Topographic axonal projection at single-cell precision supports local retinotopy in the mouse superior colliculus.

Retinotopy, like all long-range projections, can arise from the axons themselves or their targets. The underlying connectivity pattern, however, remains elusive at the fine scale in the mammalian brain. To address this question, we functionally mapped the spatial organization of the input axons and target neurons in the female mouse retinocollicular pathway at single-cell resolution using in vivo two-photon calcium imaging. We found a near-perfect retinotopic tiling of retinal ganglion cell axon terminals, with an average error below 30 μm or 2° of visual angle. The precision of retinotopy was

Beyond the labeled line: variation in visual reference frames from intraparietal cortex to frontal eye fields and the superior colliculus.

We accurately perceive the visual scene despite moving our eyes ~3 times per second, an ability that requires incorporation of eye position and retinal information. In this study, we assessed how this neural computation unfolds across three interconnected structures: frontal eye fields (FEF), intraparietal cortex (LIP/MIP), and the superior colliculus (SC). Single-unit activity was assessed in head-restrained monkeys performing visually guided saccades from different initial fixations. As previously shown, the receptive fields of most LIP/MIP neurons shifted to novel positions on the retina fo

Retinal projection to the pretectal nucleus lentiformis mesencephali in pigeons (Columba livia).

In birds, the nucleus of the basal optic root (nBOR) and the nucleus lentiformis mesencephali (LM) are retinal-recipient nuclei involved in the analysis of optic flow and the generation of the optokinetic response. The nBOR receives retinal input from displaced ganglion cells (DGCs), which are found at the margin of the inner nuclear and inner plexiform layers, rather than the ganglion cell layer. The LM receives afferents from retinal ganglion cells, but whether DGCs also project to LM remains unclear. To resolve this issue, we made small injections of retrograde tracer into LM and examined h

Irrepressible saccades from a tectal lesion in a Rhesus monkey.

We present a case of spontaneously occurring irrepressible saccades in an experimental Rhesus monkey. Though eye jerks are sometimes associated with cerebellar disease, central demyelination or brainstem lesions, there is little consensus on their neurological mechanisms. From neurological and anatomical investigation we report that these irrepressible saccades were caused by a discrete cerebrovascular accident that involved the rostral superior colliculus along with its commissure, and with minor invasion of periaqueductal gray and adjacent mesencephalic reticular formation. Other suspected s

Visual thresholds in albino and pigmented rats.

Albino rats have recently been reported to have increment thresholds against dim backgrounds that are two log units higher than those of pigmented rats. We, on the other hand, have failed to confirm these differences using electroretinogram b-waves and pupillary light reflexes. This paper reports on experiments using evoked potentials from cortex and colliculus and single-unit recordings from colliculus. We recorded visual-evoked potentials from cortex and superior colliculus in the strains of albino (CD) and pigmented (Long-Evans) rats used in the earlier studies. Thresholds were determined o

[Interactions of the superior colliculus and the sensorimotor cortex in the alert rabbit].

The tonic increased influence of the superior colliculus (SC) on the formation of visual responses of the sensorimotor cortex (SMC) was shown on the alert rabbits. It was shown that SC influence was realized through tectothalamocortical (nucleus lateralis posterior) canal relation. It was established that SMC of the alert rabbits in its turn exerts the inhibitory phasic influence on SC function. The obtained data are discussed in the light of participation of the colliculo-cortical and cortico-collicular interactions in the organization of the visual-motor coordination necessary for realizatio

Morphometric parameters of the superior colliculus of albino and pigmented rats.

The superior colliculus (SC) or optic tectum of mammals consists of seven layers, numbered I-VII from superficial to deep, each of which has distinct connectivity patterns and electrophysiological response properties. The present study is devoted to a morphometrical analysis of neuronal diameters, densities, and numbers in different layers and regions of the SC of albino as well as pigmented rats in order to present a quantitative characterization of the collicular neuronal population involved in the different connectivities and functions of these compartments. The morphometric parameters were

Organization of the visual pathways in the newborn kitten.

We have used retrograde and anterograde transport to examine the major visual pathways in newborn kittens. Retinal projections from both eyes to the dorsal lateral geniculate nucleus (dLGN) and superior colliculus (SC) are present and topographically organized. The dLGN projects topographically to areas 17 and 18 and receives reciprocal projections from cells in layer VI of areas 17, 18, 19 and suprasylvian cortex on the same side of the brain. Area 19 also has a sparse thalamic input but probably not from the dLGN. The laminar distribution of [3H]proline transported from dLGN to area 17 was q

The isthmus-tegmentum complex in the turtle and rat: a comparative analysis of its interconnections with the optic tectum.

Injections of horseradish peroxidase, wheat germ agglutinin and various amino acids into the optic tectum in both the turtle and rat, and into the nucleus isthmi magnocellularis (IsM) in the turtle were used to analyse the connections of the isthmus-tegmentum complex. The connectivities and the selective retrograde transport properties in certain tectal pathways were taken as a basis to define more accurately isthmus-tegmentum complex subdivisions. There were several main findings. In the turtle the projection from the tectum to the IsM originated in the stratum griseum periventriculare, where

Paths of axons in the visual system of perciform fish and implications of these paths for rules governing axonal growth.

The optic nerve of many perciform fish is ribbon-shaped, and axons from ganglion cells in specific parts of the retina are consistently found in specific places in this ribbon. I utilized this organization to fill selected groups of axons with horseradish peroxidase. I then traced these groups of axons through the nerve and across the tectum to their terminal arbors. The paths of the axons suggest that axons use a number of different mechanisms to guide them to their correct terminal sites. At some points they appear simply to grow along the surface created by earlier axons, but at other point

Plasticity in the developing visual system: the effects of retinal lesions made in young rats.

The central visual pathways of the rat have been used as a model for investigating the significance of axonal interactions in mammalian neural development. Attention is restricted largely to the aberrant distribution of optic axons to the ipsilateral side of the brain and their distribution in the superior colliculus after early unilateral eye damage. The normal ipsilateral retinotectal pathway in pigmented rats appears as a series of patches located anteriorly and laterally in the stratum opticum, whereas in albino animals it is a small area lying anteromedially. In both, a few axons are ofte

Hyperoxemic retinal neuronal necrosis in the premature neonate.

A cytologically distinctive type of acute hyperoxemic injury of retinal neurons occurs in premature neonates. Ganglion cells in the central well-vascularized neonate retina are susceptible to excessive oxygen and this is expressed morphologically by karyorrhexis of their nuclei. We observed retinal neuronal necrosis in neonates who had hyperoxemia of greater than 150 torr for two hours or longer in the first week of life. Neuronal necrosis was strikingly associated with immaturity as determined by gestational age and birth weight: of 30 involved neonates, all were below 2,000 g; of 47 autopsie

Clonidine in the central nervous system: site and mechanism of hypotensive action.

The data presented in this paper must be considered in conjunction with the fact that clonidine is both a partial alpha-adrenergic agonist and a partial alpha-adrenergic antagonist. In the anterior hypothalamus, clonidine, acting as an alpha-agonist, excites a pathway that inhibits excitatory cardiovascular neurons. Thus, the effect of neurons from the nucleus tractus solitarii (NTS) in inhibiting sympathetic outflow from the vasomotor center is effectively increased. In the posterior hypothalmus, clonidine, acting as an alpha-antagonist, decreases excitation of excitatory cardiovascular neuro

Physiological deficits in the visual system of mice infected with Semliki Forest virus and their correlation with those seen in patients with demyelinating disease.

Electrophysiological recordings were made from the superior colliculus of mice infected with Semliki Forest virus (SFV) which produced patchy demyelination in many different locations within the central nervous system including the optic nerve. Over a wide range of light intensities, the visual latencies of both single cell and field potential responses from the superior colliculi of infected mice were longer than those of the corresponding control mice, and there was a greater interocular difference in visual latency in the infected mice than in control mice. In addition, the collicular neuro…

Ultrastructural organisation of the projection from the superior colliculus to the ventral lateral geniculate nucleus of the rat.

Retinorecipient regions of the ventral lateral geniculate nucleus of the thalamus and the superior colliculus of the midbrain are linked by reciprocal axonal projections. In this study we have investigated the ultrastructural characteristics, the distribution, and the postsynaptic targets of the terminals of axons projecting to the ventral lateral geniculate nucleus from the superior colliculus. Horseradish peroxidase was injected into the superior colliculi of adult albino rats, and the Hanker-Yates method was used to visualize anterogradely and retrogradely transported peroxidase in the vent…

Microsaccadic eye movement generation is sufficient to enhance peripheral visual sensitivity.

Microsaccades have been convincingly linked to extrafoveal covert attention shifts for more than two decades. However, the direction of causality between individual microsaccade generation and an alteration in both extrafoveal visual sensitivity and behavior remains debated: do microsaccades merely reflect, perhaps probabilistically, an altered extrafoveal sensitivity, or is the act of generating microsaccades sufficient, on its own, to modify such sensitivity? Using a novel exploitation of real-time retinal image stabilization, behavior, and neurophysiology in the superior colliculus of rhesu

Human superior colliculus pathways represent the form and motion of looming objects.

The superior colliculus is a conserved midbrain structure critical for detecting and responding to threats, yet its role in human cognition and emotion remains poorly understood. Using task-based fMRI and computational modeling in two experiments, we investigated how the superior colliculus responds to looming threats. We found that the human superior colliculus encodes information about the motion and category of looming objects present in naturalistic videos and controlled experimental stimuli. Encoding models based on task-optimized neural networks for collision detection, object recognitio
