Impaired anorectic effect of leptin in neurotensin receptor 1-deficient mice.

Neurotensin plays a role in regulating feeding behavior. Central injection of neurotensin reduces food intake and the anorectic effect of neurotensin is mediated through neurotensin receptor 1 (Ntsr1). Ntsr1-deficient mice are characterized by mild hyperphagia and overweight without hyperleptinemia. The mechanism by which Ntsr1-deficient mice develop these metabolic abnormalities is not well understood. Leptin, secreted by adipocytes, regulates food intake by acting on hypothalamic neurons including neurotensin-producing neurons. Since the anorectic effect of leptin is blocked by neurotensin r

Assessment and management of the obese adult female: a clinical update for providers.

Obesity is epidemic in the United States, affecting 61% of adults and 14% of children. Today's health care providers should recognize obesity as a chronic disease and offer up-to-date information and management. Treatment options currently available are diet and behavior modification, medications, and surgical procedures. Implications for clinical practice include recognition of the problems associated with obesity, education of current patients as well as young women who may become patients, and appropriate diagnosis and management or referral.

Skilled reaching impairments from the lateral frontal cortex component of middle cerebral artery stroke: a qualitative and quantitative comparison to focal motor cortex lesions in rats.

The classical approach to investigating brain contributions to behavior has been to localize function to a region. In clinical investigations, however, injury is frequently multifocal, raising the question of how individual brain regions contribute to a resulting behavioral syndrome. For example, middle cerebral artery (MCA) ischemia in humans can concurrently damage a number of cortical and subcortical areas and the same areas are damaged in rat models of MCA stroke. In the rat, MCA occlusion produces severe motor deficits, but the cortical area of damage is the lateral neocortex, sparing mot

Sibutramine alters the central mechanisms regulating the defended body weight in diet-induced obese rats.

Chronic administration of sibutramine lowers body weight, presumably by altering brain monoamine metabolism. Here the effect of sibutramine on sympathoadrenal function (24-h urine norepinephrine and epinephrine levels) and arcuate nucleus (ARC) neuropeptide Y (NPY) and proopiomelanocortin (POMC) expression was assessed in diet-induced obese rats fed a low-fat diet. Chronic (10 wk) sibutramine [5 mg. kg(-1). day(-1) ip; rats fed ad libitum and injected with sibutramine (AS)] lowered body weight by 15% but only transiently (3-4 wk) reduced intake compared with vehicle-treated controls [rats fed

Relative dopamine D1 and D2 receptor affinity and efficacy determine whether dopamine agonists induce hyperactivity or oral stereotypy in rats.

The effects of a range of dopamine (DA) agonists on stereotyped behaviour in rats were analysed and compared both with the affinity of the compounds for D1 and D2 receptor binding sites in vitro and their ability to stimulate the adenylate cyclase activity in rat striatal homogenates. Full and partial agonists at the D1 receptor coupled to adenylate cyclase do not induce sterotypies when given alone, whereas full D2 agonists (e.g. quinpirole) induce hyperactivity but not oral sterotypies. Partial D2 agonists (e.g. (-)-3-PPP) only induce sedation. Mixed D1/D2 agonists (e.g. apomorphine) induce

Brain neurotransmitters in food and drug reward.

The discovery of brain peptides as neurotransmitters opens a new chapter in the study of feeding from a neural point of view. This article reviews current studies suggesting a limbic system framework of chemical neuroanatomy in which peptides play an important role in the control of eating and behavior reinforcement. In this framework, food intake and a variety of anorectic drugs (amphetamine, phenylpropanolamine, and fenfluramine) act on dopamine, norepinephrine, and serotonin systems that ascend from the hindbrain and midbrain to modulate feeding and satiety systems in the hypothalamus. A Comprehensive Approach to Outpatient Healthcare of People With Clinically Severe Obesity, According to Pathophysiological and Environmental Aspects: A Critical Review.

Severe obesity (SO), defined as a BMI ≥40 kg m-2 or ≥35 kg m-2 with comorbidities, is a complex chronic condition that poses major challenges to clinical management. Current evidence on the pathophysiological and environmental factors influencing outpatient care for people with SO (PSO) is synthesized and analyzed in this review. A literature search covering the past 10 years was conducted, including scientific articles and guidelines from scientific societies addressing these factors. The evidence suggests that conventional outpatient approaches are often insufficient for PSO, because they fa

Chocolate Processed by Extrusion Cooking Affects Glucose and Lipid Metabolism in Humans: A Randomized, Double-Blind, Crossover Trial.

Promoting positive and well-being-centered eating behavior in humans has recently garnered considerable attention. If the postprandial metabolic response of nutrients, especially sugars that are deleterious to health, can be controlled through processing, we can enjoy food without restrictions. To our knowledge, no study has reported the effect of extrusion cooking on the digestive response of sugars other than starch. Hence, we aimed to examine the effects of consuming extrusion-processed chocolate on glucose, insulin, and triglyceride blood levels through a randomized, double-blind, crossove

Hinoki Cypress Aroma Facilitates Swallowing via Premotor and Hypothalamic Down-modulation: A Randomized Within-subject Crossover fMRI Study.

Swallowing is tightly coupled with emotion-autonomic states, while olfactory pathways project to limbic and hypothalamic hubs that can enhance parasympathetic activity and reduce stress. Whether a pleasant natural-wood odor can modulate swallowing-related brain activity and behavior remains unclear. To test whether Hinoki (Japanese cypress) inhalation improves affective ratings and increases the repetitive saliva swallowing test (RSST) difference (post-pre; swallows/30 s), and whether it suppresses swallowing-related activity in the premotor cortex (Brodman area 6 (BA6)) and the hypothalamus.

The gut-brain axis in avian appetite regulation: Integrating peripheral signals with central neurocircuitry.

The precise regulation of energy homeostasis is essential for sustainable poultry production. Dysregulation of the avian gut-brain axis (GBA) contributes to metabolic disorders such as ascites and sudden death syndrome, impaired feed efficiency, and welfare problems in high-yield broilers. The avian GBA is a specialized bidirectional network that integrates hormonal, neural, and microbial signals to match feeding behavior with acute nutrient availability and long-term metabolic demands. Unique avian traits include a simplified gustatory system, lack of a functional T1R2 sweet receptor, a diver

Effect of modernized collaborative care for depression on individual depressive symptoms: A secondary analysis of the eIMPACT trial.

BACKGROUND: Depression affects over 400 million adults worldwide and is associated with substantial morbidity and mortality. Although symptoms vary in their responsiveness to treatment, clinical trials typically report total scores, obscuring symptom-level effects. Despite collaborative care's effectiveness for depression in primary care, its impact on individual depressive symptoms among racially and socioeconomically diverse patients remains unclear. METHODS: We conducted a secondary analysis of the eIMPACT trial of 216 primary care patients (Mage = 59 years; 78% women; 50% Black; 46% with

Oral administration of indole-3-lactic acid alleviates activity-based anorexia in female mice.

Anorexia nervosa (AN) is a severe psychiatric disorder characterized by extreme dietary restriction, pronounced weight loss, and disturbed energy metabolism. In recent years, increasing attention has been given to the role of the gut microbiota and its tryptophan-derived metabolites in modulating host neural function and feeding behavior via the gut-brain axis. Indole-3-lactic acid (ILA), one of the major tryptophan metabolites produced by the gut microbiota has been shown to exert neuroprotective and anti-inflammatory effects. However, its role in central appetite regulation remains unclear.

Key Signals Produced by Gut Microbiota Associated with Metabolic Syndrome, Cancer, Cardiovascular Diseases, and Brain Functions.

Gut microbiota have a significant impact neurotransmitters, short-chain fatty acids (SCFAs), immune signaling molecules, and gut hormones. These signaling molecules interact with receptors on the gut wall, immune cells, or the enteric nervous system (ENS), and reach the central nervous system (CNS) via the Vagus nerve (VN). SCFAs interact with G protein-coupled receptors (GPCRs), Toll-like receptors (TLRs), and proliferator-activated receptors (PPARs), influencing inflammatory reactions, gut motility, nutrient absorption, hormone secretion, neurochemical signaling, and brain functions. Macrotrabecular-Massive Hepatocellular Carcinoma: A Case Report.

Macrotrabecular-massive hepatocellular carcinoma (MTM-HCC) is a rare and aggressive molecular subtype of hepatocellular carcinoma (HCC) associated with a poor prognosis. Unlike typical HCC, which commonly arises in the context of cirrhosis, MTM-HCC can develop in non-cirrhotic livers, presenting unique diagnostic and therapeutic challenges. This case report describes a 35-year-old male who presented with persistent epigastric pain, fatigue, and loss of appetite. Clinical examination revealed hepatomegaly, prompting advanced imaging and laboratory investigations. Imaging studies identified a la

Variability in cannabinoid receptor genes is associated with psychiatric comorbidities in anorexia nervosa.

PURPOSE: The endocannabinoid system plays a key role in eating behavior regulating appetite and reward mechanisms, but the impact of its genetic variability has been scarcely studied in Anorexia Nervosa (AN). We aimed to analyze the association of genetic variants in cannabinoid receptors with the risk for AN and with psychiatric comorbidities that are commonplace in these patients. METHODS: We screened 221 AN patients and 396 controls for 14 tag-SNPs in the CNR1 and CNR2 genes, coding for cannabinoids receptors CB1 and CB2, respectively. Patients were diagnosed according to DSM-5 criteria an

Intracranial malignant solitary fibrous tumor metastasized to the chest wall: A case report and review of literature.

BACKGROUND: Solitary fibrous tumor (SFT) is a rare fibroblastic mesenchymal neoplasm that affects spindle cell soft tissues with broad-spectrum biological behavior; it is predominantly benign, and rarely metastasizes. SFT occurs mainly in the tissue structure of the serosa in the pleura and the thorax, and can be found throughout the body, though extra-thoracic localization, including the cephalic region, is un-common. We reported the first case of intracranial malignant SFT metastasized to the chest wall. CASE SUMMARY: An 81-year-old Japanese man was referred to our hospital due to progressi
