A comparative study of the immunohistochemical localization of a presumptive proctolin-like peptide, thyrotropin-releasing hormone and 5-hydroxytryptamine in the rat central nervous system.

A proctolin (PROC)-like peptide was studied immunohistochemically in the hypothalamus, lower brainstem and spinal cord of the rat using an antiserum against PROC conjugated to thyroglobulin. Neuronal cell bodies containing PROC-like immunoreactivity (PROC-LI) were observed in the dorsomedial, paraventricular and supraoptic nuclei of the hypothalamus and in the nucleus raphe magnus, nucleus raphe pallidus, nucleus raphe obscurus and nucleus interfascicularis nervi hypoglossi in the medulla oblongata. Fibers containing PROC-LI were seen in the median eminence and in other hypothalamic nuclei, an

Prostaglandin signaling drives peripheral inflammation-induced reduction of hypothalamic oxytocin-positive neurons.

Oxytocin (OXT) is a pleiotropic neuropeptide with diverse physiological functions, including anti-inflammatory effects. Endogenous OXT, primarily produced by neurons in hypothalamic paraventricular nucleus (PVN) and supraoptic nucleus (SON), is known to be reduced in various pathological states, particularly during peripheral inflammatory infections. However, the mechanisms by which peripheral inflammation leads to reduced OXT signaling remain poorly understood. In a mouse model of lipopolysaccharide (LPS)-induced chronic inflammation, we observed a selective reduction in the number of magnoce…

Correlation between cellular changes in supraoptic and paraventricular nuclei and water-drinking in neuroleptic-treated rats.

The neuroleptics haloperidol and thioridazine were injected intraperitoneally in rats for 81 days. During this time the rats' water consumption was measured. Drug-treated animals drank significantly less water for the first 40 days of the experiment only. On day 82 the rats were perfused for electron microscopy. The rat's paraventricular and supraoptic nuclei were embedded, sectioned, stained for electron microscopy and viewed for morphological change. There were significantly more darkly staining neurons in the haloperidol-treated rats than in control rats. Thioridazine-treated rats had more

Compensatory mechanisms underlying arginine vasopressin regulation in transient polyuria during pregnancy.

Transient polyuria during pregnancy is reportedly caused by increased arginine vasopressin (AVP) degradation due to vasopressinase produced … asopressinase was shown to be present in the placentas of pregnant mice. The Avp hnRNA level in the supraoptic nucleus, which is indicative of AVP transcriptional activity, was upregulated in wild-type and FNDI mice during late pregnancy. FNDI mice, but not wild-type mice, had a significant increase in urine volume and a decrease in urine osmolality during pregnancy …

Triphasic AVP secretion in encephalopathy.

A patient with encephalopathy developed triphasic changes in the clinical course, starting with diabetes insipidus (DI), then the syndrome of inappropriate ADH secretion (SIADH), and followed by the final phase of DI. The clinical course of encephalopathy was very rapid. The patient lost consciousness completely within only one day after the onset. During the early phase, he lapsed into a condition of "brain death". We could not identify the etiology of the encephalopathy. The triphasic change referred to above is similar to previous reports of cats model after stereotactic destruction of the

Immunocytochemical identification of the mesotocin- and vasotocin-producing systems in the brain of temperate and desert lizard species and their modifications by cold exposure.

Using immunofluorescent techniques, mesotocin (MT) and vasotocin (VT) neurosecretory systems were identified in the brain of different lizards: one temperate species living in southern France and three desert species coming from southern Algeria. In the four species, MT and VT were shown to be synthesized in specific neurons located in the anterior preoptic area (POA), the supraoptic (SON), paraventricular (PVN), and ventromedial (VMN) nuclei. The neurosecretory axons of the POA neurons terminated in the vicinity of the lamina terminalis; in the three desert species, several additional VT fibe

[Electron-microscopic changes in neurosecretory nuclei of the rat hypothalamus after total ischemia in short-term clinical death].

Hypothalamic neurosecretory nuclei of rats in which short-term clinical death was experimentally induced, were examined electron-microscopically. Material for studies was taken two weeks after resuscitation of animals following 5 min cardiac and respiratory arrest. Application of aldehyde fixatives with Alcian blue as well as fixatives containing tannic acid revealed numerous additional ultrastructural features not visualized in routine electron microscopy. The obtained results are indicative that short-term cerebral ischemia in rats results in ultrastructural abnormalities in neurosecretory h

[Increased vasopressin production in senescence and dementia].

Vasopressin (VP) is involved as a neurotransmitter in a number of central functions that are frequently disturbed during aging and dementia. Therefore, this peptide has been used in clinical trials as a 'substitution therapy' for the degenerating peptidergic neurons, aimed at improving cognitive functions in aged and demented individuals with unequivocal results. In order to investigate whether the VP systems indeed show the claimed degenerative changes during aging and dementia, we focused in the first place on the Supra Optic Nucleus (SON) and Para Ventricular Nucleus (PVN). VP cells were id

Immunohistochemical analysis of magnocellular elements in rat hypothalamus: distribution and numbers of cells containing neurophysin, oxytocin, and vasopressin.

A cell-by-cell analysis of the magnocellular elements in hypothalami of fifty Long-Evans (normal) and Brattleboro (diabetes insipidus) rats was done using the unlabeled antibody enzyme technique (PAP) with primary antisera directed against oxytocin (OXY), vasopressin (ADH), and the neurophysins. The magnocellular neurons of the hypothalamus were found in the supraoptic (SON), paraventricular (PVN), and anterior commissural (ACN) nuclei, a number of accessory nuclei, and as individual cells in the anterior hypothalamic area. SON was divided by the optic tract into the principal part and retroch

Vasopressin and oxytocin gene expression in the porcine forebrain under basal conditions and following acute stress.

This study, the first using the pig, examined expression of mRNAs for vasopressin (VP), oxytocin (OT), preproenkephalin (PENK) and pro-opiomelanocortin (POMC) in the forebrain, and of POMC and prolactin in the pituitary. High basal expression of VP and OT mRNAs was present in the paraventricular (PVN) and supraoptic (SON) nuclei. In the PVN, VP was found in magnocellular regions whereas OT was also seen in the parvocellular portion; the distribution of VP and OT mRNAs in the SON was as reported in other species. The suprachiasmatic nucleus contained VP mRNA but only OT message was present in t

Impaired regulation of neurohypophyseal vasopressin secretion in rats treated neonatally with monosodium-L-glutamate.

Monosodium-L-glutamate (MSG) lesioning of the arcuate nucleus (ARC) was utilized to elucidate the importance of its dopaminergic system for regulation of neurohypophyseal vasopressin (AVP) secretion. Water deprivation in control animals elicited a rise in plasma AVP and depletion of neurohypophyseal AVP, at unchanged AVP contents of the paraventricular (PVN) and supraoptic (SON) nuclei. Dopaminergic activity was markedly stimulated in the PVN and ARC. Neonatal MSG treatment resulted in elevated plasma AVP levels at reduced neurohypophyseal AVP due to diminished dopaminergic input from the ARC.

Immunocytochemical localization of angiotensin II and vasopressin in rat hypothalamus: evidence for production in the same neuron.

1. By immunoperoxidase technique, immunoreactive angiotensin II (ANG II) was located in the cell bodies of many magnocellular neurons in the supraoptic and paraventricular nuclei of the hypothalamus and their pathways to median eminence and posterior pituitary gland in the rat. 2. Like vasopressin and its neurophysin, but not oxytocin, ANG II was also found in parvocellular neurons in the suprachiasmatic nucleus. 3. Analysis of these peptides in the same magnocellular neurons reveals that ANG II is localized primarily in vasopressin cells. 4. Like vasopressin and its precursor, ANG II is defic
