Functional characterization of chemosensory proteins in Pharsalia antennata with special emphasis on the roles of CSP2 orthologs in pheromone detection.

Behaviorally active compounds of longhorned beetles, including sex/aggregation pheromones and plant odorants, have been extensively applied to the integrated pest management. It is largely unknown, however, how beetle's chemosensory proteins (CSPs) mediate the detection of semiochemicals. Here, we characterized six antenna-enriched PantCSPs in Pharsalia antennata, revealing the strong binding to two floral scents syringaldehyde and β-ionone (dissociation constant, Ki < 5.5 μM). In particular, PantCSP11 presented the binding preference to 11-12C aliphatic alcohols/aldehydes (Ki = 0.96 ± 0.02 μM…

Queen mandibular pheromone prevents, but does not reverse, worker reproduction in Apis mellifera, unlike in Drosophila melanogaster.

The reproductive division of labour is the defining feature of eusociality, in which reproduction is monopolised by one, or a small subset, of individuals within a colony. In the Western Honeybee Apis mellifera, workers are reproductively constrained by a suite of mechanisms, but primarily by queen mandibular pheromone (QMP). The adult developmental limits of QMP's ability to repress worker reproduction and its ability to suppress reproduction once ovaries have begun to activate remain unresolved. Here, we investigated the limits of plasticity of QMP mediated reproductive repression and compar

Maternal Pheromone Emission During Biparental Care: Evidence for Consistent Individual Differences and Links to Terminal Investment.

Pheromone production, once considered a fixed species-specific trait, is now recognized to vary plastically within individuals and consistently among them. Such variation may reflect differences in individual quality as well as adaptive allocation strategies, for example when residual reproductive value declines and individuals exhibit terminal investment in current reproduction. While most work has focused on sex pheromones, much less is known about variation in pheromones that mediate social behavior. In the burying beetle Nicrophorus vespilloides, caring females emit the pheromone methyl ge…

Pheromone circuits and transcriptional cascades modulating transcriptional and chromatin states in the Drosophila central brain with social experience.

Social experience significantly influences the behavioral and physiological responses of animals, including humans. In many animals, social isolation increases aggression, courtship, locomotion, and feeding while disrupting sleep. This occurs when peripheral neurons detect social signals, such as pheromones, which activate brain circuits involved in decision-making. However, the molecular and circuit mechanisms of how chronic social isolation or enrichment alter gene expression and affect neuronal function and behavior remain unclear. In this study, we examined how transcription patterns and c…

Phylogenetic-Pheromone Associations Obscured by Stabilising Selection and Natal Tree Effect in a Tree-Killing Bark Beetle.

Insects are highly reliant on chemical cues such as pheromones to facilitate communication and navigation. Some of the roles of pheromones include attracting and finding mates and conspecifics, and in these cases, we expect stabilising selection to dampen within-population pheromone variation. On the other hand, standing pheromone variation may lead to barriers to gene flow and saltational shifts that facilitate divergence and speciation. We investigated the relationships between pheromone variation and genetic variation in the spruce beetle, Dendroctonus rufipennis Kirby, a bark beetle that i…

Sublethal indoxacarb exposure alters pheromone production and ovarian development in the yellow peach moth, Conogethes punctiferalis.

The Conogethes punctiferalis, a major pest of peach and other fruit trees, causes significant damage by boring into fruits during its larval stage. Indoxacarb, a sodium channel blocker insecticide, has been widely applied in agriculture and horticulture for pest control, particularly against larval pests. However, its effects on adult insects remain largely unexplored. Present study employed C. punctiferalis as model to investigate the toxic effects of indoxacarb on adult moths. The results showed that sublethal exposure to indoxacarb significantly reduced the release of sex pheromones, reduce…

A C. elegans male pheromone feminizes germline gene expression in hermaphrodites and imposes life-history costs.

Sex pheromones improve reproductive success, but also impose costs. Here we show that even brief exposure to physiological amounts of the dominant C. elegans male pheromone, ascr#10, alters the expression of thousands of genes in hermaphrodites. The most dramatic effect on the transcriptome was the upregulation of genes expressed during oogenesis and downregulation of genes associated with male gametogenesis. Among the detrimental effects of ascr#10 on hermaphrodites is the increased risk of persistent infections caused by pathological pharyngeal hypertrophy. Our results reveal a way in which …

A unique variant of lymphocytic choriomeningitis virus that induces pheromone binding protein MUP: Critical role for CTL.

Lymphocytic choriomeningitis virus (LCMV) WE variant 2.2 (v2.2) generated a high level of the major mouse urinary protein: MUP. Mice infected with LCMV WE v54, which differed from v2.2 by a single amino acid in the viral glycoprotein, failed to generate MUP above baseline levels found in uninfected controls. Variant 54 bound at 2.5 logs higher affinity to the LCMV receptor α-dystroglycan (α-DG) than v2.2 and entered α-DG-expressing but not α-DG-null cells. Variant 2.2 infected both α-DG-null or -expressing cells. Variant 54 infected more dendritic cells, generated a negligible CD8 T cell respo

Synthesis, Functional Assays, Electrophysiological Activity, and Field Tests of Pheromone Antagonists of the Tomato Leafminer, Tuta absoluta.

The tomato leafminer, Tuta absoluta, is one of the major pests of tomato and other Solanaceae in many regions worldwide. In the search for new strategies to control this pest, we present herewith the stereoselective synthesis, electrophysiological activity, functional analysis, and field tests of new chemicals as possible antagonists of the sex pheromone of the leafminer. The chemicals are methyl ketone (MK) and trifluoromethyl ketone (TFMK) structural analogues of both components of the pheromone. Most of the chemicals exerted per se some electrophysiological activity and inhibited the electr

Using a proteometabolomic approach to investigate the role of Dufour's gland in pheromone biosynthesis in the social wasp Polybia paulista.

UNLABELLED: Dufour's gland is associated with the venom apparatuses of social wasps and bees. This location and its evolutionary adaptations indicate that it could be involved in the production of alarm pheromones in the social wasp Polybia paulista. To investigate this hypothesis, the volatile composition of this gland was analyzed and compared to that in the venom. Eighteen compounds were identified as secreted by Dufour's gland, and 16 of these compounds were also identified in the venom, suggesting that the compounds produced by the gland are secreted and mixed with venom in the venom rese…

A wide range of pheromone-stimulated sexual and reproductive behaviors in female mice depend on G protein Gαo.

BACKGROUND: Optimal reproductive fitness is essential for the biological success and survival of species. The vomeronasal organ is strongly … nd estrous cycle. These mutant mice are insensitive to reproductive facilitation stimulated by male pheromones that accelerate puberty and induce ovulation. Gαo-mutant females exhibit a striking reduction in sexual receptivity or lordosis behavior to males, but gender discrimination seems to be intact. These mice also show a loss in male scent preference, wh …

Accumulation of phenylpropanoid and sesquiterpenoid volatiles in male rectal pheromonal glands of the guava fruit fly, Bactrocera correcta.

The guava fruit fly, Bactrocera correcta, is widely distributed in Thailand and other surrounding Southeast Asian countries, and, like the closely related sympatric species, the oriental fruit fly, B. dorsalis, infests various fruits, including guava, peach, and mango. Males of both B. correcta and B. dorsalis are strongly attracted to, and compulsively feed on, methyl eugenol (ME). Bactrocera dorsalis males fed on ME sequester its metabolite phenylpropanoids, (E)-coniferyl alcohol and 2-allyl-4,5-dimethoxyphenol, in the rectal pheromone gland. In contrast, B. correcta males fed on ME sequeste

The male mouse pheromone ESP1 enhances female sexual receptive behaviour through a specific vomeronasal receptor.

Various social behaviours in mice are regulated by chemical signals called pheromones that act through the vomeronasal system. Exocrine gland-secreting peptide 1 (ESP1) is a 7-kDa peptide that is released into male tear fluids and stimulates vomeronasal sensory neurons in female mice. Here, we describe the molecular and neural mechanisms that are involved in the decoding of ESP1 signals in the vomeronasal system, which leads to behavioural output in female mice. ESP1 is recognized by a specific vomeronasal receptor, V2Rp5, and the ligand-receptor interaction results in sex-specific signal tran…

Locating the barnacle settlement pheromone: spatial and ontogenetic expression of the settlement-inducing protein complex of Balanus amphitrite.

Barnacles are prominent members of hard substratum benthic communities and their study has been important to advances in experimental ecology and contemporary ecological theory. Having recently characterized the cue to gregarious settlement of Balanus amphitrite, the settlement-inducing protein complex (SIPC), we use two polyclonal antibodies to examine the tissue distribution and ontogenetic expression of this glycoprotein. These antibodies were raised against two separate peptides located near the N- and C-termini of the SIPC and were used to detect the glycoprotein by western blotting and i

Attraction of male European tarnished plant bug, Lygus rugulipennis to components of the female sex pheromone in the field.

Previous work showed that females of the European tarnished plant bug, Lygus rugulipennis Poppius (Heteroptera: Miridae), produced three chemicals, hexyl butyrate, (E)-2-hexenyl butyrate, and (E)-4-oxo-2-hexenal, and that these were suspected to be components of the female sex pheromone. In field experiments, traps baited with blends of these chemicals dispensed from polyethylene vials and sachets failed to catch significant numbers of males. Here, we report more recent field experiments in which the chemicals were released from glass microcapillary tubes. A blend of hexyl butyrate and (E)-4-o

Cyclic AMP mediation of the hormonal stimulation of 14C-acetate incorporation by Heliothis armigera pheromone glands in vitro.

Evidence is presented for hormone-controlled adenosine 3',5'-cyclic monophosphate (cAMP)-mediated incorporation of 14C-acetate by in vitro pheromone gland preparations of Heliothis armigera. Basal intracellular cAMP concentrations of in vitro pheromone glands increase 28-fold in the presence of methanolic brain-complex extracts after 5 min of incubation. This intracellular cAMP increase shows a dose-dependent response. Studies on isolated pheromone gland preparations show that 8-bromo-cAMP added to the incubation media stimulates acetate incorporation thereby mimicking the hormonal response. T

Field tests of the response of female Amblyomma variegatum (Acari:Ixodidae) to the synthetic aggregation-attachment pheromone and its components.

Fifty percent of an Amblyomma variegatum female population were able to find upwind-positioned targets containing the synthetic aggregation-attachment pheromone of this species or the pheromone component o-nitrophenol alone. The ticks were not attracted to the other components of the pheromone: methyl-salicylate and pelargonic acid. The mean time required for the ticks to reach a target at 1 m from the starting point was 3 min and 20 s.

Identification of regions of the Streptococcus faecalis plasmid pCF-10 that encode antibiotic resistance and pheromone response functions.

The conjugative plasmid pCF-10 (58 kb) of Streptococcus faecalis has been mapped with restriction enzymes. By restriction mapping and Southern hybridization analysis, a 16-kb segment of the plasmid was shown to resemble closely the conjugative tetracycline resistance transposon, Tn916. Mutagenesis of the plasmid with the erythromycin resistance transposon Tn917 was used to localize a tetracycline resistance determinant and several regions involved in conjugal transfer. Fifty Tn917 insertions (outside the region of the plasmid homologous to Tn916) affecting mating behavior and the ability of do
