Endothelial lipopigment as an indicator of alpha-tocopherol deficiency in two equine neurodegenerative diseases.

Two spontaneous neurodegenerative diseases of the horse, equine motor neuron disease (EMND) and equine degenerative myeloencephalopathy (EDM), have been associated with alpha-tocopherol deficiency, and both were characterized by prominent accumulations of endothelial lipopigment in the small vessels of the spinal cord. These endothelial pigment deposits appear to be reversible. In EMND horses pasture-supplemented for 9 months or more after the progression of weakness and wasting had arrested, there was very little endothelial lipopigment. The origin and the potential effects of these endotheli

Selective block of tetramethrin-modified sodium channels by (+/-)-alpha-tocopherol (vitamin E).

Pyrethroids exert their hyperexcitatory effects by prolonging the open time of individual neuronal sodium channels. Occupational exposure to pyrethroids frequently leads to abnormal skin sensation or paresthesia. Vitamin E is known to reduce the cutaneous paresthesia. However, the mechanism of action has been totally unclear. Because the sodium channel is the major target site of pyrethroids, it is possible that vitamin E interferes with pyrethroid modification of the sodium channel. Patch clamp experiments were performed using rat dorsal root ganglion neurons and cerebellar Purkinje cells. (+

Significance of alpha-tocopherol and interleukin 8 in septic adult respiratory distress syndrome.

To elucidate the relationship between active oxygen and interleukin 8 (IL-8) in patients with septic adult respiratory distress syndrome (ARDS), we determined the serum levels of alpha-tocopherol, which has an antioxidant action, and IL-8 in seven patients with this disease. Serum alpha-tocopherol and IL-8 levels determined at the time of diagnosis of septic ARDS were 0.97 +/- 0.36 mg/dl and 0.98 +/- 0.99 ng/ml, respectively. A significant correlation was found between serum alpha-tocopherol level and IL-8 level (r = -0.758, p = 0.0473). These findings suggest that IL-8 activates neutrophils,

Alpha-tocopherol reduces doxorubicin-induced toxicity in rats--histological and biochemical evidences.

The beneficial effect of alpha-tocopherol on doxorubicin-induced toxicity was studied in rats. alpha-Tocopherol (400 mg/kg/day) was administered orally, daily for a period of 2 months along with/without doxorubicin (2.5 mg/kg, i v weekly once for 8 weeks). Histology showed liver necrosis, heart myocyte degeneration, glomerular and tubular degeneration, cellular infiltration and desquammation of intestinal mucosa in doxorubicin treated animals. There was a significant increase in lipid peroxide levels measured in terms of "TBA reactants" in all these organs. These changes were associated with e

The Emory mouse cataract: the effects on cataractogenesis of alpha-tocopherol, penicillamine, triethylenetetramine, and mercaptopropionylglycine.

The Emory mouse develops a late-onset hereditary cataract bearing some resemblances to human senile cataract. It was used as a model system for testing the effects of several drugs expected to have anticataractogenic potential. A low level of added dietary alpha-tocopherol had only a marginal effect. Penicillamine increased lens soluble protein, a good index of lens viability. Triethylenetetramine was too toxic to permit satisfactory treatment. Mercaptopropionylglycine produced several positive effects including a retardation of cataract at 6 months of age; parameters which increased under dru

HPLC method for the simultaneous determination of beta-carotene, retinol and alpha-tocopherol in serum.

A method is described for the simultaneous analysis of alpha-tocopherol, beta-carotene, and retinol in n-hexane extracts of serum. The technique uses normal-phase HPLC in a silica gel column with n-hexane-2-propanol (97:3, v/v) as the mobile phase. The compounds are eluted from the column in the order alpha-tocopherol, beta-carotene, tocol (internal standard) and retinol. The compounds are quantified by use of the following detectors connected in series: retinol and tocol by a spectrophotometer, beta-carotene by a filter photometer, and alpha-tocopherol and tocol by a spectrophotofluorimeter.

Development and Validation of a Liquid Chromatography Mass Spectrometry Method for Simultaneous Measurement of 25(OH)D3, epi-25(OH)D3, 25(OH)D2, Vitamin A, α-Tocopherol, and γ-Tocopherol.

OBJECTIVES: Fat-soluble vitamins are measured to identify deficiencies that may lead to rickets, osteomalacia, night blindness, and reversible motor and sensory neuropathies. We present a rapid liquid chromatography-mass spectrometry (LC-MS/MS) method that simultaneously measures 25-hydroxyvitamin D3 (25[OH]D3), epi-25(OH)D3, 25(OH)D2, vitamin A, α-tocopherol, and γ-tocopherol. METHODS: We mixed 100 µL serum with internal standard and extracted it by using supported liquid extraction plates. Reconstituted specimens were analyzed by LC-MS/MS with a 10-minute gradient. RESULTS: The method was

Interaction between tocopherols and phytosterols in vegetable oils and their antioxidant effects: A multi-scale investigation using molecular simulation and machine learning.

This study elucidates the interaction mechanism between tocopherols and phytosterols in vegetable oils and its influence on oxidative stability. The bond dissociation enthalpy (BDE) of α/γ-tocopherol and four phytosterols were calculated by density functional theory. It was found that the BDE of tocopherols were significantly lower than those of phytosterols, indicating the superior antioxidant activity of tocopherols. Molecular dynamics simulations show that the presence of phytosterols weakens the interaction between tocopherol and triglycerides, and the hydrogen bond density related to toco…

Comparison of Glutathione, Retinol and α- and γ-Tocopherols Concentrations Between Children with and Without Epilepsy: A Single-Center Case-Control Study.

BACKGROUND: Oxidative stress is associated with the pathogenesis of epilepsy. Long-term treatment with anti-seizure medications (ASMs) may reduce antioxidant levels, which consequently impairs the brain's ability to counteract oxidative damage. This study aimed to assess the concentrations of selected antioxidants (i.e., glutathione, retinol, and α- and γ-tocopherols) in children with epilepsy treated with polytherapy. METHODS: The study included 21 children with epilepsy treated with ≥2 ASMs for at least 6 months (mean age 7.1 ± 4.4 years) and 23 control children without epilepsy (mean age 7…

Simultaneous Determination of Pigments, Tocopherols, and Squalene in Greek Olive Oils: A Study of the Influence of Cultivation and Oil-Production Parameters.

A new facile and fast method was developed in this study for the determination of pigments (chlorophylls and carotenoids), tocopherols (α-, sum of (β + γ), and δ), and squalene in olive oil. This method consisted of a dilution of olive oil in 2-propanol, followed by reversed phase-high-pressure liquid chromatography equipped with a diode array detector (RP-HPLC-DAD). Chromatographic separation was performed using a C18 column, while the mobile phase consisted of acetonitrile and methanol using a gradient elution program. The methodology was optimized, validated, and applied to the analysis of …

Metabolomic insights into homogentisic acid-mediated regulation of tocopherol enhancement in Monascus purpureus fermentation.

Targeted enrichment of tocopherols in fungal fermentation systems remains a significant metabolic engineering challenge. This study presented a novel strategy employing homogentisic acid (HGA) modulation to efficiently enhance tocopherol production in the M. purpureus-fermented coix seed (MFCS) system. Three strategies of exogenous HGA supplementation (Exo-HGA), endogenous HGA accumulation (End-HGA), and the combination of exogenous HGA supplementation and endogenous HGA accumulation (Exo + End-HGA) were systematically investigated. While End-HGA optimally promoted lovastatin, pigment, and bio…
