In vivo and in vitro osmotic regulation of HSP-70 and prostaglandin synthase gene expression in kidney cells.

As a function of the urinary concentrating mechanism, the cells of the renal medulla are exposed to elevated and constantly varying osmolalities and adapt to this environment by selectively expressing certain mRNAs. We evaluated the expression and regulation of two RNAs that may be important in adaptation of rental medullary cells to hyperosmolality. We demonstrate selective, modulated expression in the renal medulla of heat shock protein HSP-70 mRNA and prostaglandin synthase-1 mRNA, with the abundance of these two mRNAs regulated in vivo in concert with changes in medullary sodium and urea.

Viability of Onchocerca volvulus in vitro.

The use of a selective schedule of tests to identify a viable population of isolated adult Onchocerca volvulus (Nematoda: Filarioidea) has been investigated in a large worm population. The study was initiated to develop methodology appropriate to test new candidate macrofilaricides for their in vitro activity against O. volvulus. After removal from the host the viability of isolated intact parasites was estimated by assessing the motility indices of male worms, and the colorimetric quantification of the reduction of the bioreducible tetrazolium reagent XTT and lactate output by female worms. A

Reabsorption of solutes and water from fluid-filled rabbit lungs.

Uncertainty persists concerning the mechanisms responsible for fluid clearance from the lungs after the air spaces become flooded in severe pulmonary edema. In this study, solute and water fluxes were investigated in an isolated, fluid-filled, perfused rabbit lung preparation. These lungs were perfused with physiologic 1.0 or 5.0 g/dl albumin solutions, and the air spaces were flushed and filled with the same solutions. Samples were obtained at intervals from the perfusate, and at the end of 1 or 2 h, fluid was pumped from the trachea into collection tubes. Concentrations of albumin (labeled w

Jejunal absorption and secretion of calcium in patients with chronic renal disease on hemodialysis.

10 patients with chronic renal disease on hemodialysis and 8 normals were studied by constant jejunal perfusion of calcium gluconate solutions, using polyethylene glycol as a nonabsorbable marker. Results in normals indicated that calcium absorption from 1 and 5 mM calcium solutions is mainly active. Absorption from 5, 15, and 20 mM solutions was a linear function of luminal calcium concentration, suggesting that the active transport carrier is saturated when luminal calcium concentration is greater than 5 mM and indicating that the increment in absorption at higher luminal concentrations is m

Soil column leaching of pesticides.

In this review, I address the practical and theoretical aspects of pesticide soil mobility.I also address the methods used to measure mobility, and the factors that influence it, and I summarize the data that have been published on the column leaching of pesticides.Pesticides that enter the unsaturated soil profile are transported downwards by the water flux, and are adsorbed, desorbed, and/or degraded as they pass through the soil. The rate of passage of a pesticide through the soil depends on the properties of the pesticide, the properties of the soil and the prevailing environmental conditi

Amphibian respiration and olfaction and their relationships: from Robert Townson (1794) to the present.

The present review examines the developments in the elucidation of the mechanisms of amphibian respiration and olfaction. Research in these two areas has largely proceeded along independent lines, despite the fact that ventilation of the nasobuccopharyngeal cavity is a basic element in both functions. The English naturalist Robert Townson demonstrated, in the 1790s, that amphibians, contrary to general belief, ventilated the lungs by a pressure-pump mechanism. Frogs and other amphibians respire by alternatively dilating and contracting the buccopharyngeal cavity. During dilatation, with the mo

Mechanical self-organization of the actin cytoskeleton in artificial cells.

The actin cytoskeleton generates diverse cellular functions, including intracellular positioning, symmetry breaking, and cell migration, through dynamic self-organization. Despite extensive molecular characterization of actin-binding proteins, the physical principles by which local molecular interactions give rise to cell-scale organization remain incompletely understood. Artificial cells provide a powerful platform for addressing this question because they enable precise control over molecular composition, geometry, and physical boundary conditions while minimizing the complexity inherent to

Lipid-mediated regulation of stomatal dynamics during plant stress responses.

Stomatal regulation is governed by interconnected developmental, hormonal, metabolic, and environmental signaling networks that enable plants to balance carbon acquisition with water conservation. This review highlights lipid metabolism and lipid-mediated signaling as an emerging regulatory layer integrating these processes during stomatal development and movement. We synthesize evidence that diverse lipid classes, including fatty acids, sterols, phosphoinositides, phosphatidic acid (PA), diacylglycerol (DAG), sphingolipids, and oxylipins, function not only as membrane components but also as d

Lodicule Isolation and Morphometric Analysis During Rice Floret Opening.

Rice lodicules are specialized floral organs located at the base of the ovary that undergo dynamic morphological changes during the flowering period. Water uptake-driven swelling and subsequent dehydration-induced shrinkage of the lodicules trigger floret opening and closure, respectively. Although lodicules play a central role in floret movement, standardized methods for quantitatively monitoring their temporal morphological changes remain limited. Here, we describe a detailed and reproducible workflow for lodicule sampling, dissection, imaging, and quantitative morphometric analysis. Florets

Co-exposure to positively charged polystyrene microplastics and butylparaben exacerbates intestinal injury and behavioral abnormalities in zebrafish.

Microplastics (MPs) and butylparaben (BuP) are common emerging contaminants in aquatic environments. Both can enter water bodies via domestic wastewater and wastewater treatment plant discharges, and thus the potential ecological risks posed by their combined contamination have attracted increasing attention. However, the combined toxic effects of polystyrene MPs with different surface charges and BuP on aquatic organisms remain unclear. This study used zebrafish as a model to investigate the comprehensive toxicity of single and combined exposures to negatively charged pristine polystyrene mic

Kv2.1/Kv8.2 Channels Regulate Fluid Homeostasis in the Outer Retina.

PURPOSE: Photoreceptor Kv2.1/Kv8.2 voltage-gated potassium channels carry an outward potassium current, helping to set the resting membrane potential and to shape dim light responses. Because potassium flux in the outer retina influences extracellular osmolarity and fluid distribution, we hypothesized that Kv2.1/Kv8.2 channels also contribute to fluid homeostasis in this region of the retina. METHODS: OCT imaging was performed in Kv8.2 heterozygous (Het) and knockout (KO) mice aged 4-7 weeks under dark- and light-adapted conditions. Light-dark differences in the distance between the external

Aquaporin-4 mediated glymphatic dysfunction and neuroinflammatory signaling in neurodegenerative disorders.

Aquaporin channels are the predominant fluid regulating channel found in the central nervous system (CNS) and plays a pivotal role in maintaining fluid and ion homeostasis, as well as regulating neuroinflammation, neurodegeneration, and blood-brain barrier (BBB) disruption. This protein is primarily located at astrocytes endfeet within the blood cerebral barrier and other central nervous system (CNS) junctions, facilitating the movement of water in both directions, buffering potassium levels, and aiding in the clearance of interstitial solutes, along with toxic metabolites such as amyloid-β, v

Effects of Hydroxylation and Packing Geometry on Tropocollagen Stability: Insights from Molecular Dynamics Simulations.

Collagen is the most prevalent protein in living organisms, playing diverse roles across multiple tissues. Its hierarchical structure relies on the assembly of tropocollagen, the fundamental building block of collagen fibrils. This assembly occurs in a predominantly random manner, allowing for variations in packing. This randomness can lead to regions of both tight and nontight packing within the fibrils. The mechanisms by which these regions influence collagen's packing configurations, structural organization, and functional properties remain poorly understood. This study provides a focused i

Mechanisms of pile-soil stress and deformation in excavations under the coupled effects of excavation disturbance and extreme rainfall infiltration.

Under the increasingly frequent extreme rainfall events, deep excavations are subjected to complex coupled effects of excavation-induced disturbance and rainfall infiltration, making it essential to clarify the deformation mechanisms of surrounding soils and retaining structures. In this study, a typical metro foundation pit in Zhengzhou was selected as the research object. By integrating field monitoring data with ABAQUS finite-element simulations, the deformation response and mechanical evolution of the pile-soil system under the combined influence of staged excavation and extreme rainfall i

Sea-level rise and freshwater management are reshaping coastal landscapes.

Along low-elevation coastlines, sea-level rise (SLR) threatens to salinate ecosystems. To understand the effects of SLR and freshwater management on landscape carbon (C) exchange, we measured the net ecosystem exchange (NEE) of CO2 between subtropical wetland ecosystems and the atmosphere along a dynamic salinity gradient. Ecosystems were representative of freshwater marl prairies, brackish ecotones, and saline scrub mangrove forests in the southeastern Everglades. Patterns in NEE explained the landward movement of coastal wetlands, a process observed over the last 70 years. The capacity to ca

Uncovering the core genetic programs governing plant guard cell biology.

Guard cells (GCs) regulate gas exchange and water loss in plants and have been extensively studied in Arabidopsis thaliana. However, cross-species comparisons at single-cell resolution remain limited. To address this, we aimed to define conserved and divergent transcriptomic signatures of GCs by generating a comparative single-cell atlas encompassing five species: A. thaliana, soybean (Glycine max), tomato (Solanum lycopersicum), rice (Oryza sativa), and sorghum (Sorghum bicolor). We performed single-nucleus RNA sequencing (sNucRNA-seq) on leaf tissues from each species, followed by integrativ
