Efficacy of scopolamine transdermal patch in children with sialorrhea in a pediatric tertiary care hospital.

BACKGROUND: Drooling is common in children with neurological disorders, but its management is very challenging, Scopolamine transdermal patch (STP) appears to be useful in controlling drooling, although it is not approved for this indication and there are limited clinical studies about its effectiveness. This study aimed (1) to assess the impact of STP use on the severity of drooling and on the frequency of emergency department (ED) and hospital readmission (RA) visits related to drooling, and (2) to determine the level of family satisfaction with STP when used in children with neurological di…

The involvement of monoaminergic neurotransmission in the antidepressant-like action of scopolamine in the tail suspension test.

Some clinical studies indicate that scopolamine may induce a rapid antidepressant effect. Although scopolamine is a muscarinic antagonist, it seems that not only cholinergic but also glutamatergic and GABAergic systems might be involved in the mechanism of its antidepressant activity in animal models of depression. Here, we present a set of behavioral data aimed at investigating the role of monoaminergic system activity in the mechanism of the antidepressant-like action of scopolamine in an animal model based on behavioral despair, namely, the tail suspension test (TST). It was found that AMPT…

Short-term and long-term effects of diazepam on the memory for discrimination and generalization of scopolamine.

Benzodiazepines are among the most widely prescribed and misused psychopharmaceutical drugs. Although they are well-tolerated, they are also capable of producing amnestic effects similar to those observed after pharmacological or organic cholinergic dysfunction. To date, the effect of benzodiazepine diazepam on the memory for discrimination of anticholinergic drugs has not been reported. The aim of the present study was to analyze the immediate and long-term effects of diazepam on a drug discrimination task with scopolamine. Male Wistar rats were trained to discriminate between scopolamine and…

Pretreatment Differences in BOLD Response to Emotional Faces Correlate with Antidepressant Response to Scopolamine.

BACKGROUND: Faster acting antidepressants and biomarkers that predict treatment response are needed to facilitate the development of more effective treatments for patients with major depressive disorders. Here, we evaluate implicitly and explicitly processed emotional faces using neuroimaging to identify potential biomarkers of treatment response to the antimuscarinic, scopolamine. METHODS: Healthy participants (n=15) and unmedicated-depressed major depressive disorder patients (n=16) participated in a double-blind, placebo-controlled crossover infusion study using scopolamine (4 μg/kg). Befo…

Transdermal scopolamine for the prevention of postoperative nausea and vomiting: a systematic review and meta-analysis.

BACKGROUND: Transdermal scopolamine (TDS) is a potential long-acting prophylactic antiemetic initially developed to prevent motion sickness. TDS is a centrally acting anticholinergic agent that was approved in 2001 by the US Food and Drug Administration for the prevention of postoperative nausea and vomiting (PONV). Although TDS has been reported to be clinically efficacious in the prevention of PONV, several adverse events (AEs), such as sedation, dry mouth, blurred vision, central cholinergic syndrome, and confusion (particularly in elderly patients with mild cognitive impairment), are poten…

A non-spatial, stimulus-comparison working memory task in rats and disruption by scopolamine.

Working memory is a theoretical concept referring to a set of cognitive processes that provide temporary maintenance and manipulation of the … dependent on the duration of the retention interval. The muscarinic cholinergic receptor antagonist scopolamine, but not by its quaternary analog N-methyl scopolamine, decreased the percentage of correct responding as well as the discriminability of the stimuli as measured by log d while having no effect on bias as measured by log b. The present findings are …

Effects of repeated treatment with methamphetamine plus scopolamine and methamphetamine on behavioral sensitization and conditioning.

This study compared repeated treatment with methamphetamine (4.0 mg/kg, i.p.) plus scopolamine (0.5 mg/kg, i.p.) and methamphetamine alone in behavioral sensitization and drug conditioning with respect to a reciprocal balance between the dopaminergic and cholinergic systems. Repeated methamphetamine plus scopolamine treatment induced a more progressive and enduring enhancement of stereotyped behavior than repeated methamphetamine treatment. Methamphetamine plus scopolamine-induced stereotyped behavior was reproduced by challenge injections of not only methamphetamine plus scopolamine and metha…

Effects of cholinergic agonists on regional brain energy metabolism in the scopolamine-treated rat.

The effects of scopolamine, physostigmine, RS86 and U-80816B on regional energy metabolism were studied in rodents by means of the 2-deoxyglucose autoradiographic technique. Scopolamine depressed metabolism in an area of cerebral cortex, focused around the parietal region. Rats treated with cholinergic direct agonists (U-80816B, RS86) as well as with the indirect agonist (physostigmine) all showed decreases in cortical energy metabolism, similar to scopolamine. They also induced an increase in thalamic metabolism. When these drugs were given in conjunction with scopolamine, metabolism tended t…

Relationship of the behavioral effects of aprophen, atropine and scopolamine to antagonism of the behavioral effects of physostigmine.

Behavioral effects of aprophen, atropine and scopolamine, in rats, were examined under a multiple schedule of food presentation and at different injection-test times. The effects of the varied treatments were compared to the ability of the drugs, under identical conditions, to prevent the behavioral effects of the anticholinesterase, physostigmine. Potencies of the antagonists to decrease response rates varied across three log units. All three antagonists produced dose-related attenuation of the response suppressant effects of physostigmine. In general, aprophen was a better antagonist than sc…

The effects of scopolamine and methylscopolamine on visual and auditory discriminations in male and female Wistar rats.

The present experiment was designed to investigate whether or not the administration of scopolamine hydrobromide would differentially disrupt auditory or visual discrimination performance in male and female Wistar rats. Two groups of male and female Wistar rats were trained to discriminate between a continuous and intermittent visual stimulus, while two other groups were trained to discriminate between a continuous or intermittent auditory stimulus in a discrete-trial discrimination procedure. Once discrimination performance had stabilized, subjects were treated with different doses (0.125, 0.…

The use of prostaglandin F2 alpha and scopolamine-N-butylbromide in anorectal manometric diagnosis.

To improve the accuracy of diagnosis of Hirschsprung's disease by means of anorectal manometry, prostaglandin F2 alpha (PGF2 alpha) and scopolamine-N-butylbromide (Buscopan) were used. Normal rectoanal reflex was facilitated by PGF2 alpha in 6 out of 7 children and was abolished or markedly inhibited in 12 out of 13, when Buscopan was given. Obscure rectoanal reflexes in 7 normal neonates were clarified following the administration of PGF2 alpha. On the other hand, atypical rectoanal reflexes observed in 7 children with Hirschsprung's disease were unchanged with either PGF2 alpha or Buscopan. …

Scopolamine alone or combined with ephedrine in seasickness: a double-blind, placebo-controlled study.

In seasickness, the central cholinergic-noradrenergic balance is disturbed. Capsules of identical appearance--containing scopolamine hydrobromide 0.3 mg, scopolamine hydrobromide 0.3 mg + ephedrine hydrochloride 25 mg, or placebo--were given prophylactically three times daily to 30 naval cadets during a crossing. In this randomized, double-blind trial the superiority of scopolamine and scopolamine + ephedrine over placebo was confirmed. Ephedrine did not clearly increase the effectiveness of scopolamine. One subject in the scopolamine group and one in the placebo group interrupted the treatmen…

Bavachin Attenuates Cognitive Impairment in Scopolamine-Induced Amnesia: Behavioral, Biochemical and Molecular Insights.

Cholinergic dysfunction is a key feature of cognitive impairment, and currently available acetylcholinesterase (AChE) inhibitors provide limited symptomatic benefit and are often associated with adverse effects. This study evaluated the cognitive-protective effects of bavachin (BVC), a prenylated flavonoid from Psoralea corylifolia, and its interaction with Donepezil (DNP) in a scopolamine (SCP)-induced amnesia model. Male Swiss albino mice were pretreated with BVC (10-40 mg/kg), DNP (2.5 mg/kg), or their combination for 7 days prior to SCP challenge, followed by assessment using multiple beha…

Lactobacillus delbrueckii subsp. lactis CKDB001 Ameliorates Scopolamine-Induced Cognitive Impairment Through Metabolic Modulation.

Microbiome-derived metabolites have emerged as key mediators of the gut-brain axis, influencing cognitive function and neuroprotection. This study investigated whether Lactobacillus delbrueckii subsp. lactis CKDB001 alleviates scopolamine-induced memory impairment through metabolic modulation, and how its effects compare with those of donepezil. ICR mice were administered CKDB001 or donepezil for 4-5 weeks and evaluated through behavioral, microbiome, metabolomic, and molecular analyses. CKDB001 significantly improved spatial working memory in a dose-dependent manner, with the high-dose group …

Design, Synthesis, and Activity Evaluation of Fluorine-Containing Scopolamine Analogues as Potential Antidepressants.

This study aimed to develop novel rapid-acting antidepressants with sustained efficacy and favorable safety profiles. We designed and synthesized a series of fluorine-containing scopolamine analogues and evaluated their antidepressant potential. In vitro cytotoxicity assays showed that most of these compounds exhibited minimal toxicity against neuronal and non-neuronal mammalian cell lines (IC50 > 100 μM). The antidepressant activities of the compounds were evaluated using the tail suspension test, and S-3a was identified as a lead compound with potent and sustained antidepressant effects. Beh…

Rodent models for dry eye syndrome: Standardization using benzalkonium chloride and scopolamine hydrobromide.

Dry eye disease is a highly prevalent ocular condition that significantly affects the quality of life and presents a major challenge in ophthalmology. Animal models play a crucial role in investigating the pathophysiology and developing effective treatments. The goal of this study was to compare and standardize two dry eye disease rodent models and explore their recovery aspects. We have standardized benzalkonium chloride and scopolamine-induced dry eye disease models which represents two different classes of the dry eye i.e., evaporative dry eye and aqueous deficient dry eye, respectively. Af…

Loganin enhances long-term potentiation and recovers scopolamine-induced learning and memory impairments.

Although the incidence rate of dementia is rapidly growing in the aged population, therapeutic and preventive reagents are still suboptimal. Various model systems are used for the development of such reagents in which scopolamine is one of the favorable pharmacological tools widely applied. Loganin is a major iridoid glycoside obtained from Corni fructus (Cornusofficinalis et Zucc) and demonstrated to have anti-inflammatory, anti-tumor and osteoporosis prevention effects. It has also been found to attenuate Aβ-induced inflammatory reactions and ameliorate memory deficits induced by scopolamine…

Antidepressant-like effect of low dose ketamine and scopolamine co-treatment in mice.

Current medications for depression typically require weeks of treatment before significant clinical improvement is observed, and are only effective in a relatively small subset of patients. Recent human clinical studies have demonstrated that ketamine, an NMDA receptor antagonist, and scopolamine, a muscarinic acetylcholine receptor antagonist, produce rapid antidepressant responses within hours of administration, and are effective in treatment-resistant patients. We hypothesize that efficacy and tolerability may be improved by combining lower doses of both drugs in the treatment of depression…
