Stable Tracheal Regeneration Using Organotypically Cultured Tissue Composed of Autologous Chondrocytes and Epithelial Cells in Beagles.

OBJECTIVES: In tracheal regeneration, the slow process of epithelialization is often a barrier to the stability and safety of the transplanted trachea. The aim of this study was to examine a new tracheal regeneration technique using organotypically cultured tissue composed of autologous cells. METHODS: Nine beagles were prepared. Chondrocytes from auricular cartilage and epithelial cells from buccal mucosa were isolated and cultured. Tissue-engineered cartilages were fabricated with chondrocytes at a density of 1 × 107 cells/mL (high-density group) and 1 × 106 cells/mL (low-density group). A

Cartilage tympanoplasty: a reliable technique for smokers.

The aim of this study was to report our experience with cartilage tympanoplasty (CT) in smokers and compare it with that of non-smokers using a retrospective chart review at a tertiary referral center. Between September 2008 and September 2010, cartilage tympanoplasty was performed in 52 patients. Among them, 27 were active smokers and 25 former or non-smokers. The entire tympanic membrane was replaced with conchal cartilage, shaped either as a shield for cases with intact canal wall mastoidectomies, or crashed for cases with canal wall down procedures. The mean post-operative follow-up was 12

Alkaptonuria in two Colombian patients: identification of HGD variants including a novel finding.

BACKGROUND: Alkaptonuria (AKU) is a rare autosomal recessive inborn error of metabolism caused by deficiency of homogentisate 1,2-dioxygenase (HGD), its deficiency results in homogentisic acid (HGA) accumulation, which oxidizes to form melanin-like pigments that deposit in connective tissues, leading to ochronosis and progressive multisystem complications. We describe two unrelated Colombian female patients with AKU, emphasizing clinical variability and novel genetic findings. CASE REPORTS: Case 1: Female with onset of symptoms at age 16, including scleral pigmentation, glaucoma, retinal detac

A complicated case of relapsing polychondritis: Case report.

RATIONALE: Relapsing polychondritis (RP) is a rare inflammatory disease that presents clinically with severe dyspnea and respiratory distress. Differentiating it from amyloidosis can be diagnostically challenging. In cases of severe respiratory distress, timely bronchoscopic intervention, including laser tracheobronchoplasty and glucocorticoids (GCs) spraying, is crucial. Efficacy of this combined approach has not been reported to treat onset of RP. PATIENT CONCERNS: We report a complex case of RP in a 56-year-old male. Over the past 2 years, the patient experienced recurrent episodes of dysp

CHD7 cooperates with PBAF to control multipotent neural crest formation.

Heterozygous mutations in the gene encoding the CHD (chromodomain helicase DNA-binding domain) member CHD7, an ATP-dependent chromatin remodeller homologous to the Drosophila trithorax-group protein Kismet, result in a complex constellation of congenital anomalies called CHARGE syndrome, which is a sporadic, autosomal dominant disorder characterized by malformations of the craniofacial structures, peripheral nervous system, ears, eyes and heart. Although it was postulated 25 years ago that CHARGE syndrome results from the abnormal development of the neural crest, this hypothesis remained untes

Excision and "transcutaneous" lift in patients with neurofibromatosis of the fronto-temporo-orbital and auricular regions.

Neurofibromatosis (NF) type 1 (von Recklinghausen disease) involves the face in up to 4% of the cases. Patients present with varying degrees … ures, which passed transcutaneously, supported and fixed the predetermined areas on the skin or the ear cartilage to the periosteum of the calvarium. All the patients previously had multiple debulking surgeries without any lift procedures, which resulted in resagging of the facial elements within 3 months of surgery. The transcutaneous lift was durable up to 1 …

Change of bone conduction thresholds by total footplate stapedectomy in relation to age.

INTRODUCTION: The influence of age on threshold changes of bone conduction after stapedectomy has not been thoroughly studied. Improvement of the Carhart notch by stapes surgery might be related to age as well as an increase in cochlear sensitivity to surgical trauma. PATIENTS AND METHODS: A retrospective study on the outcome of stapedectomy was undertaken. The results of surgery performed on 387 ears in 315 patients for otosclerosis between 1962 and 1989 were obtained. Stapedectomy, performed by a single surgeon, consisted of total stapedectomy, seal of the oval window with tragal perichondr

Expression of the mouse alpha 1(II) collagen gene is not restricted to cartilage during development.

The mouse alpha 1(II) collagen gene has been isolated and a 5' portion of the gene which has low homology to other collagen genes was used to study the pattern of expression during mouse embryogenesis. In situ hybridization studies show that in the mouse, like the chick, alpha 1(II) collagen is expressed in chondrogenic tissues in advance of chondrocyte differentiation. The gene is expressed early in embryogenesis at 9.5 days both in the cranial mesenchyme destined for the chondrocranium, and the sclerotome of the somites, and at 12.5 days in the primordia of the hyoid and the laryngeal cartil

Do high-frequency air-bone gaps persist after ossiculoplasty?

OBJECTIVES: Conventional reporting of postoperative hearing outcomes utilizes a pure-tone averaged air-bone gap (ABG) that is biased toward low frequencies. Consequently, a high-frequency ABG after otologic surgery may go unnoticed. In this study, we evaluate changes in low- and high-frequency ABG following ossiculoplasty. STUDY DESIGN: Retrospective review. SUBJECTS AND SETTING: Consecutive series of patients who underwent ossiculoplasty at a single tertiary care center. Patients with pre- and postoperative audiograms were included. METHODS: Low-frequency ABG was calculated as the mean ABG

Saponin from Periploca forrestii Schltr Mitigates Oxazolone-Induced Atopic Dermatitis via Modulating Macrophage Activation.

Atopic dermatitis (AD) is a relapsing, acute, and chronic skin disease featured by intractable itching, eczematous skin. Conventional therapies based on immunosuppression such as corticosteroids are associated with multiple adverse reactions. Periploca forrestii Schltr saponin (PFS) was shown to potently inhibit murine arthritis by protecting bone and cartilage injury and suppressing NF-κB activation. However, its therapeutic effect on oxazolone-induced atopic dermatitis (AD) and the underlying mechanisms on macrophage are still unclear. The AD-like dermatitis was induced by repeated oxazolone

Influence of the Size and Location of the Perforation on the Anatomical Results of Myringoplasty.

INTRODUCTION: Type 1 tympanoplasty (myringoplasty) is the surgical closure of a perforated eardrum. Its purpose is to restore the integrity of the tympanic membrane and to improve hearing in the affected ear. Nowadays, we note the increasing use of cartilage as material for the reconstruction of the tympanic membrane. The main objective of our study is to evaluate the influence of size and perforation site on the results of type 1 tympanoplasties performed in our department. MATERIALS AND METHODS: We carried out a retrospective study of a series of myringoplasties spread over a period of four

Temporomandibular Joint Disc Replacement by Autogenous Composite Tissue Transplantation (Sandwich Method) in the Treatment of TMJOA With Perforation: A Preliminary Study.

Temporomandibular joint (TMJ) disc replacement is a treatment option for severe temporomandibular joint osteoarthritis (TMJOA) with disc perforation, yet there is still a lack of an ideal disc substitute. This retrospective study evaluated the "sandwich method," using a temporal myofascial flap (TMF) and auricular cartilage graft (ACG) as a composite tissue flap for disc replacement in patients treated at Peking University School of Stomatology between January 2021 and November 2023. Clinical assessments, including a maximum interincisal opening (MIO) and joint pain changes, and MRI and CBCT i

Balloon Dilation of the Cartilaginous Portion of the Eustachian Tube.

OBJECTIVE: Studies of balloon Eustachian tuboplasty (BET) have shown encouraging results in small series with short follow-ups. Our pilot study suggested that patients with protracted otitis media with effusion (OME) or atelectasis of the tympanic membrane (TM) could benefit from BET. STUDY DESIGN: A prospective study where subjects act as their own controls. Patients from the pilot study and additional cases were enrolled in this cohort with long-term follow-up. SETTING: Regional Academic Center. SUBJECTS AND METHODS: Out of 80 patients who underwent BET, 41 consecutive Eustachian tube (ET

Combined treatment protocol for temporomandibular joint ankylosis: an observation on a clinical problem.

Various techniques have been defined for the treatment of temporomandibular joint ankylosis. However, in some cases, they are unsuccessful, resulting in continuing pain and limitation in interincisal distance after surgery. This report describes the case of a 32-year-old woman who has been experiencing temporomandibular joint ankylosis for a period of 8 years. Several surgical procedures failed. A treatment approach combining auricular cartilage interposition arthroplasty with postsurgical functional treatment using a spring activator is presented. Using this approach, pain settled and maximal

Histopathology of ossicular implants.

Ossicular and cortical bone grafts maintain their morphologic structure for long periods of time and show varying amounts of replacement of nonviable bone by new bone through a slow process of creeping substitution. Cartilage grafts develop chondromalacia, lose stiffness, and tend to be resorbed over time. Plastipore prostheses elicit foreign body giant cell responses with microscopic biodegradation of the implants. There is a great need for the study of well-documented human temporal bone cases with in situ ossicular implants.
