Physiological and behavioural responsivity to stress and anxiogenic stimuli in COMT-deficient mice.

Catechol-O-methyltransferase, an enzyme involved in regulating brain catecholamine levels, has been implicated in anxiety, pain and/or stress responsivity. Elements of this putative association remain unclarified, notably whether: (a) COMT variation modulates responses to acute and/or chronic stress equally; (b) acute pharmacological inhibition of COMT produces comparable effects on anxiety to that observed after deletion of the COMT gene; (c) COMT genotype modulates action of anxiolytic drugs. We aimed to further investigate the relationship between reduced COMT function, anxiety and stress r

Stress impairs GABAergic network function in the hippocampus by activating nongenomic glucocorticoid receptors and affecting the integrity of the parvalbumin-expressing neuronal network.

Stress facilitates the development of psychiatric disorders in vulnerable individuals. It affects physiological functions of hippocampal excitatory neurons, but little is known about the impact of stress on the GABAergic network. Here, we studied the effects of stress and a synthetic glucocorticoid on hippocampal GABAergic neurotransmission and network function focusing on two perisomatic interneurons, the parvalbumin (PV)- and the cholecystokinin (CCK)-positive neurons. In acute hippocampal slices of rat, application of the potent glucocorticoid receptor (GR) agonist dexamethasone (DEX) cause

Altered chloride homeostasis removes synaptic inhibitory constraint of the stress axis.

In mammals, stress elicits a stereotyped endocrine response that requires an increase in the activity of hypothalamic parvocellular neuroendocrine neurons. The output of these cells is normally constrained by powerful GABA-mediated synaptic inhibition. We found that acute restraint stress in rats released the system from inhibitory synaptic drive in vivo by down-regulating the transmembrane anion transporter KCC2. This manifested as a depolarizing shift in the reversal potential of GABA(A)-mediated synaptic currents that rendered GABA inputs largely ineffective. Notably, repetitive activation

Effect of acute stress on sexual behavior in female rats: participation of the central angiotensinergic system.

Stress might influence the reproductive behavior in females, and central angiotensin II (Ang II) is a peptide that plays a role in stress response and in the modulation of sexual behavior. The medial amygdala (MeA), an important structure that regulates this behavior, is strongly involved in stress response. The aim of the present study was to evaluate the effect of acute restraint stress on the night of proestrus on sexual receptivity in female rats and the participation of Ang II and MeA in this effect. Adult female Wistar rats with regular estrous cycles were utilized. The acute stress prot

Chronic restraint stress decreases the expression of glutathione S-transferase pi2 in the mouse hippocampus.

Chronic restraint stress in mice affects hippocampal structure and function. Mice were subjected to daily restraint for 3 weeks, and gene expression in hippocampus was compared to controls using large-scale cDNA microarrays. We found that 444 genes were differentially expressed, and further analysis of 6 genes by real-time reverse transcription PCR confirmed that 3 of them were downregulated by stress. These 3 genes, growth factor receptor-bound protein 2 (Grb2), phosphatidylinositol-4-phosphate 5-kinase, type 1 beta (Pip5k1b), and glutathione S-transferase, pi2 (Gstp2), were also analyzed by

Effect of Saiboku-to, an Oriental Herbal Medicine, on gastric lesion induced by restraint water-immersion stress or by ethanol treatment.

The effect of saiboku-to on gastric lesions induced by restraint water-immersion stress and ethanol has been examined in rats. Thirty minutes after oral administration of saiboku-to, the rats were placed in restraint cages and immersed in water at 23 degrees C for 7 h, or orally administered 99.5% ethanol (1 mL) and placed in normal cages for 1 h. The stress for 7 h or the ethanol treatment for 1h induced erosion in the glandular area of the stomach. Histology showed that the surface epithelial cells were desquamated and part of the lamina propria mucosae was injured. The evaluation of lesion

Expansion of extrathymic T cells as well as granulocytes in the liver and other organs of granulocyte-colony stimulating factor transgenic mice: why they lost the ability of hybrid resistance.

When we attempted to characterize the immunological state in G-CSF transgenic mice, a large number of not only granulocytes but also lymphoid cells expanded in various immune organs. Such lymphoid cells were present at unusual sites of these organs, e.g., the parenchymal space in the liver. We then determined the phenotype of these lymphoid cells by immunofluorescence tests. It was demonstrated that CD3intIL-2Rbeta+ cells (i.e., extrathymic T cells), including the NK1.1+ subset of CD3int cells (i.e., NKT cells), increased in the liver and all other tested organs. These T cells as well as NK ce

Enzyme-Instructed Self-Assembling Peptide Hydrogels for Sustained Subconjunctival Delivery of Dexamethasone: A Stereochemical Comparison of L- and D-Enantiomeric Peptides for Postoperative Ocular Anti-Inflammatory Therapy.

Sustained postoperative anti-inflammatory therapy following intraocular surgery remains an unmet clinical need, as conventional topical corticosteroid eye drops suffer from poor bioavailability, patient non-compliance, and an inability to maintain therapeutic drug levels over the multi-week inflammatory resolution period. Here we report the development and characterization of alkaline phosphatase (ALP)-responsive enzyme-instructed self-assembling (EISA) peptide hydrogels based on the ultrashort amphiphilic sequence NapFFKY(p)-OH, covalently conjugated with dexamethasone (DEX) via an ester-clea

Major Depression-Associated NEGR1 Gene is Modulated in Stress-Susceptible Male Mice.

BACKGROUND: Neuronal growth regulator 1 (NEGR1) is an IgLON cell adhesion molecule significantly associated with depression risk in genome-wide association studies. Since the role of NEGR1 in depression pathophysiology remains incompletely understood, we investigated changes in NEGR1-associated gene expression levels in stress-susceptible male mice exposed to chronic restraint stress. METHODS: Mice were subjected to 21 consecutive days of restraint stress, and stress-induced maladaptive phenotypes were evaluated by tail suspension, forced swim, splash, and open field tests. After sacrifice, t

Optimising the dose of oral midazolam sedation for dental procedures in children: a prospective, randomised, and controlled study.

BACKGROUND: Midazolam sedation poses a significant dilemma in paediatric dentistry, which is to find out the optimal dosing with minimal undesirable adverse events. In this study, we aimed to compare the effect of three doses of oral midazolam (0.5, 0.75, and 1 mg/kg) on the sedative state and cooperative behaviour of children during dental treatment. We further compared completion rates, parent satisfaction, and all adverse events. DESIGN: Ninety children aged 3-10 years were randomised to three equal groups. Groups A, B, and C received 0.5, 0.75, and 1 mg/kg of oral midazolam, respectively.

Antinociceptive effect of chronic lithium on visceral hypersensitivity in a rat model of diarrhea-predominant irritable bowel syndrome: The role of nitric oxide pathway.

BACKGROUND AND AIM: Lithium, a widely used drug in bipolar-affective disorders, plays gastro-protective roles. The effects of lithium on several tissues are mediated through nitric oxide (NO), which regulates gastrointestinal motility and mucosal integrity. The aim of this study was to investigate the protective effect of chronic lithium administration on visceral hypersensitivity and to investigate the role of NO as a potential mechanism of lithium in a rat model of irritable bowel syndrome. METHODS: Colitis was induced by the intracolonic administration of acetic acid. After subsidence of i

The serotonin transporter is required for stress-evoked increases in adrenal catecholamine synthesis and angiotensin II AT(2) receptor expression.

High numbers of serotonin transporter (SERT) binding sites and high serotonin (5-HT) content are expressed in the adrenal medulla of wild-type (SERT+/+) mice. Acute restraint stress increases adrenomedullary 5-HT, norepinephrine (NE) and epinephrine (E) release, adrenomedullary tyrosine hydroxylase (TH) mRNA, and angiotensin II AT(2) receptor expression. There are no alterations in adrenal catecholamine content during restraint. In littermate SERT-/- mice, which do not express SERT binding sites, the basal adrenomedullary 5-HT content is significantly reduced and does not increase after stress

Timing-dependent neuroprotective effects of Virgin coconut oil against chronic restraint stress-induced neurobehavioral and hippocampal dysfunction: GC-MS profiling and in vivo evaluation.

BACKGROUND: Chronic stress, often exacerbated by improper lifestyle, long-term physical confinement, and emotional distress, these are the major contributor to neuropsychiatric disorders, mostly through hippocampal dysfunction. Though synthetic drugs offer symptomatic relief, their side effects prompt a growing interest in natural plant-derived nutraceuticals. Virgin coconut oil (VCO), rich in medium-chain fatty acids, polyphenols, and antioxidants, has demonstrated preliminary neuroprotective activity; however, its pharmacological basis in hippocampal dysfunction induced by chronic restraint

Veterinary student wellbeing and its relationship with dog handling and fear responses during a clinical examination laboratory.

INTRODUCTION: Veterinary care can be a significant source of fear and stress for dogs, and handling techniques play a key role in shaping these responses. Emerging evidence suggests that clinician mental wellbeing and stress can influence patient interactions. These challenges extend to veterinary students, who experience high stress during training and represent a critical population for understanding how these influences may emerge in clinical development. During a clinical examination laboratory, we aimed to assess (1) associations among veterinary students' mental well-being and affective

Promising natural agents in psychopharmacology described by Rhazes in Al-Hawi and Liber Almansoris: a ninth-century paradigm.

The synthesis of psychopharmaceuticals in the twentieth century marked the beginning of modern psychopharmacology, replacing earlier first-line psychiatric management that relied on psychosurgery, physical restraint, or administering narcotics and sedatives, e.g., morphine and chloral, which suppressed psychotic outbreaks temporarily without addressing the origin. Chlorpromazine's effective management of psychosis in the early 1950s opened the door for non-surgical therapies, making psychopharmaceuticals the current first-line treatment. While modern psychopharmacology began in the 1950s, the

Specialized Delirium Care Environments in Hospitalized Older Adults: A Systematic Review.

Purpose: Delirium is a frequent and serious condition in older patients, associated with severe adverse outcomes and lacking proven pharmacological treatments. Non-pharmacological multicomponent strategies are recommended, and specialized delirium care environments (e.g., delirium room, delirium unit, psychogeriatric unit) have been proposed as a potential strategy to improve the management of delirium in hospitalized older adults. Our objective was to provide the first systematic review and pooled analysis on the subject, synthesizing the characteristics of specialized delirium environments i

Effects of opioids on respiration assessed by a contact-free unconstraint respiratory monitor with load cells under the bed in patients with advanced cancer.

Nocturnal periodic breathing of chronic opioid users has been predominantly documented by the use of polysomnography. No previous studies have assessed the opioid effects of respiratory rhythms throughout the day without the use of physical restraint. We recently developed a contact-free unconstraint vital sign monitoring system with four load cells placed under the bed legs, which allows continuous measurements of respiratory change at the center of gravity on the bed. We aimed to reveal details of the patient's 24-h respiratory status under a monitoring system and to test the hypothesis that

Prediction of delirium in critically ill patients with elevated C-reactive protein.

BACKGROUND AND PURPOSE: Delirium is thought to be associated with systemic inflammatory response. However, its association with the most widely used inflammatory biomarker C-reactive protein (CRP) has not been well established. We aimed to examine whether CRP on intensive care unit (ICU) entry was associated with subsequent development of delirium. DESIGN AND SETTING: This prospective observational study was conducted in a mixed 24-bed ICU in a tertiary teaching hospital. METHODS: All patients admitted to the ICU from February 2011 to June 2012 were screened for eligibility. Demographic data
