Deciphering the phosphorylation-based regulatory strategies of Haemaphysalis longicornis in heat stress.

BACKGROUND: The Asian hard tick (Haemaphysalis longicornis) is an obligate hematophagous ectoparasite belonging to the family Ixodidae (hard ticks). This species serves as a competent vector for numerous highly pathogenic agents. The number of ticks that survive the summer, particularly under high-temperature conditions, exerts immediate and lasting impacts on tick populations and tick-borne disease prevalence. Therefore, it is crucial to study how high temperatures affect ticks, as well as how ticks adopt effective behavioral strategies and physiological adaptations to cope with heat stress.

Multi-attribute monitoring (MAM) methodology for glycosylated subunit vaccines.

Many protein-based vaccines comprise viral surface proteins which are chosen for their ability to stimulate the immune system. These vaccine molecules are often heavily glycosylated, and glycosylation plays critical roles in the immunological and stability properties of vaccines. The structural characterization and product quality attribute monitoring of such complex vaccine therapeutics during process development and manufacturing is very challenging. High throughput monitoring of multiple molecular attributes, particularly glycosylation, of recombinant glycoprotein subunit vaccines are neede

Proteomic analysis of the epidermal growth factor receptor (EGFR) interactome and post-translational modifications associated with receptor endocytosis in response to EGF and stress.

Aberrant expression, activation, and stabilization of epidermal growth factor receptor (EGFR) are causally associated with several human cancers. Post-translational modifications and protein-protein interactions directly modulate the signaling and trafficking of the EGFR. Activated EGFR is internalized by endocytosis and then either recycled back to the cell surface or degraded in the lysosome. EGFR internalization and recycling also occur in response to stresses that activate p38 MAP kinase. Mass spectrometry was applied to comprehensively analyze the phosphorylation, ubiquitination, and prot

Slow-Binding and Covalent HDAC Inhibition: A New Paradigm?

The dysregulated post-translational modification of proteins is an established hallmark of human disease. Through Zn2+-dependent hydrolysis of acyl-lysine modifications, histone deacetylases (HDACs) are key regulators of disease-implicated signaling pathways and tractable drug targets in the clinic. Early targeting of this family of 11 enzymes (HDAC1-11) afforded a first generation of broadly acting inhibitors with medicinal applications in oncology, specifically in cutaneous and peripheral T-cell lymphomas and in multiple myeloma. However, first-generation HDAC inhibitors are often associated

Nitric oxide and hydrogen sulfide in plants: which comes first?

Nitric oxide (NO) is a signal molecule regarded as being involved in myriad functions in plants under physiological, pathogenic, and adverse environmental conditions. Hydrogen sulfide (H2S) has also recently been recognized as a new gasotransmitter with a diverse range of functions similar to those of NO. Depending on their respective concentrations, both these molecules act synergistically or antagonistically as signals or damage promoters in plants. Nevertheless, available evidence shows that the complex biological connections between NO and H2S involve multiple pathways and depend on the pl

Recent advances in research on novel therapeutic mechanisms and strategies for exosome-based treatment of ischemic stroke.

Ischemic stroke (IS) accounts for over 80% of all stroke cases, featuring high disability, mortality, and recurrence rates. Conventional intravenous thrombolysis and mechanical thrombectomy are limited by narrow therapeutic windows, reperfusion injury, and the blood-brain barrier (BBB), which impedes the entry of neuroprotective agents into the brain parenchyma. Exosomes (Exos) are cell-secreted nanoscale extracellular vesicles (EVs) that carry miRNAs, proteins, and lipids, with low immunogenicity, BBB permeability, and ease of genetic modification, making them promising cell-free therapeutic

m6A methylation regulatory network in pulmonary fibrosis: Molecular mechanisms from RNA stability to cellular phenotype transition (Review).

Pulmonary fibrosis (PF) is a progressive and often irreversible interstitial lung disease characterized by aberrant wound healing, excessive extracellular matrix deposition and distortion of lung architecture. N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, dynamically regulates RNA metabolism through m6A writers, erasers and readers. Evidence indicates that m6A dysregulation contributes to PF by modulating fibroblast activation, epithelial injury and senescence, epithelial-mesenchymal plasticity, macrophage-associated inflammation, oxidative stress respon

Decoding the crosstalk between ubiquitination and other post-translational modifications in cancer immunity: from mechanisms to clinical prospects.

The tumor microenvironment is dynamically regulated by complex post-translational modifications (PTMs), which play pivotal roles in cancer immunity. Ubiquitination, along with other PTMs such as phosphorylation, glycosylation, and acetylation, orchestrates immune checkpoint activity and immune cell function, shaping antitumor responses. In this review, we discuss the intricate crosstalk and regulatory mechanisms between ubiquitination and other PTMs in cancer immunity. Studies have revealed that the stability and function of immune checkpoint proteins, such as PD-L1, are dynamically regulated

The fructose-1,6-bisphosphatase deficiency and the p.(Lys204ArgfsTer72) variant.

Fructose-1,6-bisphosphatase (FBPase) deficiency is a rare inborn error of fructose metabolism caused by pathogenic variants in the FBP1 gene. As gluconeogenesis is affected, catabolic episodes can induce ketotic hypoglycemia in patients. FBP1 analysis is the most commonly used approach for the diagnosis of this disorder. Herein, a Brazilian patient is reported. The proband, a girl born to a consanguineous couple, presented with severe hypoglycemia crisis in the neonatal period. At the age 17 months, presented a new crisis accompanied by metabolic acidosis associated with a feverish episode. Ge

Aminoglycoside Conjugation for RNA Targeting: Antimicrobials and Beyond.

Natural aminoglycosides are therapeutically useful antibiotics and very efficient RNA ligands. They are oligosaccharides that contain several ammonium groups able to interfere with the translation process in prokaryotes upon binding to bacterial ribosomal RNA (rRNA), and thus, impairing protein synthesis. Even if aminoglycosides are commonly used in therapy, these RNA binders lack selectivity and are able to bind to a wide number of RNA sequences/structures. This is one of the reasons for their toxicity and limited applications in therapy. At the same time, the ability of aminoglycosides to bi

Current and emerging drugs for Parkinson's disease: mechanisms, clinical evidence, and future directions.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by dopaminergic neuronal loss and α-synuclein pathology, resulting in disabling motor and non-motor symptoms. Pharmacotherapy remains the cornerstone of PD management, yet current treatments are largely symptomatic and fail to halt disease progression. In recent years, substantial advances have been made in both dopaminergic and non-dopaminergic therapeutic strategies. Novel formulations of levodopa and dopamine agonists aim to provide more stable dopaminergic stimulation and reduce motor fluctuations. Meanwhile

Targeting GALNT7 Disrupts the TAZ O-GalNAcylation Feedback Loop to Suppress Gallbladder Cancer Progression.

Gallbladder cancer (GBC) is a lethal malignancy with limited therapeutic options and dismal prognosis. Identifying the factors driving GBC progression is crucial for developing potent preventive and therapeutic approaches. Here, using quantitative proteomics, we identify GALNT7 as the most significantly upregulated glycosyltransferase in GBC tissues, associated with adverse clinical outcomes. Mechanistically, GALNT7 physically interacts with TAZ and catalyzes O-GalNAcylation at Ser307, thereby inhibiting K48-linked ubiquitination through the recruitment of the deubiquitinase USP7 and stabilizi

Therapeutic targeting of post-translational modifications in ovarian cancer: mechanisms and clinical applications.

Ovarian cancer is a lethal disease with high mortality due to late diagnosis, recurrence, and chemotherapy resistance. There is an urgent need for better diagnostic and therapeutic strategies. Recently, protein post-translational modifications (PTMs) have gained significant attention for their role in the onset, progression, and treatment of ovarian cancer. PTMs (including phosphorylation, ubiquitination, methylation, ADP-ribosylation, and others) significantly influence tumor cell proliferation, metastasis, and drug resistance by modulating cellular signal transduction, DNA repair mechanisms,

Integrative Dynamic Proteomics and Acyl-Proteomics Analyses in Clostridium butyricum.

Clostridium butyricum (C. butyricum) is a notable butyrate-producing intestinal probiotic that has been utilized for the enhancement and treatment of various intestinal and extraintestinal diseases. Butyrate and acetate, as significant metabolites of this bacterium, are short-chain fatty acids (SCFAs) that play crucial roles in maintaining intestinal stability and safeguarding host health. However, the regulatory functions of lysine butyrylation (Kbu) and acetylation (Kac), mediated by these metabolites, remain inadequately understood. In this study, we performed a comprehensive dynamic multio

Melatonin Rescues Hyperacetylation of Liver and Impaired Enzymatic Activities of Mitochondrial in IVF Offspring.

Increased risks of obesity and abnormal glucose metabolism were observed in IVF offspring. However, the underlying molecular mechanism was still unclear. As an important post-translational modification (PTM), lysine acetylation changed with the changes in the metabolic environment and usually occurred on metabolic enzymes to regulate metabolic pathways and enzyme activities and participated in the regulation of downstream metabolites. In our previous study, we proved that supplementation of melatonin in the culture medium improved obesity and metabolic dysfunction in IVF mice. In this study, w

Exposure to diethyl phthalates impairs ovarian reserve and oocyte maturation in female mice.

Diethyl phthalates (DEP) are commonly used as a plasticizer and have been found to cause male reproductive defects and metabolic disease in mammals as a potential environmental endocrine disruptor. However, the effects and underlying mechanisms of DEP exposure on female follicle development and oocyte maturation were still unclear. In this study, we found that mice exposed to DEP had significantly reduced primordial follicles. DEP exposure also led to a decrease in the number of ovulated oocytes, the rate of the first polar body, and an increase in fragmentation. Transcriptome analysis showed
