NMDA and glycine receptors provide dual presynaptic regulation on climbing fibre inputs to cerebellar Purkinje cells.

Presynaptic receptors are important regulators of CNS activity. We have examined presynaptic receptor populations using a vibrodissociated r … when applying a high concentration of glycine (100 µM) alone, a response that was blocked by 500 nM strychnine, but not by 1 mM Mg2+. Both glycine and NMDA stimulation produced many large amplitude excitatory postsynaptic currents of ∼200-600 pA. The size of these events suggests that they arise from climbing fibre, rather than parallel fibre, connections. I …

Schisandrin B, a dual positive allosteric modulator of GABAA and glycine receptors, alleviates seizures in multiple mouse models.

Epilepsy is a prevalent and severe neurological disorder and approximately 30% of patients are resistant to existing medications. It is of utmost importance to develop alternative therapies to treat epilepsy. Schisandrin B (SchB) is a major bioactive constituent of Schisandra chinensis (Turcz.) Baill and has multiple neuroprotective effects, sedative and hypnotic activities. In this study, we investigated the antiseizure effect of SchB in various mouse models of seizure and explored the underlying mechanisms. Pentylenetetrazole (PTZ), strychnine (STR), and pilocarpine-induced mouse seizure mod…

Types of pesticides involved in domestic and wild animal poisoning in Italy.

Data obtained from samples of poisoned domestic and wild animals sent for toxicological evaluation during the period between 2005 and 2014 h … Even if some pesticides are frequently involved in domestic animal poisoning (i.e. metaldehyde and strychnine), they did not show the same diffusion in wild animals. In particular, unlike domestic species, cyanide and pyrethroids have not been found responsible of intoxications in wild animals. Interestingly, a great number of positive samples involved bann …

The central nervous effects of Mitragyna africanus (Willd) stembark extract in rats.

The acute toxicity and the central effects of Mitragyna africanus (M. africanus) stembark methanol extract were studied in rats. The extract did not produce any death in the treated rats even at the highest dose (6400 mg kg(-1)) used. It produced depressant effects on the central nervous system. The stembark extract potentiated amylobarbitone sleeping time in rats dose-dependently, induced sleep in rats and also produced significant local anaesthetic effect on rabbits, the effects being comparable to that of xylocaine. The extract protected rats treated with a convulsive dose of strychnine (2 …

d-cycloserine adjuvant therapy to conventional neuroleptic treatment in schizophrenia: an open-label study.

D-cycloserine, a partial agonist at the strychnine-insensitive glycine site of the NMDA receptor complex, was tested as adjuvant treatment to conventional neuroleptics in chronic schizophrenic volunteers. The drug was administered, o.a.d., at the daily dose of 250 mg for six weeks. Mental status outcome measures were completed at the end of each week of treatment. The major finding was a deterioration of the patients' clinical condition, specifically of their psychotic symptoms. These preliminary results are discussed among others in view of d-cycloserine pharmacologic properties and recent fi…

Action of vinblastine on myenteric neurones of guinea-pig ileum.

Motor activity of guinea-pig ileum in vitro was monitored by recording isometric tension. Superfusion of the serosal surface with vinblastine (22 microM) increased the motility of ileum but failed to do so in the presence of local anaesthetics and also in preparations depolarised with K+-Tyrode. Atropine (6 microM) or pentolinium (186 microM) reduced the motor response of vinblastine but higher concentration of atropine (60 microM) or pentolinium (930 microM) did not further attenuate the response. Strychnine (58 microM) reversibly blocked the atropine-resistant motor response elicited by vinb…

[Mechanism of formation of epileptic activity complexes in the cerebral cortex under the influence of a determinant focus].

It was shown in the experiments on cats anesthetized by nembutal that by forming a hyperactive focus with a high level of excitation in the temporal cortex and a number of foci with a less higher level of the excitation in other parts of neocortex, it is possible to create a functional complex, working in a single routine, determined by the activity of hyperactive focus. The latter plays a role of a determinant structure. The inhibition of the determinant focus results in the disturbance of the epileptic complex. Focus nature has no sufficient importance in the realization of the following int

The Anticonvulsant Effect of Hydroethanolic Leaf Extract of Calotropis procera (Ait) R. Br. (Apocynaceae).

A number of currently used drugs have been obtained from medicinal plants which are a major source of drugs. These drugs are either used in their pure form or modified to a semisynthetic drug. Drug discovery through natural product research has been fruitful over the years. Traditionally, Calotropis procera is used extensively in the management of epilepsy. This study is conducted to explore the anticonvulsant effect of a hydroethanolic leaf extract of Calotropis procera (CPE) in murine models. This effect was evaluated using picrotoxin-induced convulsions, strychnine-induced convulsions, and …

Resolution and chiral recognition of muscone as well as actions on neural system.

In this letter, (R)-muscone and (S)-muscone were prepared by optical resolution of dl-muscone using N,N'-dibenzyl-l-tartaramide or N,N'-dibenzyl-d-tartaramide, according to the method reported by Kunihiko Takabe. But we separated the diastereomers using EtOH instead of MeOH in the recrystallization step to get the goal product with higher optical purity and yield. The regulatory effect of muscone and its enantiomer on the neural system showed that they could prolong mouse hypoxia tolerance and dose-dependently enhance mouse spinal cord stimulation induced by strychnine nitrate. (R)-muscone and…

The death of Alexander the Great: malaria or typhoid fever?

Alexander the Great had a profound effect on world history. His conquests covered the entire known world at the time, and he was responsible for the spread of Greek culture throughout the ancient world. In Babylon in 323 BC, Alexander died when he was nearly 33 years old. Possible explanations for his death have included alcoholic liver disease and strychnine poisoning, but little data support either condition as the cause of his death. Alexander most likely died from malaria or typhoid fever, which were rampant in ancient Babylon. The description of his final illness from the royal diaries is…
