Effects of solar-simulated (UVB plus UVA) radiation on the skin microbiome: An exploratory study.

This exploratory in vivo study investigated the impact of solar-simulated ultraviolet (UV) radiation (UVB plus UVA) on the composition of the human skin microbiome in healthy male volunteers. Thirty Caucasian men were exposed to suberythemal and erythemal doses of UV radiation (0.5, 0.7, and 1.0 minimal erythema dose, MED) on defined areas of the lower back. Skin swabs were collected from both irradiated (n = 243) and nonirradiated control sites (n = 81) 30 min, 24 h, and 96 h postexposure. The microbial profiles were generated using flow cytometry, and the data were analyzed via the open-acce

The skin microbiome as a key regulator of psoriasis: From dysbiotic drivers to host-directed therapeutics.

The skin harbors diverse microbial communities. Compared with gut microbiome dysbiosis, perturbations in the skin microbiome may be more directly implicated in the onset and progression of psoriasis (PSO). More than twenty studies reveal microbial dysbiosis within psoriatic lesions, which is manifested by elevated abundances of pro-inflammatory taxa such as Corynebacterium and Staphylococcus aureus, declined levels of commensal microbes including Cutibacterium, increased total bacterial load and decreased microbial community diversity. Such cutaneous microbial dysbiosis accelerates disease pro

Skin microbiome variation in people living with HIV: associations with antiretroviral therapy and host factors.

INTRODUCTION: The skin microbiome plays a key role in cutaneous immunity and is shaped by host immune status. HIV infection is associated with immune dysfunction and dermatological disease, yet its impact on the skin microbiome and the modifying effect of antiretroviral therapy (ART) remain incompletely defined. This prospective observational study conducted in Indonesia aimed to characterize differences in skin microbiome composition across HIV status and ART exposure and relate these profiles to clinical parameters. METHODS: Skin swabs were obtained from sebaceous (posterior neck) and dry (

Microbiome diversity of low biomass skin sites is captured by metagenomics but not 16S amplicon sequencing.

Established workflows for microbiome analysis work well for high microbial biomass samples, like stool, but often fail to accurately define microbial communities when applied to low microbial biomass samples. Here, we systemically compare microbiome analysis methods -16S rRNA sequencing, shallow metagenomics, and qPCR PMP™ panels-as well as extraction methods across skin swab samples and mock community dilutions. While extraction method minimally impacted results, with no significant signal for method-specific contamination or bias, we observed critical differences in inferred composition acro

The ecology and plasticity of fish skin and gill microbiomes: seeking what matters in health and disease.

The microbiomes of skin and gill mucosal surfaces are critical components in fish health and homeostasis by competitively excluding pathogens, secreting beneficial compounds, and priming the immune system. Disruption of these microbiomes can compromise their capacity for disease resilience and maintaining host homeostasis. However, the extent and nature of microbiome disruption required to impact fish health negatively remains poorly understood. This review examines how various stressors influence the community composition and functionality of fish skin and gill microbiomes, and the subsequent

Alternations in the human skin, gut and vaginal microbiomes in perimenopausal or postmenopausal Vulvar lichen sclerosus.

Vulvar lichen sclerosus (VLS) is a chronic and progressive dermatologic condition that can cause physical dysfunction, disfigurement, and impaired quality of life. However, the etiology of VLS remains unknown. The vulvar skin, intestinal and vaginal microbiomes have been postulated to play important roles in the pathogenesis of this disease. The aim of this study was to compare the compositional characteristics of the vulvar skin, vagina, and gut microbiota between perimenopausal or postmenopausal VLS patients and healthy controls. The study involved six perimenopausal or postmenopausal VLS pa

Evaluating the Impact of Laundry Detergents on the Skin Microbiome of Atopic Dermatitis Patients-A Clinical Study.

BACKGROUND AND AIMS: Remnants of laundry detergent ingredients are suspected to be in constant contact with the skin. Allergy sufferers need assurance that the chemicals in everyday products do not cause such disruption. The purpose of this study was to evaluate the effects and tolerability of common laundry detergents on the skin microbiome in individuals with atopic dermatitis. METHODS: Two laundry detergents for sensitive skin were tested on subjects with atopic dermatitis by having to wear detergent-washed socks for 7 days. The bacterial population from volunteers' skin swabs was determin

Temporal Changes in the Skin Microbiome of Epidermolysis Bullosa Patients following the Application of Wound Dressings.

OBJECTIVE: Epidermolysis bullosa (EB) is a group of rare hereditary skin disorders characterized by the formation of painful blisters, erosions, and ulcers. In addition, the wounds can easily become infected with different pathogens. Therefore, the dynamics in the microbial populations across the various stages of EB can shed light on pathophysiology, the effect of treatment, and the factors involved in its recovery, but they are understudied. We thus sought to characterize the skin microbiome among patients with EB over time. METHODS: A prospective study conducted in the pediatric dermatolog

Skin dysbiosis in the microbiome in atopic dermatitis is site-specific and involves bacteria, fungus and virus.

BACKGROUND: Microbial dysbiosis with increased Staphylococcus aureus (S. aureus) colonization on the skin is a hallmark of atopic dermatitis (AD), however most microbiome studies focus on bacteria in the flexures and the microbial composition at other body sites have not been studied systematically. OBJECTIVES: The aim of the study is to characterize the skin microbiome, including bacteria, fungi and virus, at different body sites in relation to AD, lesional state, and S. aureus colonization, and to test whether the nares could be a reservoir for S. aureus strain colonization. METHODS: Using

Yin and Yang: A disrupted skin microbiome and an aberrant host immune response in hidradenitis suppurativa.

The skin microbiome plays an important role in maintaining skin homeostasis by controlling inflammation, providing immune education and maintaining host defense. However, in many inflammatory skin disorders the skin microbiome is disrupted. This dysbiotic community may contribute to disease initiation or exacerbation through the induction of aberrant immune responses in the absence of infection. Hidradenitis suppurativa (HS) is a complex, multifaceted disease involving the skin, innate and adaptive immunity, microbiota and environmental stimuli. Herein, we discuss the current state of HS skin

Staphylococcus epidermidis protease EcpA can be a deleterious component of the skin microbiome in atopic dermatitis.

BACKGROUND: Staphylococcus aureus and Staphylococcus epidermidis are the most abundant bacteria found on the skin of patients with atopic dermatitis (AD). S aureus is known to exacerbate AD, whereas S epidermidis has been considered a beneficial commensal organism. OBJECTIVE: In this study, we hypothesized that S epidermidis could promote skin damage in AD by the production of a protease that damages the epidermal barrier. METHODS: The protease activity of S epidermidis isolates was compared with that of other staphylococcal species. The capacity of S epidermidis to degrade the barrier and i

Scabies mites alter the skin microbiome and promote growth of opportunistic pathogens in a porcine model.

BACKGROUND: The resident skin microbiota plays an important role in restricting pathogenic bacteria, thereby protecting the host. Scabies mites (Sarcoptes scabiei) are thought to promote bacterial infections by breaching the skin barrier and excreting molecules that inhibit host innate immune responses. Epidemiological studies in humans confirm increased incidence of impetigo, generally caused by Staphylococcus aureus and Streptococcus pyogenes, secondary to the epidermal infestation with the parasitic mite. It is therefore possible that mite infestation could alter the healthy skin microbiota

Urban Exposome and Skin Microbiome in Atopic Dermatitis: A Comparative Study Between Two Brazilian Municipalities.

INTRODUCTION: Atopic dermatitis (AD) is thought to arise from the interplay of skin barrier dysfunction, type 2 immune dysregulation, and cutaneous dysbiosis, the last of these characterized by reduced bacterial diversity and predominance of Staphylococcus aureus. The urban exposome, including air pollution and reduced environmental microbial diversity, may modulate this dysbiosis. We hypothesized that patients with AD residing in a metropolitan area with a higher pollutant burden would show greater skin dysbiosis, reflected by a higher relative abundance of S. aureus, than patients residing i

Regulatory effects of natural polysaccharides on skin wound repair through modulation of immune-microbiome interactions.

Chronic non-healing wounds pose a significant clinical challenge, driven by dysregulation of the "inflammation-immune-microbiome" triad. Traditional "debridement-anti-infection-coverage" approaches fail to break the vicious cycle of dysbiosis and immune dysfunction. Leveraging structural diversity and bioactivity, natural polysaccharides provide a versatile platform for multi-targeted intervention. This review systematically explores the mechanisms through which polysaccharides modulate the wound immune microenvironment, restructure microbial communities, and facilitate barrier repair. This in

Integrated Host Genetics and Skin Microbiome Profiling Suggest an HLA-C-Peptostreptococcus Axis in Psoriasis.

Psoriasis is a chronic immune-mediated disease driven by genetic susceptibility and environmental factors, including microbial exposure. While HLA-C-linked variants represent the strongest genetic risk factors, their relationship with the cutaneous microbiome remains incompletely understood. This study aimed to investigate host-microbiome interactions in psoriasis through integrative multi-omics analysis. Skin microbiome profiling using 16S rRNA sequencing and targeted genotyping of psoriasis-associated single-nucleotide polymorphisms (SNPs) was performed in lesional and non-lesional skin from
