Reply to: Bridging predictive algorithms and clinical practice in DBS contact selection.

We thank the authors (Weide et al.1) for their evaluation of our algorithm, which predicts optimal deep-brain stimulation contact points in Parkinson's disease. Their analysis on an independent dataset highlights practical challenges of translating predictive tools into routine clinical use. Here, we clarify key methodological points, discuss the robustness of the algorithm, and emphasize the need for broader validation across diverse clinical centers to fully demonstrate its utility.

Infectious disease prediction model based on optimized deep learning algorithm.

Since the end of 2019, a novel coronavirus known as COVID-19 has caused a severe outbreak worldwide. Due to the complexity of epidemic data, traditional algorithms have struggled to accurately predict the development of the pandemic. The Autoregressive Integrated Moving Average (ARIMA) model is capable of capturing time-based trends in epidemic data, including seasonality, cyclic patterns, and long-term trends, which helps improve the accuracy of forecasting future epidemic trajectories. The Bidirectional Long Short-Term Memory (BiLSTM) network, a variant of the Recurrent Neural Network (RNN),

Machine-learning algorithms for predicting colchicine resistance in Familial Mediterranean Fever.

OBJECTIVES: FMF is a genetic autoinflammatory condition marked by recurrent fever and serositis. Colchicine is the mainstay treatment. However, 5% to 10% of patients exhibit colchicine resistance, requiring alternative therapies. Early identification of resistance is vital. While some scoring systems exist for paediatric FMF, artificial intelligence's (AI's) potential to predict colchicine resistance in adult patients with FMF has not been systematically explored. This study aimed to utilize machine-learning (ML) and deep-learning (DL) algorithms to predict colchicine resistance in adult patie

An Accelerated Maximum Flow Algorithm with Prediction Enhancement in Dynamic LEO Networks.

Efficient data transmission in low Earth orbit (LEO) satellite networks is critical for supporting real-time global communication, Earth observation, and numerous data-intensive space missions. A fundamental challenge in these networks involves solving the maximum flow problem, which determines the optimal data throughput across highly dynamic topologies with limited onboard energy and data processing capability. Traditional algorithms often fall short in these environments due to their high computational costs and inability to adapt to frequent topological changes or fluctuating link capaciti

Predictive Algorithm for Thiopurine-Induced Hepatotoxicity in Inflammatory Bowel Disease Patients.

BACKGROUND: Approximately 25% of patients with inflammatory bowel disease (IBD) discontinue azathioprine (AZA) or mercaptopurine (MP) therapy within 3 months of treatment initiation because of adverse drug reactions. Of these side-effects, about half are because of hepatotoxicity. The aim of this study was to validate and (subsequently) optimize a previously reported predictive algorithm for thiopurine-associated hepatotoxicity by increasing the number of patients with IBD benefitting from conventional thiopurine therapy. METHODS: This multicenter observational study included consecutive thio

Predicting phenotype transition probabilities via conditional algorithmic probability approximations.

Unravelling the structure of genotype-phenotype (GP) maps is an important problem in biology. Recently, arguments inspired by algorithmic information theory (AIT) and Kolmogorov complexity have been invoked to uncover simplicity bias in GP maps, an exponentially decaying upper bound in phenotype probability with the increasing phenotype descriptional complexity. This means that phenotypes with many genotypes assigned via the GP map must be simple, while complex phenotypes must have few genotypes assigned. Here, we use similar arguments to bound the probability P(x → y) that phenotype x, upon r
