Berberine chloride hydrate impairs Streptococcus mutans biofilm formation via inhibiting sortase A activity.

Dental caries is a biofilm-associated chronic progressive disease, results from the dissolution of mineralized tooth tissue by acidic generated from bacterial sugar fermentation. S. mutans, a prominent pathogen of dental caries, is acknowledged for its role in cariogenic biofilm formation, utilizing Sortase A (SrtA) to catalyse surface proteins, thus promoting biofilm formation. In our previous studies, the inhibitory effect of the berberine chloride hydrate (BH) on S. mutans biofilms was confirmed. Here, we further investigate the influence of BH on S. mutans biofilm-induced bovine enamel car

Anandamide Alters Glycolytic Activity in Streptococcus mutans: Metabolomics and Stable Isotope Labeling Study.

Streptococcus mutans (S. mutans) is a cariogenic bacterium in the oral cavity that plays a significant role in plaque formation and dental caries. In previous research by our group, we showed that the endocannabinoid anandamide (AEA) has anti-bacterial and anti-biofilm activities against S. mutans. Here, we aimed to investigate its effects on S. mutans through metabolomics analyses. S. mutans was cultivated in the absence or presence of AEA at a sub-minimum inhibitory concentration (MIC), and changes in metabolites and metabolic pathways were assessed through liquid chromatography-mass spectro…

Polyketides/nonribosomal peptides from Streptococcus mutans and their ecological roles in dental biofilm.

Streptococcus mutans is the major etiological agent of dental caries in humans. S. mutans overgrowth within dental biofilms can trigger biofilm dysbiosis, ultimately leading to the initiation or progression of dental caries. Polyketides and nonribosomal peptides (PKs/NRPs) are secondary metabolites with complex structures encoded by a cluster of biosynthetic genes. Although not essential for microbial growth, PKs/NRPs play important roles in physiological regulation. Three main classes of hybrid PKs/NRPs in S. mutans have been identified, including mutanobactin, mutanocyclin, and mutanofactin,…

Targeted elimination of cariogenic Streptococcus mutans biofilms via Cu,Fe-doped chitosan nanozyme.

Human dental caries is an intractable biofilm-associated disease caused by the symbiotic cariogenic bacteria, but how to target effectively eliminate cariogenic bacterial and their biofilms without affecting normal bacteria still remains great challenges. To address this issue, we reported Cu,Fe-doped chitosan-based nanozyme (i.e. CS@Cu,Fe) that exhibits well peroxidase-like activity at acidic environment of caries, and kill S. mutans and S. sanguinis without impacting the normal S. oralis. The synergistic interaction between Cu and Fe could effectively enhance the efficiency of electron trans

Investigation of Components in Roasted Green Tea That Inhibit Streptococcus mutans Biofilm Formation.

Streptococcus mutans form oral biofilms (BFs) and cause dental caries. Roasted green tea (RGT) is prepared by roasting the tea plant, and RGT-specific polyphenols are produced during the roasting process. Catechins, polyphenols in green tea, have BF inhibitory activity against S. mutans; therefore, RGT-specific polyphenols are also expected to have this activity. However, there are few reports on the structural and functional properties of RGT. This study aimed to investigate the inhibitory activity of RGT against S. mutans BF formation and to investigate the active compounds. RGT extract frac…

Deciphering the mechanism of anti-quorum sensing post-biotic mediators against Streptococcus mutans.

OBJECTIVE: Glucosyltransferases (Gtfs) and quorum sensing (QS) mediated transduction genes play critical roles in the pathogenesis of Streptococcus mutan-mediated dental caries. Therefore, targeting gtfs and QS-mediated virulence genes have therefore emerged as an intriguing goal for efficient therapeutic approaches that block cariogenic biofilms. METHODS: Post-biotic mediators (PMs) obtained from our previously isolated and characterized beneficial bacteria Enterobacter colacae PS-74 was assessed for its antibiofilm potential against S. mutans. According to the transcriptome method, qRT-PCR

Oxidative stress response: a critical factor affecting the ecological competitiveness of Streptococcus mutans.

Oral microecological balance is closely associated with the development of dental caries. Oxidative stress is one of the important factors regulating the composition and structure of the oral microbial community. Streptococcus mutans is linked to the occurrence and development of dental caries. The ability of S. mutans to withstand oxidative stress affects its survival competitiveness in biofilms. The oxidative stress regulatory mechanisms of S. mutans include synthesis of reductase, regulation of metal ions uptake, regulator PerR, transcription regulator Spx, extracellular uptake of glutathio…

Oxidative stress response plays a role in antibiotic tolerance of Streptococcus mutans biofilms.

Knowledge about biofilm-associated antibiotic tolerance mechanisms is warranted in order to develop effective treatments against biofilm infections. We performed a screen of a Streptococcus mutans transposon mutant library for mutants with reduced biofilm-associated antimicrobial tolerance, and found that the spxA1 gene plays a role in tolerance towards gentamicin and other antibiotics such as vancomycin and linezolid. SpxA1 is a regulator of genes involved in the oxidative stress response in S. mutans. The oxidative stress response genes gor and ahpC were found to be up-regulated upon antibio…

Association of Streptococcus mutans infection and oral developmental nodules in pre-dentate infants.

Since dental caries may present soon after tooth eruption, we hypothesized that colonization of Streptococcus mutans can occur in the predentate stages. In this study, we examined S. mutans colonization and its association with oral developmental nodules (Bohn's nodules) in 60 pre-term and 128 full-term, three-month-old infants. Overall, S. mutans was cultured from 30% (56/188) of the infants, and oral developmental nodules were noted in 55% (103/188). Compared with the pre-term, full-term infants showed a higher prevalence of S. mutans (34% vs. 20%, p < 0.02) as well as developmental nodules …

Impact of Modified Lactoperoxidase Systems on Glycolytic Metabolism and Virulence Factors in Streptococcus mutans.

The lactoperoxidase system (LpoS) is an enzymatic antimicrobial mechanism of saliva that oxidizes (pseudo)halide substrates to reactive compounds capable of limiting microbial growth. This study evaluated how different LpoS variants-utilizing iodide (LpoS-I-), thiocyanate (LpoS-SCN-), selenocyanate (LpoS-SeCN-), and a thiocyanate-iodide mixture (LpoS-SCN- + I-)-affect virulence, metabolism, and biofilm structure in Streptococcus mutans. Using qRT-PCR, pyruvate assays, MTT reduction, and confocal microscopy, we found that LpoS-I- most effectively reduced atpD and ldh expression, impaired acid t…

Characterization of the Nicotinamide Adenine Dinucleotide Biosynthesis Pathway and Regulatory Mechanisms in Streptococcus mutans.

NAD+ and its derivatives, which act as redox coenzymes, are crucial for cellular metabolism and energy production. Nevertheless, the processes by which Streptococcus mutans, a bacterium known for causing dental caries, synthesizes NAD+ are not well elucidated. Through a genome-wide screen, we identified the nicotinic acid salvage pathway and the evolutionarily incomplete PnuC-NadR pathway involved in NAD+ biosynthesis in S. mutans UA159. The nicotinic acid pathway is regulated by SmNiaR, a nicotinic acid-responsive transcription regulator featuring an N-terminal DNA-binding winged helix-turn-h…

Methionine biosynthesis as a key metabolic pathway for antimicrobial drug discovery in Streptococcus mutans.

Streptococcus mutans, a Gram-positive, facultative anaerobic bacterium, is a key contributor to dental caries. It forms biofilms to colonize the tooth surface, has stress resistance mechanisms to survive the fluctuating oral environment, and produces virulence factors, all of which cause enamel demineralization, acid production, and caries development. Traditional antimicrobial strategies target central metabolic pathways in S. mutans, but most enzymes are conserved between humans and bacteria. However, one prospective approach is to target methionine biosynthesis. This pathway is essential fo…

FRUCTOSE ACTIVATES A STRESS RESPONSE SHARED BY METHYLGLYOXAL AND HYDROGEN PEROXIDE IN STREPTOCOCCUS MUTANS.

Fructose catabolism by Streptococcus mutans is initiated by three PTS transporters yielding either fructose-1-phoshate (F-1-P) or fructose-6-phosphate (F-6-P). Deletion of one such F-1-P-generating PTS, fruI, has been shown to reduce the cariogenicity of S. mutans in rats fed a high-sucrose diet. Moreover, a recent study linked fructose metabolism in S. mutans to a reactive electrophile species (RES) methylglyoxal. Here, we conducted a comparative transcriptomic analysis of exponentially grown S. mutans shocked with 50 mM fructose, 50 mM glucose, 5 mM methylglyoxal, or 0.5 mM hydrogen peroxide…
