Design of 4-(piperazin-1-yl)thieno[2,3-d]pyrimidine derivatives targeting the DFG-out conformation of VEGFR-2: Synthesis, biological evaluations, and molecular modeling approaches.

Novel series of piperazinyl-thienopyrimidine acetamide derivatives (10a-l, 13a-b, and 16a) were rationally designed and synthesized as potential type II VEGFR-2 inhibitors based on the essential pharmacophoric features of sorafenib and the co-crystallized ligand in the VEGFR-2 crystal structure (PDB: 2OH4). The target compounds were prepared via a convergent synthetic route and confirmed by 1H NMR, 13C NMR, HRMS, and elemental analyses. In vitro VEGFR-2 kinase assays revealed inhibitory activity ranging from 0.056 to 1.346 μM, with compound 10d (4-methoxyphenyl) emerging as the most potent mem

Structural analysis of ITS 1 gene of Leishmania tropica and evaluation of a novel ligand, benzo[d][1,3]dioxol-5-yl 4-acetamidobenzenesulfonate, via molecular modeling methods.

INTRODUCTION: Leishmaniasis, a prevalent tropical disease caused by intracellular protozoa of the genus Leishmania, poses significant health challenges globally, exacerbated by migration waves from endemic regions. Despite its widespread impact, an effective vaccine for leishmaniasis remains elusive. Historically, antimony compounds have been employed in its treatment, but the emergence of resistant strains necessitates the development of new therapeutic agents. Addressing this need, our study focused on the structural characterization of a previously uncharacterized protein from Leishmania tr

Molecular modelling of diterpene compounds from copaifera oleoresin as potential bioactive compounds against SARS‑CoV‑2: calculations of density functional theory and in silico ADME-Tox properties.

Trees of the Copaifera genus produce oleoresins in their trunks, known as copaiba oil, widely used for their therapeutic properties. Copaiba oils contain diterpenic acids, rare compounds present in low quantities but with significant potential for treating various diseases. SARS-CoV-2, the causative agent of COVID-19, remains a major public health threat, emphasising the need for antiviral drug development despite vaccine availability. In this study, in silico tests were conducted on diterpenic compounds found in copaiba oil to evaluate their interaction with the receptor-binding domain (RBD)

Berberine chloride nanoemulgel enriched with clove oil attenuated monobenzone-induced experimental vitiligo in C57BL/6 mice: Pharmaceutical, molecular modeling, biochemical, protein, and mRNA expression analysis.

Vitiligo causes progressive loss of pigmentation owing to the destruction of melanocytes. Berberine chloride (BRB) protects melanocytes from oxidative stress and enhances melanophores. However, BRB exhibited reduced skin permeation efficiency due to poor (pKa-2.47 and 15.7; log P--1.5) physicochemical attributes. Therefore, in the present investigation, berberine chloride nanoemulsion (BRB-NE) enriched with clove oil, a well-known source of eugenol (EUG), was prepared with a droplet size of 20.50 ± 0.10 nm and a Polydispersity index (PDI) of 0.178 ± 0.022. Later, BRB-NE was transformed into be

Multi-conformational frame from molecular dynamics as a structure-based pharmacophore model for mapping, screening and identifying ligands against PPAR-γ: a new protocol to develop promising candidates.

Despite intensive research on clinical and molecular factors, the development of antidiabetic drugs in the last few decades is decelerating and as a result, the number of drugs approved by the US FDA is reduced. Hence, there is a persistent need for the innovative development of novel anti-diabetic drugs. Recent studies have provided ample proof that the peroxisome proliferator-activated receptor gamma (PPARγ), a ligand-activated transcription factor and its co-activator PGC-1 alpha may serve as good candidates for the treatment of several metabolic disorders. Therefore, in this study, 50 ns m

Interaction between TNF and BmooMP-Alpha-I, a Zinc Metalloprotease Derived from Bothrops moojeni Snake Venom, Promotes Direct Proteolysis of This Cytokine: Molecular Modeling and Docking at a Glance.

Tumor necrosis factor (TNF) is a major cytokine in inflammatory processes and its deregulation plays a pivotal role in several diseases. Here, we report that a zinc metalloprotease extracted from Bothrops moojeni venom (BmooMP-alpha-I) inhibits TNF directly by promoting its degradation. This inhibition was demonstrated by both in vitro and in vivo assays, using known TLR ligands. These findings are supported by molecular docking results, which reveal interaction between BmooMP-alpha-I and TNF. The major cluster of interaction between BmooMP-alpha-I and TNF was confirmed by the structural align

Anti-oxidation treatment of ultra high molecular weight polyethylene components to decrease periprosthetic osteolysis: evaluation of osteolytic and osteogenic properties of wear debris particles in a murine calvaria model.

Wear debris-induced osteolysis remains the greatest limitation of long-term success for total joint replacements with ultra-high molecular weight polyethylene (UHMWPE) bearings. To address oxidative degradation post-gamma irradiation, manufacturers are investigating the incorporation of antioxidants into PE resins. Similarly, larger molecular weight monomers have been developed to increase crosslinking and decrease wear debris, and ultimately osteolysis. However, the effects of modifying monomer size, crosslink density, and antioxidant incorporation on UHMWPE particle-induced osteoclastic bone

Molecular cytogenetic characterization of canine histiocytic sarcoma: A spontaneous model for human histiocytic cancer identifies deletion of tumor suppressor genes and highlights influence of genetic background on tumor behavior.

BackgroundHistiocytic malignancies in both humans and dogs are rare and poorly understood. While canine histiocytic sarcoma (HS) is uncommon in the general domestic dog population, there is a strikingly high incidence in a subset of breeds, suggesting heritable predisposition. Molecular cytogenetic profiling of canine HS in these breeds would serve to reveal recurrent DNA copy number aberrations (CNAs) that are breed and/or tumor associated, as well as defining those shared with human HS. This process would identify evolutionarily conserved cytogenetic changes to highlight regions of particula
