Optimization of corn, rice and buckwheat formulations for gluten-free wafer production.

Gluten-free baked products for celiac sufferers are essential for healthy living. Cereals having gluten such as wheat and rye must be removed from the diet for the clinical and histological improvement. The variety of gluten-free foods should be offered for the sufferers. In the study, gluten-free wafer formulas were optimized using corn, rice and buckwheat flours, xanthan and guar gum blend as an alternative product for celiac sufferers. Wafer sheet attributes and textural properties were investigated. Considering all wafer sheet properties in gluten-free formulas, better results were obtaine

Profilin as a severe food allergen in allergic patients overexposed to grass pollen.

BACKGROUND: Profilins are ubiquitous proteins that act as panallergens in sensitized patients, considered to be mild or incomplete food allergens. The aim of the study was to evaluate the role of profilins as severe food allergens in allergic patients overexposed to grass who were referred for severe food reactions and were sensitized to profilins. METHODS: After a careful in vitro screening, 26 patients were included, classified into two groups, mild (17) and severe reactors (9), based on clinical history and subsequently provoked orally with purified profilin in a double-blind placebo-contr

The plant pathogen Streptomyces scabies 87-22 has a functional pyochelin biosynthetic pathway that is regulated by TetR- and AfsR-family proteins.

Siderophores are high-affinity iron-chelating compounds produced by bacteria for iron uptake that can act as important virulence determinants for both plant and animal pathogens. Genome sequencing of the plant pathogen Streptomyces scabies 87-22 revealed the presence of a putative pyochelin biosynthetic gene cluster (PBGC). Liquid chromatography (LC)-MS analyses of culture supernatants of S. scabies mutants, in which expression of the cluster is upregulated and which lack a key biosynthetic gene from the cluster, indicated that pyochelin is a product of the PBGC. LC-MS comparisons with authent

The expression of analgesic-antitumor peptide (AGAP) from Chinese Buthus martensii Karsch in transgenic tobacco and tomato.

The present study aimed to obtain analgesic-antitumor peptide (AGAP) gene expression in plants. The analgesic-antitumor peptide (AGAP) gene was from the venom of Buthus martensii Karsch. Previous studies showed that AGAP has both analgesic and antitumor activities, suggesting that AGAP would be useful in clinical situations as an antitumor drug. Given that using a plant as an expression vector has more advantages than prokaryotic expression, we tried to obtain transgenic plants containing AGAP. In the present study, the AGAP gene was cloned into the plasmid pBI121 to obtain the plant expressio

Enhanced microbial degradation of cadusafos in soils from potato monoculture: demonstration and characterization.

Rapid degradation of cadusafos was evident in soils collected from previously-treated field sites from a potato monoculture area in northern Greece. The slower degradation of cadusafos observed in corresponding antibiotic-treated soils as well as in soils from an adjacent previously-untreated field demonstrated the microbial involvement in the rapid degradation of cadusafos in the soils from the previously-treated sites. Application of the non-specific antibacterial antibiotic chloramphenicol or of the Gram+ bacteria-inhibiting antibiotics penicillin + lyncomycin + vancomycin significantly inh

cDNA-AFLP analysis of differential gene expression in the prokaryotic plant pathogen Erwinia carotovora.

For studies of differential gene expression in prokaryotes, methods for synthesizing representative cDNA populations are required. Here, a technique is described for the synthesis of cDNA from the potato pathogens Erwinia carotovora subsp. atroseptica (Eca) and Erwinia carotovora subsp. carotovora (Ecc) using a combination of short oligonucleotide (11-mer) primers that were known to anneal to conserved sequences in the 3' regions of enterobacterial genes. Specific PCR amplifications with primers designed to anneal to 14 known genes from either Eca or Ecc revealed the presence of the correspond

Enhancement of butyrate production in the rat caecocolonic tract by long-term ingestion of resistant potato starch.

Some data suggest that the colonic microflora may adapt to produce more butyrate if given time and the proper substrate. To test this hypothesis, we investigated the effect of prolonged feeding of resistant potato starch on butyrate production. Rats were fed on either a low-fibre diet (basal) or the same diet supplemented with 90 g resistant potato starch/kg (PoS) for 0.5, 2 and 6 months. Short-chain fatty acid (SCFA) concentrations were determined in caecal and colonic contents at the end of each ingestion period. Total SCFA concentration increased over time throughout the caecocolonic tract

Both Conserved Aromatic Nature and Critical RNA Motif of Residue 67 in the ToBRFV Movement Protein May Be Essential for Evading Tm-22-Mediated Resistance.

The 68th amino acid residue (cysteine, C68) in tobacco mosaic virus (TMV) movement protein (MP) and the corresponding position in some tobamoviruses are required to trigger Tm-22-mediated resistance. In contrast, the 67th amino acid residue (histidine, H67) in tomato brown rugose fruit virus (ToBRFV) MP enables evasion, although its key property remains unclear. To assess the key property of this site, a saturated mutant pool was generated at ToBRFV MP H67 and TMV MP C68, along with two single mutants, H67C and C68H. Infection assays were conducted using two tomato cultivars, Jingpeng No. 1 (J

Boosting genome editing in perennial plants by CRISPR-Combo mediated morphogenic gene activation.

A major bottleneck in genome editing of many perennial plants is their recalcitrance to transformation and regeneration. To boost genome editing in such perennial crops, transcriptional reprogramming of morphogenic genes is introduced by CRISPR-Combo, a versatile system for simultaneous genome editing and transcriptional activation in plant cells. In potato, we screen 17 morphogenic genes and identify 4 genes (WOX11/12, ARF5, ABI3-1, and ABI3-2) that promote regeneration of genome-edited hairy roots, and 3 of the 4 genes are also found to boost shoot regeneration by Agrobacterium-mediated stab

Description of Streptomyces borealis sp. nov. and Streptomyces saintjeani sp. nov., associated with potato common scab.

Six Streptomyces strains were isolated from scab-infected potato tubers collected in the province of Quebec, Canada. All isolates produced disease symptoms characteristic of potato common scab. Multilocus sequence analysis showed that the strains clustered into two putative novel species-level groups represented by strains LBUM1542T and LBUM3076T. Strains LBUM1542T and LBUM3076T shared the highest 16S rRNA gene sequence similarity with Streptomyces rishiriensis NBRC 13407T (99.73%) and Streptomyces acidiscabies ATCC 49003T (99.67%), respectively. Synergistic micro-nano bubble treatment via high-speed vortex and ultrasound enhances quality and safety of lettuce and cherry tomatoes.

This study aimed to address quality deterioration and microbial contamination in fresh produce during postharvest storage by exploring efficient and safe cleaning technologies. The effects of three micro-nano bubble (MNB) treatments generated via ultrasound (US), high-speed vortex (HV), and their synergistic system (HV-US) on the quality and safety of lettuce and cherry tomatoes were compared. Results showed that the HV-US synergistic system significantly outperformed the individual treatments (p < 0.05), which was attributed to its smaller bubble size, higher dissolved oxygen content, stronge

Pathogenicity factor p22 of tomato chlorosis virus interferes with abscisic acid-mediated antiviral defenses by disrupting the function of WRKY81 in tomato.

Abscisic acid (ABA) is essential for plant resistance to both biotic and abiotic stresses, but its regulation during virus infection and role in antiviral defense remain poorly understood. Here, we report that overexpression of SlWRKY81 led to increased ABA accumulation, whereas silencing SlWRKY81 reduced ABA levels in tomato plants. Further analysis revealed that SlWRKY81 directly binds the promoters of ABA biosynthesis genes SlABA2 and SlABA3, thereby activating their expression. Infection by tomato chlorosis virus (ToCV) inhibited ABA content significantly. Consistently, heterologous expres

Spd-CDs-driven respiratory burst oxidase homolog/polyamine oxidase-dependent H2O2 signaling molecule engineering for salt tolerance in tomato.

Carbon dots, are now considered safe, environment-friendly materials. Spermidine carbon dots (Spd-CDs) have been used as new agrochemicals for abiotic stress, but in-depth studies of salt stress remain scarce. Here, foliar application of Spd-CDs improved salt stress tolerance in tomatoes, and the beneficial effects were concentration-dependent. Tomato seedlings supplied with Spd-CDs (3.0 mg/L) had a greater height, a higher maximum quantum yield of PSII, and a higher net photosynthetic rate than controls after being exposed to 120 mM NaCl for 7 d. Molecular evidence showed that Spd-CDs promote

Mimosine facilitates metallic cation uptake by plants through formation of mimosine-cation complexes.

Iron deficiency conditions as well as iron supplied as a Fe(III)-mimosine complex induced a number of strategy I and strategy II genes for iron uptake in leucaena. Leucaena leucocephala (leucaena) is a tree-legume that can grow in alkaline soils, where metal-cofactors like Fe(III) are sparingly available. Mimosine, a known chelator of Fe(III), may facilitate Fe(III) uptake in leucaena by serving as a phytosiderophore. To test if mimosine can serve as a phytosiderophore, three sets of experiments were carried out. First, the binding properties and solubility of metal-mimosine complexes were ass

Use of LAMP Detection to Identify Potential Contamination Sources of Plant-Pathogenic Pythium Species in Hydroponic Culture Systems of Tomato and Eustoma.

Hydroponic culture systems are subject to high risks of diseases caused by zoosporic plant pathogens. Control is generally difficult because of the rapid spread of zoospores in the nutrient solutions. In Japan, tomato and eustoma, which are cultivated using the D-tray and nutrient film techniques, respectively, are susceptible to diseases caused by Pythium aphanidermatum and P. irregulare. We used loop-mediated isothermal amplification to identify potential contamination sources of these two pathogens by monitoring their presence in the water supply wells, seedling terraces, nutrient solutions
