Cadmium toxicity assessment based on a climbing behaviour test with intertidal periwinkles, Littorina littorea (L.).

The present investigation was carried out to develop a climbing behaviour test with periwinkles, Littorina littorea. Sublethal exposure to pollutants may jeopardize the ability of this intertidal mollusc to climb up to below the water surface and remain attached to the substrate to defend against waves and predators, which is an ecologically relevant effect. The total climbing distance covered for 8 hr (TCD-8), the cumulative climbing activity (CCA) and the tenacity of attachment (ToA) index were shown to exhibit dose-dependent changes on exposure to sublethal concentrations of Cd. Periwinkles

Ontogeny of Placental FcRn Protein During Human and Animal Pregnancy to Inform Developmental Toxicity Testing.

INTRODUCTION: Species differences exist in Fc domain-containing molecule transfer/biodistribution to the embryo-fetus during pregnancy. Placentation was reviewed and placental neonatal Fc receptor (FcRn) quantitation in the chorioallantoic placenta (CAP) and/or inverted yolk sac placenta (InvYSP) was investigated across species throughout gestation to further understand and inform developmental toxicity testing of Fc domain-containing molecules. METHODS: Quantitative mass spectrometry was used to quantify FcRn protein in CAP and/or InvYSP throughout gestation in mouse, rat, guinea pig, rabbit

Rapid Volumetric Bioprinting Coupled with Dynamic Perfusion Enhances Human Hepatic Organoid Toxicity Testing.

Drug-induced liver injury (DILI) remains a major cause of acute liver failure and drug withdrawal from the market. Recently developed three-dimensional (3D) hepatic in vitro systems exhibit improved functionality and drug sensitivity compared with conventional two-dimensional cultures. These 3D models range from simple physiologic-like culture systems to advanced bioreactors with dynamic flow to provide sufficient nutrients and consistent drug exposure. However, whether dynamic perfusion improves sensitivity and reproducibility of hepatotoxicity testing remains unclear. Here, we developed a ta

Daphnids can safeguard the use of alternative bioassays to the acute fish toxicity test: a focus on neurotoxicity.

Assessment of potential impacts of chemicals on the environment traditionally involves regulatory standard data requirements for acute aquatic toxicity testing using algae, daphnids, and fish (e.g., Organisation for Economic Co-operation and Development [OECD] test guidelines 201, 202, and 203, respectively), representing different trophic levels. In line with the societal goal to replace or reduce vertebrate animal testing, alternative bioassays were developed to replace testing with fish: the fish cell line RTgill-W1 acute toxicity assay (OECD test guideline 249) and the zebrafish embryo acu

Toxicity assessment of Eucomis autumnalis and Merwilla plumbea using the in vivo zebrafish embryotoxicity test (ZET).

ETHNOPHARMACOLOGICAL RELEVANCE: Eucomis autumnalis (Mill.) Chitt. and Merwilla plumbea (Lindl.) Speta are popularly used in southern African healing practices to treat a wide range of diseases such as gastrointestinal, respiratory and skin conditions, as well as spiritual protectants against witchcraft. The pharmacological properties of the two species are well documented; however, studies that investigate the safety profiles using in vivo models are limited. AIM OF THE STUDY: This study evaluated the toxicity profiles of the two plant species independently using the in vivo zebrafish embryot

Development of a high-throughput spore germination test to assess the toxicity of pesticides on arbuscular mycorrhizal fungi.

Pesticides are essential agricultural inputs that help securing crop yields. However, they can affect non-target soil microorganisms, including arbuscular mycorrhizal (AM) fungi, that are potential indicators of the toxicity of pesticides on the soil microbiota. Here, we developed a fast-track high-throughput spore germination test, for AM fungi produced in vitro. This test allows the determination of EC50 values and the nature of the effects of pesticides on AM fungal spores (fungicidal or fungistatic). First, 19 active ingredients were tested on Rhizophagus intraradices MUCL 49410. Secondly,

StreptInCor, a Group A Streptococcal Adsorbed Vaccine: Evaluation of Repeated Intramuscular Dose Toxicity Testing in Rats.

Streptococcus pyogenes infections continue to be a worldwide public health problem, causing various diseases in humans, with rheumatic fever and rheumatic heart disease being the most harmful manifestations. Impetigo and post-streptococcal glomerulonephritis are also important sequelae of skin infections. We have developed a candidate vaccine epitope (StreptInCor) that presents promising results in diverse animal models. To assess whether the StreptInCor alum-adsorbed vaccine could induce undesirable effects, a certified independent company conducted a repeated intramuscular dose toxicity eval

Health Misinformation about Toxic-Site Harm: The Case for Independent-Party Testing to Confirm Safety.

Health misinformation can cause harm if regulators or private remediators falsely claim that a hazardous facility is safe. This misinformation especially threatens the health of children, minorities, and poor people, disproportionate numbers of whom live near toxic facilities. Yet, perhaps because of financial incentives, private remediators may use safety misinformation to justify reduced cleanup. Such incentives exist in nations like the United States, where most toxic-site testing/remediation is semi-privatized or voluntary, conducted by private parties, commercial redevelopers, who can inc

Could the environmental toxicity of diclofenac in vultures been predictable if preclinical testing methodology were applied?

Diclofenac, a non-steroidal anti-inflammatory pharmaceutical agent was responsible for the death of millions of Gyps vulture's in the Indian sub-region with the safety of the other non-steroidal anti-inflammatory drugs (NSAIDs) being questionable. With preclinical safety testing not well established for avian species unlike for mammalian and environmental toxicity, we ask the question if a preclinical model could have predicted the toxic effect of the drug. For this study, we test an Organisation for Economic Co-operation and Development (OECD) guideline 223 for assessing the acute toxic poten

Development and validation of an OECD reproductive toxicity test guideline with the mudsnail Potamopyrgus antipodarum (Mollusca, Gastropoda).

Mollusks are known to be uniquely sensitive to a number of reproductive toxicants including some vertebrate endocrine disrupting chemicals. However, they have widely been ignored in environmental risk assessment procedures for chemicals. This study describes the validation of the Potamopyrgus antipodarum reproduction test within the OECD Conceptual Framework for Endocrine Disrupters Testing and Assessment. The number of embryos in the brood pouch and adult mortality serve as main endpoints. The experiments are conducted as static systems in beakers filled with artificial medium, which is aerat

PAH toxicity at aqueous solubility in the fish embryo test with Danio rerio using passive dosing.

As part of the risk assessment process within REACh, prior to manufacturing and distribution of chemical substances their (eco)toxicological impacts have to be investigated. The fish embryo toxicity test (FET) with the zebrafish Danio rerio has gained a high significance as an in vitro alternative to animal testing in (eco)toxicology. However, for hydrophobic organic chemicals it remains a technical challenge to ensure constant freely dissolved concentration at the maximum exposure level during such biotests. Passive dosing with PDMS silicone was thus applied to control the freely dissolved co

(13)C-uracil breath test to predict 5-fluorouracil toxicity in gastrointestinal cancer patients.

PURPOSE: Up to 30 % of patients undergoing 5-fluorouracil (5FU)-based chemotherapy experience severe toxicity. Dihydropyrimidine dehydrogenase (DPD) deficiency explains 36-61 % of cases. Predicting toxicity is an unmet challenge. Uracil breath test (UraBT) consists of measuring (13)CO2 in exhaled breath after ingestion of 2-(13)C-uracil to evaluate pyrimidine (and 5FU) catabolism. METHODS: We studied 33 gastrointestinal cancer patients previously exposed to 5FU: Thirteen had grade 3-4 and 20, grade 0-1 toxicity. The following tests were used to evaluate pyrimidine catabolism: (1) sequencing o

Development and application of whole-sediment toxicity test using immobilized freshwater microalgae Pseudokirchneriella subcapitata.

A method for a whole-sediment toxicity test using alginate immobilized microalgae Pseudokirchneriella subcapitata was developed using spiked sediments and applied to contaminated field sediment samples. For method development, a growth inhibition test (72 h) with algal beads was conducted for the sediments spiked with Cu or diuron. The method was validated by determining dose-response relationships for Cu and diuron in both fine-grained and coarse-grained sediments. The results of a spiked sediment toxicity test suggested that sediment particle size distribution (clay content) had a significan

Reproductive toxicity of bisphenol A and cadmium in Potamopyrgus antipodarum and modulation of bisphenol A effects by different test temperature.

An OECD initiative for the development of mollusc-based toxicity tests for endocrine disrupters and other chemicals has recommended three test species with respective test designs for further standardisation. Preparing a subsequent pre-validation study we performed a reproduction test with Potamopyrgus antipodarum, determining the concentration range of the selected test substances, bisphenol A (BPA) and cadmium (Cd). At 16 °C, the recommended test temperature, the number of embryos in the brood pouch was increased by BPA and decreased by Cd (NOEC: 20 μg BPA/L and 1 μg Cd/L). Coinstantaneous B…
