Defence sugarcane glycoproteins disorganize microtubules and prevent nuclear polarization and germination of Sporisorium scitamineum teliospores.

Microtubules (MTs) are involved in the germination of Sporisorium scitamineum teliospores. Resistant varieties of sugar cane plants produce defence glycoproteins that prevent the infection of the plants by the filamentous fungi Sporisorium scitamineum. Here, we show that a fraction of these glycoproteins prevents the correct arrangement of MTs and causes nuclear fragmentation defects. As a result, nuclei cannot correctly migrate through the growing hyphae, causing germinative failure. Arginase activity contained in defence glycoproteins is already described for preventing fungal germination. N

Sensitive methodology for simultaneous determination of residues of imidacloprid and its metabolites in sugarcane leaves and soil.

An analytical method to quantify imidacloprid and its metabolites in sugarcane leaves and soil using HPLC has been developed. The samples were extracted with acetonitrile + water (80 + 20, v/v), soil samples partitioned with dichloromethane, and leaf samples with hexane + ethyl acetate (9 + 1, v/v) and dichloromethane. Further, the extracts were dried, filtered, and concentrated under vacuum into HPLC-grade acetonitrile. Residues were estimated using an HPLC equipped with a photodiode array detector system, C18 column with a mobile phase of acetonitrile + water (40 + 60, v/v) at 0.3 mL/min to

Genome-wide identification of the RLCK gene family in sugarcane and functional analysis of ScRLCK53 in salt tolerance.

Sugarcane (Saccharum spp.) is a major sugar crop, yet its growth and yield are frequently limited by soil salinization. Receptor-like cytoplasmic kinases (RLCKs) act as central signaling hubs in plants, regulating growth, development, immunity, and abiotic stress responses. However, the evolution of the RLCK gene family in sugarcane and its role in salt tolerance remain unclear. Using the high-quality genome of elite cultivar 'Zhongzhe No.1', we identified 262 ScRLCK genes, grouped into 15 subfamilies, all of which contain a typical protein kinase domain. Synteny analysis revealed that whole-g…

Phenotype-genotype association for the identification of resistance to stem borers (Diatraea spp., Lepidoptera: Crambidae) in sugarcane.

Sugarcane (Saccharum spp.) is a major crop for sugar production, but its productivity is affected by stem borers (Diatraea spp.). These insects cause significant losses by damaging the stem and disrupting the transport of photosynthates and nutrients. Although biological control has been effective in Colombia, the identification of genetic resources for the resistance would provide complementary long-term crop protection. This study aimed to identify markers associated with the resistance to insects. For this purpose, a population of 348 genotypes was evaluated by infesting 30 stalks per genot…

Root-associated Bacterial Communities of Sugarcane Cultivated for Traditional Wasanbon Sugar differ among Cultivation Sites in Kagawa, Japan.

Sugarcane for traditional Wasanbon sugar has long been cultivated in Higashi-Kagawa. Three sugarcane cultivars were cultivated at three sites in Kagawa Prefecture, and root-associated bacterial communities were analyzed using 16S rRNA gene amplicon sequencing. Bacterial community compositions varied primarily according to the cultivation site rather than the cultivar, and soil bacterial communities showed similar site-dependent clustering patterns. These results indicate that the cultivation site affects the root-associated bacterial community structure.

Effect of phosphate-solubilizing bacteria on phosphorus dynamics and the bacterial community during composting of sugarcane industry waste.

Sugarcane processing generates a large quantity of residues, such as filter cake and ashes, which are sometimes composted prior to their amendment in soil. However, important issues still have to be addressed on this subject, such as the description of bacterial succession that occurs throughout the composting process and the possibilities of using phosphate-solubilizing bacteria (PSB) during the process to improve phosphorus (P) availability in the compost end product. Consequently, this study evaluated the bacterial diversity and P dynamics during the composting process when inoculated with

Insights of cellular and molecular changes in sugarcane response to oxidative signaling.

Significant changes in the proteome highlight essential metabolic adaptations for development and oxidative signaling induced by the treatment of young sugarcane plants with hydrogen peroxide. These adaptations suggest that hydrogen peroxide acts not only as a stressor but primarily as a signaling molecule, triggering specific metabolic pathways that regulate growth and plant resilience. Sugarcane is a crucial crop for sugar and ethanol production, often influenced by environmental signals. Hydrogen peroxide (H2O2) is increasingly recognized as an important signaling molecule that regulates pl

Nanoparticle-induced enzyme pretreatment method for increased glucose production from lignocellulosic biomass under cold conditions.

BACKGROUND: Appropriate pretreatment strategies that fractionate sugarcane bagasse (SB) are essential for the successful use of this feedstock in ethanol production. In this paper, we investigate a purely enzymatic process to achieve increased production of reducing sugars (RS) from SB in the presence of MgO nanoparticles (MgN) subjected to a three-step sequential enzyme treatment. RESULT: Pretreatment of SB with protease activated by magnesium oxide nanoparticles (MgN-pro) at 95 °C showed an increase in amino acid production by 6.18-fold compared to the untreated enzyme set at the same tempe

[Risk of exposure to heatstroke in sugarcane workers in São Paulo State, Brazil].

Environmental heat is an aggravating factor for sugarcane workers, and manual cane cutting is one of the heavy work activities that favor the generation of metabolic heat. This can combine with other precarious working conditions to produce heatstroke, potentially leading to serious complications and even death. The study aimed to quantify the risk of overheating in sugarcane cutters in São Paulo, Brazil. Drawing on a specific methodology, the study used four years of data to estimate the Globe Thermometer Wet Bulb Index based on light, moderate, and heavy activities. The results showed that t

Combining treatments to improve the fermentation of sugarcane bagasse hydrolysates by ethanologenic Escherichia coli LY180.

Inhibitory side products from dilute acid pretreatment is a major challenge for conversion of lignocellulose into ethanol. Six strategies to detoxify sugarcane hydrolysates were investigated alone, and in combinations (vacuum evaporation of volatiles, high pH treatment with ammonia, laccase, bisulfite, microaeration, and inoculum size). High pH was the most beneficial single treatment, increasing the minimum inhibitory concentration (measured by ethanol production) from 15% (control) to 70% hydrolysate. Combining treatments provided incremental improvements, consistent with different modes of
