DAX-1 in sex determination and gonadal development: revisiting the anti-testis hypothesis.

DAX-1 (NR0B1) is a pivotal regulator of mammalian reproductive and endocrine development. Discovered as a gene whose dosage imbalance can reverse sex in XY individuals, DAX-1 is now recognized as a key component of the network controlling gonadal formation and function. Despite decades of research, the molecular mechanisms of its function in gonad development and function remain only partially understood. We combine genetic, developmental, molecular and biochemical perspectives to present how DAX-1 regulates gene expression from early sex determination and gonadal differentiation to the mainte

Multi-omics integration in malignant pleural mesothelioma: from molecular evolution and immune ecosystems to precision therapy.

Malignant pleural mesothelioma (MPM) exhibits substantial molecular and cell-state heterogeneity, together with marked spatial variation in its immune and stromal microenvironment. Histological classification and individual biomarkers alone cannot adequately explain its evolution or heterogeneous treatment responses. This review critically integrates genomic, epigenomic, transcriptomic, single-cell, and spatial omics evidence along a conceptual continuum encompassing tumor-intrinsic molecular evolution, microenvironmental ecosystem remodeling, candidate stratification, and precision therapy. From forest floor to doctor's office: the immunological journey of Borrelia burgdorferi through vertebrate hosts.

Lyme disease, caused by the spirochete Borrelia burgdorferi, is the most prevalent vector-borne infection in the Northern Hemisphere and continues to expand geographically. Although B. burgdorferi has a streamlined genome and minimal virulence repertoire, it establishes persistent infection through coordinated modulation of mammalian host immune responses. Here, we synthesize recent advances in the immunobiology of B. burgdorferi using a stage-structured framework that traces a tick-mediated vertebrate infection through systemic dissemination, tissue colonization, and chronic immune engagement

SuperAging is not the inverse of common-variant Alzheimer's risk: evidence across genetic ancestries.

BACKGROUND: As longevity increases and the population over age 65 expands, advancing age remains the most reliable predictor of cognitive decline, highlighting the need to identify biological mechanisms that support exceptional cognitive aging. We tested whether lower inherited risk of Alzheimer's disease (AD) dementia predicts SuperAger status (adults ≥ 80 years with episodic memory at least as good as middle-age adults) using prospectively enrolled SuperAgers and Cognitively Average Controls (Controls) from the multisite SuperAging Research Initiative. METHODS: We studied 231 participants (

Whole genome sequencing approach for the detection of rare protein-altering variants associated with familial male infertility.

BACKGROUND: Whole-genome sequencing (WGS) is an efficient, accurate, and high-throughput technique used for the discovery of exonic as well as non-exonic variants causing different genetic anomalies, improving clinical management for the disease. This study was designed to evaluate pathogenic genetic variants related to male infertility in male infertile families in remote rural areas of Pakistan. METHODS: For the five families recruited, WGS was conducted on 14 patients and 6healthy controls. WGS was performed using the Illumina TruSeq DNA Nano Library Prep workflow protocol. DNA was assesse

Targeted short read NGS of HCV 5'-UTR for genotyping and identification of Sub-variants using Ion Torrent platform.

Hepatitis C Virus (HCV) is highly a mutable RNA virus, primarily due to lack of proofreading activity in RNA-dependent RNA polymerase. This intrinsic feature leads to the generation of substantial intra-host variation, resulting in the formation of viral quasispecies. The 5' untranslated region (5'UTR) of the HCV genome is highly conserved across different genotypes and serves as a widely adopted target for genotyping in clinical practice. In this study, the performance of targeted short-read next-generation sequencing of the HCV 5' UTR using Ion Torrent technology was evaluated, with the dual

Comparative Phylogeography of West African Rainforest Frogs Reveals Regional Variation in Refugia Dynamics.

Understanding how species responded to climatic change in the past can help predict the long-term implications of contemporary climate change. The Upper Guinean rainforests of West Africa are a global biodiversity hotspot, and it is well documented that climatic fluctuations in the Pleistocene drove the expansion and contraction of rainforest cover in this region. West African slippery frogs, genus Conraua, are rainforest specialists and present an excellent opportunity to study the effects of climate-driven landscape changes on contemporary phylogeographic patterns and population dynamics. Cytogenomic characterization of karyotypes with additional autosomal material.

Chromosomal rearrangements involving additional material in individuals with phenotypic alterations usually result in partial trisomy, often accompanied by partial monosomy. To characterize chromosomal rearrangements and analyze genomic characteristics in the breakpoint regions in 31 patients with additional material on an autosomal chromosome. Different tests were performed to characterize these patients, including karyotyping, chromosomal microarray analysis (CMA), and fluorescent in situ hybridization (FISH). In silico analyses evaluated A/B chromosomal compartments, segmental duplications,

Cross-continental transmission and host adaptation of Mycobacterium tuberculosis in China unveiled by population history reconstruction and adaptive evolution signal detection.

The transmission history and adaptive evolution of Mycobacterium tuberculosis complex (MTBC) in China remain underexplored despite its remarkably low diversity and enduring public health burden. Here, we analyzed 23,873 whole genome sequences of MTBC to reconstruct its spread timeline and routes, population dynamics, and host adaptation patterns in China. The Bayesian coalescent models revealed the recurrent introductions of four MTBC sub-lineages (L2.2, L4.2, L4.4, and L4.5) during 1000-400 years ago, by European-Chinese transmission networks, which might have triggered the formation of their

Molecular characterization of JC virus in progressive multifocal leukoencephalopathy cases from India.

Progressive Multifocal Leukoencephalopathy (PML) is a rare, often fatal demyelinating disease of the central nervous system caused by reactivation of the John Cunningham virus (JCV) in immunocompromised individuals. Despite an estimated 2.4 million people living with HIV in India, the reported incidence of PML remains lower than in Western countries, likely due to underdiagnosis, underreporting, and distinct host genetic and viral factors. The rising number of individuals on immunosuppressive therapies, including organ transplant recipients and those with autoimmune disorders, further emphasiz

Structure of ClfA002 in Complex With Neutralizing Antibody AZD7745 Provides Insight into Its Broad Neutralization Mechanism in Staphylococcus aureus Infection.

BACKGROUND: Complications due to Staphylococcus aureus infections are a major health concern as current therapies lack broad coverage; therefore, development of a broad neutralizing therapy is critical. We previously showed that a monoclonal antibody combination comprising suvratoxumab and AZD7745, targeting respectively alpha toxin and clumping factor A (ClfA), 2 S aureus virulence factors, provided broad strain coverage in a mouse model. To understand the basis for the broad strain coverage of AZD7745, the epitope was determined. Moreover, we surveyed a global collection of bacteremia isolat

Cell-type specific and differential expression of LINC-RSAS long noncoding RNA declines in the testes during ageing of the rat.

Long noncoding RNAs (lncRNAs) have emerged as major regulators of gene expression, chromatin structure, epigenetic changes, post-transcriptional processing of RNAs, translation of mRNAs into proteins as well as contributing to the process of ageing. Ageing is a universal, slow, progressive change in almost all physiological processes of organisms after attaining reproductive maturity and often associated with age-related diseases. Mammalian testes contain various cell-types, vast reservoir of transcriptome complexity, produce haploid male gametes for reproduction and testosterone for developme

B chromosome and its non-Mendelian inheritance in Atractylodes lancea.

Supernumerary B chromosomes contribute to intraspecific karyotypic variation. B chromosomes have been detected in more than 2000 organisms; they possess unique and diverse features, including non-Mendelian inheritance. Here, we report one or more B chromosomes in the gynodioecious plant Atractylodes lancea. Among 54 A. lancea lines, 0-2 B chromosomes were detected in both hermaphroditic and female plants, with the B chromosomes appearing as DAPI-bright regions within the nuclei. Genomic in situ hybridization revealed that the B chromosomes had no conserved A chromosome DNA sequences, confirmed

Deep Screening for X Chromosome Parent-of-Origin Effects on Neurobehavioral and Neuroanatomical Phenotypes in 47,XXY Klinefelter Syndrome.

BACKGROUND: X chromosome parent of origin (POX) has been proposed as a source of phenotypic variation within sex chromosome aneuploidies such as Klinefelter syndrome (XXY/KS) and between XX and XY individuals. However, previous studies have yielded conflicting results regarding the presence and nature of POX effects, which we sought to clarify in an expanded sample with deeper neurobehavioral phenotyping. METHODS: A cohort of 58 individuals with XXY/KS underwent duo or trio genome sequencing with parents (n = 151), measurement of 66 neurobehavioral phenotypes by standardized research assessme

Genetic diversity of Rhombomys opimus and Meriones meridianus with potential divergence of plague resistance in the Junggar Basin plague focus based on RT1-Db1*exon1.

In the Junggar Basin plague focus, Rhombomys opimus and Meriones meridianus live together, and their parasitic fleas have a similar community structure. However, R. opimus has significantly higher positive rates of Yersinia pestis and anti-F1 antibody compared with M. meridianus. In this study, Y. pestis- and antiF1 antibody-negative R. opimus and M. penicilliger were collected in Qitai county, Fukang city and Mulei county of the eastern part of the Junggar Basin. The genomic DNA was extracted from their spleen tissues, and RT1-Db1*exon1 was amplified through PCR procedure and then sequenced.
