-enantiomer were found to be 5

Chiral separation of Frovatriptan isomers by HPLC using amylose based chiral stationary phase.

A stereospecific HPLC method for separation of Frovatriptan enantiomers in bulk drug and pharmaceutical formulations was developed and validated on a normal-phase amylose derivertized chiral column. The effects of the organic modifiers namely 2-propanol, ethanol and diethyl amine (DEA) in the mobile phase were optimized to obtain the best enantiomeric separation. Calibration curves were linear over the range of 200-6150 ng/mL, with a regression coefficient (R(2)) of 0.9998. The limit of detection (LOD) and limit of quantification (LOQ) were 65 ng/mL and 200 ng/mL, respectively. The method was

Enantiomeric separation of drugs and herbicides on a beta-cyclodextrin-bonded stationary phase.

A chemically bonded beta-cyclodextrin chiral stationary phase for HPLC was prepared in a "one pot" process by the reaction of a phenylated beta-cyclodextrin with silica gel. Various racemic analytes such as drugs (aminoalcohol adrenergic beta-blockers, benzodiazepine anxiolytics, arylpropionic acid antiinflammatory agents) and herbicides (aryloxypropionic acids and esters) were separated on the prepared material. The column showed good chiral recognition ability for most of the solutes tested when using heptane and either 2-propanol or chloroform as organic mobile phase modifiers.

Separation of tocopherol and tocotrienol isomers using normal- and reverse-phase liquid chromatography.

This optimization study for tocopherols and tocotrienols involved both normal- and reverse-phase liquid chromatography using various columns and mobile phases. Normal-phase systems showed elution of the homologs in order of increasing polarity with separation based on methyl substituents on the chromanol moiety. Reverse-phase systems showed class separation based on the saturation of the phytyl side chain; the more saturated tocopherols were retained on the column longer. When the Zorbax ODS was used with an isocratic ternary acetonitrile:methanol:methylene chloride (60:35:5) mixture, the opti

Liquid-liquid phase separation in neurodegenerative diseases: Mechanisms, pathological transitions, and therapeutic opportunities.

Liquid-liquid phase separation (LLPS) provides a biophysical framework for understanding biomolecular organization, yet its pathological relevance in neurodegeneration remains incompletely resolved. In this narrative review, we critically synthesize current evidence on LLPS in Huntington's disease, Parkinson's disease, Alzheimer's disease, and amyotrophic lateral sclerosis, focusing on mutant huntingtin, α-synuclein, amyloid-β, tau, TDP-43, and FUS. We first outline the molecular principles-including intrinsic disorder, multivalent interactions, and environmental sensitivity-that govern their …

Structural Phase Separation Couples to Charge-Density-Wave Formation in Kagome Metal FeGe.

The intertwining of charge, spin, and lattice degrees of freedom underlies the emergent properties of correlated materials. A recent prominent example is the kagome metal FeGe, which hosts a charge density wave (CDW) within a preexisting antiferromagnetic state. Although partial Ge-Ge dimerization is known to accompany CDW formation, the thermodynamic pathway through which long-range CDW order emerges remains unclear. Using temperature-dependent high-resolution x-ray diffraction measurements, we observed a robust splitting of the lattice reflection into two coexisting peaks with distinct latti

Novel chitosan-based phase-selective organogelator for efficient separation of oil/water mixtures.

Phase-selective organogelators (PSOGs) can selectively gel the oil phase in oil/water biphasic systems, demonstrating significant research value in organic solvent leakage and oil spill emergency response. In this study, N-cinnamoyl-6-dodecanoyl chitosan (L1.75C1CS) oil gelling agent was successfully prepared using natural biopolymer chitosan as the backbone through a two-step grafting modification process. The gelling mechanism of L1.75C1CS to form gels in solvents was investigated via UV-vis, FT-IR, and XRD. The experimental results showed that the formation of gel mainly depended on the syn

Phase-separation mode in HPLC: Theoretical prediction and experimental evaluation of elution times.

A novel high-performance liquid chromatography (HPLC) approach, termed the phase-separation mode, was developed by employing a phase-separation multiphase flow (PSMF) as the mobile phase. Unlike conventional modes, this system exploits two coexisting liquid phases that generate distinct liquid-liquid interfaces in packed columns. In this study, we established a theoretical framework for predicting analyte elution times in the phase-separation mode and validated them experimentally. A theoretical equation was derived on the basis of the proposed separation mechanism, incorporating capacity fact

Hierarchical Epoxy Structures via Tunable Polymerization-Induced Phase Separation Combined with Additive Manufacturing.

Polymerization-induced phase separation (PIPS) allows for the control of thermoset morphologies and properties, enabling the tuning of domain sizes and thermomechanical response. However, its use in generating substructural features in additively manufactured materials has been limited. In this work, we combine epoxy PIPS with UV curable acrylate and rheological modifiers to print nano- to macro-phase separating materials via a two-step, dual-cure approach. This method enables direct ink write printing of hierarchical structures with both controlled morphologies through phase separation and ma…

Scalable Fabrication of MOF Glass Membranes for Gas Separation via Stepwise Phase Transition.

MOF glasses have emerged as promising membrane materials, yet the scalable fabrication of macroscopically continuous and crack-free MOF glass membranes remains a critical bottleneck that hinders their industrial translation. Here, we report a stepwise phase transition strategy that enables the formation of large-area MOF glass membranes. By constructing a crystalline-amorphous composite precursor, this strategy mitigates electrostatic repulsion and leverage graded viscoelastic flow to promote early-stage interparticle bonding. This approach effectively suppresses cracking and pinhole defect fo

Structural organization of HBV pgRNA genome driven by phase separation in capsid confinement.

Viruses rely on the precise packaging of their genomes within a capsid to execute essential life-cycle events, yet the principles governing genome structural organization in this confined environment remain elusive. Here, we reveal that hepatitis B virus (HBV) pregenomic RNA (pgRNA) exploits liquid-liquid phase separation (LLPS) inside the capsid to sculpt its architecture. Multiscale molecular dynamics (MD) simulations, supplemented by biochemical assays, show that pgRNA coalesces into a hollow, shell-like condensate along the inner capsid surface, with coexisting low- and high-density region…

Nucleolar migration regulates meiotic sex chromosome inactivation via phase separation during mammalian spermatogenesis.

During spermatogenesis, the unsynapsed XY chromosomes undergo meiotic sex chromosome inactivation (MSCI) and form a heterochromatic XY body. … ly, SENP3-mediated deSUMOylation of NPM1 promotes its interaction with rRNA, enabling liquid-liquid phase separation, via which they exclude Pol II from the XY body. Together, these data reveal a critical role of nucleolar components in the transcriptional regulation of MSCI in mammalian spermatogenesis.

A Brownian dynamics study of liquid-liquid phase separation in multiscale chromatin networks.

In living cells, proteins involved in specialized biochemical functions are often spatially organized within biomolecular condensates. Increasing evidence suggests that some of these condensates, including DNA repair condensates, emerge through liquid-liquid phase separation (LLPS). In the nucleus, however, condensates form within a highly heterogeneous environment composed of chromatin fibers, RNA, and additional protein scaffolds such as PAR chains, all of which may interact with phase-separating proteins. Moreover, condensate formation is frequently associated with specific chromatin confor…

Chirality-induced phase separation and collective dynamics in binary mixtures of chiral particles.

Transverse interactions between particles can disrupt thermodynamic equilibrium, giving rise to distinctive nonequilibrium dynamics. However, the collective behavior of mixed particles with varying chiralities remains largely unexplored. In this paper, we use numerical simulations to investigate the separation and collective dynamics of binary mixtures with differing chiralities. Our results show that, even without attractive forces, chirality-induced nonhomogeneous bubble phases drive the clustering of similar particles, facilitating phase separation. Binary mixtures with opposing chirality s…

Molecular mechanisms mediated by liquid-liquid phase separation in chronic liver disease progression.

Liquid-liquid phase separation (LLPS) is a cellular process driven by multivalent interactions, forming dynamic biomolecular condensates containing proteins, RNAs, and other molecules. LLPS plays a pivotal role in processes such as signal transduction, gene expression, autophagy, and cellular stress responses. The dysregulation of LLPS is linked to chronic liver diseases (CLDs), particularly non-alcoholic fatty liver disease (NAFLD), liver fibrosis, and hepatocellular carcinoma (HCC). LLPS profoundly mediates the pathological evolution of these diseases by regulating key mechanisms, including …
