Formation of α-helical and β-sheet structures in membrane-bound human IAPP monomer and the resulting membrane deformation.

The amyloid formation of human islet amyloid polypeptide (hIAPP)-an intrinsically disordered peptide, is associated with type II diabetes. Cellular membranes, especially those composed of negatively-charged lipids, accelerate the hIAPP amyloid fibrillation, and their integrity is disrupted during the aggregation process, leading to cell apoptosis. However, the underlying molecular mechanism is not well understood. Herein, we investigated the conformational dynamics during the interactions of hIAPP monomer with POPG membrane bilayer, by carrying out μs-long all-atom molecular dynamics simulatio

Multiple poloxamers increase plasma membrane repair capacity in muscle and nonmuscle cells.

Various previous studies established that the amphiphilic tri-block copolymer known as poloxamer 188 (P188) or Pluronic-F68 can stabilize the plasma membrane following a variety of injuries to multiple mammalian cell types. This characteristic led to proposals for the use of P188 as a therapeutic treatment for various disease states, including muscular dystrophy. Previous studies suggest that P188 increases plasma membrane integrity by resealing plasma membrane disruptions through its affinity for the hydrophobic lipid chains on the lipid bilayer. P188 is one of a large family of copolymers th

Structural basis of Toxoplasma gondii perforin-like protein 1 membrane interaction and activity during egress.

Intracellular pathogens must egress from the host cell to continue their infectious cycle. Apicomplexans are a phylum of intracellular protozoans that have evolved members of the membrane attack complex and perforin (MACPF) family of pore forming proteins to disrupt cellular membranes for traversing cells during tissue migration or egress from a replicative vacuole following intracellular reproduction. Previous work showed that the apicomplexan Toxoplasma gondii secretes a perforin-like protein (TgPLP1) that contains a C-terminal Domain (CTD) which is necessary for efficient parasite egress. H

Membrane protein oxidation determines neuronal degeneration.

Oxidative stress is an early hallmark in neurodegenerative disorders such as Alzheimer's and Parkinson's diseases. However, the critical biochemical effector mechanisms of oxidative neurotoxicity have remained surprisingly elusive. In screening various peroxides and potential substrates of oxidation for their effect on neuronal survival, we observed that intramembrane compounds were significantly more active than aqueous or amphiphilic compounds. To better understand this result, we synthesized a series of competitive and site-specific membrane protein oxidation inhibitors termed aminoacyllipi

Auto-oxidation and a membrane-associated 'Fenton reagent': a possible explanation for development of membrane lesions in sickle erythrocytes.

Sickle erythrocytes spontaneously generate approximately twice-normal amounts of activated oxygen species (superoxide, peroxide and the highly reactive hydroxyl radical). In addition, they contain excessive amounts of membrane-bound haemichromes, which presumably accumulate because of some degree of sickle haemoglobin instability, perhaps involving interactions between haemoglobin and the lipid bilayer. Since most erythrocyte antioxidant mechanisms are located in the cytoplasm, it is hypothesized that a physiologically significant hydroxyl radical generator would be located within or directly

Shaping membrane interfaces in lipid vesicles mimicking the cytoplasmic leaflet of myelin through variation of cholesterol and myelin basic protein contents.

Myelin basic protein (MBP) is an intrinsically disordered protein and in the central nervous system (CNS) mainly responsible for connecting the cytoplasmic surfaces of the multilamellar, compact myelin. Increased posttranslational modification of MBP is linked to both, the natural development (from adolescent to adult brains) of myelin, and features of multiple sclerosis. Here, we study how a combination of this intrinsically disordered myelin protein with varying the natural cholesterol content may alter the characteristics of myelin-like membranes and interactions between these membranes.

Membrane interactions and biological activity of antimicrobial peptides from Australian scorpion.

UyCT peptides are antimicrobial peptides isolated from the venom of the Australian scorpion. The activity of the UyCT peptides against Gram positive and Gram negative bacteria and red blood cells was determined. The membrane interactions of these peptides were evaluated by dye release (DR) of the fluorophore calcein from liposomes and isothermal titration calorimetry (ITC); and their secondary structure was determined by circular dichroism (CD). Three different lipid systems were used to mimic red blood cells, Escherichia coli and Staphylococcus aureus membranes. UyCT peptides exhibited broad

Helix stabilization precedes aqueous and bilayer-catalyzed fiber formation in islet amyloid polypeptide.

Islet amyloid polypeptide (IAPP) is an unstructured polypeptide hormone that is cosecreted with insulin. In patients with type 2 diabetes, IAPP undergoes a transition from its natively disordered state to a highly ordered, all-beta-strand amyloid fiber. Although predominantly disordered, IAPP transiently samples alpha-helical structure in solution. IAPP adopts a fully helical structure when bound to membrane surfaces in a process associated with catalysis of amyloid formation. Here, we use spectroscopic techniques to study the structure of full-length, monomeric IAPP under amyloidogenic condit

Differential expression of porins OmpP2A and OmpP2B of Haemophilus ducreyi.

Haemophilus ducreyi is a strict human pathogen and the causative agent of the sexually transmitted disease chancroid. The genome of the human-passaged strain of H. ducreyi (35000HP) contains two homologous genes whose protein products have estimated molecular masses of 46 and 43 kDa. A comparative analysis of the deduced amino acid sequences revealed that these proteins share 27 to 33% identity to the outer membrane protein P2 (OmpP2), a major porin of Haemophilus influenzae. Therefore, these proteins have been designated OmpP2A and OmpP2B, respectively. The detection of ompP2A and ompP2B tran

Heterogeneous amyloid-formed ion channels as a common cytotoxic mechanism: implications for therapeutic strategies against amyloidosis.

The amyloidoses consist of human and animal chronic, progressive, and sometimes fatal diseases that are characterized by the deposition of insoluble proteinaceous amyloid fibrils in various tissues. Despite the biochemical diversity of amyloids, they share certain properties. The amphipathic and the charged nature of many amyloid-forming peptides point to their intrinsic ability to form diverse beta-sheet-based aggregates and channel types in negatively charged membranes. We hypothesize that the formation of heterogeneous channels represents a common cytotoxic mechanism that accentuates the ch

Leptinotoxin-h action in synaptosomes, neurosecretory cells, and artificial membranes: stimulation of ion fluxes.

Leptinotoxin-h (LPTx), a neurotoxin (otherwise designated beta-leptinotarsin-h) known to stimulate the release of neurotransmitters from synapses, was purified from the hemolymph of the potato beetle, Leptinotarsa haldemani, by a simplification of the procedure originally developed by Crosland et al. [Biochemistry 23, 734-741, (1984)]. Highly and partially purified preparations of the toxin were applied to guinea pig synaptosomes and neurosecretory (PC12) cells. When applied in a Ca2+-containing Ringer medium, at concentrations in the 10(-11) - 10(-10) M range, the toxin induced: (a) rapid dep

Catalysis of electron transfer across phospholipid bilayers by iron-porphyrin complexes.

Phospholipid vesicles containing K3Fe(CN)6 were prepared form egg yolk phosphatidylcholine. Hemin dimethyl ester was incorporated into these vesicles during preparation in ratios of phospholipid to hemin dimethyl ester that varied from 200 : 1 to 45 000: 1. Electron transfer across the bilayer was measured anaerobically after injecting the vesicles into a solution containing reduced indigotetrasulfonic acid. Vesicles containing hemin dimethyl ester exhibited high rates of electron transfer (240 electrons/molecule hemin dimethyl ester per min). Conditions could be selected where the rate-limiti

Research Progress on Mesenchymal Stem Cells-Derived Small Size Vesicles for the Treatment of Liver Diseases.

Liver disease is a major public health challenge, accounting for a substantial burden of mortality and morbidity worldwide. Currently, liver transplantation is the gold standard for treating end-stage liver diseases. However, it is affected by the shortage of organ sources and immune rejection. Mesenchymal stem cells (MSCs) can be used as an alternative therapy for liver diseases, especially in cases of liver cirrhosis, liver failure and complications of liver transplantation. However, MSCs may have potential effects on tumor occurrence. MSCs derived small size vesicles are nanoscale extracell

Plant-derived extracellular vesicles (PDEVs) in anti-aging research: A review of biomedical characteristics and therapeutic applications.

Aging is a complex, irreversible physiological process characterized by gradual deterioration of tissue structure and function, accompanied by impaired regenerative capacity, dysregulated immune homeostasis, and increased susceptibility to chronic age-related diseases (e.g., neurodegenerative disorders, metabolic syndromes, age-related skin lesions, and inflammatory bowel disease). In recent years, plant-derived extracellular vesicles (PDEVs), the lipid bilayer membrane vesicles released by plant cells, have emerged as innovative candidates for anti-aging therapy because of their inherent bioc

Functional louts leaf extracellular vesicles for targeted delivery of curcumin through PPAR against obesity.

Obesity is a serious global epidemic and is a risk factor for diseases such as cancer and cardiovascular disease, having a profound negative impact on human health. Obesity is caused by factors such as adipocyte differentiation, lipolysis, and cell cycle, and the number of fat cells continues to increase, ultimately leading to obesity. Curcumin (cur) is a food additive that has a role in obesity control and is a safe and effective active molecule for the treatment of obesity. However low solubility and rapid metabolic clearance limit its bioavailability. Plant-derived extracellular vesicles (P

Bright NIR-II fluorescence lipid membrane probes for labelling of biomimetic nanovesicles with image-guided targeted NIR-II mild photothermal-immunotherapy of orthotopic glioblastoma.

Cell-derived nanovesicles possess the capacity to cross the blood-brain barrier (BBB) and modulate the immunosuppressive tumor microenvironment (TME); however, the lack of efficient labelling probes limits their application in tracing biodistribution and achieving synergistic glioblastoma (GBM) theranostics. Herein, we report bright NIR-II fluorescence membrane probe (CL1)-labelled nanovesicles (CL1-L-TV) composed of liposomes (L) and activated autologous T cell-derived extracellular vesicles (TV) for image-guided NIR-II mild photothermal immunotherapy of GBM. Phosphatidylinositol diphosphate binding by ESCRT-III filaments.

Different inositol phospholipids (PIPs) distribute to distinct subcellular organelles, creating an addressing system that dictates the sites of action of PIP-binding proteins, including components of the Endosomal Sorting Complexes Required for Transport (ESCRT). The ESCRT machinery is recruited to remodel many different cellular membranes through combinatorial binding interactions made by the early-acting ESCRT-I and ESCRT-II complexes with PIPs, ubiquitin modifications, and membrane-specific adaptors. Membrane remodeling, constriction, and fission are then mediated by membrane-associated fil
