Di (2-ethylhexyl) phthalate induced lipophagy-related renal ferroptosis in quail (Coturnix japonica).

Di(2-ethylhexyl) phthalate (DEHP) is a synthetic chemical applied as a plasticizer. As an environmental toxicant, DEHP poses a serious health threat. Many studies have revealed that DEHP can cause lead to various degrees of damage to the kidney. However, the evidence of DEHP-induced renal ferroptosis has not been reported. The purpose of this work was to probe the specific role of lipophagy in DEHP-induced renal injury and to investigate the relationship between lipophagy and ferroptosis. Quail were treated with DEHP (250 mg/kg BW/day, 500 mg/kg BW/day and 750 mg/kg BW/day) for 45 days. Micros

Protocol for inducing hippocampal formation lesions and associated behavioral testing in Japanese quail.

Here, we present a protocol for inducing selective lesions in the hippocampal formation of Japanese quail (Coturnix japonica), coupled with associated behavioral testing. We first describe the surgical procedure for aspiration lesions in Japanese quail. We then detail two well-known hippocampus-dependent behavioral tests adapted to birds-foraging array (FA) and spontaneous object recognition (SOR). This protocol is adapted from those used in mammals and can be used to study the involvement of Japanese quail memory centers in declarative memory. For complete details on the use and execution of

Trans-ovo permethrin exposure affects growth, brain morphology and cardiac development in quail.

Permethrin is a commonly used, highly effective pesticide in poultry agriculture, and has recently been trialed in conservation efforts to protect Galápagos finch hatchlings from an invasive ectoparasite. Although permethrin is considered safe for adults, pesticides can have health consequences when animals are exposed during early life stages. The few studies that have examined permethrin's effects in embryonic chicks and rats have shown hydrocephaly, anencephaly, reduced cellular energy conversion, and disruption of developing heart muscle. To test whether trans-ovo exposure of permethrin af

Muscle Hyperplasia in Japanese Quail by Single Amino Acid Deletion in MSTN Propeptide.

Mutation in myostatin (MSTN), a negative regulator of muscle growth in skeletal muscle, resulted in increased muscle mass in mammals and fishes. However, MSTN mutation in avian species has not been reported. The objective of this study was to generate MSTN mutation in quail and investigate the effect of MSTN mutation in avian muscle growth. Recently, a new targeted gene knockout approach for the avian species has been developed using an adenoviral CRISPR/Cas9 system. By injecting the recombinant adenovirus containing CRISPR/Cas9 into the quail blastoderm, potential germline chimeras were gener

Hypothalamic mechanisms associated with neuropeptide K-induced anorexia in Japanese quail (Coturnix japonica).

Central administration of neuropeptide K (NPK), a 36-amino acid peptide, is associated with anorexigenic effects in rodents and chickens. The mechanisms underlying the potent anorexigenic effects of NPK are still poorly understood. Thus, the aim of the present study was to identify the hypothalamic nuclei and neuropeptides that mediate anorexic effects of NPK in 7 day-old Japanese quail (Coturnix japonica). After a 6 h fast, intracerebroventricular (ICV) injection of NPK decreased food and water intake for 180 min post-injection. Quail injected with NPK had more c-Fos immunoreactive cells in t

Mesocestoides sp. in Wild Northern Bobwhite ( Colinus virginianus) and Scaled Quail ( Callipepla squamata).

Mesocestoides spp. have a cosmopolitan distribution with zoonotic potential. Mesocestoides tetrathyridia were found under the pericardial sac, on the surface of the crop, and in the peritoneal cavity of a hunter-harvested Northern Bobwhite ( Colinus virginianus) and a Scaled Quail ( Callipepla squamata) collected during the 2016-17 quail hunting season in northwest and southern Texas, US respectively. Molecular analysis indicated that the tetrathyridia from the birds likely belonged to an undescribed species and are identical to pretetrathyridium stages recently found in Scincella lateralis sk

Immunotoxicity and disease resistance in Japanese quail (Corturnix coturnix japonica) exposed to malathion.

The purpose of this study was to evaluate the impact of malathion on the immune system of wild birds, using Japanese quail (Coturnix coturnix japonica) as a model. Quail were exposed to malathion in drinking water at environmentally realistic concentrations (0 ppm, 1 ppm, and 10 ppm). In the fifth week, several arms of the immune response were tested using the T-cell based phytohemagglutinin (PHA) skin test, the B-cell mediated antibody response, and the chemiluminescence assay measuring innate immunity. After the sixth week of malathion exposure, quail were challenged with E. coli O2. The bur

Sexual fetishism in a quail (Coturnix japonica) model system: test of reproductive success.

In the present study, the authors explored the reproductive consequences of fetishistic behavior in a previously developed animal model of sexual fetishism (F. Köksal et al., 2004). Male domesticated quail (Coturnix japonica) received sexual conditioning trials in which a terrycloth object (the conditioned stimulus [CS]) was paired with the opportunity to copulate with a female quail (the unconditioned stimulus). Approximately half of the male quail came to copulate with the CS object and were considered to have developed fetishistic behavior. Each of the male quail was then tested with a fema

Evaluation of adult quail and egg production following exposure to perchlorate-treated water.

Twenty-three adult female northern bobwhite (Colinus virginianus) quail were exposed to 0, 0.01, 0.1, and 1 mM ammonium perchlorate (AP) in drinking water for 30 d. Eggs laid in all treatment groups, including control, were collected, dated, given an identification number, and weighed. On day 30 of exposure, 10 birds were euthanized by carbon dioxide asphyxiation. Gross toxicological endpoints and thyroid histology were evaluated in 10 birds. Egg production and accumulation of perchlorate in the eggs (n = 10) and liver (n = 5) were determined. Perchlorate did not affect body or organ weights s

Immediate early gene expression associated with induction of brooding behavior in Japanese quail.

Certain species can be induced to foster infant or neonatal animals through the process of sensitization. We induced brooding behavior in adult Japanese quail through repeated exposure to foster chicks across five 20-min trials. Brooding behavior was characterized by a bird allowing chicks to approach and remain underneath its wings while assuming a distinctive stationary crouching posture, preening, and feather fluffing. Birds who did not show brooding behavior actively avoided chicks. Among the birds that brooded chicks, females brooded chicks for longer durations compared to males. Brooding

Regulation of hydroxyindole-O-methyltransferase gene expression in Japanese quail (Coturnix coturnix japonica).

Hydroxyindole-O-methyltrasferase (HIOMT) plays an important role as the final enzyme in the synthesis of melatonin. In this study, the expression of the HIOMT gene in Japanese quail was investigated with respect to tissue distribution and the effects of light and vitamin A deficiency. HIOMT mRNA in the pineal gland and eye had a clear daily rhythm with peak values in daytime. The testis also contained a detectable amount of HIOMT mRNA, which did not display a rhythmic change over a 24-h period. When birds were rendered vitamin A deficient through feeding with a vitamin A-free diet, the daily r

Shotgun proteomic analysis of quail liver under feed deprivation and low-energy diet conditions.

Feed intake determines nutrient utilization and production efficiency in poultry, yet hepatic mechanisms underlying responses to acute nutritional challenges remain insufficiently characterized. This study aimed to identify the liver proteins and pathways associated with feed intake regulation in Japanese quails (Coturnix japonica) subjected to different nutritional challenges. Eighteen 12-wk-old male quails (245.20 g ± 0.213) were housed individually and allocated to ad libitum control, 24 h feed-deprived, or 24 h low metabolizable energy groups (n = 6/group). The basal diet contained 18% cru

Impact of family-based selection on growth performance and immune response of Japanese quail.

The present study was aimed to evaluate the growth performance and immune response of three genetic lines of Japanese quails. These lines i.e., selected for 4-week body-weight group (WBS), selected for egg number (EBS), and random-bred control (RBC), were selected for three consecutive generations from a base population of 1125 quails. In total, 2700 four-week-old quails from three selected groups were slaughtered in total of four generations (G0 to G3). Effects of selection and generations as well as their interactions were assessed for growth performance and immune response by applying a two

Action spectrum for photoperiodic control of thyroid-stimulating hormone in Japanese quail (Coturnix japonica).

At higher latitudes, vertebrates exhibit a seasonal cycle of reproduction in response to changes in day-length, referred to as photoperiodism. Extended day-length induces thyroid-stimulating hormone in the pars tuberalis of the pituitary gland. This hormone triggers the local activation of thyroid hormone in the mediobasal hypothalamus and eventually induces gonadal development. In avian species, light information associated with day-length is detected through photoreceptors located in deep-brain regions. Within these regions, the expressions of multiple photoreceptive molecules, opsins, have

Female Japanese quail with high levels of estradiol demonstrate cocaine-induced conditioned place preference.

Preclinical research has indicated that females may be more sensitive to the rewarding properties of cocaine. However, the majority of this research has been done in rodent species. Environmental cues associated with human drug-taking behavior tend to be visual. Because rodents do not rely on the visual system as their primary sense modality, the use of a visually oriented species may add to our understanding of cue-elicited drug cravings and relapse. The present study examined the potential role of the steroid hormone, estradiol, in the rewarding properties of cocaine in female Japanese quail

Characterization of Newcastle disease viruses isolated from chicken, gamefowl, pigeon and quail in Mexico.

Velogenic Newcastle disease has threatened the Mexican poultry industry since 1946. Seven strains of velogenic Newcastle disease virus were isolated from poultry and other avian species in central and northern Mexico from 1998 to 2006 and subjected to phylogenetic analysis and biological characterization using standard pathogenicity tests and challenge studies. Phylogenetic analysis showed that all velogenic strains belonged to genetic group V and are clearly divided in two lineages, since phylogenetic similarities between groups are of only 93-94%. Isolates from 1998 to 2001 are closely relat
