The development of psychotic disorders in adolescence: a potential role for hormones.

This article is part of a Special Issue "Puberty and Adolescence". The notion that adolescence is characterized by dramatic changes in behavior, and often by emotional upheaval, is widespread and longstanding in popular western culture. In recent decades, this notion has gained increasing support from empirical research showing that the peri- and post-pubertal developmental stages are associated with a significant rise in the rate of psychiatric symptoms and syndromes. As a result, interest in adolescent development has burgeoned among researchers focused on the origins of schizophrenia and ot

Family-based association of YWHAH in psychotic bipolar disorder.

YWHAH is a positional and functional candidate gene for both schizophrenia and bipolar disorder (BP). This gene has been previously shown to be associated with both disorders, and the chromosome location (22q12.3) has been repeatedly implicated in linkage studies for these disorders. It codes for the eta subtype of the 14-3-3 protein family, is expressed mainly in brain, and is involved in HPA axis regulation. We investigated the association of YWHAH with BP in a large sample, consisting of 1211 subjects from 318 nuclear families including 554 affected offspring. We tested for association with

Correlates of the affective impact of auditory hallucinations in psychotic disorders.

While many who hear auditory hallucinations (AHs) experience them as unpleasant, some do not. Little is known about the correlates of AHs that are not unpleasant, or of the characteristics of those who hear them. To better understand this symptom, we used a comprehensive structured interview schedule to study 199 subjects who had experienced AHs. Subjects' responses to AHs were combined into two indexes: one assessing total affective impact and the other assessing the affective direction (positive or negative). Subjects who had grandiose delusions experienced their AHs more positively. AHs tha

Body dysmorphic disorder: does it have a psychotic subtype?

BACKGROUND: Although body dysmorphic disorder (BDD) is classified in DSM-III-R as a nonpsychotic somatoform disorder, controversy exists as to whether BDD can present with psychotic features. If it can, this raises the possibility that its DSM-III-R psychotic counterpart-delusional disorder, somatic type--may not be a separate disorder. The purpose of this study was to determine whether patients with nonpsychotic BDD (defined according to DSM-III-R criteria, i.e., with maintenance of some insight) were different from patients with psychotic BDD (those whose preoccupation was without insight an

Schizophrenia and psychotic disorders in precision medicine and body-brain axis.

The concept of precision medicine has emerged over the past decade: it emphasizes the utilization of human genomic data, demonstrating that molecular variations could predict therapeutic responses and clinical outcomes. This approach has been highly successful in oncology and other medical conditions. Nevertheless, given the heavy impact of environmental stressors and longitudinal life-course experiences on the molecular signatures that directly underlie the pathophysiology beyond genetic factors in neuropsychiatric and psychiatric disorders, post-genomic precision medicine is also highly warr

Structural connectomic signatures of childhood maltreatment across affective and psychotic disorders.

Exposure to childhood maltreatment (CM) is a crucial risk factor for affective disorders and schizophrenia, which are linked to structural brain network alterations. We hypothesized a significant association between CM and brain structural connectivity in a transdiagnostic sample. Participants included patients with major depressive disorder (n = 827), bipolar disorder (n = 134), schizophrenia spectrum disorders (n = 118), and healthy controls (n = 932) aged 18-65 from the Marburg-Muenster Affective Disorders Cohort Study. Structural brain networks were reconstructed from structural and diffus

Precision medicine for psychotic disorders: objective assessment, risk prediction, and pharmacogenomics.

Psychosis occurs inside the brain, but may have external manifestations (peripheral molecular biomarkers, behaviors) that can be objectively and quantitatively measured. Blood biomarkers that track core psychotic manifestations such as hallucinations and delusions could provide a window into the biology of psychosis, as well as help with diagnosis and treatment. We endeavored to identify objective blood gene expression biomarkers for hallucinations and delusions, using a stepwise discovery, prioritization, validation, and testing in independent cohorts design. We were successful in identifying

Glucose-6-phosphate dehydrogenase deficiency and psychotic disorders: A systematic review.

OBJECTIVES: Glucose-6-Phosphate Dehydrogenase Deficiency (G6PD) is the most common enzymopathy globally. Early studies suggested an association with severe psychotic illness; however, changes to laboratory testing and diagnostic classification renders the association unclear. This study aims to explore the interaction between G6PD deficiency and psychotic symptoms, in particular to identify specific patterns of presentation or impact on outcomes. METHODS: Pubmed, Embase, and PsycInfo databases were searched from inception to May 2023. Descriptive statistics and narrative review of were used t

Test-retest reliability of the Childhood Trauma Questionnaire in psychotic disorders.

BACKGROUND: Childhood trauma is common and associated with worse psychiatric outcomes. Yet, clinicians may not inquire about childhood trauma due to a misconception that patients cannot provide reliable reports. The goal of this study was to examine the reliability of self-reports of childhood trauma in psychotic disorders. METHODS: We examined the test-retest reliability of the Childhood Trauma Questionnaire (CTQ) in schizophrenia (SZ, n = 19), psychotic bipolar disorder (BD, n = 17), and healthy control (HC, n = 28) participants who completed the CTQ on ≥2 occasions over variable time perio

A neuropsychological approach to differentiating cannabis-induced and primary psychotic disorders.

AIM: Rates of cannabis use are elevated in early psychosis populations, rendering it difficult to determine if an episode of psychosis is related to cannabis use (e.g., cannabis-induced psychosis), or if substance use is co-occurring with a primary psychotic disorder (e.g., schizophrenia). Clinical presentations of these disorders are often indistinguishable, hindering assessment and treatment. Despite substantial research identifying cognitive deficits, eye movement abnormalities and speech impairment associated with primary psychotic disorders, these neuropsychological features have not been

Differing functional mechanisms underlie cognitive control deficits in psychotic spectrum disorders.

BACKGROUND: Functional underpinnings of cognitive control deficits in unbiased samples (i.e., all comers) of patients with psychotic spectrum disorders (PSD) remain actively debated. While many studies suggest hypofrontality in the lateral prefrontal cortex (PFC) and greater deficits during proactive relative to reactive control, few have examined the full hemodynamic response. METHODS: Patients with PSD (n = 154) and healthy controls (n = 65) performed the AX continuous performance task (AX-CPT) during rapid (460 ms) functional neuroimaging and underwent full clinical characterization. Sauna Bathing and Risk of Psychotic Disorders: A Prospective Cohort Study.

OBJECTIVE: Sauna bathing has been suggested to promote mental well-being and relaxation, but the evidence is uncertain with respect to mental disorders. We aimed to assess the association of frequency of sauna bathing with risk of psychosis in the Kuopio Ischemic Heart Disease prospective population-based study. SUBJECTS AND METHODS: Baseline sauna bathing habits were assessed in 2,138 men aged 42-61 years who had no history of psychotic disorders. Participants were classified into three groups based on the frequency of sauna bathing (once, 2-3, and 4-7 times per week). RESULTS: During a med

Psychotic disorders in sexually abused youth: A prospective matched-cohort study.

Child sexual abuse has been identified as a potential risk factor for developing a psychotic disorder. However, little is known about the prevalence of psychotic disorders in youth who were sexually abused during adolescence and young adulthood. Gender differences also remain unclear. This study used administrative databases from a Child Protection Agency and a public health care system. It aimed to investigate the prevalence of psychotic disorders in sexually abused youth between the first substantiated report of sexual abuse and the beginning of adulthood. A second objective was to assess ge

The Bidirectional Associations Between Psychotic Experiences and DSM-IV Mental Disorders.

OBJECTIVE: While it is now recognized that psychotic experiences are associated with an increased risk of later mental disorders, we lack a detailed understanding of the reciprocal time-lagged relationships between first onsets of psychotic experiences and mental disorders. Using data from World Health Organization World Mental Health (WMH) Surveys, the authors assessed the bidirectional temporal associations between psychotic experiences and mental disorders. METHOD: The WMH Surveys assessed lifetime prevalence and age at onset of psychotic experiences and 21 common DSM-IV mental disorders a

Neuroimaging and emerging psychotic disorders: the Melbourne ultra-high risk studies.

Although the underlying neurobiology of emerging psychotic disorders is not well understood, evidence from structural imaging and other studies support the notion that schizophrenia arises as a consequence of both an 'early neurodevelopmental' disturbance, as well as 'late neurodevelopmental' changes occurring during the initial stages of a psychotic illness, including around the time of transition to illness. In line with this, our longitudinal MRI findings in individuals at ultra-high risk for developing a psychotic illness show that there are excessive neuroanatomical changes in those who c

The Intersection Between Childhood Trauma, the COVID-19 Pandemic, and Trauma-related and Psychotic Symptoms in People With Psychotic Disorders.

INTRODUCTION: People with psychotic disorders may be disproportionately affected by the traumatic effects of the COVID-19 pandemic. Childhood trauma, which also increases vulnerability to subsequent stressors, is common in individuals with psychosis. In this study, we investigated the intersection of the pandemic, childhood trauma, and psychotic and trauma-related symptoms in individuals with psychotic disorders. METHODS: We administered a cross-sectional survey to 151 participants [47 schizophrenia (SZ), 53 psychotic bipolar disorder (BP)], 51 healthy control (HC)] during the COVID-19 pandem

Associations between the Five-Factor Model personality traits and psychotic experiences in patients with psychotic disorders, their siblings and controls.

Earlier studies indicated that personality characteristics contribute to symptomatic outcome in patients with psychotic disorders. The aim of the present study was to further explore this connection by examining the relationship between the Five-Factor Model (FFM) personality traits and a dimensional liability for psychosis. FFM traits according to the NEO-FFI and levels of subclinical psychotic symptoms according to the CAPE were assessed in 217 patients with psychotic disorders, 281 of their siblings and 176 healthy controls. Psychotic symptoms according to the PANSS were assessed in the pat

Associations between PTSD and psychotic symptoms: A network analysis of patients with psychotic disorders in Uganda: Psychosis-PTSD network analysis.

INTRODUCTION: Individuals with psychosis often endorse a history of trauma, yet the symptom-level interplay between trauma-related and psychotic symptoms is understudied globally and largely absent in Sub-Saharan Africa. We used network analysis to examine associations within and between these domains. METHODS: We conducted a network analysis of 807 participants with a psychotic disorder and trauma history in the Ugandan cohort of the NeuroGAP-Psychosis program.
