Adjunct antiseizure effect of clotrimazole in a rotenone corneal kindling mouse model of mitochondrial drug-resistant epilepsy.

This study aimed to investigate the therapeutic potential of clotrimazole, an inhibitor of the transient receptor potential cation channel, for treating mitochondrial drug-resistant epilepsy and to understand its underlying neurochemical mechanisms. Adult albino mice underwent rotenone-corneal kindling, receiving daily electric shocks (15 mA, 20 V, 6-Hz for 3 s) through a corneal electrode, to induce mitochondrial drug-resistant epilepsy. The onset of drug resistance was confirmed by the significant (p < 0.05) lack of seizure control with standard antiseizure medications including levetiraceta…

Rotenone induced olfactory deficit in Parkinson's disease rat model: The protective role of adenosine A2A receptors antagonist.

BACKGROUND: Parkinson's disease is both a motor and non-motor disorder. Despite the non-motor being an intrinsic feature of PD, it has been poorly researched and understood in clinical practices; olfactory deficit is one of the first established non motor symptom and nearly all ∼90 % of sporadic PD cases are associated with olfactory dysfunction and there is inconsistency in various pharmacological approaches. Hence this study aimed to evaluate the impact of caffeine at the A2A receptors of the olfactory bulb of a rotenone rat model of Parkinson's disease. MATERIALS AND METHODS: About 50 male…

Rotenone impairs learning and memory in mice through microglia-mediated blood brain barrier disruption and neuronal apoptosis.

Rotenone is a neurotoxic pesticide widely used in agriculture. Dopaminergic neuron has long been considered as the target of rotenone. We recently reported that rotenone exposure also resulted in hippocampal and cortical neurodegeneration and cognitive dysfunction in mice. However, the mechanisms remain unknown. Here, we elucidated whether blood brain barrier (BBB) disruption and subsequent neuronal apoptosis mediated by microglial activation were involved in rotenone-elicited cognitive impairments. Results showed that rotenone dose-dependently elevated evens blue extravasation, fibrinogen acc…

Ginsenoside Rg3 exerts a neuroprotective effect in rotenone-induced Parkinson's disease mice via its anti-oxidative properties.

Ginsenoside Rg3, extracted from Panax ginseng C.A. Meyer, has been shown to possess neuroprotective properties. The present study aims to investigate the neuroprotective effects of ginsenoside Rg3 on rotenone-induced Parkinson's disease mice. Rotenone, a mitochondrial complex I inhibitor, leads to the augmentation of reactive oxygen species production in cells. Male C57/BL6 mice were intragastrically administered rotenone (30 mg/kg) and then treated with ginsenoside Rg3 (5, 10, or 20 mg/kg). Pole, rotarod, and open field tests were performed to evaluate motor function. Ginsenoside Rg3 decrease…

Downregulation of metabotropic glutamate receptor 5 inhibits hepatoma development in a neurotoxin rotenone-induced Parkinson's disease model.

Clinical epidemiological studies have shown that there is a link between Parkinson's disease (PD) and cancer, but how PD regulates cancer development remains unknown. In our study, the effect of metabotropic glutamate receptor 5 (mGlu5) on hepatoma was explored in a rotenone-induced PD model both in vitro and in vivo. We found that conditioned media derived from MN9D dopaminergic neuronal cells by rotenone-induced toxicity inhibited the growth, migration, invasion and promoted apoptosis of Hepa1-6 cells, which corresponded with decreased expression of mGlu5. Furthermore, treatment with 2-methy…

Behavioral differences in a rotenone-induced hemiparkinsonian rat model developed following intranigral or median forebrain bundle infusion.

A mitochondrial complex-I inhibitor, rotenone was unilaterally infused into the substantia nigra pars compacta (SNpc) or median forebrain bundle (MFB) to create hemiparkinsonian animal models and investigated spontaneous and drug-induced stereotypic rotations, as well as certain postural behaviors in Sprague-Dawley rats. Animals infused intranigrally, but not intra-MFB, with rotenone exhibited spontaneous contralateral rotations immediately after recovery from anesthesia. Head position bias and elevated body swing test showed insignificant contralateral bias in animals with nigral damage but a…

Regulation of HSP90AA1/NRF2 by BDNF Contributes to the Attenuation of Rotenone-Induced Oxidative Stress in Parkinson's Disease Models.

Parkinson's disease, a neurodegenerative disorder, is characterized by the degeneration of dopaminergic neurons and the accumulation of α-synuclein, both of which are aggravated by oxidative stress. This study utilized rotenone-treated SH-SY5Y cells to assess cell viability, ROS levels, and mitochondrial function. RNA-seq, mass spectrometry, and Co-IP analyses identified BDNF-regulated proteins linked to oxidative stress. In rotenone-induced PD mice, evaluations were made of motor performance, neuronal degeneration, and protein expression. Results showed that a 36-h exposure to 0.5 µM rotenone…

Rotenone Induces Morphological Damage and Reduction in Catecholamine Levels in the Central Nervous System of the Ascidian Styela plicata.

Ascidians are invertebrates that occupy a key phylogenetic position as a sister group of the vertebrates. The organization of their central nervous system (CNS), with cortex and medulla, indicates a plesiomorphic character of the gray and white matter of vertebrates, making this group of animals useful for studies of neurodegenerative events. Rotenone, a natural pesticide extracted from the roots of specific plants, can be used as a neurotoxin to promote parkinsonism in various organism models, leading to degeneration of the dopaminergic neurons, mitochondrial dysfunction, and locomotor defici…

The Effect of Neuronal CoQ10 Deficiency and Mitochondrial Dysfunction on a Rotenone-Induced Neuronal Cell Model of Parkinson's Disease.

Parkinson's disease (PD) is the second most prevalent neurodegenerative disorder currently affecting the ageing population. Although the aetiology of PD has yet to be fully elucidated, environmental factors such as exposure to the naturally occurring neurotoxin rotenone has been associated with an increased risk of developing PD. Rotenone inhibits mitochondrial respiratory chain (MRC) complex I activity as well as induces dopaminergic neuronal death. The aim of the present study was to investigate the underlying mechanisms of rotenone-induced mitochondrial dysfunction and oxidative stress in a…

Effects of Catechin, Quercetin and Taxifolin on Redox Parameters and Metabolites Linked with Renal Health in Rotenone-toxified Rats.

Nephrotoxicity, with the attendant risk of progression to kidney failure, is a growing problem in many parts of the world. Current orthodox treatment options for nephrotoxicity and kidney failure are limited and there is need for alternative or complementary approaches. This study aimed at evaluating the effect of three structurally related flavonoids, catechin, quercetin and taxifolin on renal redox and metabolite biochemical disturbances in rotenone intoxicated animals. Male Wistar rats were administered 1.5 mg/kg rotenone (s.c.) for ten days followed by post-treatment with catechin (5, 10 o…

Controversies on new animal models of Parkinson's disease pro and con: the rotenone model of Parkinson's disease (PD).

A general complex I deficit has been hypothesized to contribute to neurodegeneration in Parkinson's disease (PD) and all toxins used to destroy dopaminergic neurons are complex I inhibitors. With MPTP or 6-OHdopamine, this hypothesis can not be tested since these toxins selectively accumulate in the dopaminergic neurons. However with rotenone, which penetrates all cells, the hypothesis can be tested. Thus, the proof of the hypothesis is whether or not rotenone-induced neurodegeneration mimics the degenerative processes underlying PD. Low doses of rotenone (1.5 or 2.5 mg/kg in oil i.p.) were ad…

The Neuroameliorative Effects of Enzymatically Modified Isoquercitrin and Sodium R-lipoate on the Rotenone rat Model of Parkinson's Disease.

Parkinson's disease (PD) is a neurodegenerative condition distinguished by both motor and non-motor signs. Currently, therapeutic interventions effectively mitigate motor symptoms; however, they fail to impede the progression of neurodegeneration. Enzymatically modified isoquercitrin (EMIQ) is a variant of isoquercitrin with enhanced bioavailability and potentially greater health benefits. Sodium R-Lipoate (NaRLA) is a modified form of lipoic acid with improved stability and efficacy. The current work assessed EMIQ and NaRLA neuroameliorative properties in a rotenone (ROT) model of PD in rats.…

Rescue of a Rotenone Model of Parkinson's Disease in C. elegans by the Mitochondrial Na+/Ca2+ Exchanger Inhibitor CGP37157.

We have previously shown that the compound CGP37157, a mitochondrial Na+/Ca2+ exchanger inhibitor, increases lifespan and improves muscle and mitochondrial structure during aging in wild-type C. elegans nematodes. We used here a rotenone model of Parkinson's disease in C. elegans to test the ability of CGP37157 to rescue the alterations induced by the toxicant. Rotenone, a mitochondrial respiratory chain complex I inhibitor, reduced worm lifespan and muscle activity, measured as worm mobility, pharyngeal pumping, and defecation rate. It also increased ROS production, decreased mitochondrial me…

PROTECTIVE EFFECTS OF CURCUMA LONGA IN A ROTENONE-INDUCED RAT MODEL OF PARKINSON'S DISEASE: ELECTROPHYSIOLOGICAL AND BEHAVIORAL EVIDENCE.

Parkinson's disease (PD) is characterized by progressive dopaminergic neurodegeneration leading to motor deficits, yet effective neuroprotective therapies remain limited. Curcuma longa (turmeric), a traditionally cultivated spice in Southeast Asia, exhibits potent antioxidant properties that may counteract neurodegenerative processes. This study investigated the neuroprotective potential of orally administered turmeric extract (1100 mg/kg) in a rotenone-induced rat model of PD. Rats received rotenone (2.5 mg/kg) for 21 days to induce PD-like pathology, followed by concurrent turmeric treatment…
