Maresin1 alleviates inflammation and improves extracellular matrix regeneration after intervertebral disc degeneration.

Intervertebral disc degeneration (IVDD) is characterized by excessive inflammation and extracellular matrix (ECM) degradation in the nucleus pulposus (NP), with limited current therapeutic options. Specialized pro-resolving mediators (SPMs) such as Maresin 1 (MaR1) have emerged as key regulators of inflammation resolution and tissue repair. Here, we report that MaR1 and its receptor LGR6 are significantly downregulated in degenerated human NP tissues, suggesting a loss of endogenous resolution capacity. Exogenous MaR1 administration effectively attenuated IL-1β-induced inflammation and ECM deg

Eicosanoid Derivative, Lipoxin A4, Guards Against Testicular Ferroptosis in Rat Model of Type II Diabetes by Regulating Nrf2/SLC7A11/GPX4 Pathway.

Ferroptosis, a type of iron overload-induced cell death, is involved in diabetes-induced testicular dysfunction. Hence, this study was designed to investigate, for the first time, the impact of lipoxin A4 (LXA4) administration on testicular tissue in diabetic rats and explore its probable role in regulating ferroptosis in comparison with the standard ferroptosis inhibitor (ferrostatin-1, Fer-1). Albino rats of Wistar strain were divided into a control group, a type II diabetes mellitus (DM) group, a DM + Fer-1group, and a DM + LXA4 group. Serum levels of iron, insulin, glucose, total cholester

Restoration of epigenetic impairment in the skeletal muscle and chronic inflammation resolution as a therapeutic approach in sarcopenia.

Sarcopenia is an age-associated loss of skeletal muscle mass, strength, and function, accompanied by severe adverse health outcomes, such as falls and fractures, functional decline, high health costs, and mortality. Hence, its prevention and treatment have become increasingly urgent. However, despite the wide prevalence and extensive research on sarcopenia, no FDA-approved disease-modifying drugs exist. This is probably due to a poor understanding of the mechanisms underlying its pathophysiology. Recent evidence demonstrate that sarcopenia development is characterized by two key elements: (i)

Exploiting oxidized lipids and the lipid-binding GPCRs against cardiometabolic diseases.

Lipids govern vital cellular processes and drive physiological changes in response to different pathological or environmental cues. Lipid species can be roughly divided into structural and signalling lipids. The former is essential for membrane composition, while the latter are usually oxidized lipids. These mediators provide beneficial effects against cardiometabolic diseases (CMDs), including fatty-liver diseases, atherosclerosis, thrombosis, obesity, and Type 2 diabetes. For instance, several oxylipins were recently found to improve glucose homeostasis, increase insulin secretion, and inhib

Tocotrienol metabolites redirect lipid mediator production in innate immune cells towards inflammation resolution.

Restoring homeostasis in persistent inflammation requires suppressing the inflammatory response and promoting resolution. Currently, no clinically used small molecules intentionally achieve both effects. Endogenous long-chain vitamin E metabolites (LCMs) address inflammation at multiple sites, including pro-inflammatory leukotriene production and cytokine release, thus limiting inflammation. Among LCMs, we here identified ω-carboxylates and ω-alcohols that suppress leukotriene biosynthesis while enhancing the biosynthesis of prostaglandin E2 and specialized pro-resolving mediators, specificall

Fatty Acid Metabolic Reprogramming in Rheumatoid Arthritis: Integrating Inflammation, Resolution Failure, and Joint Destruction.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent synovitis and progressive structural joint damage. Its pathology involves immune-cell infiltration, pannus formation, cartilage degradation, and bone destruction. Although autoantibodies, inflammatory cytokines, and aberrant immune activation are central to RA pathogenesis, they do not fully explain the persistence of synovitis, defective inflammation resolution, or structural joint damage in some patients. Emerging evidence indicates that fatty acid metabolic reprogramming may actively shape RA-related cellu

Maresin 1 ameliorates myocardial ischaemia‒reperfusion injury by promoting tissue resident macrophage efferocytosis.

AIMS: Myocardial ischaemia‒reperfusion (I/R) injury triggers a robust inflammatory storm cascade that critically compromises reperfusion efficacy following acute myocardial infarction. Enhanced efferocytosis by cardiac resident macrophages (RMs) has therapeutic potential for inflammation resolution. The unsaturated long-chain fatty acid Maresin1 (MaR1) exhibits potent anti-inflammatory properties that is devoid of immunosuppressive effects. However, its therapeutic potential in myocardial I/R injury and regulatory mechanisms in cardiac RMs remains unexplored. METHODS AND RESULTS: A clinical c

Lipid-mediated resolution of inflammation and survival in amyotrophic lateral sclerosis.

Neuroinflammation impacts on the progression of amyotrophic lateral sclerosis (ALS), a fatal neurodegenerative disorder. Specialized pro-resolving mediators trigger the resolution of inflammation. We investigate the specialized pro-resolving mediator blood profile and their receptors' expression in peripheral blood mononuclear cells in relation to survival in ALS. People living with ALS (pwALS) were stratified based on bulbar versus limb onset and on key progression metrics using a latent class model, to separate faster progressing from slower progressing ALS. Specialized pro-resolving mediato

Resolvin D1 requires TLR2-FPR2 crosstalk for inflammation resolution and protection during ocular bacterial infection.

The eye is highly susceptible to inflammation-induced tissue damage; however, the mechanisms that drive inflammation resolution during ocular infections remain unclear. In this study, we utilize a murine model of intraocular bacterial infection (S. aureus-induced endophthalmitis) and lipidomics analysis to uncover a critical role of pro-resolving lipid mediators, particularly resolvin D1 (RvD1), in resolving inflammation and restoring ocular tissue homeostasis and vision. RvD1 protects mouse eyes from severe endophthalmitis by enhancing bacterial clearance, suppressing intraocular inflammation

Circulating Maresin-1 and cartilage remodeling biomarkers in rheumatoid arthritis and osteoarthritis.

Rheumatoid arthritis (RA) and osteoarthritis (OA) are prevalent chronic joint diseases characterized by persistent inflammation, progressive cartilage degeneration, and impaired tissue repair mechanisms. While inflammatory pathways have been extensively studied, increasing attention has been directed toward the failure of inflammation resolution as a key contributor to chronic joint pathology. Specialized pro-resolving mediators (SPMs), including Maresin-1 (MaR1), play an active role in terminating inflammation and promoting tissue homeostasis. However, the relationship between MaR1 and cartil

Resolvin D5 limits inflammatory lung injury and enhances antibacterial host defense in severe pneumococcal pneumonia.

Streptococcus pneumoniae remains a leading cause of lower respiratory tract infections and pneumonia-related mortality worldwide. Although neutrophil recruitment is essential for pneumococcal containment, excessive and dysregulated neutrophilic inflammation can drive severe lung injury and acute respiratory distress syndrome. Specialized pro-resolving mediators, such as Resolvin D5 (7S,17S-dihydroxy-4Z,8E,10Z,13Z,15E,19Z-docosahexaenoic acid; RvD5), are produced in the lungs and have the potential to promote pathogen clearance and limit inflammatory injury. Neuroinflammation in hypertension: the renin-angiotensin system versus pro-resolution pathways.

Overstimulation of the pro-inflammatory pathways within brain areas responsible for sympathetic outflow is well evidenced as a primary contributing factor to the establishment and maintenance of neurogenic hypertension. However, the precise mechanisms and stimuli responsible for promoting a pro-inflammatory state are not fully elucidated. Recent work has unveiled novel compounds derived from omega-3 polyunsaturated fatty acids (ω-3 PUFAs), termed specialized pro-resolving mediators (SPMs), which actively regulate the resolution of inflammation. Failure or dysregulation of the resolution proces

Allosteric Activation of 15-Lipoxygenase-1 by Boswellic Acid Induces the Lipid Mediator Class Switch to Promote Resolution of Inflammation.

Specialized pro-resolving mediators (SPM), primarily produced in innate immune cells, exert crucial bioactions for resolving inflammation. Among various lipoxygenases (LOX), 15-LOX-1 is key for SPM biosynthesis, but cellular activation principles of 15-LOX-1 are unexplored. It was shown that 3-O-acetyl-11-keto-β-boswellic acid (AKBA) shifts 5-LOX regiospecificity from 5- to 12-lipoxygenation products. Here, it is demonstrated that AKBA additionally activates cellular 15-LOX-1 via an allosteric site accomplishing robust SPM formation in innate immune cells, particularly in M2 macrophages. Compa

Potent Anti-Inflammatory and Pro-Resolving Effects of Anabasum in a Human Model of Self-Resolving Acute Inflammation.

Anabasum is a synthetic analog of Δ8 -tetrahydrocannabinol (THC)-11-oic acid that in preclinical models of experimental inflammation exerts potent anti-inflammatory actions with minimal central nervous system (CNS) cannabimimetic activity. Here we used a novel model of acute inflammation driven by i.d. UV-killed E. coli in healthy humans and found that anabasum (5 mg) exerted a potent anti-inflammatory effect equivalent to that of prednisolone in terms of inhibiting neutrophil infiltration, the hallmark of acute inflammation. These effects arose from the inhibition of the neutrophil chemoattra

The Involvement of Resolvins in Pathological Mechanisms of Periodontal Disease Associated with Type 2 Diabetes: A Narrative Review.

Resolvins are specialized pro-resolving mediators (SPMs) derived from omega-3 fatty acids that play a critical role in resolving inflammation and restoring tissues to a state of health after an immune response. Their role in chronic inflammatory conditions highlights their importance in maintaining a balance between an effective immune response and the resolution of inflammation to prevent tissue damage. Periodontal disease is a chronic inflammatory condition affecting the tissues surrounding the teeth, leading to gum damage and bone loss. Chronic inflammation in periodontal disease can exacer

Beneficial Modulation of Lipid Mediator Biosynthesis in Innate Immune Cells by Antirheumatic Tripterygium wilfordii Glycosides.

Tripterygium wilfordii glycosides (TWG) is a traditional Chinese medicine with effectiveness against rheumatoid arthritis (RA), supported by numerous clinical trials. Lipid mediators (LM) are biomolecules produced from polyunsaturated fatty acids mainly by cyclooxygenases (COX) and lipoxygenases (LOX) in complex networks which regulate inflammation and immune responses and are strongly linked to RA. The mechanism by which TWG affects LM networks in RA treatment remains elusive. Employing LM metabololipidomics using ultra-performance liquid chromatography-tandem mass spectrometry revealed strik

Neuroimmune communication in infection and pain: Friends or foes?

Clinically, a variety of micro-organisms cause painful infections. Before seen as bystanders in the context of infections, recent studies have demonstrated that, as immune cells, nociceptors can sense pathogen-derived products. Nociceptors and immune cells, therefore, have evolved to communicate with each other to control inflammatory and host responses against pathogens in a complementary way. This interaction is named as neuroimmune communication (or axon-axon immune reflex) and initiates after the release of neuropeptides, such as CGRP and VIP by neurons. By this neurogenic response, nocice

Eicosanoids and Inflammation: A delicate balance of Pro-Inflammatory and Pro-Resolving mediators.

Inflammation represents a fundamental biological process essential for host defense and tissue integrity. However, its dysregulation, frequently characterized by a failure of active resolution, underpins a vast array of chronic pathologies. Central to this intricate process is the superfamily of eicosanoids, which are potent bioactive lipid mediators derived from 20-carbon polyunsaturated fatty acids (PUFAs). Historically, specific eicosanoids, such as prostaglandins (PGs) and leukotrienes (LTs), were regarded primarily as pro-inflammatory agents purported to be responsible for mediating vasod

Resolution Failure in Multiple Sclerosis: Linking Lipid Mediators to Neuroinflammation and Disease Progression.

Multiple sclerosis (MS) is a chronic immune-mediated and neurodegenerative disorder of the central nervous system characterized by highly heterogeneous clinical courses and progressive accumulation of disability. Traditionally viewed as a relapsing inflammatory disease driven by peripheral autoimmunity, emerging evidence instead supports a continuum in which early immune attacks evolve into compartmentalized, chronic "smoldering" pathology that persists independently of overt relapses. This chapter integrates current knowledge of MS pathogenesis, focusing on the central emerging concept that M

Forging resilient warriors within: exercise's epic role in training innate immunity and taming inflammation's storm.

The concept of "trained immunity," which refers to a type of long-term immunological memory within innate cells, has significantly challenged the traditional division between the non-specific innate and antigen-specific adaptive immune systems. Simultaneously, it is now understood that the resolution of inflammation is not just a passive return to homeostasis but rather an active, strictly regulated biological process. This comprehensive review synthesizes these evolving concepts by hypothesizing that exercise, through its pleiotropic effects on cellular metabolism, may strongly stimulate both
