The purification and characterization of kallikrein-like proteases from rat submandibular glands is described. The proteolytic activity of each fraction during purification was monitored on the synthetic substrate N-alpha-tosyl-L-arginine methyl ester (TAME). The purification scheme involved ammonium sulfate precipitation, chromatography on columns of DEAE-Sepharose and Sephadex G-100 and chromatofocusing. Fibronectin was detected by indirect immunofluorescence on the cell surfaces of HUH6 C15 cells, established from a human hepatoblastoma and maintained with serum-free RPMI 1640 medium. Fibronectin synthesized by HUH6 Cl5 was purified by gelatin-Sepharose affinity chromatography and compared with human plasma fibronectin in respect to molecular weight, electrophoretic mobility and antigenicity. Fibronectin synthesized by this cell line was proved to be identical with human plasma fibronectin.

N-vinylpyrrolidone-N(2-hydroxy) acrylamide copolymer-modified porous glass was studied as a support for gel permeation chromatography. Influenza, Sendai, fowl plague viruses and rota-viruses were purified by chromatography on the support. As compared with Sepharose 4B the support shows better hydrodynamic properties and yields higher amounts of purified viruses. The support can be also applied to the t-RNA and ribosomes separation.

Cell-free extracts were prepared from Pseudomonas pseudomallei cells by freezing-thawing, sonication, and differential ultracentrifugation. The extracts were subjected to column chromatography with DEAE-sepharose to obtain glycoprotein fractions. The fractions showed acid phosphatase activity to p-nitrophenyl phosphate, tyrosine phosphate, serine phosphate, but not to threonine phosphate. They were highly antigenic when tested by immunofluorescence assay with the sera of melioidosis patients.

Calmodulin-binding proteins in fractions purified from human submandibular glands by calmodulin-Sepharose were phosphorylated with [gamma-32P]ATP, in the absence of exogenous protein kinase. The major proteins phosphorylated had molecular masses of 45, 51 and 61 kDa. Phosphorylation was increased by activators of protein kinase C and inhibited by H-7. Phosphorylation of the 61 kDa band was markedly decreased in cystic-fibrosis submandibular glands.

The optimized proline-rich antimicrobial peptide B7-005 was loaded on bone scaffolds based on polysaccharides and hydroxyapatite. A novel protocol to reuse agarose following agarose gel electrophoresis was established in this study. By repeated freeze-and-thaw of the agarose gel, ethidium bromide and other buffer components in the gel were safely removed from the gel without generation of any toxic fume. The agarose recovered using this method can be used for further electrophoretic experiments without any issues.

By using PEG (polyethylen glycol 6000) in the gel immunodiffusion tests (ID), the precipitin lines were increased in 25.5% of the 192 sera reactions and the titers were increased from one to four serial dilutions in 44.6% of the 139 serum samples. Owing to its sensitivity, easy interpretation of the results and low cost, the use of 2% PEG incorporated into the gel in ID tests is recommended for the diagnosis and serological follow-up of paracoccidioidomycosis infections.
