Peroxidase and kallikrein in atropine-resistant secretion of submandibular saliva on parasympathetic nerve stimulation in anaesthetized rats.

Flow of submandibular saliva and the constituents secreted during chorda-lingual nerve stimulation at 40 Hz (3 x 10 min) were studied after alpha- and beta-adrenergic blockade, and in the absence (control) or presence of cholinergic blockade in anaesthetized rats. Peroxidase and true tissue kallikrein (rK1) were assessed to gain insight into the effects of the non-adrenergic, non-cholinergic (NANC) transmitters on protein secretion by acini and granular tubules, respectively. From control glands there was an overall mean flow of 181 +/- 15 microliters g-1 min-1, with no significant differences

Cultured epithelial cells release cyclooxygenase-dependent and cyclooxygenase-independent factors that inhibit cholinergic contraction of canine airway smooth muscles.

We observed the effects of supernatants from cultured epithelial cells on the contraction of tracheal smooth muscle evoked by acetylcholine (ACh) or by electrical field stimulation (EFS). Cultured canine tracheal epithelial cells were incubated in Krebs solution with or without indomethacin (10(-5)M) for 30 and 120 min. The amplitude of the tracheal smooth muscle contractions evoked by EFS or exogenously applied ACh were measured before and after the application of each supernatant in the combined presence of both indomethacin (10(-5)M) and propranolol (10(-6)M). The control supernatant incuba

Potentiation of acetylcholine release from tracheal parasympathetic nerves by cAMP.

We tested the hypothesis that increasing intracellular levels of adenosine 3', 5'-cyclic monophosphate (cAMP) increases acetylcholine (ACh) release from airway parasympathetic nerves. Muscle strips from equine trachea were preincubated for 60 min with 10(-7)M atropine, 10(-6)M neostigmine, and 10(-5) M guanethidine. The ACh release was evoked by electrical field stimulation (EFS, 20 V, 0.5 ms, 0.5 Hz) and measured by high-performance liquid chromatography with electrochemical detection. Agents known to increase cAMP, i.e., forskolin (10(-6) - 10(-4) M), 8-bromoadenosine 3', 5'-cyclic monophosp

Calcitonin gene-related peptide: mechanisms of modulation of antral endocrine cells and cholinergic neurons.

Actions of human calcitonin-gene related peptide (hCGRP) on acetylcholine (ACh) discharge and gastrin and somatostatin release from rat antral mucosal-submucosal fragments were examined in both dynamic perifusion experiments and short-term static incubation studies. The principal findings of the dynamic perifusion experiments were that hCGRP exerted a dual or biphasic effect on ACh discharge and gastrin release. Initial exposure of antral tissues to hCGRP (1 x 10(-8) M) resulted in stimulation of both ACh and gastrin release that was of brief duration. Continued hCGRP perifusion caused subsequ

Ciliary muscle muscarinic binding sites, choline acetyltransferase, and acetylcholinesterase in aging rhesus monkeys.

Choline acetyltransferase (ChAT) and acetylcholinesterase (AChE) activity, and the affinity and number of muscarinic binding sites (as reflected by specific 3H-quinuclidinyl benzilate binding) were determined in the ciliary muscle of rhesus monkeys ranging in age from 1-34 years. No age dependence was evident for any of these parameters. Within the limits of their specificity and precision, the data indicate that biochemical alterations in ciliary neuromuscular mechanisms do not account for the age-related loss of ciliary muscle configurational responses to topical pilocarpine and electrical s

Role of cholinergic, vagal reflexes on the bronchoconstrictor responses to histamine during carbon dioxide inhalation in conscious guinea-pigs.

Histamine-induced bronchoconstriction in conscious guinea-pigs involves a cholinergic, bronchoconstrictor reflex, but the role of this reflex during elevated levels of inspired carbon dioxide is unknown. In this study we examined the role of cholinergic, vagal reflexes on the bronchoconstrictor responses to histamine during CO2 inhalation. Guinea-pigs were placed inside a whole body plethysmograph for measurement of tidal volume (VT), respiratory rate (f) and minute volume (V) and a head chamber was used to deliver a hypercapnic gas mixture (10% CO2, 21% O2, 69% N2) and for inhalation of aeros

Permeability increase in rat submandibular gland by glandular substance P.

When the chorda tympani was stimulated at 20 Hz, infused horseradish peroxidase (HRP) entered the saliva through the tight junctions in the rat submandibular gland. This response was not inhibited by atropine but was reduced by a substance P(SP)-antagonist. The increase in junctional permeability for HRP never occurred in the gland that was stimulated at 5 Hz or in the SP-depleted gland. Our data suggest that the effect of parasympathetic stimulation on the increase in junctional permeability for macromolecules was due to the release of SP in the gland.

Cholinergic neuromodulation by prostaglandin D2 in canine airway smooth muscle.

To determine whether prostaglandin D2 (PGD2) modulates cholinergic neurotransmission in airway smooth muscle and, if so, what the mechanism of action is, we studied bronchial segments from dogs under isometric conditions in vitro. PGD2 (10(-8)-10(-5) M) elicited dose-dependent muscle contraction, which was reduced after blockade of muscarinic receptors, so that 50% effective dose (ED50) increased from 1.3 +/- 0.3 X 10(-6) to 3.9 +/- 1.0 X 10(-6) M by atropine (10(-6) M) (mean +/- SE, P less than 0.05). Physostigmine, at a concentration insufficient to alter base-line tension (10(-8) M), enhanc

Absence of the enteric nervous system in the newborn: presentation of three patients and review of the literature.

Three patients suffering from an absence of the enteric nervous system are reported. Two sisters presented with severe vomiting shortly after birth and dilatation of the intestine proximal to a stenosis. There was an absence of the enteric nervous system throughout the entire length of the intestine distal to the duodenum. A boy presenting an ileus was found to suffer from an aganglionosis of the entire colon. There was also an absence of neuronal bodies and nerve fibers in the small intestine. The final diagnosis was made by histochemical and immunocytochemical stains for acetylcholinesterase

Vasoactive intestinal peptide and substance P in salivary glands of the rat following denervation or duct ligation.

Immunoreactive vasoactive intestinal peptide (VIP) and substance P (SP) were studied in parotid, submaxillary and sublingual glands of the rat. The concentration of VIP was highest in the submaxillary gland and lowest in the parotid gland. The concentration of SP was highest in the parotid gland; it was at, or below the limit of detection in the sublingual gland. In the parotid gland the total amounts of VIP and SP were reduced by 95% after parasympathetic denervation (section of the auriculo-temporal nerve). In the submaxillary gland the total amounts of the peptides were unchanged after para

Cyclic GMP accumulation during cholinergic stimulation of eccrine sweat glands.

The possibility that guanosine 3'5'-cyclic monophosphate (cGMP) may be an intracellular mediator of cholinergic stimulation [methacholine chloride (MCh)] was explored by comparing the relationship between the time course of cGMP accumulation and sweat secretion by use of isolated monkey palm eccrine sweat glands. Isolated sweat glands were incubated with MCh or other agents, and tissue levels of cGMP were determined by radioimmunoassay. In parallel experiments, sweat secretion was induced from cannulated single sweat glands in vitro. Stimulation with MCh produced a Ca-dependent transient eleva

Pulmonary response to antigen infusion in the sensitized guinea pig: modification by atropine.

Alterations in pulmonary conductance, dynamic compliance, respiratory frequency, minute volume, mean arterial pressure, pulse rate, relaxation volume-to-dry weight ratio, and wet-to-dry weight ratio resulting from antigen infusion in sensitized guinea pigs was examined with and without atropine treatment. In untreated animals 3 min after antigen infusion there were significant decreases in dynamic compliance and pulmonary conductance with an increase in relaxation volume-to-dry weight ratio while other parameters were not altered. In atropine-treated animals antigen infusion resulted in a decr

Cephalic phase response in systemic glucose metabolism: How taste triggers glucose homeostasis.

BACKGROUND: Cephalic phase responses (CPRs) are anticipatory and conditioned responses elicited by the stimulation of food-related cues in the oral and nasal cavities. By initiating autonomic and endocrine reactions before absorption of nutrients, CPRs can blunt acute-postprandial hyperglycemia. Taste-related information is critical for triggering CPRs, but the neural mechanisms underlying taste-driven CPRs remain incompletely elucidated. HIGHLIGHT: Current evidence supports a model in which taste-driven CPRs arise from coordinated processing across peripheral taste receptors, the rostral com

Vagus nerve stimulation in musculoskeletal diseases.

The vagus nerve is the main nerve of the parasympathetic autonomic nervous system. Beyond its vegetative functions, the vagus nerve possesses anti-inflammatory and analgesic properties. Initially developed in the treatment of refractory epilepsy, vagus nerve stimulation (VNS) is currently being evaluated in several musculoskeletal diseases. VNS can be invasive by placing an electrode around the cervical vagus nerve and connected to a generator implanted subcutaneously or non-invasive stimulating the cervical vagus nerve branch percutaneously (auricular or cervical). In rheumatoid arthritis (RA

Neural mechanisms underlying lower urinary tract dysfunction.

This article summarizes anatomical, neurophysiological, and pharmacological studies in humans and animals to provide insights into the neural circuitry and neurotransmitter mechanisms controlling the lower urinary tract and alterations in these mechanisms in lower urinary tract dysfunction. The functions of the lower urinary tract, to store and periodically release urine, are dependent on the activity of smooth and striated muscles in the bladder, urethra, and external urethral sphincter. During urine storage, the outlet is closed and the bladder smooth muscle is quiescent. When bladder volume

Maturation of the cardiac autonomic regulation system, as function of gestational age in a cohort of low risk preterm infants born between 28 and 32 weeks of gestation.

OBJECTIVES: The maturation of the sympathetic nervous system (SNS) occurs steadily throughout gestation while the myelinated vagus has accelerated maturation periods, between 25 and 32 weeks of gestation and a further increase around 37-38 weeks of gestation. The aim was to quantify the cardiac autonomic regulation maturation, as a function of gestational age (GA) in a cohort of low risk preterm infants born between 28 and 32 weeks of gestation by assessing heart rate variability (HRV) at week 32, and at week 35 postmenstrual age (PMA). METHODS: Forty preterm infants were recruited, 24 h reco
