STUDY PURPOSE: Botulinum toxin (BoNT) is commonly used for both cosmetic and medical purposes. However, improper use can lead to iatrogenic botulism. This scoping review aimed to evaluate the systemic and skin-related effects, identify risk factors, and highlight safety gaps in BoNT-related botulism.
METHODS: We followed PRISMA-ScR guidelines to search for literature from 1997 to 2024 across PubMed, Scopus, EMBASE, and Google Scholar. Ten case reports (113 patient cases) met the inclusion criteria.
RESULTS: Females accounted for 78.8% of cases. Systemic symptoms typically developed within fo

Campylobacter is one of the leading bacterial causes of gastroenteritis worldwide. It frequently contaminates poultry and other raw meat products, which are the primary sources of Campylobacter infections in humans. Plasmids, known as important mobile genetic elements, often carry genes for antibiotic resistance, virulence, and self-mobilization. They serve as the main vectors for transferring genetic material and spreading resistance and virulence among bacteria. In this study, we identified 34 new plasmids from 43 C. jejuni and C. coli strains isolated from retail meat using long-read and sh

UNLABELLED: Metabolism plays an important role in the persister phenotype, as evidenced by the number of strategies that perturb metabolism to sabotage this troublesome subpopulation. However, the absence of techniques to isolate high-purity populations of native persisters has precluded direct measurement of persister metabolism. To address this technical challenge, we studied Escherichia coli populations whose growth had been inhibited by the accumulation of the MazF toxin, which catalyzes RNA cleavage, as a model system for persistence. Using chromosomally integrated, orthogonally inducible

1. Agkistrodon piscivorus venom and E. coli endotoxin were shown to be immunologically distinct, and to differ in certain biologic properties: effects on immune response, body temperature, and circulating leukocyte count, and capacity to prepare for and provoke the local and generalized Shwartzman reaction. 2. Neither a single prior injection of venom nor the existence of hyperimmunity to lethal doses of venom protected rabbits against the local and generalized Shwartzman reaction. 3. Serial intravenous injections of sublethal doses of venom produced enhanced susceptibility to venom rather tha

BACKGROUND AND OBJECTIVE: KRAS mutation is an early event in colorectal cancer carcinogenesis. We previously reported that a recombinant adenovirus, carrying a pro-apoptotic gene (PUMA) under the regulation of Ets/AP1 (RAS-responsive elements) suppressed the growth of cancer cells harboring hyperactive KRAS. We propose to exploit the hyperactive RAS pathway, rather than to inhibit it as was previously tried and failed repeatedly. We aim to improve efficacy by substituting PUMA with a more potent toxin, the bacterial MazF-MazE toxin-antitoxin system, under a very tight regulation.

Hemolytic Uremic Syndrome (HUS) is a severe clinical manifestation primarily triggered by Shiga toxin-producing Escherichia coli (STEC). While Shiga toxins (Stx) are central to the development of systemic endothelial damage, current recombinant antibody developments have overwhelmingly focused on neutralizing the Stx2 subtype. However, numerous STEC isolates produce Stx1 either independently or alongside Stx2, revealing a critical need to diversify the antibody repertoire for comprehensive antitoxin therapies. To address this, we characterized two novel, fully human recombinant Fabs targeting

In late September 1986, we found 7 patients from a printing factory in Chang-Hwa city who developed an endemic disease manifested by general malaise, ptosis, double vision, dysarthria, dysphagia, and proximal limb weakness. After clinical, epidemiological, microbiological, and toxicological investigations, an outbreak of botulism was confirmed 2 weeks later, Commercially canned peanuts made by an unlicensed cannery were identified as the vehicle of botulinum toxin transmission. Antitoxin was given to 2 patients who needed ventilator support. One of the 7 victims died from medical complications

INTRODUCTION: Providencia species are emerging opportunistic pathogens associated with multidrug-resistant infections, yet their molecular defense mechanisms against phage or mobile genetic elements remain poorly characterized.
METHODS: We present a comprehensive pan-genomic analysis of antiviral defense systems across 73 complete genomes (or chromosomes) of Providencia stuartii (n = 31) and Providencia rettgeri (n = 42), using DefenseFinder and CRISPRCasFinder. We further expanded analysis of contig/scaffold assemblies to confirm conservation of core defense profiles across assembly types. B

Acinetobacter baumannii and Acinetobacter nosocomialis are the imperious pathogens in the intensive care units. We aimed to explore the genomic features of these pathogens to understand the factors influencing their plasticity. Using next-generation sequencing, two carbapenem-resistant A. baumannii (AbaBS-3, AbaETR-4) isolates and a pan-susceptible A. nosocomialis (AbaAS-5) isolate were characterised. All genomes exhibited 94% similarity with a degree of heterogeneity. AbaBS-3 and AbaETR-4 harboured antibiotic resistance gene (ARG) repertoire to most antibiotic classes. Carbapenem resistance w

Streptococcus pneumoniae (pneumococcus) is a leading cause of severe invasive infectious diseases such as sepsis and meningitis. Understanding how pneumococcus adapts and survive in the human bloodstream environment and cerebrospinal fluid (CSF) is important for development of future treatment strategies. This study investigates the global transcriptional response of pneumococcus to human blood components and CSF acquired from discarded and anonymized patient samples. Extensive transcriptional changes to human blood components were observed during early stages of interaction. Plasma-specific r

The plasmid of emerging Salmonella Infantis (pESI) and pESI-like plasmids have been reported as a major cause of the dissemination of S. Infantis worldwide, and these plasmids have also been reported in Korea. In this study, eight pESI or pESI-like plasmids, including two pESI-like plasmids of S. Infantis from the poultry industry in Korea, harbored yersiniabactin operon, toxin/antitoxin systems, and fimbria clusters. Interestingly, various resistance genes were identified in the pESI-like plasmids but not in the pESI plasmid. In particular, dfrA14, which confers resistance to trimethoprim, wa

Persistent and viable but non-culturable (VBNC) Pseudomonas aeruginosa cells are mainly responsible for the recurrence and non-responsiveness to antibiotics of cystic fibrosis (CF) lung infections. The sub-inhibitory antibiotic concentrations found in the CF lung in between successive therapeutic cycles can trigger the entry into the VBNC state, albeit with a strain-specific pattern. Here, we analyzed the VBNC cell induction in the biofilms of two CF P. aeruginosa isolates, exposed to starvation with/without antibiotics, and investigated the putative genetic determinants involved. Targeted-Antibacterial-Plasmids (TAPs) offer a precise approach to combat multidrug-resistant bacteria by selectively removing resistant strains while preserving commensals. Here, we assess TAPs that deliver CRISPR/Cas systems via conjugation to kill or resensitize extended-spectrum β-lactamase (ESBL)-producing Escherichia coli carrying the blaCTX-M-15 gene. We systematically tested multiple variables in the TAP approach, including two commensal E. coli donors, three distinct helper plasmids encoding the transfer machineries, and six recipient strains harbouring the resistance gene either chro

Human beings release thousands of antibiotics into the environment, which could generate the related transformation products (TPs), most of which have yet to be identified and lack rigorous microbial risk information. This study aimed to investigate the impact and mechanisms of 4-nitro sulfamethoxazole, N4-acetylated sulfamethoxazole, and N4-acetylated sulfadiazine, three typical sulfonamide (SAs) metabolites, on the risk of antibiotic resistance genes (ARGs) transmission. The results revealed that TPs significantly enhance the risk of conjugative transfer of RP4 plasmid at clinically and envi

INTRODUCTION: The emergence of extended-spectrum β-lactamases (ESBLs)-producing Escherichia coli clones are a public health concern worldwide. Scarce information does exist about the spread of ESBLs-producing E. coli in pediatric patients from developing countries.
METHODOLOGY: E. coli strains were analyzed by multilocus-sequence-typing, pulsed-field-gel-electrophoresis and phylogenetic group. The antimicrobial-resistance genes were detected by PCR, and plasmid content by the PCR-based replicon-typing. Horizontal transfer was tested by conjugation and the location of the blaCTX-M-15 gene by S

Persisters are phenotypically antibiotic-tolerant cells which can survive antibiotic exposure without acquiring antibiotic resistance. A proposed important factor in persistence is low intracellular ATP levels, which are thought to reduce the activity of antibiotic targets. However, previous studies demonstrated that persisters have comparable DNA damage as drug-sensitive bacteria after fluoroquinolone treatment. Furthermore, recent studies reported that endogenous prophages can reduce levels of antibiotic persistence in Salmonella after fluoroquinolone treatment. In this study, we examined pr

Infectious bacteria remain among the most plausible agents for deliberate biological attack because they combine environmental robustness, low infectious dose, and the capacity for rapid, severe disease. This review synthesizes current knowledge on major bacterial biowarfare threats including Bacillus anthracis, Yersinia pestis, Francisella tularensis, Brucella spp., and Clostridium botulinum across three levels: transmission routes, molecular pathogenesis, and defense strategies. We summarize how aerosol, food-water, and vector-borne pathways, together with globalization and urban crowding, s

Hepatitis C virus (HCV) infection is a major cause of chronic liver disease and has become a global health threat. No HCV vaccine is currently available and treatment with antiviral therapy is associated with adverse side effects. Moreover, there is no preventive therapy for recurrent hepatitis C post liver transplantation. The NS3 serine protease is necessary for HCV replication and represents a prime target for developing anti HCV therapies. Recently we described a therapeutic approach for eradication of HCV infected cells that is based on protein delivery of two NS3 protease-activatable rec

BACKGROUND: Respiratory diphtheria is a toxin-mediated disease caused by Corynebacterium diphtheriae. Diphtheria-like illness, clinically indistinguishable from diphtheria, is caused by Corynebacterium ulcerans, a zoonotic bacterium that can also produce diphtheria toxin. In the United States, respiratory diphtheria is nationally notifiable: specimens from suspected cases are submitted to the Centers for Disease Control and Prevention (CDC) for species and toxin confirmation, and diphtheria antitoxin (DAT) is obtained from CDC for treatment. We summarize the epidemiology of respiratory diphthe

Bacterial flagella are known for facilitating motility to support nutrient acquisition and predator evasion, but can also serve as receptors for phages. Here, we characterize a dual role of flagella during infection of Yersinia enterocolitica by phage X1: functioning as a conduit for phage DNA entry and as a trigger for bacterial defense. X1 employs a "contraction-driven" mechanism, using its contractile tail to inject DNA into Yersinia flagella. Unlike other characterized flagellotropic phages that rely exclusively on flagella or less efficiently on secondary surface receptors, X1 can infect
