Concanavalin A, lectin from Canavalia ensiformis seeds has Leishmania infantum antipromastigote activity mediated by carbohydrate recognition domain.

Leishmaniases, caused by Leishmania parasites, are widespread and pose significant health risks globally. Visceral leishmaniasis (VL) is particularly prevalent in Brazil, with high morbidity and mortality rates. Traditional treatments, such as pentavalent antimonials, have limitations due to toxicity and resistance. Therefore, exploring new compounds like lectins is crucial. Concanavalin A (ConA) has shown promise in inhibiting Leishmania growth. This study aimed to evaluate its leishmanicidal effect on L. infantum promastigotes and understand its mechanism of action. In vitro tests demonstrat

Lectin cytochemical demonstration of glucose- and mannose-containing glycoconjugates on human reactive astrocytes.

Lectin cytochemistry of the human autopsy cases of Creutzfeldt-Jakob disease and old cerebral infarction was performed on formalin-fixed, paraffin-embedded sections using 15 lectins. Reactive astrocytes showed a positive reaction with concanavalin A (Con A), Lens culinaris agglutinin (LCA) and Pisum sativum agglutinin (PSA), with common specificity for both mannose and glucose. However normal astrocytes demonstrated no or little cytochemical reaction for any of the lectins. Reactivity for the lectins in control sections was obviously reduced using the blocking sugars (0.1 mol/l D-mannose and 0

The influence of diet on the mucin carbohydrates in the chick intestinal tract.

The presence of nutrients in the intestinal lumen is a major factor influencing bacterial colonization in poultry. This study was conducted to investigate the effects of poultry feed on viscosity of intestinal contents and on mucin carbohydrates by comparing jejunal supernatants and the histochemical composition of goblet cells in chicks reared to 5 weeks of age on either a conventional maize-based diet or a wheat-based diet or a wheat diet supplemented with 0.1% xylanase. Regional differences in the distribution of the neutral, carboxylated and sulphated mucins were demonstrated using convent

Sublines of the Burkitt line Daudi, selected on the basis of reactivity with soybean agglutinin, differ in tumorigenicity in athymic mice.

Two subpopulations were isolated on the basis of soybean agglutinin (SBA) binding, from the human Burkitt lymphoma line Daudi. The low- and high-binder sublines maintained this characteristic in continuous passages. Their surface marker profiles, antibodies, scanning electron microscope (SEM), cytochemical reactions and binding of other lectins (concanavalin A and wheatgerm agglutinin) were not different. They differed, however, in growth potential in athymic mice. The low-binder subline had lower frequency of takes, tumor weight and volume, and did not metastasize as compared to the high-bind

Ultrastructural demonstration of Maclura pomifera agglutinin binding sites in the membranocystic lesions of membranous lipodystrophy (Nasu-Hakola disease).

This paper reports three cases of membranous lipodystrophy (Nasu-Hakola disease) in two families and studies the carbohydrate components of membranocystic lesions in all three cases, using twelve kinds of lectins labelled by horseradish peroxidase (HRP). Maclura pomifera agglutinin (MPA), which specifically binds alpha-D-galactose residues, strongly stained typical membranocystic lesions, whereas the other lectins did not. However, Helix pomatia agglutinin (HPA), which specifically binds to N-acetyl-D-galactosamine (GalNAc), stained the membranes of degenerated adipose cells. These were though

Thrombospondin in essential thrombocythemia.

Essential thrombocythemia is a myeloproliferative disorder characterized by frequent bleeding and thrombotic complications. On a molecular level, two abnormalities of platelet thrombospondin have been identified: abnormal glycosylation of the intact 185,000-dalton chain has been detected and a shortened form of the thrombospondin chain is present. We have used two monoclonal antibodies and Lens culinaris lectin to probe the structure of thrombospondin in the platelets from three patients with essential thrombocythemia; one patient with polycythemia vera and two patients with secondary thromboc

Cytofluorometric quantification of somatopetally transported FITC-labelled lectins: enhanced uptake of concanavalin A and wheat germ agglutinin from the periphery in regenerating facial nerve.

Cytofluorometric quantification of the accumulation of somatopetally transported fluoroisothiocyanate (FITC)-labelled wheat germ agglutinin (WGA) and Concanavalin A (Con A) were performed on sections of the mouse facial nucleus. The lectins were injected bilaterally in the snout region of six-week-old mice eleven days after one of the facial nerves had been crushed. Analysis of variance revealed a significantly higher uptake of both WGA and Con A in the regenerating than in the control nerve. WGA appeared to be taken up to a higher degree than Con A, both in normal and regenerating nerve. Afte

The toxin-agglutinin fold. A new group of small protein structures organized around a four-disulfide core.

The three-dimensional structures of the snake venom postsynaptic neurotoxins and of the domains in wheat germ agglutinin show a remarkably similar overall folding pattern, consisting of equivalently placed, but variably sized loops which are held together by four similarly positioned disulfide bonds. Furthermore, occurrence of this wheat germ agglutinin-neurotoxin domain fold is predicted not only in the snake venom cardiotoxins and cytotoxins with neurotoxin-matched half-cystine sequence positions, but also for two small plant proteins, hevein and ragweed pollen allergen Ra5, on the basis of

The Use of Soybean Agglutinin (SBA) for Bone Marrow (BM) Purging and Hematopoietic Progenitor Cell Enrichment in Clinical Bone Marrow Transplantation.

Soybean agglutinin (SBA) is a plant lectin, a glycoprotein of nonimmune origin that binds specifically to cell surface carbohydrates through noncovalent combinations and thus provokes agglutination of the bound cells. SBA has been used, therefore, to fractionate a variety of cell types. SBA binds approx 60-90% of bone marrow mononuclear cells, including mature myeloid, erythroid, and lymphoid cells, but has very low binding affinity and no toxic effect to the human hematopoietic progenitor cells. In addition, it binds very effectively to certain tumor cells, including neuroblastoma, breast can

Expression of the L-fucose moiety on infrainfundibular follicular keratinocytes of terminal follicles, its decreased expression on vellus and indeterminate follicles of androgenetic alopecia, and re-expression in drug-induced hair regrowth.

The distribution of various glycoprotein molecules on the surface of follicular keratinocytes was studied with a panel of lectins with specificity for various sugar moieties on biopsy specimens from both bald/balding scalp and normal occipital scalp, of 23 patients with androgenetic alopecia as well as on biopsies of normal forearm skin of four patients. The most significant differences between bald and normal scalp biopsy were noted with Ulex europaeus agglutinin I (UEA I). We noted an increased (91.8% +/- 3.1; mean +/- SE) expression of UEA I binding sites on the infra-infundibular follicula

Hepatic sinusoidal endothelium: Ulex lectin binding.

The sinusoidal endothelial cells of human liver can be identified by light and electron microscopy, but there appear to be no specific immunocytochemical markers of these cells. Among specific markers available for vascular endothelial cells in general, Ulex europaeus I lectin (UEA I) is the most sensitive. In the present study, 37 liver biopsies were examined for UEA I binding and for Factor VIII related antigen (F VIII RAg) to determine if sinusoidal endothelial cells were positive. The material included normal liver, biopsies from patients with cirrhosis and biopsies in a variety of other l

Ulex europeus agglutinin I-reactive high molecular weight glycoproteins of adenocarcinoma of distal colon and rectum and their possible relationship with metastatic potential.

A Ulex europeus agglutinin I (UEAI)-reactive glycoprotein(s) with molecular weight higher than 300,000 was detected by direct binding of 125I-labeled UEAI to lysates of rectal or sigmoid colon cancer tissues separated by polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. This was the major UEAI-reactive molecule in tumor tissues and different tumors possessed varying reactivities. Very little UEAI binding was detected to lower molecular weight components. Histochemical localization of UEAI confirmed that the UEAI-reactive molecules were mostly localized to the surfac

Engineering cotton with ASAL and MYB genes confer dual benefits of pest resistance and fiber improvement.

The MYB-ASAL strategy is a promising integrated approach for developing cotton cultivars with enhanced fiber quality and reduced pesticide resistance, strengthening the textile security. The quality of cotton fiber is a critical determinant in the textile industry, significantly influencing the overall quality of textile products. Environmental stressors, particularly extreme climatic conditions, adversely affect fiber quality. This study aims to investigate the enhancement of fiber traits by expressing the transcription factor MYB involved in fiber quality along with the implementation of cro

Toxicity studies of detoxified Jatropha meal (Jatropha curcas) in rats.

Jatropha curcas, a tropical plant introduced in many Asian and African countries is presently used as a source of biodiesel. The cake after oil extraction is rich in protein and is a potential source of livestock feed. In view of the high toxic nature of whole as well as dehulled seed meal due to the presence of toxic phorbol esters and lectin, the meal was subjected to alkali and heat treatments to deactivate the phorbol ester as well as lectin content. After treatment, the phorbol ester content was reduced up to 89% in whole and dehulled seed meal. Toxicity studies were conducted on male gro

Cell surface carbohydrates reveal heterogeneity in olfactory receptor cell axons in the mouse.

Cell surface carbohydrates, both in the olfactory system and elsewhere, have been proposed to play critical roles in axon guidance and targeting. Recent studies have used plant lectins to study the heterogeneous distribution of carbohydrates in the olfactory system. One lectin, Dolichos biflorus agglutinin (DBA), heterogeneously labels subsets of glomeruli. In the olfactory epithelium DBA labeled a subset of olfactory sensory neurons (OSNs) including their cilia, dendrites, and somata. OSN axons were also labeled and readily observed in the olfactory nerve and bulb. The patterns of glomerular …

Beta 1-6 branched oligosaccharides as a marker of tumor progression in human breast and colon neoplasia.

Malignant transformation of murine and human cells is commonly associated with increased--GlcNAc beta 1-6Man alpha 1-6Man beta--branching in asparagine-linked oligosaccharides. Somatic mutations and drugs which block expression of the beta 1-6 branched oligosaccharides are potent inhibitors of tumor cell invasion and metastasis in animal models. This suggests that the oligosaccharides are required for metastasis to occur and therefore their increased presence in primary tumors may be diagnostic of metastatic disease. Although antibodies to the beta 1-6 branched portion of the oligosaccharides
