Plant Virus Intratumoral Immunotherapy with CPMV and PVX Elicits Durable Antitumor Immunity in a Mouse Model of Diffuse Large B-Cell Lymphoma.

Plant viruses are naturally occurring nanoparticles and adjuvants that interact with the mammalian immune system. This property can be harnessed in vaccines and immunotherapy. We have previously demonstrated that intratumoral immunotherapy with cowpea mosaic virus (CPMV) stimulates systemic and durable antitumor immunity in mouse tumor models and canine cancer patients. Here we compared the antitumor efficacy of CPMV with potato virus X (PVX) using a mouse model B-cell lymphoma (A20 and BALB/c mice). Despite their diverse morphologies and physiochemical properties, both plant viruses elicited

Novel cinnamic acid derivatives enable the induction of chloroplast-associated immunity and interaction with viral coat proteins for plant viral disease control.

BACKGROUND: Viral diseases remain persistent global challenges to public health systems and agricultural productivity, while current antiviral strategies are constrained by their narrow therapeutic spectrum and overreliance on unimodal mechanisms. These critical limitations highlight the imperative to develop new antiviral agents with innovative chemical scaffolds and multimodal mechanisms of action. RESULTS: In this study, 31 novel cinnamic acid derivatives were synthesized and their inhibitory activities against tobacco mosaic virus (TMV) in tobacco hosts were evaluated. Compound B7, which

Induction of toxin-specific neutralizing immunity by molecularly uniform rice-based oral cholera toxin B subunit vaccine without plant-associated sugar modification.

Plants have been used as expression systems for a number of vaccines. However, the expression of vaccines in plants sometimes results in unexpected modification of the vaccines by N-terminal blocking and sugar-chain attachment. Although MucoRice-CTB was thought to be the first cold-chain-free and unpurified oral vaccine, the molecular heterogeneity of MucoRice-CTB, together with plant-based sugar modifications of the CTB protein, has made it difficult to assess immunological activity of vaccine and yield from rice seed. Using a T-DNA vector driven by a prolamin promoter and a signal peptide ad

Calcium-dependent protein kinase signalling in plant immune responses: current status and future prospects.

This review focuses on the critical role of CDPKs in advancing plant immunity research, highlighting how their functions can uncover key defence mechanisms and bridge critical knowledge gaps in the field. Calcium serves as the cornerstone of immune signalling, propagating pathogen-induced signals through specific sensors and decoders. Among these, calcium-dependent protein kinases (CDPKs) stand out as plant-specific signal transducers, directly converting calcium fluxes into defence activation. By modulating key immune components, CDPKs act as key hubs in plant immunity. Deciphering their role…

PELOTA at the Crossroads of Ribosome-associated quality control and Silencing: Coordinating Development, Stress, and Immunity in Plants.

Ribosome-associated quality control (RaQC) pathways, including no-go decay (NGD) and non-stop decay (NSD), are essential for maintaining translational fidelity and regulating gene expression in eukaryotes. Central to these pathways is the conserved ribosome rescue factor PELOTA, which resolves stalled ribosomes and promotes the clearance of aberrant mRNAs and nascent polypeptides. While NGD and NSD have been extensively characterized in yeast and animals, our understanding of these processes in plants remains limited. Nevertheless, emerging evidence indicates that PELOTA plays a pivotal role i

Reframing Pathogenesis-Related proteins: from plant immune drivers to stress resilience integration nodes.

Plants are constantly exposed to biotic and abiotic stresses, but the molecular mechanisms integrating these signals remain underexplored. Traditionally viewed as defense proteins against pathogen infection, pathogenesis-related (PR) proteins comprise 19 diverse families encompassing defense-related enzymes (glucanases, chitinases, nucleases, proteases), antimicrobial peptides, enzyme inhibitors and reactive oxygen species (ROS)-generating proteins. While their significance during biotic stresses is well-documented, research has started to emerge on their dynamic regulation and functional role

Immunity priming and biostimulation by airborne nonanal increase yield of field-grown common bean plants.

INTRODUCTION: Stress-induced volatile organic compounds (VOCs) that induce plant immunity bear potential for biocontrol. Here, we explore the potential of nonanal to enhance the seed yield of common bean (Phaseolus vulgaris) under open field conditions that are realistic for smallholder farmers. METHODS AND RESULTS: Using plastic cups with a nonanal-containing lanolin paste as low-cost dispensers, we observed that exposure of Flor de Junio Marcela (FJM) plants over 48h to airborne nonanal was followed by a 3-fold higher expression of pathogenesis-related (PR) genes PR1 and PR4. Both genes fur…

Natural Plant Products Mediated Prevention of Cancer Facilitated through Immune Suppression of Treg Cells.

Cancer is one of the leading causes of death, and numerous methods have been tested and used to figure out an optimum way of treatment. Besides targeted therapy, immunotherapy has proven to be effective by controlling certain immune cells. Traditional cancer therapy is met with the consequences of adverse side effects that have been a major issue for treatment; hence, a leap towards naturally occurring immunomodulators was taken to develop safer methods of treatment. One of the major immune cells responsible for the growth of tumors is regulatory T cells (Tregs). To maintain immunological home

The Bacillus cereus Strain EC9 Primes the Plant Immune System for Superior Biocontrol of Fusarium oxysporum.

Antibiosis is a key feature widely exploited to develop biofungicides based on the ability of biological control agents (BCAs) to produce fungitoxic compounds. A less recognised attribute of plant-associated beneficial microorganisms is their ability to stimulate the plant immune system, which may provide long-term, systemic self-protection against different types of pathogens. By using conventional antifungal in vitro screening coupled with in planta assays, we found antifungal and non-antifungal Bacillus strains that protected the ornamental plant Kalanchoe against the soil-borne pathogen Fu

Bacterial medium-chain 3-hydroxy fatty acid metabolites trigger immunity in Arabidopsis plants.

In plants, cell-surface immune receptors sense molecular non-self-signatures. Lipid A of Gram-negative bacterial lipopolysaccharide is considered such a non-self-signature. The receptor kinase LIPOOLIGOSACCHARIDE-SPECIFIC REDUCED ELICITATION (LORE) mediates plant immune responses to Pseudomonas and Xanthomonas but not enterobacterial lipid A or lipopolysaccharide preparations. Here, we demonstrate that synthetic and bacterial lipopolysaccharide-copurified medium-chain 3-hydroxy fatty acid (mc-3-OH-FA) metabolites elicit LORE-dependent immunity. The mc-3-OH-FAs are sensed in a chain length- and

A plant-produced antigen elicits potent immune responses against West Nile virus in mice.

We described the rapid production of the domain III (DIII) of the envelope (E) protein in plants as a vaccine candidate for West Nile Virus (WNV). Using various combinations of vector modules of a deconstructed viral vector expression system, DIII was produced in three subcellular compartments in leaves of Nicotiana benthamiana by transient expression. DIII expressed at much higher levels when targeted to the endoplasmic reticulum (ER) than that targeted to the chloroplast or the cytosol, with accumulation level up to 73  μ g DIII per gram of leaf fresh weight within 4 days after infiltration.

Genetic requirements for signaling from an autoactive plant NB-LRR intracellular innate immune receptor.

Plants react to pathogen attack via recognition of, and response to, pathogen-specific molecules at the cell surface and inside the cell. Pathogen effectors (virulence factors) are monitored by intracellular nucleotide-binding leucine-rich repeat (NB-LRR) sensor proteins in plants and mammals. Here, we study the genetic requirements for defense responses of an autoactive mutant of ADR1-L2, an Arabidopsis coiled-coil (CC)-NB-LRR protein. ADR1-L2 functions upstream of salicylic acid (SA) accumulation in several defense contexts, and it can act in this context as a "helper" to transduce specific

Plant phytochemicals as new potential drugs for immune disorders and cancer therapy: really a promising path?

Plant phytochemicals represent an exciting opportunity to maintain best health conditions through a balanced and properly administered daily nutrition or dietary supplement and have often been considered a good option for obtaining a few promising, expensive new drugs from plants. Several polyphenolic compounds, such as resveratrol, tea catechins and flavonoids, which are commonly found in vegetables, fruits and plant-derived juices or beverages, exert well-evidenced cardioprotective, neuroprotective, chemopreventive and anti-inflammatory properties, but, nevertheless, further clinical and epi

Trichoderma-induced plant immunity likely involves both hormonal- and camalexin-dependent mechanisms in Arabidopsis thaliana and confers resistance against necrotrophic fungi Botrytis cinerea.

BACKGROUND: High-intensity sprint exercises (HIS) are central to sprinter training and require careful monitoring of athlete muscle fatigue to improve performance and prevent injury. While the countermovement jump (CMJ) may be used to monitor neuromuscular fatigue (NMF), little is known about the specific effects from HIS. The purpose of this study is to investigate the effects of HIS on the CMJ to assess its utility for assessing NMF following HIS. METHODS: Ten male collegiate 400 m sprinters completed a 400 m sprint fatigue protocol and underwent five CMJ-testing sessions (baseline, 3 minut

Plant-Based Oral Vaccines: Molecular Biotechnology Approaches Toward Functional Food-Based Immunization.

Plant-based oral vaccines are now becoming a more transformative medical form in immunotherapy; they are cheap and needle-free. These systems use natural bioencapsulation in plant cell walls, which preserves antigen stability within the gastrointestinal tract and promotes targeted release to mucosal immune tissues. Tests indicate that plant-derived antigens preserved their structure after exposure and they produced robust mucosal IgA and systemic IgG responses. From the early trial, such as seroconversion among human subjects consuming hepatitis B surface antigen (HBsAg)-expressing potatoes an

Redox and immune response in early-lactating dairy cows supplemented with vitamin E or plant extract.

In dairy cows, early lactation is a period prone to oxidative stress, inflammation and health problems. Our objective was to investigate the effects of 2 strategies of nutritional supplementation on physiological and immunological status of early lactating cows. The first strategy consisted of supplementing vitamin E (all-rac-α-tocopheryl acetate) from 3 weeks before calving (3,000 IU/d before and 1,000 IU/d after calving). The other strategy consisted of supplementing plant extracts (Sambucus nigra, Salix alba, Laurus nobilis, Harpagophytum procumbens, Silybum marianum and Arctium lappa, QUAL

TOR Inhibition Enhances Autophagic Flux and Immune Response in Tomato Plants Against PSTVd Infection.

Viroids are small, non-coding RNA pathogens known for their ability to cause severe plant diseases. Despite their simple structure, viroids like Potato Spindle Tuber Viroid (PSTVd) can interfere with plant cellular processes, including transcriptional and post-transcriptional mechanisms, impacting plant growth and yield. In this study, we have investigated the role of the Target Of Rapamycin (TOR) signaling pathway in modulating viroid pathogenesis in tomato plants infected with PSTVd. Our findings reveal that PSTVd infection induces the accumulation of the selective autophagy receptor NBR1, p
