Intrinsic network activity in tinnitus investigated using functional MRI.

Tinnitus is an increasingly common disorder in which patients experience phantom auditory sensations, usually ringing or buzzing in the ear. Tinnitus pathophysiology has been repeatedly shown to involve both auditory and non-auditory brain structures, making network-level studies of tinnitus critical. In this magnetic resonance imaging (MRI) study, two resting-state functional connectivity (RSFC) approaches were used to better understand functional network disturbances in tinnitus. First, we demonstrated tinnitus-related reductions in RSFC between specific brain regions and resting-state netwo

Network dynamics in dyslexia: Review and implications for remediation.

Extant neurobiological theories of dyslexia appear fractional in focusing on isolated brain regions, mechanisms, and functional pathways. A synthesis of current research shows support for an Interactive Specialization (IS) model of dyslexia involving the dysfunctional orchestration of a widely-distributed, attentionally-controlled, hierarchical, and interhemispheric circuit of intercommunicating neuronal networks. This circuitry is comprised principally of the frontostriatal-parietal cognitive control system of networks, the posterior corpus callosum, and the left arcuate fasciculus. During de

Meta-connectomics: human brain network and connectivity meta-analyses.

Abnormal brain connectivity or network dysfunction has been suggested as a paradigm to understand several psychiatric disorders. We here review the use of novel meta-analytic approaches in neuroscience that go beyond a summary description of existing results by applying network analysis methods to previously published studies and/or publicly accessible databases. We define this strategy of combining connectivity with other brain characteristics as 'meta-connectomics'. For example, we show how network analysis of task-based neuroimaging studies has been used to infer functional co-activation fr

Distinct relationships of parietal and prefrontal cortices to evidence accumulation.

Gradual accumulation of evidence is thought to be fundamental for decision-making, and its neural correlates have been found in several brain regions. Here we develop a generalizable method to measure tuning curves that specify the relationship between neural responses and mentally accumulated evidence, and apply it to distinguish the encoding of decision variables in posterior parietal cortex and prefrontal cortex (frontal orienting fields, FOF). We recorded the firing rates of neurons in posterior parietal cortex and FOF from rats performing a perceptual decision-making task. Classical analy

Eyeblink conditioning: a non-invasive biomarker for neurodevelopmental disorders.

Eyeblink conditioning (EBC) is a classical conditioning paradigm typically used to study the underlying neural processes of learning and memory. EBC has a well-defined neural circuitry, is non-invasive, and can be employed in human infants shortly after birth making it an ideal tool to use in both developing and special populations. In addition, abnormalities in the cerebellum, a region of the brain highly involved in EBC, have been implicated in a number of neurodevelopmental disorders including autism spectrum disorders (ASDs). In the current paper, we review studies that have employed EBC a

Origins of multisynaptic projections from the basal ganglia to the forelimb region of the ventral premotor cortex in macaque monkeys.

The ventral premotor cortex (PMv), occupying the ventral aspect of area 6 in the frontal lobe, has been implicated in action planning and execution based on visual signals. Although the PMv has been characterized by cortico-cortical connections with specific subregions of the parietal and prefrontal cortical areas, a topographical input/output organization between the PMv and the basal ganglia (BG) still remains elusive. In the present study, retrograde transneuronal labelling with the rabies virus was employed to identify the origins of multisynaptic projections from the BG to the PMv. The vi

The role of the right temporoparietal junction in attention and social interaction as revealed by ALE meta-analysis.

The right temporoparietal junction (rTPJ) is frequently associated with different capacities that to shift attention to unexpected stimuli (reorienting of attention) and to understand others' (false) mental state [theory of mind (ToM), typically represented by false belief tasks]. Competing hypotheses either suggest the rTPJ representing a unitary region involved in separate cognitive functions or consisting of subregions subserving distinct processes. We conducted activation likelihood estimation (ALE) meta-analyses to test these hypotheses. A conjunction analysis across ALE meta-analyses del

Does resting-state connectivity reflect depressive rumination? A tale of two analyses.

Major Depressive Disorder (MDD) is characterized by rumination. Prior research suggests that resting-state brain activation reflects rumination when depressed individuals are not task engaged. However, no study has directly tested this. Here we investigated whether resting-state epochs differ from induced ruminative states for healthy and depressed individuals. Most previous research on resting-state networks comes from seed-based analyses with the posterior cingulate cortex (PCC). By contrast, we examined resting state connectivity by using the complete multivariate connectivity profile (i.e.

Etiology, triggers and neurochemical circuits associated with unexpected, expected, and laboratory-induced panic attacks.

Panic disorder (PD) is a severe anxiety disorder that is characterized by recurrent panic attacks (PA), which can be unexpected (uPA, i.e., no clear identifiable trigger) or expected (ePA). Panic typically involves an abrupt feeling of catastrophic fear or distress accompanied by physiological symptoms such as palpitations, racing heart, thermal sensations, and sweating. Recurrent uPA and ePA can also lead to agoraphobia, where subjects with PD avoid situations that were associated with PA. Here we will review recent developments in our understanding of PD, which includes discussions on: sympt

Control of urinary drainage and voiding.

Urine differs greatly in ion and solute composition from plasma and contains harmful and noxious substances that must be stored for hours and then eliminated when it is socially convenient to do so. The urinary tract that handles this output is composed of a series of pressurizable muscular compartments separated by sphincteric structures. With neural input, these structures coordinate the delivery, collection, and, ultimately, expulsion of urine. Despite large osmotic and chemical gradients in this waste fluid, the bladder maintains a highly impermeable surface in the face of a physically dem

Visual interhemispheric communication and callosal connections of the occipital lobes.

Callosal connections of the occipital lobes, coursing in the splenium of the corpus callosum, have long been thought to be crucial for interactions between the cerebral hemispheres in vision in both experimental animals and humans. Yet the callosal connections of the temporal and parietal lobes appear to have more important roles than those of the occipital callosal connections in at least some high-order interhemispheric visual functions. The partial intermixing and overlap of temporal, parietal and occipital callosal connections within the splenium has made it difficult to attribute the effe

Noninvasive brain stimulation in Huntington's disease.

Several important advances in the pathophysiology of Huntington's disease (HD) have been achieved by means of neurophysiological techniques designed to investigate the excitability and plasticity of brainstem and cortical circuits in patients with the condition. Studies designed to investigate brainstem reflexes, with paired-pulse and repetitive stimulation of the supraorbital nerve (blink reflex), have demonstrated abnormal excitability and plasticity of brainstem interneurons. In addition, several authors have tested the excitability of the primary motor cortex (M1) with the transcranial mag

Important casual association of carotid body and glossopharyngeal nerve and lung following experimental subarachnoid hemorrhage in rabbits. First report.

OBJECT: The glossopharyngeal nerves (GPNs) and carotid bodies (CBs) have an important role in the continuation of the cerebral autoregulation and cardiorespiratory functions. The relationship between degenerative injury of CB and the GPN in subarachnoid hemorrhage (SAH) was studied. METHODS: Twenty rabbits were included in this study. Five of them (n=5) were used as control group. The remaining animals (n=15) were exposed to experimental SAH. In the six animals of the SAH group, severe signs of illness were observed, and these six animals were killed in the first week after SAH. Others animal

Motor circuits in action: specification, connectivity, and function.

Mammalian motor behavior is enabled by a hierarchy of interleaved circuit modules constructed by interneurons in the spinal cord, sensory feedback loops, and bilateral communication with supraspinal centers. Neuronal subpopulations are specified through a process of precisely timed neurogenesis, acquisition of transcriptional programs, and migration to spatially confined domains. Developmental and genetic programs instruct stereotyped and highly specific connectivity patterns, binding functionally distinct neuronal subpopulations into motor circuit modules at all hierarchical levels. Recent wo

Altered connectivity and action model formation in autism is autism.

Internal action models refer to sensory-motor programs that form the brain basis for a wide range of skilled behavior and for understanding others' actions. Development of these action models, particularly those reliant on visual cues from the external world, depends on connectivity between distant brain regions. Studies of children with autism reveal anomalous patterns of motor learning and impaired execution of skilled motor gestures. These findings robustly correlate with measures of social and communicative function, suggesting that anomalous action model formation may contribute to impair

Functional connectivity between the left and right inferior frontal lobes in a small sample of children with and without reading difficulties.

Individuals with dyslexia often demonstrate bilateral inferior frontal lobe activation while performing basic reading tasks. To investigate these findings, functional connectivity analyses were conducted on fMRI data collected from children with dyslexia, who did and did not respond well to treatment, and from non-impaired readers. Analysis of active and resting-state fMRI data across 15 participants revealed functional connections between the inferior frontal regions in non-impaired readers and treatment responders, but not in treatment non-responders. Analyses incorporating DTI data revealed

Sleep abnormalities in schizophrenia may suggest impaired trans-thalamic cortico-cortical communication: towards a dynamic model of the illness.

Schizophrenia is associated with a wide range of symptoms. These include auditory hallucinations, delusions, and experiences that one is not in control of one's own thoughts and actions, but that they are inserted by an outside agency. It has been proposed that a disturbance in the sense of self may account for many of these symptoms. This disturbance in turn may be associated with source monitoring deficits. In other words, individuals with schizophrenia may misattribute the source of their own thoughts and actions to an outside agency, which then results in the experience of psychosis such a

Postnatal refinement of proprioceptive afferents in the cat cervical spinal cord.

Proprioceptive afferent (PA) information is integrated with signals from descending pathways, including the corticospinal tract (CST), by spinal interneurons in the dorsal horn and intermediate zone for controlling movements. PA spinal projections, and the reflexes that they evoke, develop prenatally. The CST projects to the spinal cord postnatally, and its connections are subsequently refined. Consequently, the tract becomes effective in transmitting control signals from motor cortex to muscle. This suggests sequential development of PAs and the CST rather than co-development. In this study w

Respiratory circuits: function, mechanisms, topology, and pathology.

Neuroscientists have long sought to understand how circuits in the nervous system are organized to generate the precise neural outputs that underlie particular behaviors. Recent studies deepened our understanding of the mechanisms responsible for the generation of the rhythmic output for breathing. Here, the author focuses on issues that are pertinent for the respiratory network and considers its organization and how it derives the functional output. The author discusses pacemaker and network mechanisms of rhythm generation, which are now combined into a novel concept of emergent network activ
