Absence of mutations associated with sulfa resistance in Pneumocystis carinii dihydropteroate synthase gene from non-human primates.

The dihydropteroate synthase (DHPS) gene from Pneumocystis carinii isolated from non-human primates was amplified using a polymerase chain reaction (PCR) and sequenced to analyse point mutations associated with sulfa resistance. P. carinii DHPS gene amplification was obtained from eight lung samples from five New World primate species and one Old World primate species. None of the animals had been exposed to sulfa drugs and only the wild-type P. carinii DHPS sequence at codons 55 and 57 was observed. These data support the hypothesis that high rates of DHPS mutants in P. carinii f. sp. hominis…

Cloning, sequencing, and functional characterization of the vitamin D receptor in vitamin D-resistant New World primates.

New World primates (NWPs) have high circulating 1,25-dihydroxyvitamin D (1,25-(OH)2D) levels. Comparable levels would be harmful to Old World primates (OWPs) and humans. Thus, NWPs must have developed mechanisms of 1,25-(OH)2D resistance to survive. In humans, patients with hypocalcemic vitamin D-resistant rickets type II have high circulating vitamin D levels and vitamin D resistance due to expression of a dysfunctional vitamin D receptor (VDR). To examine if this could wholly or in part explain vitamin D resistance in NWPs, VDR from Saguinus oedipus (cotton top tamarin) NWP B95-8 cells was c…

In vivo proton magnetic resonance spectroscopy for metabolic changes in brain during chronic cerebral vasospasm in primates.

OBJECTIVE: To study how neuronal cells are affected by development of chronic cerebral vasospasm after subarachnoid hemorrhage (SAH), the changes in neuronal metabolites during development of vasospasm were evaluated by in vivo localized proton magnetic resonance spectroscopy (MRS) in primates. METHODS: SAH was produced by introduction of a blood clot around the right middle cerebral artery and the right side of the circle of Willis. MRS experiments were performed before SAH and on Days 7 and 14 after SAH. Multislice magnetic resonance images were obtained to locate the volume of interest (1.…

Time course of the impairment of cerebral autoregulation during chronic cerebral vasospasm after subarachnoid hemorrhage in primates.

The time course of the impairment of cerebral autoregulation during chronic cerebral vasospasm after subarachnoid hemorrhage was studied in 18 monkeys. Changes in cerebral blood flow (CBF) at the regional level and central conduction times during either graded hypo- or hypertension were evaluated in these animals at three stages (3, 7, and 14 days) following the introduction of an autologous blood clot around the right middle cerebral artery (MCA). Angiograms revealed a reduction in vessel caliber (compared to the baseline level in the involved MCA) of 30% at 3 days, 50% at 7 days, and 10% at

Effects of systemic administration of 6-hydroxydopamine on the circumventricular organs in nonhuman primates. I. Area postrema.

6-Hydroxydopamine (6-OH-DA) has been shown to produce degenerative changes in noradrenergic nerve terminals and preterminals in the CNS following intracisternal, intraventricular or direct injection into the brain parenchyma. Systemic injection of 6-OH-DA is known to result in degenerative changes in noradrenergic terminals in the peripheral nervous system. However, only a few studies have been carried out on the effects of systemic injections of 6-OH-DA on noradrenergic terminals in the CNS. In the present study cynomolgus and squirrel monkeys were injected intravenously on two successive day

Sequential production of fatty liver, hepatitis, and cirrhosis in sub-human primates fed ethanol with adequate diets.

This study reproduces in experimental animals the sequential development of all the liver lesions seen in the human alcoholic: in 15 baboons fed ethanol, all developed fatty liver, five progressed to hepatitis, and five had cirrhosis. Maintenance of a nutritionally adequate regimen despite the intake of inebriating amounts of ethanol (50% of total calories) was achieved by incorporation of the ethanol in a totally liquid diet. Upon ethanol withdrawal, signs of physical dependence, such as seizures and tremors, developed. Ultrastructural changes of the mitochondria and the endoplasmic reticulum

Despotism promotes dyadic cooperation through enhanced interdependencies in non-human primate societies.

Cooperation is the cornerstone of human societies, and its emergence is firmly linked to enhanced tolerance and egalitarianism. However, evidence of profuse cooperation in less tolerant and despotic societies challenges this predominant view. The overarching interdependency hypothesis may resolve the conundrum. It posits that group-level interdependencies, like strength in numbers in colonially nesting species or allomaternal care in cooperatively breeding species, promote indiscriminate cooperation through enhanced tolerance. Crucially, this hypothesis also predicts that dyadic interdependenc

Multiscale structural evolution of proline-rich protein genes shapes primate salivary defenses.

Saliva forms the first biochemical interface with the environment, mediating dietary exposure and oral defense. Proline-rich proteins (PRPs) … xpression in saliva. Humans retain abundant PRP expression, but at reduced levels relative to other primates, potentially reflecting a lineage-specific regulatory shift. We further identify widespread exonic tandem repeats that provide a flexible mechanism for modulating protein composition. Notably, colobine PRH genes independently evolved trinucleotide r …

Deficiency of primate-specific TFDP3 causes male infertility with oligoasthenoteratozoospermia in humans and cynomolgus monkeys.

Primate-specific genes (PSGs), important contributors to the origin of adaptive evolutionary novelties, are abundantly expressed in the testis. However, the specific roles of PSGs in the male reproductive system of humans and other primates are largely unknown. Here, we employed whole-exome sequencing and identified deleterious variants of TFDP3, an X-linked PSG, in eight infertile men with oligoasthenoteratozoospermia. All of the male subjects harboring TFDP3 variants presented dramatic reductions in sperm concentration, motility, and abnormal sperm morphology. Furthermore, Tfdp3-knockdown (K…

Reversible cystogenesis in juvenile primate ADPKD models: evidence from PKD1 heterozygous monkeys.

Autosomal dominant polycystic kidney disease (ADPKD) is a common inherited disorder caused predominantly by heterozygous mutations in the PKD1 gene, leading to progressive renal cyst formation. While PKD1 mutant mouse models have provided mechanistic insights, PKD1 heterozygous mice fail to replicate the early cystogenesis observed in human patients. To address this gap, we conducted a longitudinal study using PKD1 heterozygous cynomolgus monkeys. Serial renal ultrasonography from birth to five years of age-corresponding to human childhood-revealed progressive cyst development. Remarkably, a s

Application of Chorionic Villus Sampling to Longitudinal Studies in Pregnant Non-Human Primate Models.

The rhesus macaque (Macaca mulatta) is a valuable model for pregnancy research due to its physiological similarity to humans and the ability to conduct studies in a controlled environment. Our previous work used non-invasive imaging methods to assess placental hemodynamics across gestation with correlative tissue analysis post-delivery. Here, we expand access to longitudinal timepoints from ongoing pregnancies by obtaining placental biopsies using ultrasound-guided needle aspiration. This approach aligns with New Approach Methods (NAMs) and supports animal welfare by reducing the number of ani

Liver Xenotransplantation: From Early Primate Trials to the First-in-Human Porcine Bridging Therapies.

Liver transplantation remains the definitive treatment for end-stage liver disease and acute liver failure, yet a critical and persistent shortage of donor organs results in thousands of preventable deaths annually worldwide. Xenotransplantation has emerged as a potential solution to this structural deficit. This narrative review traces the evolution of liver xenotransplantation, from early non-human primate trials in the 1960s through the application of CRISPR/Cas9-driven multi-gene editing platforms in contemporary porcine donors. The immunological barriers that drove the transition from pri

Comparative primate analysis shows that humans are not unique in having a tight cephalopelvic fit at birth.

Human childbirth is regarded as uniquely difficult among primates, due to a tight cephalopelvic fit thought to result from an evolutionary trade-off between adaptations to bipedal locomotion and increasing brain size. This impression, however, may be an artefact of past adoption of anthropocentric measurements that underestimate birth challenges in non-human primates. Here we re-evaluate cephalopelvic proportions using species-specific three-dimensional data of the pelvic inlet along with neonatal cranial dimensions from a broad sample of extant primates. Results reveal that maternal body size…

High-resolution in utero SV2A PET imaging of the nonhuman primate brain using the NeuroEXPLORER.

The NeuroEXPLORER (NX), a next generation brain-dedicated PET scanner, has demonstrated improved spatial resolution and sensitivity compared with previous PET scanners. In this study, we performed synaptic vesicle glycoprotein 2A (SV2A) PET, an in vivo marker of synaptic density, to evaluate the performance of the NX in both fetal and maternal brains of four pregnant rhesus macaques. Results were compared with the previously acquired PET images using miniEXPLORER and Biograph mCT for fetal and maternal brains (four pregnant monkeys per scanner). The NX demonstrated superior performance compare

Rethinking non-human primate models for emerging viral infections after COVID-19.

Non-human primates (NHPs) retain particular value in emerging viral infection research when questions require integrated analysis of systemic viral kinetics, protective immunity, tissue pathology, or longitudinal outcomes. However, their cost, limited availability, specialized husbandry, and ethical constraints preclude their routine use as default screening platforms. Using severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) as a case study, we compare how rhesus macaques, cynomolgus macaques, African green monkeys, baboons, and marmosets have contributed to studies of pathogenesis, …

A spinal origin for the obligate flexor synergy in the non-human primate: Implications for control of reaching.

Stroke survivors frequently develop the flexor synergy, an obligate co-contraction of shoulder abductors and elbow flexors; the neural substrate has proven elusive. Here we trained healthy monkeys to generate isometric elbow and shoulder torques to move an on-screen cursor, and recorded neuron firing from motor cortical areas and the reticular formation. In all regions we found cells correlated with activity around a single joint. Neurons coding co-contractions showed a bias towards combinations orthogonal to the post-stroke flexor synergy, e.g. shoulder abduction with elbow extension. Thresho

SRSF12 is a primate-specific splicing factor that induces a tissue-specific gene expression program.

Alternative splicing expands proteomic diversity and is tightly regulated by splicing factors, including the serine/arginine-rich (SR) protein family. Here, we analyze the poorly characterized protein SRSF12. Although SRSF12 is conserved across vertebrates, it is lowly expressed in most mammals, and we find that SRSF12 knockout mice do not display overt physiological or transcriptomic alterations. In contrast, SRSF12 is more highly expressed in primates, where it is predominantly transcribed in the testes, oocytes, and brain. SRSF12 colocalizes with other splicing components to nuclear speckle…
