Mycalolides are marine-derived trisoxazole macrolides that exhibit potent actin-depolymerizing activity and inhibit the proliferation, migration, and invasion of various cancer cells. The C25-C35 side-chain moiety of mycalolides is important for their actin-binding properties; however, the biological function of the C1-C24 trisoxazole macrolactone moiety remains largely unexplored. In this study, using a photoaffinity biotin probe, we identified tubulin as a new, actin-independent target of mycalolide C (MyC). Ultracentrifugation, fluorescence-based tubulin polymerization assays, and transmiss

A 46-year-old man, with HIV infection A2 stage and chronic C hepatitis was diagnosed with stage 4 prostate cancer with bone metastases in September 2019. His treatment was started with docetaxel, receiving five cycles with good tolerance, after which he received hormonal treatment. Currently, he presents progression of the disease and docetaxel is reintroduced. After infusing the first 5 mL, he showed respiratory distress with thoracic pain and low oxygen saturation (88%) together with lumbar pain. Both tryptase and interleukin 6 (IL-6) levels in blood were measured at the time of the reaction

PURPOSE: Treatment for metastatic castration-resistant prostate cancer (mCRPC) includes chemotherapy and inhibition of the androgen receptor pathway. However, the optimal treatment sequence in this scenario is not yet fully understood. Therefore, we conducted a systematic review and meta-analysis comparing cabazitaxel versus abiraterone or enzalutamide for efficacy and safety outcomes as second-line therapy in mCRPC patients after docetaxel failure.
METHODS: We searched PubMed, Embase, and Cochrane databases for interventional studies comparing cabazitaxel versus abiraterone or enzalutamide f

BACKGROUND: Cabazitaxel frequently causes severe neutropenia. A higher cabazitaxel systemic exposure is related to a lower nadir absolute neutrophil count (ANC).
OBJECTIVE: To describe the effect of cabazitaxel systemic exposure on ANC by a population pharmacokinetic/pharmacodynamic (POP-PK/PD) model, and to identify patients at risk of severe neutropenia early in their treatment course using a PK threshold.
DESIGN, SETTING, AND PARTICIPANTS: Data from five clinical studies were pooled to develop a POP-PK/PD model using NONMEM, linking both patient characteristics and cabazitaxel systemic ex

BACKGROUND: Little is known about the characteristics of oncological patients, cancer therapy-induced cardiotoxicity, and guidelines-directed interventions in the Caribbean; analysis of cardio-oncology services may shed light on this and clarify links between ethnicity, cultural, and local socioeconomic factors.
OBJECTIVES: This study compared patients' phenotypes, adherence to guidelines recommendations, and patterns of cardiotoxicity between two cardio-oncology programs: one in the Dominican Republic (DR) and the other in Chicago IL, United States (US).
METHODS: Patients being considered f

Chemotherapy-induced peripheral neuropathy (CIPN) is a common and debilitating condition associated with a number of chemotherapeutic agents. Drugs commonly implicated in the development of CIPN include platinum agents, taxanes, vinca alkaloids, bortezomib, and thalidomide analogues. As a drug response can vary between individuals, it is hypothesized that an individual's specific genetic variants could impact the regulation of genes involved in drug pharmacokinetics, ion channel functioning, neurotoxicity, and DNA repair, which in turn affect CIPN development and severity. Variations of other

Prostate cancer (PC) is a heterogeneous disease, whose growth is driven by androgens and androgen receptors. Androgen deprivation therapy (ADT) is the standard treatment of hormone-naïve metastatic disease. The majority of patients are treated with medical castration with GnRH agonists or antagonists, which usually determines a profound PSA decline and a radiological and clinical benefit. However, essentially all patients experience progression to castration-resistant prostate cancer (CRPC), and overall prognosis remains disappointing. Early targeting of cells that survive hormonal therapy may

Taxanes are an established option in the standard treatment paradigm for patients with metastatic breast cancer (MBC). Neuropathy is a common, dose-limiting side effect of taxane therapy that is often managed by dose reductions and delays. The severity, time to onset, and improvement in neuropathy are important considerations for patient management and vary among currently approved taxanes. The rate of grade ≥3 neuropathy with taxanes has been shown to be dose and schedule dependent; however, time to improvement to grade ≤1 is typically shorter for nab-paclitaxel than for other taxanes in pati

Anagen effluvium due to chemotherapy is usually reversible with complete hair regrowth. However, there is increased evidence that certain chemotherapy regimens can cause dose-dependent permanent alopecia. The histological features of this type of alopecia and the mechanisms of its origin are not known yet. We discuss the histological features of 10 cases of permanent alopecia after systematic chemotherapy with taxanes (docetaxel) for breast cancer (6 patients), busulfan for acute myelogenous leukemia (3 patients), and cisplatin and etoposide for lung cancer (1 patient). All patients had modera

BACKGROUND: A 51-year-old woman experienced refractory chemotherapy-induced nausea and vomiting (CINV) in spite of extensive antiemetic therapy, including 5-HT3 antagonists, corticosteroids, dopamine antagonists and antihistamines.
CASE REPORT: We administered the patient clonazepam. After taking clonazepam, the patient fully recovered from the nausea and vomiting and never experienced them again.
CONCLUSION: Clonazepam may be useful in the control of CINV. We believe that clonazepam contributed to the favorable outcome by expressing an anxiolytic and an anticonvulsant effect on myoclonus. T

BACKGROUND: We have revealed in a pre-clinical study that the combination of adriamycin (ADR) and docetaxel (DOC) in which ADR was administered 12 h after DOC injection not only significantly reduced leukopenia and toxic death but also significantly increased the antitumor effect compared with the dosing schedule without an interval between each injection used commonly in clinical practice. The purpose of this study was to clarify in mice whether the toxic death caused by ADR was reduced by administering ADR after DOC injection when the doses and dosing-interval of ADR and DOC were changed.

Chemotherapeutic agents used for most common cancers are frequently associated with neurotoxicity, which often include debilitating side effects such as seizures. Docetaxel, one of the most widely and effectively used chemotherapeutic drugs, is associated with an array of symptoms referred to as Docetaxel-Induced Peripheral Neuropathies (DIPNs), including motor neuropathy, tingling, muscle weakness, and numbness. In this study, we use the electroconvulsive assay to model DIPN-related muscle weakness in C. elegans, via shock induction. We show that acutely or chronically exposing nematodes to d

The development of simple, efficient, and actively targeted drug delivery systems remains a significant challenge for clinical translation. Here, we propose a carrier-free nanodrug platform based on polyphenol-modified prodrugs with self-assembly and ligand-adsorption capabilities. Specifically, two gallic acid-modified Cabazitaxel (CTX) prodrugs were synthesized: a reduction-responsive prodrug (CSG) and a non-responsive control (CCG). These prodrugs spontaneously assembled into nanocomplexes, and transferrin (Tf) was non-covalently adsorbed onto their surfaces via polyphenol-protein interacti
